Abstract:In the past decade, polymer vesicles prepared by self-assembly techniques have attracted increasing scientific interest based on their unique features highlighted with tunable membrane properties, versatility, stability, and capacity of transporting hydrophilic as well as hydrophobic species. Polymersomes exhibit intriguing potential applications such as cell mimicking dimensions and functions, tunable delivery vehicles, for the templating of biomineralization, nanoreactors, and as scaffolds for biological conjugation. In this Feature Article, an overview of the preparation and application of recently developed smart polymer vesicles, which can respond to the novel external stimuli, including carbon dioxide (CO2), electrochemical potential, ultrasound, enzyme, near-infrared light, and magnetic field is given. The response mechanism and morphology change are explored with specific focus on the functionalization of various domains of the polymer vesicles. In addition, the current limitations are explored as well as the challenges facing the development of these nanostructures toward real-world applications.

Abstract:Poly(N,N-dimethylaminoethyl methacrylate) (PDMAEMA) with a beta-cyclodextrin (beta-CD) at the chain end was synthesized via atom transfer radical polymerization (ATRP); poly(epsilon-caprolactone) (PCL) with a -C=C- segment and an adamantane (Ada) group at two ends, respectively, was prepared through ring-opening polymerization (ROP), and poly(N-isopropylacrylamide) (PNIPAM) with a -S-C(S)-S- segment, which can be converted into a thiol group, was yielded by reversible addition-fragmentation chain transfer polymerization (RAFT). A supramolecular triblock stimuli-responsive copolymer PNIPAM-b-PCL-b-PDMAEMA having good biocompatibility with PNIPAM and PDMAEMA hydrophilic segments and PCL hydrophobic segment was constructed by thiol-ene Michael addition and host- guest interaction. The triblock copolymer could self-assemble into vesicles and respond to carbon dioxide (CO2) gas and temperature reversibly. Under the stimulation of CO2, the vesicular assemblies swelled obviously; while raising the temperature from 25 to 40 degrees C, the assemblies displayed a conversion between vesicles and spherical micelles.

Abstract:CO2-responsive bowl-shaped polymersomes caused by osmotic pressure have been successfully designed and constructed. Upon CO2-stimulus, these polymersomes can transform from bowl-shape into wrinkled structure because of the restricted hydration effect. These promising polymersomes with special structure upon exposure to biological gas are expected to be useful in nanomedicine.

Abstract:This paper reviews liposomes with crystalline phase and polymersomes exhibiting crystalline and thermotropic liquid crystalline phases in the membrane. Intriguing morphologies of vesicles are described, including spherical, ellipsoidal and faceted vesicles, produced by a large variety of amphiphilic molecules and polymers with nematic phase, smectic phase or crystalline phase. It is highlighted how the phase transitions and the phase grain boundaries could be used ingeniously to destabilize the vesicular structure and to achieve cargo-release under the action of external stimulation. These liposomes and polymersomes are responsive to physical stimuli, such as temperature variation, shear stress, light illumination, and magnetic and electric fields. These stimuli-responsive properties make them promising candidates as new smart drug delivery systems.

Abstract:Stimuli-responsive polymeric vesicles have attracted great attention in drug delivery due to their intrinsic hollow structures and "on demand" release of drugs upon environmental stimuli. The drug-release kinetics from polymeric vesicles, which is usually dependent on the stimuli-responsive behaviors of the polymeric vesicle, has great impacts on the therapeutic efficacy. Over the past decade, polymerization-induced self-assembly (PISA) has been demonstrated to be a powerful strategy to prepare the polymeric vesicles. However, fabrication of stimuli-responsive vesicles with adjustable drug release kinetics via PISA has been rarely reported, which may be due to the poor selectivity of functional membrane-forming polymers in the PISA system. Herein, a series of vesicles with different pH-responsive behaviors were fabricated via RAFT dispersion copolymerization of (diisopropylamino)ethyl methacrylate (DIPEMA) and benzyl methacrylate (BzMA). Both the content of DIPEMA units in hydrophobic P(DIPEMA-co-BzMA) blocks and the DP of P(DIPEMA-co-BzMA) have great influence on the pH-responsive behaviors of the vesicles, which can be used to adjust the responsive release rate of RhB from the vesicles upon the same pH switch.

Abstract:CO2-responsive polymers are a class of polymers that can respond to CO2 gases and solutions. The reaction with CO2 changes the properties of the polymer such as hydrophobicity and charge. By introducing argon, nitrogen and air, or by slightly heating, CO2 can be removed and the properties of the polymer and composite material can be fully or partially restored. This review summarizes the recent development of CO2-responsive polymers and composites, introduces different types of responsive polymers from chemical structures and responsive groups, including amidines, tertiary amines, guanidines and imidazoles, and summarizes the monomers of common CO2-responsive polymers. The different functions of CO2-responsive polymers are introduced. Different properties of vesicles, spherical micelles and composite assemblies and other morphologies are discussed from the viewpoint of assemblies, while the construction and effects of emulsions, aerogels and hydrogels, functional surfaces and nanofiber membranes, organic-inorganic hybrid materials and composites, and fluorescence sensing and monitoring systems are summarized progressively. Finally, the problems that need to be explored and optimized in this field are discussed, and the future development direction is foreseen. [Graphics] .

