Abstract:Metal-organic frameworks (MOFs), as drug delivery carriers, with high loading capacity and controllable release behavior can provide a more efficacious therapy in cancer treatments. In our work, a novel biocompatible zinc MOF Zn-cpon-1 with the (3,6)-connected rtl 3D topological network was designed and synthesized via employing ClO4- anion as template. The optically and chemically stable Zn-cpon-1 could be verified as a pH-responsive dual-emission platform and excellent drug delivery carrier with higher 5-fluorouracil (5-FU) (44.75 wt %) loading behavior than 6-mercaptopurine (6-MP) (4.79 wt %) ascribed to the influence of size and shape matching. The multiple interactions between Zn-cpon-1 and 5-FU drug molecules have been discussed and evidenced, which could be quantitatively estimated via the rate constant related to the topological structure. Specially, the gust release behavior of 5-FU@Zn-cpon-1 microcrystal was described and programmed via the Weibull distribution model and could be dual-triggered by the temperature and pH stimulus. This study illustrates that the Zn-cpon-1 without any postmodification performs a favorable potential of being used as biomedical drug delivery alternative carriers in effective drug payload, flexible release administration, and superior dual-stimuli responsiveness.
Controlled Zn2+-Triggered Drug Release by Preferred Coordination of Open Active Sites Within Functionalization Indium Metal Organic Frameworks.

Abstract:Drug delivery in target regions could make extraordinary progress in chemoselective therapies. A novel preferred coordination (PC) strategy referring to proactive interacting with open active sites to replace previous occupation by ion-exchange for controlling release of drug molecules is well-constructed. Two topological types of MOF-In1 (Schläfli symbol: (4,8)-connected of (410·615·83)(45·6)2) and MOF-In2 (Schläfli symbol: (4,4)-connected of (66)) show the specific way. Increasing node connectivity as well as the trapping of guest OH- anions, 5-fluorouracil (5-FU) is preferentially captured into the MOF-In1, which exhibits an outstanding loading capacity around 34.32 wt %. 19F NMR spectroscopy was further employed to investigate host-guest interaction and reveal the binding constant (Ka = 3.84 × 102 M-1). Meanwhile, the controlled release of 5-FU in a simulated human body with liquid phosphate-buffered saline solution by biofriendly Zn2+-triggered is realized. With an elevated Zn2+ concentration, the drug release will be enhanced. This efficient strategy for MOFs as multifunctional drug carrier opens a new avenue for biological and medical applications.

Abstract:The stimulus-responsive drug delivery system has attracted increasing attention due to its ability to enhance therapeutic efficacy and reduce side effects. Herein, a pH and glutathione (GSH) dually responsive drug carrier, hollow silica–-polyelectrolyte composite nanoparticle, was successfully prepared by using a template of spherical polyelectrolyte brush (SPB) which consists of a polystyrene (PS) core and a densely grafted linear poly(acrylic acid) (PAA) shell. The existence of PAA chains and introduction of disulfide bonds in silica framework endow the composite nanoparticles with pH and GSH dually responsive properties which were confirmed by dynamic light scattering (DLS) and small-angle X-ray scattering (SAXS). With doxorubicin hydrochloride (DOX) as the model drug, the loading content and encapsulation efficiency could reach up to 43% and 96%, respectively. The drug release behavior was investigated under various environments, showing that the drug release rate increased with the decrease in pH value and the increase in GSH concentration. The prepared hollow SiO2–PAA composite nanoparticles possess a great potential as carriers for controlled drug delivery.

Abstract:A novel nanocontainer, which has silica nanotube (SNT) core and pH-sensitive polymer shell attaching on the exterior surface of silica nanotube, is presented in this paper. Polymer nanorods, which are conveniently fabricated though polymerization-induced self-assembly and reorganization method, are used as templates for the deposition of silica to fabricate hybrid nanorods. Calcination of as-synthesized silica hybrid nanorods leads to hollow SNTs. SNTs are functionalized with reversible additionfragmentation chain transfer (RAFT) agent, then surface RAFT polymerization is conducted to get poly(2-(diethylamino)ethyl methacrylate)-b-poly(oligo(ethylene glycol) methacrylate)-coated SNTs (SNT-PDEAEMA-b-POEGMA). Doxorubicin (DOX) can be encapsulated in SNT-PDEAEMA-b-POEGMA, and controlled release of loaded DOX is achieved by adjusting pH of the medium. In vitro cell viability and cellular internalization study confirm the potential application of this nanocontainer in drug and gene delivery.

