Abstract:Abstract Three new sesquiterpenes, namely arvestolides A–C ( 1 – 3 ), were isolated from the aerial parts of Artemisia vestita . Their planar structures were determined by extensive analysis of spectroscopic data including MS, 1D, and 2D NMR, and their absolute configurations were resolved by single-crystal X-ray diffraction. Compound 1 is a sesquiterpene with a rare 6/8/3 skeleton, while compounds 2 and 3 possess a rare 5/8/3 sesquiterpene skeleton. Compound 2 exhibited a moderate inhibition effect on the lipopolysaccharide (LPS)-induced nitric oxide (NO) production in BV-2 microglial cells with an IC 50 value of 38.59 μM.

Abstract:Eleven previously undescribed sesquiterpenoids, arvestolides D-J, arvestonates A-C, and arvestonol were isolated from the aerial parts of Artemisia vestita W. Their structures were elucidated by extensive analysis of HRMS, UV, IR, and NMR spectroscopic data, and the absolute configurations were determined by single crystal X-ray diffraction, empirical rules, and comparison of calculated and experimental ECD data. Arvestolides H and I showed inhibitory effects on nitric oxide production in BV-2 cells induced by lipopolysaccharide with IC50 values of 43.2 and 39.9 mu M, respectively. (C) 2018 Elsevier Ltd. Abstract:Five undescribed eudesmane methyl esters ( 1 − 5 ), three undescribed eudesmane-12,6-olides ( 6 − 8 ), and 21 known analogues ( 9 − 29 ) were isolated from the aerial part of Artemisia princeps Pamp. Their structures were established by detailed analysis of the NMR and HRESIMS data. The absolute configurations of 1 − 8 were determined based on single-crystal X-ray diffraction analysis and ECD calculations. Moreover, the inhibitory effects on LPS-induced NO production in BV-2 microglial cells of all the isolated compounds were assessed. Except for compounds 2 , 4 , 10 , and 11 , the others showed significant inhibitory activities, with IC 50 values of 0.73–18.66 μM, wherein the potential structure-activity relationship was also discussed.

Abstract:A chemical investigation of the aerial parts of Artemisia vestita Wall. led to the isolation of 12 known sesquiterpenes, including2 furan-containing sesquiterpenoids and 10 eudesmane sesquiterpene lactones. Their structures were identified as negunfurol(1),schensianol A(2), artemine(3), erivanin(4), 1,5-diepi-artemin(5), acetylartemin(6), naphtho[1,2-b]furan-2(3H)-one, 6-(acetyloxy)decahydro-9a-hydroxy-3,5a-dimethyl-9-methylene-(3S,3aS,5aS,6S,9aS,9bS)(7), naphtho[1,2-b]furan-2(3H)-one, 6-(acetyloxy)-3a,4,5,5a,6,7,8,9b-octahydro-8-hydroxy-3,5a,9-trimethyl-(3S,3aS,5aR,6S,8S,9bS)(8), isoerivanin(9), barrelierin(10),(11S)-1-oxoeudesm-4(14)-eno-13,6α-lactone(11), 1-epi-dehydroisoeranin(12), respectively. All of these compounds were isolated from Artemisia vestita for the first time, and compounds 1 and 2 were isolated from the genus Artemisia for the first time.
Sesquiterpenoids and Triterpenoids with Anti-Inflammatory Effects from Artemisia Vulgaris L

Abstract:In this study, eight undescribed sesquiterpenoids (artemvulactone A-G and artemvulemdiol A), and two undescribed triterpenoids, (3S)-dammar-20,25-diene-3-hydroxy-24-one and (3S,23E)-dammar-20,23-diene-25- methoxy-3-ol were isolated from the leaves of Artemisia vulgaris L., together with ten known sesquiterpenoids and three known triterpenoids. The structures of these undescribed terpenoids were determined by extensive spectroscopy methods, including 1D and 2D-NMR, HRESIMS, IR, UV, X-ray diffraction, and ECD. The absolute configurations of artemvulactone A, artemvulactone D, and artemvulactone E were determined by X-ray diffraction (Cu Kα). All isolates were evaluated for their anti-inflammatory efficacy by detecting the expression of inflammatory mediator NO in LPS-induced RAW264.7 cells, and the results indicated that artemvulactone E exhibited significant anti-inflammatory effect with an IC50 value of 0.9 ± 0.2 μM. Furthermore, artemvulactone E could reduce LPS-induced COX-2 protein expression dose-dependently by western blotting experiments.

