Abstract:Insect cuticular hydrocarbons (CHCs) are the main components of the surface lipid layer that function as a barrier against water loss and xenobiotics penetration, and serve as chemical signals. Plasticity of CHC profiles can vary depending upon numerous biological and environmental factors. Understanding these factors associated with variation in CHC profiles is important for identifying potential vulnerabilities relating to pest ecology and developing novel pest management strategies. In this study, we investigated potential sources of variation in CHC profiles of Nilaparvata lugens, Laodelphax striatellus and Sogatella furcifera which are important rice pests in Asia. We quantified CHC profiles among three planthoppers by GC/MS, and explored factors associated with variation in CHC profiles by using principal component analysis (PCA). We further compared the transcriptomes of the planthoppers under different environmental conditions. The results showed that there were significant differences in CHC profiles among the three species. CHC profiles can change with different developmental stage, sex, temperature, humidity and host plant. Genes involving in cuticular lipid biosynthesis pathway are modulated, which might explain why CHC profiles are varying among species; other differentially expressed genes related with amino acid metabolism, cuticle proteins and heat shock proteins might also play roles in regulating lipid metabolism of planthoppers for adapting to different environments. Our study illustrates some of the biological and ecological variations in modifying CHC profiles and the underlying molecular regulation mechanisms of the planthoppers in coping with changes of environmental conditions, and serves to better understanding of molecular ecology regarding with CHCs plasticity.
