Effects of Initial Super-saturation on the Polymorphism and Size Distribution of Glutamic Acid

Abstract:The effect of initial glutamic acid (GA) super-saturation on GA crystallization was investigated by using monosodium glutamate (MSG) solution to replace GA fermentation broth under pH-shift operation mode. The results show that the super-saturation degree of the solution influences both crystal habit and crystal size distribution (CSD). The crystal habit is mainly in α-form and CSD exhibits a symmetric distribution with logarithmic form when super-saturation degree (S) is below 7.15 (S≤7.15). With the increase of super-saturation degree, theβ-form crystal percentage increases and medium size crystallization decreases but with a wider CSD. When S exceedes 14.17 (S≥14.17), all of the crystal habit are in β-form, and CSD becomes dispersive and irregular. Based on the classical nucleation theory, the GA polymorphism nucleation rates under different super-saturation degrees and MSG concentrations are calculated, and the results demonstrates that the polymorphism nucleation rates both increase with the increases of super-saturation degree and MSG concentration. The α-form crystal nucleation rate is faster than that of β-form at the same super-saturation degree, whileβ-form crystal nucleation rate is faster than that ofα-form at the same MSG concentration. The results suggest that the major reason accounting for CSD of glutamic acid is due to the nucleation rate increases with the increase of super-saturation degree, while the crystal habit mainly depends on the difference of the two competitive nucleation rates. A C-pH phase diagram model forα-andβ-GA crystals was proposed based on the existing reports and the results obtained in this study, and this model could theoretically interpret the mechanism of super-saturation degree affecting GA crystal habit.

Abstract:Supersaturation is an important controlling factor for crystallization process and polymorphism. Droplet-based microchannels and conventional crystallization were used to investigate polymorphs of l-gluatamic acid in this work. The results illustrate that it is easy to realize the accurate and rapid control of the crystallization temperature in the droplets, which is especially beneficial to heat and mass transfer during crystallization. It is also noted that higher degree of supersaturation favors the nucleation of α crystal form, while lower degree of supersaturation favors the nucleation of β crystal form under droplet-based microchannels for l-gluatamic acid. In addition, there is a different nucleation behavior to be found under droplet-based microchannels both for the β form and α form of l-glutamic acid. This new finding can provide important insight into the development and design of investigation meanings for drug polymorph.

Abstract:Solution-mediated polymorphic transformation of L-glutamic acid (L-Glu) from alpha to beta form was studied by combining experiments and molecular simulation. Experimental results show that the beta form mainly nucleates on the three surfaces of the alpha form during the transformation, and the corresponding-probability follows the order of {011} > {111} > {001}. Molecular simulation of the adsorption of molecule initially with alpha or beta conformation on the three surfaces indicates that regardless-of the initial conformation, the adsorption energies decrease in the order of {011} > {111} > {001}, which is consistent with the experimental results. Moreover, it reveals that the three surfaces have different abilities to induce the conformational change of alpha to beta. Especially, for a molecule adsorbed on the {011} surface initially with alpha conformation, it will evolve into the beta conformation. These findings provide insights about the crucial, role of alpha form Surfaces in the polymorphic transformation of L-Glu.