Abstract:The development of efficient multiresponsive drug delivery systems (DDSs) to control drug release has been widely explored. Herein, a facile strategy is reported that enables the micelles of the selenium‐containing polymer with the drug to be encapsulated in metal‐organic frameworks (MOFs), which serves as multiresponsive drug release by employing the selenium‐containing polymers with redox‐triggered property and the MOFs with pH‐triggered property in DDS. In this case, the micelles of selenium‐containing polymers, as core easily disassembles in the presence of redox agents, can then release the drug in MOFs matrixes. The ZIF‐8 (one type of MOFs) crystal frameworks serving as shell can collapse only under low pH conditions, and the drug can be further released. In the presence of external redox agents as well as the pH stimuli, the prepared nanocomposite (P@ZIF‐8) drug system exhibits the capability of multiresponsive release of the doxorubicin (DOX) and possesses good selectivity in releasing the DOX under low pH conditions instead of normal pH conditions. In addition, the merits of P@ZIF‐8 such as good biocompatibility, multiresponsive release properties, and especially the selective release properties under different pH conditions make the materials highly promising candidates for the realization of controlled drug delivery in tumor tissue systems.

Abstract:A cylindrical dosage form comprising a laminated composite polymer core and a hydrophobic polycarbonate coating was proposed for programmable drug delivery. In the core, poly[(ethyl glycinate) (benzyl amino acethydroxamate) phosphazene] was synthesized as drug-loaded layers for its strong pH-sensitive degradation (eroded after 1.5 days at pH 7.4 and more than 20 days at pH 5.0 and 6.0). Poly(sebacic anhydride)-b-polyethylene glycol or poly(sebacic anhydride-co-trimellitylimidoglycine)-b-poly(ethylene glycol) was selected as isolating layers for their good processing properties at room temperature and suitable erosion duration. The in vitro drug release studies of these devices were conducted under physiological conditions (pH 7.4). The results revealed that the model drugs (brilliant blue, FITC-dextran, myoglobin) could be released in typical pulsatile manner. Moreover, the duration time of drug release (24–40 h) and the lag time (18–118 h) could be separately regulated by the mass of polyphosphazene and the type or mass of polyanhydride. In this experiment, the cooperative effect of polyanhydrides and pH-sensitive degradable polyphosphazene was specially demonstrated, which offers a new idea to develop a programmable drug delivery system for single dose vaccine and other related applications.
Fluorine-containing pH-responsive core/shell microgel particles: preparation, characterization, and their applications in controlled drug release

Abstract:A kind of novel fluorine-containing pH-responsive core/shell microgels poly(DMAEMA- co -HFMA)- g -PEG were prepared via surfactant-free emulsion polymerization using water as the solvent. The well-defined chemical structure of the copolymers was characterized by FTIR, 1 H-NMR, 19 F-NMR, and elemental analysis. The microgel particles were studied by florescence probe technique, dynamic light scattering, and zeta potential measurement; the microgels displayed a significant pH-responsive behavior. Furthermore, the cytotoxicity assay indicated that the copolymer microgels had low toxicity, and 5-FU-loaded microgels offered a certain killing potency against cancer cells. In addition, the drug loading and in vitro drug release demonstrated that 5-FU was successfully incorporated into polymeric microgels, and the drug-loaded microgels showed a marked pH-dependent drug release behavior. This study suggests that the poly(DMAEMA- co -HFMA)- g -PEG microgels play an important role in the release mechanism stimulated by the change in the pH and have potential applications as a controlled drug release carrier.

