Abstract:Atom- and step-economy in IBX assisted diversity-oriented synthesis is achieved with a versatile AQ auxiliary α-amino acid analogs offering rapid access to polycyclic spiro-quinolines featuring a quaternary stereocenter in 20%–91% yields under mild conditions via 7,8-dearomatization of quinolines. Free of a preinstalled activation group is highlight of this intramolecular oxidation spiroannulation tandem reaction. This type of N-heterospirocycles, traditionally difficult to access, may open the door to a potentially interest scaffold for synthetic and medicinal chemistry.

Abstract:Quinoline synthesis from easily accessible raw materials such as anilines is a valuable and meaningful task. Herein, we communicate an iodide- and silver-mediated C-H/C-H oxidative annulation-aromatization between anilines and allyl alcohols. This protocol provides a direct route to the synthesis of quinoline derivatives from inexpensive commodities. Various kinds of anilines, even heterocyclic anilines, were shown to be workable substrates, generating the corresponding multi-substituted quinolines in good yields.

Abstract:2-Perfluoroalkylated quinolines were efficiently synthesized via the Michael addition of perfluoroalk-2-ynoates with isatins, followed by the reaction with alcohols through an intramolecular rearrangement cyclization in the presence of Na2CO3. Under the mild and metal-free reaction conditions, a broad scope of quinolines was synthesized and the mechanism was proposed.

Abstract:A highly enantioselective rhodium(I)-catalyzed dearomative arylation or alkenylation of easily available N-alkylquinolinium salts is reported, thus providing an effective and practical approach to the synthesis of dihydroquinolines in up to 99 % ee. This reaction tolerates a wide range of functional groups with respect to both the organic boronic acids and the quinoline starting materials. Moreover, the synthetic utility of this protocol is demonstrated in the formal asymmetric synthesis of bioactive tetrahydroquinoline and the total syntheses of (-)-angustureine and (+)-cuspareine.

Abstract:An efficient method for the synthesis of substituted quinolines is described. By FeCl3-promoted C–C bond formation, 2,3-disubstituted and 2,3,4-trisubstituted quinolines were successfully synthesized from the reaction between imines and electron-deficient alkynes in good yields. The reaction can also be performed in a sequential three-component manner by using aromatic amines and aromatic aldehydes instead of the corresponding imines.

Abstract:We describe here an approach for synthesizing quinolines either from N-alkyl anilines or from anilines and aldehydes. A dual-catalyst system consisting of a photocatalyst and a proton reduction cocatalyst is employed. Without the use of any sacrificial oxidant and under extremely mild conditions, the reactions afford quinolines in excellent yields and produce H2 as a byproduct.

Abstract:Disclosed herein is a novel one pot synthesis of C3-sulfonate esters and C4-chloro of quinolines from quinoline N-oxides with sulfonyl chloride. Sulfonyl chlorides play the dual role of sulfonylation and chlorination. Both arenesulfonyl chlorides and aliphatic sulfonyl chlorides are suitable precursors for sulfonation and chloration reagent. Therefore, a wide variety of C3-sulfonate esters and C4-chloride of quinolines were afforded in manners under metal-free conditions. This procedure features with practicality, high efficiency, environmental friendliness and atomic economy.

Abstract:We developed an efficient and convenient method for metal‐free bromination of 8‐aminoquinoline scaffolds on the C5 position that is geometrically inaccessible. And this bromination step can be followed in a one‐pot manner with five kinds of C−X (X=C, O, S) cross‐coupling reactions without any extra workup, such as removal of the solvent or filtration for the separation of intermediate. The reaction tolerates a wide scope of alkyl and aryl amides as well as (hetero)aryl boronic acids, generating the corresponding arylation products in good to excellent yields. Furthermore, the one‐pot alkenylation, alkynylation, thiolation and phenoxylation of quinolines on the C5 position can be efficiently realized using this brominated quinoline amide molecular platform. Thus, the method shows high practical potential in industrial organic synthesis.

Abstract:C-N bond formation is crucial for the preparation of synthetic organic nitrogen-containing heterocyclic compounds. This work provides a facile and highly efficient strategy (ca. similar to 92 %) for the synthesis of quinazoline compounds, which involves tandem C-H/N-H bond functionalization using metal-free conditions without the need for an external oxidant. Meanwhile, the reaction system exhibits good functional group tolerance and a wide substrate scope. Furthermore, our corresponding isotope tracking experiments have shown that dimethyl sulfoxide acts as a carbon source. Finally, this work affords a new strategy toward the further design and synthesis of heteroaromatic rings.

Abstract:An electrochemical protocol for the synthesis of 2,3-disubstituted quinolines by reacting benzoxazinones with arylsulfonyl hydrazides was established under simple and mild conditions (room temperature and an undivided cell). The methodology does not require noble catalysts or external oxidants, providing a green and mild pathway for de novo synthesis of functional quinolines with excellent regioselectivity. Moreover, the method tolerated a variety of functional groups. Notably, the scaled-up experiment and follow-up procedures demonstrate the practicality of electrochemical synthesis.

Abstract:Al‐based MOF catalyst with a typical spatial distance of Lewis and Brønsted acid active sites can efficiently catalyse Friedländer synthesis with high yields of physiologically active quinoline. We report on the use of heterogeneous metal organic framework (MOF) catalysts for the Friedländer synthesis of 2‐aminoaryl ketone with α‐methylene carbonyl derivatives, a versatile and widely used method for the synthesis of quinoline derivatives. The importance of the Lewis and Brønsted acid sites and their spatial proximity have been explored. Postsynthetic modifications were made to introduce the Brønsted −SO3H acid sites in the pristine MIL‐53(Al). Two modifications were carried out, resulting in two different MOFs: MIL‐53(Al)−NH−(CH2)3−SO3H, in which there is a significant spatial distance between the Lewis and Brønsted sites, and MIL‐53(Al)−SO3H, where the two sites are in close proximity. The availability of sulfonic acid groups has a significant impact on their catalytic performance. The sulfonated MIL‐53(Al) material exhibited a high yield of quinoline derivatives under solventless and mild reaction conditions, with a wide substrate scope and good reusability. Mechanistic studies revealed that the dual acidic sites of the catalysts play an important role in the reaction mechanism.

Abstract:A new and metal-free three-component method for the synthesis of 2,4-disubstituted quinolines via the reactions of anilines, α-keto acids and alkyl lactates is reported. The reactions proceed in the presence of p-toluene sulfonic acid (p-TSA) and tert-butyl peroxybenzoate (TBPB) to provide diverse quinoline products via the construction of new CC double, C-C single and CN double bonds without producing any organic mass-based side product. Notably, the anti-inflammatory activity of the quinolines has been investigated by measuring their ability to inhibit NO release by lipopolysaccharide (LPS) induced RAW264.7 cells, leading to the identification of 4i, 4t and 4x as potent anti-inflammatory compounds in vitro.
