Abstract:Natural enzymes are used as special reagents for the preparation of electrochemical (bio)sensors due to their ability to catalyze processes, improving the selectivity of detection. However, some drawbacks, such as denaturation in harsh experimental conditions and their rapid de- gradation, as well as the high cost and difficulties in recycling them, restrict their practical applications. Nowadays, the use of artificial enzymes, mostly based on nanomaterials, mimicking the functions of natural products, has been growing. These so-called nanozymes present several advantages over natural enzymes, such as enhanced stability, low cost, easy production, and rapid activity. These outstanding features are responsible for their widespread use in areas such as catalysis, energy, imaging, sensing, or biomedicine. These materials can be divided into two main groups: metal and carbon-based nanozymes. The latter provides additional advantages compared to metal nanozymes, i.e., stable and tuneable activity and good biocompatibility, mimicking enzyme activities such as those of peroxidase, catalase, oxidase, superoxide dismutase, nuclease, or phosphatase. In this review article, we have focused on the use of carbon-based nanozymes for the preparation of electrochemical (bio)sensors. The main features of the most recent applications have been revised and illustrated with examples selected from the literature over the last four years (since 2020).

Comprehensive Study of the Effects of Nanopore Structures on Enzyme Activity for the Enzyme Based Electrochemical Biosensors Based on Molecular Simulation.

Abstract:Assembly of biocompatible nanostructures to retain the enzyme activity and improve the biocatalytic ability is a decisive factor for enhancing the performance of enzyme biosensors. However, there is still a lack of molecular level understandings of the physicochemical interaction mechanism at the interface of biosensor electrodes and enzymes. Here, for the first time at molecular level, the effects of two classic biosensor electrode materials with different electrical properties and morphologies and glucose oxidase (GOD) on retaining the enzyme conformation were analyzed by molecular dynamics simulation. First, for the immobilization of GOD, the interfaces of zinc oxide (ZnO) with different electrical properties and 10 nm diameter ZnO nanopore were studied. Then, to simulate the sensing process when electric voltages are applied, positively charged gold planes and 10 nm diameter gold nanopore were investigated as well. The results showed that the nanopore structure was confirmed to be well adapted for the enzyme conformation retaining compared to the plane structure for both ZnO and gold materials, and they almost fit well with the sensitivity measurement results from many previously reported experimental studies. This study also indicates that molecular modeling of the interactions between biomolecules and functional nanostructures is helpful for developing high performance enzyme nanobiosensors.

Abstract:Nanozymes are nanomaterials with enzyme-like catalytic activities. The unique features of nanozymes (such as high stability, low cost, large surface area for bioconjugation, ease of storage, and multi-functionalities) offer unprecedented opportunities for designing electrochemical biosensors. Recent years have witnessed the rapid development of nanozyme-based electrochemical biosensors. To highlight these achievements, this review first discusses the representative nanozymes including peroxidase mimics, oxidase mimics, hydrolase mimics, and superoxide dismutase mimics used in electrochemical biosensors. Then, it summarizes the bioanalytical applications for the detection of various analytes. Finally, current challenges and future research directions are summarized.

Abstract:Nanozyme, a term defined for nanomaterial with enzyme-like properties, has attracted significant research attention owing to its striking merits. Recently, a surge of nanozymes have been demonstrated to catalyze some typical enzymatic reactions mimicking oxidase, peroxidase and catalase. Especially, nanozymes with peroxidase-like activity have grown into a big family due to their broad range of applications in the field of biosensing and immunoassay. Since inorganic nanoparticles possess the advantages of high stability and easy surface modification, nanozymes have been emerging alternatives to natural enzymes to some extent. In this Review, we briefly summarize several typical nanozymes and then focus our attention on their enormous applications with respect to analytical chemistry. Representative examples would be discussed in detail from the literatures of last 10 years. Additionally, the current challenges and future directions about nanozymes are speculated at the end of this review.