Abstract:Multi-responsive drug carriers have great advantages in the targeted delivery of drugs in heterogeneous and complicated biological micro-environments. However, the sophisticated fabrication process is a great obstacle for their further application. Herein, ROS/GSH/pH triple responsive polycarbonate was rationally synthesizedviathe one-pot ring-opening copolymerization of the diselenide and tertiary amine macrocyclic carbonate monomers with methoxy poly(ethylene glycol) as the initiator. The micellar nanoparticles were formed by the amphiphilic block copolymers composed of a PEG segment and diselenide and tertiary amine containing the polycarbonate blockviaself-assembly. The diselenide and tertiary amine groups allowed for rich ROS/GSH/pH responsiveness, which caused these nanoparticles to undergo changes in the size and morphology. These micellar nanoparticles exhibited a stimuli-responsive drug release profile upon the stimulation of acidic pH, GSH, and ROS, and the release mechanism was elucidated. The blank nanoparticles were cyto-compatible, whereas the triple responsive drug-loaded nanoparticles exhibited an improved concentration-dependent and synergistic cytotoxicity towards the A549 cells. More importantly, the micellar nanoparticles could facilitate cell uptake and realize controlled intracellular release. Thus, the newly developed micellar nanocarriers could open a new avenue for smart antitumor drug delivery applications.

Abstract:The high storage capacities and excellent biocompatibilities of zinc(II) metal-organic frameworks (Zn-MOFs) have made them outstanding candidates as drug delivery carriers. Recent studies on the pH-responsive processes based on carrier-drug interactions have proven them to be the most efficient and effective way to control the release profiles of drugs. To satisfy the ever-growing demand in cancer therapy, great efforts are being devoted to the development of methods to precisely control drug release and achieve targeted use of an active substance at the right time and place. In this review article, we discuss the diverse stimuli based on Zn-MOFs carriers that have been achieved upon external activation from single pH-stimulus-responsive or/and multiple pH-stimuli-responsive viewpoints. Also, the perspectives and future challenges in this type of carrier system are discussed.
Controllable microfluidic fabrication of Janus and microcapsule particles for drug delivery applications

Abstract:Janus and microcapsule particles are very attractive for drug delivery applications due to their capability of targeted and/or programmed drug release. In this paper, a facile yet robust strategy is reported for the first time to fabricate both Janus and microcapsule particles in a simple microfluidic device via the same protocol. According to the classic spreading and partial wetting theory, the key point to fabricate Janus and microcapsule particles in single emulsions is that the two immiscible components should undergo a precisely controlled phase separation, i.e., dewetting for Janus particles and wetting for microcapsule particles. Herein we show that this requirement can be satisfied simply by a subtle choice of organic solvents for the dispersed phase. As a model hybrid material, PLGA/PCL Janus and microcapsule particles are obtained via switching the organic solvent between dimethyl carbonate and dichloromethane. ATR-FTIR spectra and acetone treatment demonstrate that PLGA and PCL occupy their own hemisphere in the Janus particles. In contrast, the shell on the microcapsule particle surface is composed of only PLGA, and the core is composed of PCL in which tiny PLGA beads are embedded. The applications of PLGA/PCL Janus and microcapsule particles in drug delivery are characterized via an in vitro degradation test. The results prove that Janus and microcapsule particles exhibit distinct degradation behaviors, implying their capability of programmable drug delivery in different manners.
Facile Synthesis of Ph Sensitive Polymer-Coated Mesoporous Silica Nanoparticles and Their Application in Drug Delivery

Abstract:pH-responsive polymer shell chitosan/poly (methacrylic acid) (CS-PMAA) was coated on mesoporous silica nanoparticles (MSN) through the facile in situ polymerization method. The resultant composite microspheres showed a flexible control over shell thickness, surface charges and hydrodynamic size by adjusting the feeding amount of MSN and the molar ratio of [–NH2]/MAA. The MSN/CS-PMAA composite microspheres were stable in the pH range of 5–8 as well as in the physiological saline (0.15M NaCl). Doxorubicin hydrochloride (DOX) was applied as a model drug to investigate the drug storage and release behavior. The results demonstrated that DOX could be effectively loaded into the composite microspheres. The cumulative release of DOX-loaded composite microspheres was pH dependent and the release rate was much faster at low pH (5.5) than that of pH 7.4. The cytotoxicity test by MTT assay showed that the blank carrier MSN/CS-PMAA microspheres were suitable as drug carriers. The cellular uptake of composite microspheres was investigated by confocal laser scanning microscopy (CLSM), which indicated that MSN/CS-PMAA could deliver the drugs into HeLa cell. The above results imply that the composite microspheres are a promising drug delivery system for cancer therapy.
Coordination polymer nanocapsules prepared using metal–organic framework templates for pH-responsive drug delivery