Abstract:CO2-stimuli responsive polymers are a class of newly developed smart stimuli responsive polymers, which usually refers to the polymers possessing reversible changes upon admission and emission of CO2. Since the regulation process only involves CO2 and inert gases, without the introduction of other impurities, there are many potential applications in this field. This article summarized recent research progress on the preparation of CO2-stimuli responsive polymers, followed by the discussion of their self-assembly, classifying in accordance with the CO2-switchable groups,such as primary amine, amide and some specific polymer chains. The development prospect of this research field and its potential applications are also discussed.
Photo- and Reduction-Responsive Polymersomes for Programmed Release of Small and Macromolecular Payloads

Abstract:We report on the preparation of photo- and reduction-responsive diblock copolymers through reversible addition fragmentation chain transfer (RAFT) polymerization of a coumarin-based disulfide-containing monomer (i.e., CSSMA) using a poly(ethylene oxide) (PEO)-based macro RAFT agent. The resulting amphiphilic PEO-b-PCSSMA copolymers self-assembled into polymersomes with hydrophilic PEO shielding coronas and hydrophobic bilayer membranes. Upon irradiating the polymersomes with visible light (e.g., 430 nm), the coumarin moieties within the bilayer membranes were cleaved with the generation of primary amine groups, which spontaneously underwent inter/intrachain amidation reactions with the ester moieties, thereby tracelessly cross linking and permeating the bilayer membranes. Notably, this process only gave rise to the release of small molecule payloads (e.g., doxorubicin hydrochloride, DOX) while large molecule encapsulants (e.g., Texas red-labeled dextran, TR-dextran) were retained within the cross-linked polymersomes due to the preservation of the integrity of the vesicular nanostructures. However, cross-linked polymersomes undergo further structural disintegration upon incubation with glutathione (GSH) due to the scission of disulfide linkages, resulting in the release of macromolecular payloads. Thus, dual-stimuli responsive polymersomes with tracelessly cross-linkable characteristics enable sequential release of payloads with spatiotemporal precision, which could be of promising applications in synergistic loading and programmed release of therapeutics.

Abstract:Despite numerous studies on utilizing polymeric vesicles as nanocapsules, fabrication of tunable molecular pathways on transportable vesicle walls remains challenging. Traditional methods for building penetrated channels on vesicular membrane surface often involve regulating the solvent polarity or photo-cross-linking. Herein, we developed a neat, green approach of stimulation by using CO2 gas as "molecular drill" to pierce macroporous structures on the membrane of polymersomes. By simply introducing CO2/N2 gases into the aqueous solution of self-assemblies without accumulating any byproducts, we observed two processes of polymeric shape transformation: "gas breathing" and "gas piercing." Moreover, the pathways in terms of dimension and time were found to be adjustable simply by controlling the CO2 stimulation level for different functional encapsulated molecules in accumulation, transport, and releasing. CO2-breathing and piercing of polymersomes offers a promising functionality to tune nanocapsules for encapsulating and releasing fluorescent dyes and bioactive molecules in living systems and also a unique platform to mimic the structural formation of nucleus pore complex and the breathing process in human beings and animals.

Abstract:The amphiphilic diblock copolymer composed of poly(ethylene oxide) as the hydrophilic chain and polymethacrylate with azopyridine moieties in the side groups as the hydrophobic chain self-assembled to vesicles in the mixture of THF/water. The dual photo- and pH-responsive behavior of the copolymeric vesicles was investigated by UV–Vis absorption spectra and optical microscope. It was found that the trans–cis photoisomerization of azopyridine in the vesicles was influenced by the water content in the mixture and the intensity of light irradiation. Regard to the pH-responsive behavior, the copolymer vesicles dissociated into solution gradually triggered by acid, while they reformed in a basic environment. Furthermore, the UV light irradiation accelerated the process of pH-induced dissociation of the vesicles.