Abstract:Nanoparticles exhibiting diverse shapes, high porosity and chemical stability reveal, upon appropriate chemical engineering, enzyme-like catalytic activities, "nanozymes", providing a plethora of nanomaterials for diverse applications. The present review article addresses the sensing applications of the catalytic functions of nanozymes consisting of metal nanoparticles, metal oxides, metal sulfides and cyanometallate nanoparticles, carbon-based nanomaterials and metal-organic-framework nanoparticles. The nanozymes emulate catalytic functions of oxidases or peroxidases and are employed as amplifying agents for sensing diverse analytes such as glucose, dopamine, NADH, thiols, phosphates and more. Moreover, the immobilization of nanozymes on electrodes provides versatile means to develop electrochemical sensing platforms. Different principles of the electrochemical sensing platforms, synthetic methodologies to deposit nanozymes on electrodes, and methods to establish electrical communication between the bulk conductive support and nanozyme particles are introduced. Electrochemical sensing platforms applying nanozyme-modified electrodes for the detection of analytes such as organophosphates, glucose and more are discussed. In particular, the application of nanozymes as amplifying labels for biosensor devices detecting proteins, DNA and microRNAs are addressed. Finally, the uses of nanozymes as functional constituents to design sense-and-treat systems are discussed. This is exemplified with the assembly of a bioreactor system for the sensing of glucose, the nanozyme-promoted generation of reactive oxygen species as cytotoxic agents towards cancer cells, and the autonomous nanozyme-based glucose-controlled release of insulin from nanocarrier devices. The future challenges in developing nanozyme-based sensors and sense-and-treat systems are presented.

Abstract:Natural enzymes are crucial catalysts that not only control over the biotransformation in intracellular environments with high efficiency but also are widely harnessed in vitro for diverse applications, among which are used as a core element to engineer biosensor attributing to their high sensitivity and specificity in recognition. However, the vulnerable conformation of a native enzyme arises a series of unavoidable issues associated with lifetime and cost effectiveness, greatly limiting their large-scale applications. The insight into the enzyme mimics using programmable and structurally stable nanomaterial, known as nanozyme, offers an efficient scheme to circumvent the obstacles of natural enzymes, yet, render the biocatalytic functions. In the various categories of nanozymes, porous organic frameworks (POFs) including metal organic frameworks (MOFs), covalent organic frameworks (COFs) and hydrogen-bonded organic frameworks (HOFs) arise enormous attention due to their unique features of ultrahigh surface area, high porosity, and intercommunicating channels. Most importantly, POFs have unambiguous yet predictable topologies, which are structurally favorable for mimicking the active sites as well as microenvironments of natural enzymes. In this review, we overview the advances of POFs nanozymes in which different oxidoreductase- and hydrolase-mimicking activities are involved, with special emphasis on harnessing these diverse activities for conceiving next-generation biosensor targeting micromolecules, metal ions, peptides and proteins, nucleic acids and circulating tumor cells (CTCs), etc. Finally, the current challenges and future perspectives on the design and biosensing applications of POFs nanozymes are discussed. We believe that this review can offer new insight into the access of enzyme mimics with user-defined bioactivity and high stability, and may inspire more interesting biosensing applications.

Abstract:Nanozymes are a class of artificial enzymes that have dimensions in the nanometer range and can be composed of simple metal and metal oxide nanoparticles, metal nanoclusters, dots (both quantum and carbon), nanotubes, nanowires, or multiple metal-organic frameworks (MOFs). They exhibit excellent catalytic activities with low cost, high operational robustness, and a stable shelf-life. More importantly, they are amenable to modifications that can change their surface structures and increase the range of their applications. There are three main classes of nanozymes including the peroxidase-like, the oxidase-like, and the antioxidant nanozymes. Each of these classes catalyzes a specific group of reactions. With the development of nanoscience and nanotechnology, the variety of applications for nanozymes in diverse fields has expanded dramatically, with the most popular applications in biosensing. Nanozyme-based novel biosensors have been designed to detect ions, small molecules, nucleic acids, proteins, and cancer cells. The current review focuses on the catalytic mechanism of nanozymes, their application in biosensing, and the identification of future directions for the field.