Abstract:A facile, efficient, and versatile approach is presented to synthesize pH-responsive nanocapsules (∼120 nm) by combining the advantages of metal-organic frameworks (MOFs) and metal-organic thin films. ZIF-8 nanoparticles are used as templates on which a thin film coating of iron(III)-catechol complexes is derived from the coordination between dopamine-modified alginate (AlgDA) and iron(III) ions. After the template removal, nanocapsules with a pH-responsive wall are obtained. Doxorubicin (Dox), a typical anticancer drug, is first immobilized in ZIF-8 frameworks through coprecipitation and then encapsulated in nanocapsules after the removal of ZIF-8. The structure of the iron(III)-catechol complex varies with pH value, thus conferring the Dox@Nanocapsules with tailored release behavior in vitro. Cytotoxicity tests illustrate the highly effective cytotoxicity of Dox@Nanocapsules towards cancer cells. This study provides a new method for preparing smart nanocapsules and offers more opportunities for the controlled delivery of drugs.

Abstract:Novel multi-stimuli-responsive microcapsules with adjustable controlled-release characteristics are reported by X.-J. Ju, L.-Y. Chu, and co-workers on page 3312. The microcapsules can achieve targeted delivery at specific sites by applying an external magnetic field, pH-responsive drug release at acidic environments and flexibly adjustable controlledrelease rate depending on temperature regulation, which provide a new mode for designing “intelligent” controlled-release systems and for realizing more rational drug administration.

Abstract:Herein, for rate-tunable controlled release, pH and redox dual responsive polymeric vesicles were constructed based on host-guest interaction between a water soluble pillar[5]arene (WP5) and a paraquat-containing block copolymer (BCP) in water. The yielding polymeric vesicles can be further applied in the controlled release of a hydrophilic model drug, doxorubicin hydrochloride (DOX). The drug release rate is regulated depending on the type of single stimulus or the combination of two stimuli. Meanwhile, DOX-loaded polymeric vesicles present anticancer activity in vitro comparable to free DOX under the studied conditions, which may be important for applications in the therapy of cancers as a controlled-release drug carrier.
Preparation of ternary hybridized chitosan microspheres with photothermal effect for pH-sensitive long-lasting controlled release system and its release mechanism

Abstract:To enhance the effective utilization of drugs and diminish the number of medications for patients, it is vital to investigate controlled long-acting drug release systems and their release mechanisms. In this study, chitosan microspheres were synthesized by cross-linking pH-sensitive Schiff bases, and Zif-8, a pH-sensitive component, was encapsulated within them. Furthermore, MXene was incorporated as a third component to augment the strength and photothermal synergistic characteristics of the microspheres. By adjusting the ratio of each component, the optimal ternary heterogeneous functional pH-controllable long-lasting drug release system was obtained. The optimal coating and the most effective adsorption-release effect were achieved. The release mechanism was examined under varying pH and temperature conditions. The release process was found to be divided into three stages. The initial phase was predominantly governed by the Fick diffusion mechanism, whereas the subsequent phases were primarily regulated by the pH-induced co-decomposition of CS and Zif-8, and the individual decomposition of CS, respectively. The drug was observed to be released in a linear manner during the latter two stages of the experiment. This was evidenced by the persistent release of ciprofloxacin hydrochloride in response to pH changes, and further supported by the results of tests on its antimicrobial properties. The drug release persisted for over three days at pH 5.4 and could be utilized repeatedly, exhibiting promising applications in drug delivery. Moreover, the drug's photothermal properties offer the possibility of developing synergistic therapeutic programs.