Abstract:Online powder X-ray diffraction, employing a flow-through cell [Hammond et al. Cryst. Growth Des. 2004, 4, 943] and a previously developed chemometric method [Chen et al. Anal. Chem. 2005, 77, 6563], was applied for quantitative analysis of the polymorphic phase transformation from the metastable α-form to the stable β-form of l-glutamic acid (LGA). The process of interconversion was monitored in aqueous slurries, using a jacketed 400 mL magnetically stirred reactor as a function of temperature. Calibration studies revealed that the current detection limits are ca. 0.8 and 0.2 wt%, respectively, for α- and β-forms of LGA as determined for slurries of LGA in methanol and good signal detection can be accomplished using a data acquisition time of 10 s albeit with increased noise levels. Abstract:The inconsistency between solution chemistry and the crystal structure indicates the potential conformational evolution and rearrangement during the nucleation process. However, the mechanism of how the conformation is selected to form the corresponding polymorph is still not clear due to the complicated crystallization environment. In this work, conformation and polymorph selectivity of three anhydrous forms of gabapentin were investigated. Quantum chemical computation was used to analyze gabapentin conformations in the solution state and explore their transformation pathway. Besides, visual molecular dynamics was adopted to calculate the structural difference of the conformers in solution and crystal states, which was closely related to conformational selectivity. Moreover, the effects of solvent types and the supersaturation on solute–solvent interactions were investigated by Fourier transform infrared spectra and molecular dynamics simulations. Combined with the outcome of rapid cooling experiments, it was found that the solute–solvent interaction may affect the difficulty of desolvation so as to affect the conformational selectivity before nucleation. A special evaporation experiment was designed to obtain the unstable polymorph of gabapentin. Finally, the possible mechanism of selective polymorph crystallization was proposed and analyzed.

Abstract:The dependence of crystal growth rate of L-alanine on solution supersaturation was investigated by combining experiments and molecular dynamics (MD) simulations. The experimental results show that lower supersaturated solution yields more elongated L-alanine crystals along the c-axis, i.e., the aspect ratio (c/b) of the crystal decreases with the increase of solution supersaturation, which is due to the higher supersaturation inducing a rise in the relative growth rate between the main side surface (the (120) surface) and the main end surface (the (011) surface). MD simulations on the two surfaces in contact with different supersaturated solutions revealed that the solute molecules tend to be more efficiently attached to the (011) surface than to the (120) surface at both supersaturations studied, as the interaction between the solute molecules and the L-alanine molecules in the first layer of the (011) surface is stronger than that of the (120) surface. However, higher supersaturation leads to larger relative interaction energy between the (120) and (011) surfaces, suggesting an increase in the relative growth rate of the two surfaces (R-(120)/R-(011)) with super saturation, which is in agreement with the experimental results.

Abstract:In this study, L-carnosine was chosen as the model compound to systematically study solution-mediated polymorphic transformation by online experiment and theoretical simulation. Form II, a new polymorph of L-carnosine, was developed using an antisolvent crystallization method. The properties of form I and form II L-carnosine were characterized by powder X-ray diffraction, polarizing microscope, thermal analysis, and Raman spectroscopy. In order to explore the relative stability, the solubility of L-carnosine form I and form II in a (water + DMAC) binary solvent mixture was determined by a dynamic method. During the solution-mediated polymorphic transformation process of L-carnosine in different solvents, Raman spectroscopy was employed to detect the solid-phase composition of suspension in situ, and the gravimetric method was used to measure the liquid concentration. In addition, the effect of the solvent on the transformation process was evaluated and analyzed. Finally, a mathematical model of dissolution–precipitation was established to simulate the kinetics of the polymorphic transformation process based on the experimental data. Taking the simulation results and the experimental data into consideration, the controlling step of solution-mediated polymorphic transformation was discussed.
Solution Thermodynamics of L-Glutamic Acid Polymorphs from Finite-Sized Molecular Dynamics Simulations

Abstract:Efficiently obtaining atomic-scale thermodynamic parameters characterizing crystallization from solution is key to developing the modeling strategies needed in the quest for digital design strategies of industrial crystallization processes. Based on the thermodynamics of crystal nucleation in confined solutions, we develop a simulation framework to efficiently estimate the solubility and surface tension of organic crystals in solution from a few unbiased molecular dynamics simulations at a reference temperature. We then show that such a result can be extended with minimal computational overhead to capture the whole solubility curve. This enables an efficient and self-consistent estimate of the solubility and limit of solution stability associated with crystal nucleation in molecular systems from equilibrium molecular dynamics without requiring sophisticated free energy calculations. We apply our analysis to investigate the relative thermodynamics stability and aqueous solubility of the α and β polymorphs of L-Glutamic acid. Our analysis enables an efficient appraisal of emergent ensemble properties associated with the thermodynamics of nucleation from solutions against experimental data, demonstrating that while the absolute solubility is still far from being quantitatively captured by an off-the-shelf point charge transferable forcefield, the relative polymorphic stability and solubility obtained from finite temperature simulation are consistent with the experimentally available information on glutamic acid. We foresee the ability to efficiently obtain solubility information from a limited number of computational experiments as a key component of high-throughput computational polymorph screenings.