Abstract:Multistimuli-responsive precise morphological control over self-assembled polymers is of great importance for applications in nanoscience as drug delivery system. A novel pH, photoresponsive, and cyclodextrin-responsive block copolymer were developed to investigate the reversible morphological transition from micelles to vesicles. The azobenzene-containing block copolymer poly(ethylene oxide)-b-poly(2-(diethylamino)ethyl methacrylate-co-6-(4-phenylazo phenoxy)hexyl methacrylate) [PEO-b-P(DEAEMA-co-PPHMA)] was synthesized by atom transfer radical polymerization. This system can self-assemble into vesicles in aqueous solution at pH 8. On adjusting the solution pH to 3, there was a transition from vesicles to micelles. The same behavior, that is, transition from vesicles to micelles was also realizable on addition of beta-cyclodextrin (beta-CD) to the PEO-b-P(DEAEMA-co-PPHMA) solution at pH 8. Furthermore, after beta-CD was added, alternating irradiation of the solution with UV and visible light can also induce the reversible micelle-to-vesicle transition because of the photoinduced trans-to-cis isomerization of azobenzene units. Abstract:To well adapt to the complicated physiological environments, it is necessary to engineer dual- and/or multi-stimuli responsive drug carriers for more effective drug release. For this, a novel temperature responsive lateral chain photosensitive block copolymer, poly[(N-isopropylacrylamide-co-N,N-dimethylacrylamide) -block-propyleneacylalkyl-4-azobenzoate] (P(NIPAM-co-DMAA)-b-PAzoHPA), is synthesized by atom transfer radical polymerization. The structure is characterized by 1 H nuclear magnetic resonance spectrometry and laser light scattering gel chromatography system. The self-assembly behavior, morphology, and sizes of micelles are investigated by fluorescence spectroscopy, transmission electron microscope, and laser particle analyzer. Dual responsiveness to light and temperature is explored by ultraviolet-visible absorption spectroscopy. The results show that the copolymer micelles take on apparent light and temperature dual responsiveness, and its lower critical solution temperature (LCST) is above 37 °C, and changes with the trans-/cis- isomerization of azobenzene structure under UV irradiation. The blank copolymers are nontoxic, whereas the paclitaxel (PTX)-loaded counterparts possessed comparable anticancer activities to free PTX, with entrapment efficiency of 83.7%. The PTX release from the PTX-loaded micelles can be mediated by changing temperature and/or light stimuli. The developed block copolymers can potentially be used for cancer therapy as drug controlled release carriers.

Abstract:pH-Responsive amphiphilic graft macromolecules consisting of a polyphosphazene backbone, hydrophilic PEG branches and pH-sensitive DPA were successfully synthesized and characterized. The copolymer can self-assemble into vesicles in an aqueous solution with unique inner structure and homogeneously encapsulate both lipophilic and hydrophilic molecules. The pH-dependent structure change of vesicles was also observed by DLS and TEM. Dox-loaded vesicles exhibit a sharp pH-responsive drug release profile and dramatically enhance the cytotoxicity of Dox against Dox-resistant MCF-7/adr cells. These results suggest such vesicles based on pH-responsive polyphosphazene hold great potential for specific drug therapy.

Abstract:A spiropyran-based amphiphilic random copolymer was synthesized and self-assembled into photo-, pH-, and thermo-responsive micellar nanoparticles. The triple-stimuli triggered morphological changes of the nanoparticles were revealed by TEM and DLS. Highly efficient controlled release of encapsulated molecules, coumarin 102, from the nanoparticles under stimulation of UV light, acid and the combined stimuli could be realized.

Abstract:A new concept of designing a photocontrollable supramolecular polymer nanocontainer through the electrostatic association between an azobenzene-containing surfactant (AzoC10) and a double-hydrophilic block ionomer, poly(ethylene glycol)-b-poly(acrylic acid) (PEG(43)-PAA(153)), is described. Such a block ionomer complex can self-assemble in aqueous solution and form vesicle-like aggregates, which are composed of a poly(ethylene glycol) corona and a poly(acrylic acid) shell associated with azobenzene-containing surfactant. The photoisomerization of azobenzene moieties in the block ionomer complex can reversibly tune the amphiphilicity of the surfactants, inducing the disassembly of the vesicles. Such block ionomer complex vesicles are further evaluated as nanocontainers capable to encapsulate and release guest solutes on demand controlled by light irradiation. For example, the vesicles encapsulating the fluorescein sodium display clear spherical images observed by fluorescence microscopy. However, such fluorescence-marked images disappear after releasing the solute from the vesicles triggered by the UV light. Such novel materials are of both basic and practical significance, especially as prospective nanocontainers for cargo delivery.

Abstract:A novel photo and thermo double-responsive block copolymer was developed to fabricate micelles and reverse micelles in aqueous solution. The block copolymer was synthesized by ATRP block copolymerization of a spiropyran-containing methacrylate (SPMA) with di(ethylene glycol) methyl ether methacrylate (DEGMMA). By facile control of the photo irradiation and solution temperature, PSPMA-core and PDEGMMA-core micelles can be obtained, respectively. The thermo- and photo-responsive micelles were used as smart polymeric nanocarriers for controlled encapsulation, triggered release, and re-encapsulation of model drug coumarin 102. Abstract:Two end-decorated homopolymers, poly(styrene)-beta-cyclodextrin (PS-beta-CD) and poly(ethylene oxide)-ferrocene (PEO-Fc), can orthogonally self-assemble into a supramolecular diblock copolymer (PS-beta-CD/PEO-Fc) in aqueous solutions based on the terminal host guest interactions. These assemblies can further form supramolecular vesicles, and their assembly and disassembly behaviors can be reversibly switched by voltage through the reversible association and disassociation of the middle supramolecular connection. The vesicles possess an unprecedented property that their assembly or disassembly speed can be controlled by the applied voltage strength. Luminescence spectroscopy demonstrates that the vesicles act as nanocapsules carrying molecules within their hollow cavities and that the external voltage strength accurately regulates the drug release time.