Nanozymes: From New Concepts, Mechanisms, and Standards to Applications

Abstract:Nanozymes are nanomaterials with intrinsic enzyme-like characteristics that have been booming over the past decade because of their capability to address the limitations of natural enzymes such as low stability, high cost, and difficult storage. Along with the rapid development and ever-deepening understanding of nanoscience and nanotechnology, nanozymes hold promise to serve as direct surrogates of traditional enzymes by mimicking and further engineering the active centers of natural enzymes. In 2007, we reported the first evidence that Fe3O4 nanoparticles (NPs) have intrinsic peroxidase-mimicking activity, and since that time, hundreds of nanomaterials have been found to mimic the catalytic activity of peroxidase, oxidase, catalase, haloperoxidase, glutathione peroxidase, uricase, methane monooxygenase, hydrolase, and superoxide dismutase. Uniquely, a broad variety of nanomaterials have been reported to simultaneously exhibit dual- or multienzyme mimetic activity. For example, Fe3O4 NPs show pH-dependent peroxidase-like and catalase-like activities; Prussian blue NPs simultaneously possess peroxidase-, catalase-, and superoxide dismutase-like activity; and Mn3O4 NPs mimic all three cellular antioxidant enzymes including superoxide dismutase, catalase, and glutathione peroxidase. Taking advantage of the physiochemical properties of nanomaterials, nanozymes have shown a broad range of applications from in vitro detection to replacing specific enzymes in living systems. With the emergence of the new concept of "nanozymology", nanozymes have now become an emerging new field connecting nanotechnology and biology. Since the landmark paper on nanozymes was published in 2007, we have extensively explored their catalytic mechanism, established the corresponding standards to quantitatively determine their catalytic activities, and opened up a broad range of applications from biological detection and environmental monitoring to disease diagnosis and biomedicine development. Here we mainly focus on our progress in the systematic design and construction of functionally specific nanozymes, the standardization of nanozyme research, and the exploration of their applications for replacing natural enzymes in living systems. We also show that, by combining the unique physicochemical properties and enzyme-like catalytic activities, nanozymes can offer a variety of multifunctional platforms with a broad of applications from in vitro detection to in vivo monitoring and therapy. For instance, targeting antibody-conjugated ferromagnetic nanozymes simultaneously provide three functions: target capture, magnetic separation, and nanozyme color development for target detection. We finally will address the prospect of nanozyme research to become "nanozymology". We expect that nanozymes with unique physicochemical properties and intrinsic enzyme-mimicking catalytic properties will attract broad interest in both fundamental research and practical applications and offer new opportunities for traditional enzymology.

Abstract:Because of the high catalytic activities and substrate specificity, natural enzymes have been widely used in industrial, medical, and biological fields, etc. Although promising, they often suffer from intrinsic shortcomings such as high cost, low operational stability, and difficulties of recycling. To overcome these shortcomings, researchers have been devoted to the exploration of artificial enzyme mimics for a long time. Since the discovery of ferromagnetic nanoparticles with intrinsic horseradish peroxidase-like activity in 2007, a large amount of studies on nanozymes have been constantly emerging in the next decade. Nanozymes are one kind of nanomaterials with enzymatic catalytic properties. Compared with natural enzymes, nanozymes have the advantages such as low cost, high stability and durability, which have been widely used in industrial, medical, and biological fields. A thorough understanding of the possible catalytic mechanisms will contribute to the development of novel and high-efficient nanozymes, and the rational regulations of the activities of nanozymes are of great significance. In this review, we systematically introduce the classification, catalytic mechanism, activity regulation as well as recent research progress of nanozymes in the field of biosensing, environmental protection, and disease treatments, etc. in the past years. We also propose the current challenges of nanozymes as well as their future research focus. We anticipate this review may be of significance for the field to understand the properties of nanozymes and the development of novel nanomaterials with enzyme mimicking activities.

Abstract: Accurate, rapid, inexpensive, and selective analysis is required today for use in clinical diagnostics and the food industry. The majority of known electrochemical biosensors are based on immobilized specific biomolecules, such as proteins, enzymes, nuclear acids, antibodies, and antigens on the modified electrodes [1–8]. These resulting biomolecule-based devices usually show high sensitivity and specificity due to the high loading of enzymes on the nanoparticles [9–12]. However, their stability is limited due to easy denaturation [13] and leakage of biomolecules during their storage and immobilization procedure. Furthermore, the preparation and purification of biomolecules are usually time-consuming and expensive [14]. Therefore, the syntheses of artificial biomolecules with highly catalytic properties and a wide range of practical applications are becoming a significant field for different purposes.