Abstract:Supersaturation control (SSC) is a simple yet effective technique to control, among other properties, the purity, shape, and polymorphic form of crystals during the crystallization process. However, for the enantiotropic polymorph systems it is difficult to efficiently operate the traditional batch SSC because of the intersecting solubility lines and hence the temperature dependence of polymorph stability. From a thermodynamic perspective, the transition temperature in a batch crystallization is the minimum final temperature for the polymorph that is stable at high temperature, and it is the maximum initial temperature for the polymorph that is stable at lower temperatures. The existence of a transition point between the two polymorphs often makes it difficult to produce consistently one or the other form with good yield in a batch crystallization process. Due to the constant-temperature operation, continuous tank crystallizers could overcome this problem, but the scale of production and other technology challenges often does not justify their application. This paper proposes a semibatch cooling SSC strategy, having as the manipulated variable the feed flow rate of a high-temperature, concentrated inlet stream in the same solvent system as used in the crystallization process. The crystallizer temperature is fixed under the transition temperature, and the concentration corresponding to the supersaturation set point is undersaturated for the counter polymorph, ensuring in this way thermodynamic polymorph selection. The system is presented through the case study of p-aminobenzoic acid (PABA) crystallization from ethanol for SSC of both enantiotropic polymorphs: alpha-PABA can be obtained in both batch and semibatch operation. According to the literature, beta-PABA cannot be easily obtained in batch cooling crystallization, but the semibatch setup was able to produce it consistently without yield limitations due to the transition temperature.

Abstract:Controlling the solid-state properties is particularly important in the pharmaceutical field, where polymorphism and morphology have a significant impact on drug properties. Sulfathiazole (ST) is a highly and concomitantly polymorphic model system. We found that pure form II cauliflower-like ST spherulites can be obtained by cooling crystallization by only adjusting the gelatin concentration in water. The ST polymorphism and morphology were characterized by PXRD, Raman and SEM. The results showed that the gelatin additive (>= 0.02 g per 100 g H2O) exhibited high selectivity to ST form II. We investigated the evolution of ST morphology by varying the gelatin concentration and initial supersaturation. The gelatin additive had the effect of inducing branching and inhibiting nucleation and growth, and promoted the formation of cauliflower-like spherulites. The mechanism for simultaneous control of polymorphism and morphology via gelatin induction was proposed, and the gelatin concentration was a dominant factor in this system. These findings provide new strategies for understanding polymorphic nucleation pathways and inducing organic spherulites.

Abstract:The most stable conformers of L-monosodium glutamate and D-monosodium glutamate in the gas phase were selected by density functional approaches.The calculated results show that the neutral structures of L-monosodium glutamate and D-monosodium glutamate are more stable than that of dipolar ions in the gas phase.The results of NBO analysis are in accord with the electronegativity analysis.The analyses of frontier orbital energies suggest that the L-monosodium glutamate as an electron donor is more active than D-monosodium glutamate.The topological properties indicate that both L-and D-monosodium glutamates have two intramolecular hydrogen bondings.The calculation of electronic circular dichroism exhibits that electron transition from ground state to singlet excited states of L-and D-monosodium glutamate is chiral optically active.