Abstract:Therapeutic platforms such as chemotherapy that respond to physical and biological stimuli are highly desirable for effective cancer therapy. In this study, pH-responsive charge-reversal, polymer-coated mesoporous silica nanoparticles [PAH-cit/APTES-MSNs; PAH-cit refers to poly(allylamine)-citraconic anhydride; APTES refers to (3-aminopropyl)triethoxysilane] were synthesized for application as drug-delivery systems for the treatment of malignant cells. Confocal laser scanning microscopy (CLSM) revealed that the PAH-cit/APTES-MSNs nanocomposite effectively delivered and released doxorubicin hydrochloride to the nucleus of HeLa (human cervical carcinoma) cells. Additionally, the real-time dynamic drug-release process was monitored by CLSM. The current pH-controlled-smart-release platform holds promise in drug-delivery and cancer therapy-related applications.

Abstract:A novel nanocontainer, which could regulate the release of payloads, has been successfully fabricated by attaching zwitterionic sulfobetaine copolymer onto the mesoporous silica nanoparticles (MSNs). RAFT polymerization is employed to prepare the hybrid poly(2-(dimethylamino)ethyl methacrylate)-coated MSNs (MSN-PDMAEMA). Subsequently, the tertiary amine groups in PDMAEMA are quaternized with 1,3-propanesultone to get poly(DMAEMA-co-3-dimethyl(methacryloyloxyethyl)ammonium propanesulfonate)-coated MSNs [MSN-Poly(DMAEMA-co-DMAPS)]. The zwitterionic PDMAPS component endows the nanocarrier with biocompatibility, and the PDMAEMA component makes the copolymer shell temperature-responsive. Controlled release of loaded rhodamine B has been achieved in the saline solutions.

In this work, a pH and H 2 O 2 -responsive drug delivery platform with high doxorubicin (DOX) loading capacity has been established through coordination interaction between DOX and phenylboronic acid containing block polymer. A composited drug nanocarrier is further fabricated by growing a zeolitic imidazolate framework 8 (ZIF-8) on the surface of drug-loaded polymer micelles. The study verifies that ZIF-8 shell can act as intelligent "switch" to prevent DOX leaking from core-shell nanoparticles upon H 2 O 2 stimulus. However, a burst drug release is detected upon pH and H 2 O 2 stimuli due to the further disassociation of ZIF-8 in acid solution. Moreover, the in vitro anti-cancer experiments demonstrate that the DOX-loaded core—shell nanoparticles provide effective treatment towards cancer cells but have negligible effect on normal cells, which results from the high concentration of H 2 O 2 and low pH in the microenvironment of tumor cells.

Ph-Responsive Hollow Polymeric Microspheres from Irradiated Cyclic Ether Aqueous Solution

Abstract:Smart hollow polymeric microspheres have been widely applied in various fields such as controlled release, drug delivery, catalysis, and so on. Herein, a facile, green and one-step template-free method is introduced for preparing pH-responsive hollow polymeric microspheres via gamma irradiation of cyclic ether aqueous solution. The hollow polymeric microspheres are synthesized by radiation-induced polymerization and following the self-assembly and self-organization of amphiphilic polymer with cyclic ethers as monomers in water. SEM, TEM, micro-FTIR, and NMR confirmed the morphology and structures of the resultant microspheres. The confocal laser scanning microscope was used to investigate the stimuli-responsiveness and release behavior of hollow microspheres using 1-pyrene carboxaldehyde as a hydrophobic molecule model. The well-defined hollow polymeric microspheres with an average diameter of ca. 2.6 μm or 1.6 μm were prepared directly from dicyclohexal-18-crown-6 or tetraphydropyrane aqueous solution, respectively. The prepared hollow microspheres exhibit obvious pH stimuli-responsiveness and can release the encapsulated hydrophobic molecules when pH is higher than 5.0. Moreover, the reversible morphology transition between hollow microspheres and micelles makes the prepared hollow polymeric microspheres potentially suitable for a wide range of applications, including removal of dyes, oil field engineering, and biomedical fields.