Abstract:The latest advances and current challenges of enzyme‐mimicking nanoparticles and their use in biosensing are highlighted, with a special focus on point‐of‐care (PoC) devices and their translational path for use in biomedical devices. These materials exhibit innate advantages compared to biological enzymes and nanoparticle probes currently utilized and hold great promise for implementation in biotechnology and clinical diagnosis. Nanozymes represent a class of nanosized materials that exhibit innate catalytic properties similar to biological enzymes. The unique features of these materials have positioned them as promising candidates for applications in clinical sensing devices, specifically those employed at the point‐of‐care. They notably have found use as a means to amplify signals in nanosensor‐based platforms and thereby improve sensor detection limits. Recent developments in the understanding of the fundamental chemistries underpinning these materials have enabled the development of highly effective nanozymes capable of sensing clinically relevant biomarkers at detection limits that compete with "gold‐standard" techniques. However, there remain considerable hurdles that need to be overcome before these nanozyme‐based sensors can be utilized in a platform ready for clinical use. An overview of the current understandings of nanozymes for disease diagnostics and biosensing applications and the unmet challenges that must be considered prior to their translation in clinical diagnostic tests is provided.

Construction of Bio-Nano Interfaces on Nanozymes for Bioanalysis

Abstract:Nanomaterials with enzyme-like activity (nanozymes) have been of great interest in broad applications ranging from biosensing to biomedical applications. Despite that much effort has been devoted to the development of the synthesis and applications of nanozymes, it is essential to understand the interactions between nanozymes and most commonly used biomolecules, i.e., avidin, streptavidin (SA), bovine serum albumin (BSA), immunoglobulin G (IgG), and glutathione (GSH), yet they have been rarely explored. Here, a series of bio-nano interfaces were constructed through direct immobilization of proteins on a variety of iron oxide and carbon-based nanozymes with different dimensions, including Fe3O4 nanoparticles (NPs, 0D), Fe3O4@C NPs (0D), Fe3O4@C nanowires (NWs, 1D), and graphene oxide nanosheets (GO NSs, 2D). Such interfaces enabled the modulation of the catalytic activities of the nanozymes with varying degrees, which allowed a good identification of multiplex proteins with high accuracy. Given the maximum inhibition on Fe3O4@C NP by BSA, we established molecular switches based on aptamer and toehold DNA, as well as Boolean logic gates (AND and NOR) in response to both DNA and proteins. Also importantly, we developed an on-particle reaction strategy for colorimetric detection of GSH with ultrahigh sensitivity and good specificity. The proposed sensor achieved a broad dynamic range spanning 7 orders of magnitude with a detection limit down to 200 pg mL–1, which was better than that of an in-solution reaction-based biosensor by 2 orders of magnitude. Furthermore, we explored the mechanisms of the interactions at bio-nano interfaces by studying the interfacial factors, including surface coverage, salt concentration, and the curvature of the nanozyme. Abstract:Over the past few decades, researchers have established artificial enzymes as highly stable and low-cost alternatives to natural enzymes in a wide range of applications. A variety of materials including cyclodextrins, metal complexes, porphyrins, polymers, dendrimers and biomolecules have been extensively explored to mimic the structures and functions of naturally occurring enzymes. Recently, some nanomaterials have been found to exhibit unexpected enzyme-like activities, and great advances have been made in this area due to the tremendous progress in nano-research and the unique characteristics of nanomaterials. To highlight the progress in the field of nanomaterial-based artificial enzymes (nanozymes), this review discusses various nanomaterials that have been explored to mimic different kinds of enzymes. We cover their kinetics, mechanisms and applications in numerous fields, from biosensing and immunoassays, to stem cell growth and pollutant removal. We also summarize several approaches to tune the activities of nanozymes. Finally, we make comparisons between nanozymes and other catalytic materials (other artificial enzymes, natural enzymes, organic catalysts and nanomaterial-based catalysts) and address the current challenges and future directions (302 references).