Analysis of the diffraction peaks arising from the (111) reflection, 2 theta = 18 degrees for the a-forth and (102) reflection, 2 theta = 31 degrees for the beta-form, was used to obtain the rates of phase interconversion. Phenomenologically, the rate law observed for dissolution of the alpha-form was -D-alpha = k(d alpha) and the rate law observed for growth of the beta-form was D-beta = k(g beta)C(beta) where C-beta is the concentration of the beta-form expressed in weight percent. At 45 degrees C, the values of the rate constants k(d alpha) and k(g beta) were 18 x 10(-4) wt% s(-1) and 3 x 10(-4) s(-1), respectively. Activation energies calculated, using the Arrhenius relationship, were found to be 43.9 kJ/mol with k(d alpha 0) calculated to be 25.9 x 10(3) wt% s(-1) for the dissolution of alpha-form and 47.0 kJ/mol with k(g beta 0) calculated to be 16.2 x 10(30) s(-1) for the growth of the beta-form. The activation energy calculated from a different, characteristic peak of the beta-form at 2 theta = 21 degrees corresponding to the reflection from (101) planes, was 32.1 kJ/mol suggesting that there might be some texture effects manifested in the flow field of the crystalline slurry. The overall quantitative accuracy of this method together with potential for improvement is also discussed.

Abstract:From a physicochemical viewpoint, amyloid fibril formation is a phase transition from soluble to crystal-like sates limited by supersaturation. It occurs only above solubility (i.e., the solubility limit) coupled with a breakdown of supersaturation. Although many studies have examined the role of molecular chaperones in the context of proteostasis, the role of supersaturation has not been addressed. Moreover, although molecular chaperone-dependent disaggregations have been reported for preformed amyloid fibrils, amyloid fibrils will not dissolve above the solubility of monomers, even if agitations fragment long fibrils to shorter amyloid particles. On the other hand, on considering a reversible and coupled equilibrium of interactions, folding/unfolding and amyloid formation/disaggregation, molecules stabilizing native states can work as a disaggregase reversing the amyloid fibrils to monomers. It is likely that the proteostasis network has various intra- and extracellular components which disaggregate preformed amyloid fibrils as well as prevent amyloid formation. Further studies with a view of solubility and supersaturation will be essential for comprehensive understanding of proteostasis.

Abstract:Nucleation is a combined result of thermodynamics and kinetics. Calcium sulfate dihydrate (DH), alpha-calcium sulfate hemihydrate (alpha-HH), and calcium sulfate anhydrite (AH) all have the potential to precipitate when the solution is supersaturated with them at the same time. The effect of supersaturation on CaSO4 phase's occurrence was investigated in a 3.00 m CaCl2 solution at 91 degrees C to better understand the kinetics of competitive nucleation. In a wide range of supersaturations, DH and alpha-HH precipitated concomitantly without AR forming. The molar fraction of alpha-HH in the precipitate increased gradually from 47.23% to a summit of 98.91% when the supersaturation (S-HH) varied from 1.90 to 3.76 and then followed a dramatic decrease to 18.77% with a further increase in S-HH to 6.26. The predominant nucleation shifted from alpha-HH at the lower supersaturations to DH at the higher supersaturations. The crystallization rate evolution with supersaturation was responsible for the growth and decline of DH and alpha-HH in the precipitate.

Abstract:The in situ measurement of solution supersaturation associated with the batch cooling crystallization Of L-glutamic acid (LGA) at 500 mL and 20 L scale sizes is assessed via ATR-FTIR spectroscopy. A partial least squares chemometric calibration model was developed for the online prediction of LGA concentration from measured FTIR absorbance spectra overcoming some significant challenges related to the low sensitivity of LGA in the mid-IR frequency range, its low solubility in water, and its complex speciation chemistry. The solubility data of LGA in water over the temperature range from 40 to 80 degrees C, using ATR-FTIR, reveals excellent agreement with those obtained both from using a gravimetric method and literature data. The metastable zone width determined using the turbidimetric methods as a function of heating/cooling rates and solute concentration is found to increase with increasing cooling rate while it decreases with increasing solution concentration. Monitoring online crystallization via both spontaneous and seeded in 500 mL and 20 L crystallizers reveals good concentration predictions for seeded crystallization, while fouling of the ATR crystal prevents its routine use for unseeded crystallization studies. Higher supersaturation levels are found for the larger crystallizer scale-size consistent with enhancement of secondary nucleation at the smaller scale-size.

Abstract:Penicillin sulfoxide is the intermediate for the synthesis of 7-amino-3-desacetoxycephalosporanic acid which is one of the most important nucleuses of cephalosporin antibiotic. In this contribution, two crystal structures of penicillin sulfoxide (forms I and II) were determined by X-ray diffraction, and their thermotropic properties were investigated by differential scanning calorimetry (DSC). Furthermore, the transformation of form II to form I was studied quantitatively by Raman spectroscopy, and its rates at different temperatures were determined. The results indicate that penicillin sulfoxide is more stable as form I, and the temperature plays an important role in the crystal transformation.

Abstract:Glutathione (GSH) is a bioactive oligopeptide which was widely used in sports, pharmaceutical and food industry. This paper compared the properties of two crystalline forms of GSH (Form α and Form β) in thermodynamics and explored the transformation of corresponding polymorph. The solubility data of GSH were determined by using the dynamic method in binary mixed solvents (methanol + water, ethanol + water, isopropanol + water, ethylene glycol + water) at temperatures ranging from 278.15 to 313.15 K. The solubilities of the two forms both increased with the increasing of mole fraction of water and temperature. The modified Apelblat equation, the CNIBS/R-K model and the Jouyban-Acree model were used to correlate the experimental solubility. The mixing thermodynamic properties of GSH in the binary mixed solvents were calculated by Van't Hoff equation. Form β was supposed to be the stable crystalline form based on the thermodynamics analysis, which corresponded to the solvent-mediated transformation from Form α to Form β.

Is there a correlation between the glass forming ability of a drug and its supersaturation propensity?

Abstract:The use of an enabling formulation technique, such as amorphization of a poorly water-soluble crystalline drug, can result in supersaturation with respect to the crystalline form of the drug and thus potentially in a higher degree of absorption after oral administration. The ease with which such drugs can be amorphized is known as their glass forming ability (GFA). In this study, a potential correlation between GFA and supersaturation propensity is investigated. The GFA of 23 different drugs was determined by melt quenching or milling the crystalline drugs to obtain their respective amorphous forms. The inherent propensity of the drug to supersaturate, i.e. the maximal apparent degree of supersaturation (aDS), and the time until precipitation at a given aDS were determined. Supersaturation was induced via a solvent shift method where the drug was initially dissolved in dimethyl sulfoxide and then added to a biorelevant medium (fasted state simulated intestinal fluid).The study showed that drugs which are good glass formers also have the potential to supersaturate to a high degree (high maximal aDS) whereas drugs that are modest glass formers supersaturate only to a low degree. This correlation was confirmed by principal component analysis, which also indicated that melt enthalpy inversely correlated with both GFA and maximal aDS. However, no correlation between GFA of a drug and the time until precipitation at a given aDS was found.

Abstract:The high risk of solution-mediated phase transformation (SMPT) from the metastable monohydrate to stable Form I makes it difficult to produce pure metastable monohydrate of calcium d-gluconate. In this work, we explored the effect of various operating parameters on the SMPT of calcium d-gluconate in water and proposed an effective approach to obtain the desired monohydrate. First, the two forms of calcium d-gluconate were characterized and compared using powder X-ray diffraction (PXRD), thermal analysis, and Raman spectroscopy. The lower solubility of Form I in water illustrates its higher thermodynamic stability than monohydrate when the temperature is higher than 292 K. Afterward, the SMPT of calcium d-gluconate from monohydrate to Form I was investigated in water using in situ Raman spectroscopy combined with scanning electron microscopy and PXRD. Results showed that the nucleation and growth of Form I was the rate-limiting step in the SMPT from monohydrate to Form I. The phase transformation from monohydrate to Form I was delayed to produce pure monohydrate by decreasing temperature and agitation rate, reducing the amount of solid loading, and increasing the particle size of solid loading. Furthermore, the transformation kinetics were studied by the JMA model to explore how temperature influences the SMPT process. This study enriches the study of the calcium d-gluconate SMPT mechanism, and also provides guidance for obtaining high-quality injection-grade calcium gluconate monohydrate.