Abstract:This review highlights the recent advancements in the field of nanozymes and their applications in the development of point-of-care biosensors. The use of nanozymes as enzyme-mimicking components in biosensing systems has led to improved performance and miniaturization of these sensors. The unique properties of nanozymes, such as high stability, robustness, and surface tunability, make them an attractive alternative to traditional enzymes in biosensing applications. Researchers have explored a wide range of nanomaterials, including metals, metal oxides, and metal–organic frameworks, for the development of nanozyme-based biosensors. Different sensing strategies, such as colorimetric, fluorescent, electrochemical and SERS, have been implemented using nanozymes as signal-producing components. Despite the numerous advantages, there are also challenges associated with nanozyme-based biosensors, including stability and specificity, which need to be addressed for their wider applications. The future of nanozyme-based biosensors looks promising, with the potential to bring a paradigm shift in biomolecular sensing. The development of highly specific, multi-enzyme mimicking nanozymes could lead to the creation of highly sensitive and low-biofouling biosensors. Integration of nanozymes into point-of-care diagnostics promises to revolutionize healthcare by improving patient outcomes and reducing costs while enhancing the accuracy and sensitivity of diagnostic tools.

ZnO nanostructures in enzyme biosensors

Abstract:Biosensing has developed tremendously since it was demonstrated by Leland C. Clark Jr. in 1962. ZnO nanomaterials are attractive candidates for fabricating biosensors, because of their diverse range of nanostructures, high electron mobility, chemical stability, electrochemical activity, high isoelectric points which promote enzyme adsorption, biocompatibility, and piezoelectric properties. This review covers ZnO nanostructures applied in enzyme biosensors, in the light of electrochemical transduction and field effect transduction. Different assembly processes and immobilization methods have been used to load enzymes into various ZnO nanostructures, providing enzymes with favorable micro-environments and enhancing their sensing performance. We briefly describe recent trends in ZnO syntheses, and the analytical performance of the fabricated biosensors, summarize the advantages of using ZnO nanostructures in biosensors, and conclude with future challenges and prospects.

Abstract:Most natural enzymes are composed of hundreds of amino acid molecules, and widely used in many fields such as biosensor, cancer therapy, immunoassay, food safety and environmental management due to their efficient catalytic ability. However, on account of intrinsic drawbacks of natural enzymes, such as ease of inactivated in extreme conditions, difficulty preparation and high cost, limit the application of enzymes, people make great efforts to develop diverse artificial enzymes of high catalytic activity and stability. With the development of nanomaterials, their catalytic performance has been gradually discovered. These nanomaterials, known as nanozymes, have similar activities with natural enzymes. Due to their unique catalytic activities and the inherent properties, nanozymes have cause great attention in recent decades. So far, more than 50 kinds of nanoparticles have been found to have oxidoreductase activity such as peroxidase (POD), catalase (CAT), superoxide dismutase (SOD), glucose oxidase (GOD). In addition, the catalytic activity can be regulated and designed by changing their size, structure, composition and surface modification. Those nanozymes can be mainly divided into metal oxide nanozymes, noble metal nanozymes and carbon-based nanozymes. Metal oxide nanozymes usually refer to the transition metal oxide nanoparticles. The generation of oxidoreductase activity by integration the nanozymes and the substrates results in valence changes and electron transport. Unlike the catalytic mechanism of metal oxide nanozyme, noble metal nanozymes and carbon-based nanozymes have no valence change of metal elements in the catalytic reaction. They have also been found to exert their activities by the adsorption, activation and surface electron transfer of catalytic substrates. Although the nanozymes have advantage of broader tolerable p H and widely temperature ranges, the enzyme activity generally has p H and temperature dependency similar to the natural enzyme. However, a great advantage of nanozymes is that their activity can be tuned by the size, composition, structure and surface modification of nanomaterials. In general, the smaller the nanozyme size, the greater the specific surface area, the more substrate binding, resulting in the stronger enzyme catalytic activity. Under the same conditions, the 30 nm IONPs have stronger POD activity than IONPs of the 150 and 300 nm size, the smaller Pt NPs have stronger ascorbate oxidase activity and POD activity. Similarly, smaller Au NPs also have stronger POD activity. Moreover, the morphology and structure of nanomaterials also have a considerable impact on the nanozyme activity. Since the catalytic reaction takes place on the surface of the nanozyme, the modification of the nanozyme is also very important for enhancing the sensitivity and selectivity of the nanozymes. Surface-modified molecules include small molecules, synthetic polymers and nucleic acids. In this review, the catalytic mechanisms of the nanozymes in recent years are sorted and classified. In addition, the regulatory factors of the activities and the catalytic specificity of the nanozymes are summarized and analyzed. It is highly expected that establishment of a novel nanozymes as highly stable and low-cost alternatives to natural nanozymes in a wide range of applications in multiple field.

Abstract:In recent years, with the development of nanomaterials, a slice of nanomaterials has been demonstrated to possess high catalytic activity similar to natural enzymes and counter the dilemmas including easy inactivation and low yield natural of enzymes, which are labeled as nanozymes. The catalytic activity of nanozymes could be easily regulated by size, structure, surface modification and other factors. In comparison with natural enzymes, nanozymes featured with a more stable structure, economical preparation and preservation, diversity of functions and adjustable catalytic activity, thus becoming the potentially ideal substitute for natural enzymes. Generally, the are mainly three types containing metal oxide nanozymes, noble metal nanozymes and carbon-based nanozymes, owing various applications in biomedical, energy and environmental fields. In this review, to summarize the recent representative applications of nanozymes, and potentially explore the scientific problems in this field at the same time, we are going to discuss the catalytic mechanisms of diverse nanozymes, with the emphasis on their applications in the fields of tumor therapy, anti-inflammatory and biosensing, hoping to help and guide the future development of nanozymes.

Abstract:The recent discovery of a new class of nanomaterials called nanozymes, which have the action of enzymes and are thus of tremendous significance, has altered our understanding of these previously believed to be biologically inert nanomaterials. As a significant and exciting class of synthetic enzymes, nanozymes have distinct advantages over natural enzymes. They are less expensive, more stable, and easier to work with and store, making them a viable substitute. This practical advantage of nanozymes over natural enzymes reassures us about the potential of this new technology. Peroxidase-like nanozymes have been investigated for the purpose of creating adaptable biosensors via the use of molecularly imprinted polymers (MIPs) or particular bio recognition ligands, including enzymes, antibodies, and aptamers. This review delves into the distinctions between synthetic and natural enzymes, explaining their structures and analytical applications. It primarily focuses on carbon-based nanozymes, particularly those that contain both carbon and hydrogen, as well as metal-based nanozymes like Fe, Cu, and Au, along with their metal oxide (FeO, CuO), which have applications in many fields today. Analytical chemistry finds great use for nanozymes for sensing and other applications, particularly in comparison with other classical methods in terms of selectivity and sensitivity. Nanozymes, with their unique catalytic capabilities, have emerged as a crucial tool in the early diagnosis of COVID-19. Their application in nanozyme-based sensing and detection, particularly through colorimetric and fluorometric methods, has significantly advanced our ability to detect the virus at an early stage.

Abstract:In comparison with natural enzymes, nanozymes show many advantages such as high stability, ease to mass production and tunable enzymatic properties and possess great potentials for application in detection and biosensing fields. Especially, noble metal based nanozymes usually exhibit superior catalytic activity and stability. The achievements in precisely synthesizing noble metal -based nanomaterials at atomic levels enable investigation of interaction mechanisms between structures and enzymatic performances. In this review, the development of noble metal based nanozymes were sumarized, including noble metal nanocrystals and noble metal based single -atom nanozymes. The preparation strategies, regulation methods of enzymatic properties and underlying mechanisms were mainly discussed. The applications for detection and biosensing were also elucidated. At last, the challenges and prospects of this area were briefly discussed.

Abstract:In comparison with bioenzymes, nanozymes exhibit excellent robustness against extreme conditions, a low production cost, and easy-to-adjust properties, as well as potential versatility. These superiorities have attracted abundant interest in the last 15 years, to develop various nanozymes for applications including analytical sensing, environmental engineering, and biomedicine. In particular, for analytical sensing, a lot of nanozyme-involved principles and methods have been explored and applied to clinical diagnosis, environmental monitoring, food safety detection, and forensic analysis. Moreover, rational exploitation and use of nanozyme materials promote the performance of analytical methods. To highlight the latest progress in this attractive field, recent design concepts of nanozymes for advanced biochemical sensing are summarized. The development of single-atom nanozymes, self-cascade nanozymes, structurally biomimetic nanozymes, molecularly imprinted nanozymes, nanozymes breaking the pH limit, and multifunctional nanozymes is discussed in detail, to enhance detection sensitivity and selectivity, as well as expand application scenarios. Finally, some challenges and trends related to nanozyme-based sensors are reported, to satisfy the increasing needs of biochemical analysis with nanozymes.