Abstract:Two new disesquiterpenoids (1 and 2) and 11 new (3-13) and 10 known (14-23) sesquiterpenoids were isolated from the whole plants of Artemisia freyniana. Their structures were elucidated by spectroscopic data analysis and comparison with published NMR data. The absolute configurations of the new isolates (1-13) were assigned based on single-crystal X-ray diffraction data and comparison of the experimental and calculated ECD data. The eremophilane derivatives 8 and 9 possess an unprecedented 2-isopropyl-3,7,7a-trimethyl-2,4,5,6,7,7a-hexahydro-1 H-indene scaffold, and a putative biosynthetic pathway for these compounds is proposed. Compounds 4, 5, and 9 exhibited inhibitory effects against LPS-stimulated nitric oxide (NO) production in RAW 264.7 macrophage cells with IC50 values of 10.8, 12.6, and 11.7 μM, respectively.

Abstract:Two new eudesmane sesquiterpenoids artanoate (1) and eudesmanomolide (2) were isolated from the aerial parts of Artemisia anomala S. Moore. Their structures were elucidated as methyl (4R, 5S, 6S, 7S, 10R)-1-oxo-4, 6-dihydroxy-eudesma-2, 11 (13)-dien-12-oate (1) and (1R, 5R, 6R, 10R)-3, 13-diacetoxy-1-hydroxy-3, 7(11)-diene-12, 6-olide (2) on the basis of extensive spectroscopic analyses. Compound 1 showed cytotoxicity against HCT-8 cell lines with IC50 value of 9.13μM, and compound 2 exhibited inhibitory activities against HCT-8 and A549 cell lines with IC50 values of 3.76 and 5.49μM, respectively.

Abstract:The present study is to investigate the chemical constituents from the aerial parts of Artemisia myriantha. The chemical constituents were isolated by column chromatographies over silica gel, Sephadex LH-20, ODS and Semi-prep HPLC, and the structures were identified by NMR and MS data. Thirteen known compounds were isolated and identified as: blumenol A (1),(+)-dehydrovomifoliol (2),(+)-3-hydroxy-β-ionone (3),(3R, 6R, 7E)-3-hydroxy-4, 7-megastigmadien-9-one (4),(-)-10-oxo-isodauc-3-en-15-oic acid (5),isoerivanin (6),eudesmafraglaucolide (7), artanomalide A (8),13-acetoxy-3β-hydroxy-germacra-1(10) E,4E,7(11)-trien-12,6α-olide (9),13-acetoxy-3β-tigloyl-germacra-1(10) E, 4E, 7(11)-trien-12, 6α-olide (10),13-acetoxy-3β-(3-methylbutanoyl)-germacra-1(10)E, 4E, 7(11)-trien-12, 6α-olide (11),3,9-diacetoxy-13-hydroxy-1(10), 4, 7(11)-germacratrien-12,6α-olide (12), and 8α-angeloyloxycostunolide (13). Compounds 1-6 and 13 were obtained from the genus Artemisia for the first time, and 7-9 and 12 were isolated from this plant for the first time. Compound 8 exhibited selective cytotoxicity against human colon cancer (HCT-8) and human gastric cancer (BGC-823) with IC₅₀ values of 2.33 and 4.53 μmol•L ⁻¹, respectively.

Abstract:One new highly oxygenated nortriterpene, named sieverlactone (1), one new sesquiterpene, 1 beta, 10 beta-epoxy-8 alpha-acetoxyachillin (2), one new natural product, 5-propinyl-thiophene-2-carboxylic acid (3), and one new thiophene, 3-hydroxy-5-propinyl-2-acetyl-thiophene (4), together with 10 other known compounds (5 - 14), were isolated from the dried aerial parts of Artemisia sieversiana. Their structures were elucidated by a combination of extensive spectroscopic analysis, including 1D, 2D NMR spectroscopic and mass spectrometric data. Meanwhile, the stereochemistry of 1 and 2 was confirmed by single-crystal X-ray diffraction technique using Cu radiation. All the isolates were evaluated for their anti-neuroinflammatory effects on the lipopolysaccharide-induced nitric oxide production in BV-2 murine microglial cells. Compounds 2, 5, and 6 exhibited the significant activities with IC50 values of 6.5 +/- 0.5, 11.9 +/- 0.7, and 10.1 +/- 0.3 mu M, respectively, comparable to the positive control, quercetin, with an IC50 value of 16.3 +/- 0.4 mu M. Abstract:Two new ditetrahydrofuran lignans, named sieverlignans A and B (1 and 2), together with six known ones (3-8), were isolated from the aerial parts of Artemisia sieversiana. Their structures were established on the basis of spectroscopic analysis including HRMS, NMR spectra and circular dichroism experiments. All the compounds were evaluated for their anti-neuroinflammatory effects on the lipopolysaccharides (LPS)-induced nitric oxide production in BV-2 murine microglial cells. Compound 2 exhibited the significant activity with its IC50 value of 11.9 +/- 0.8 mu M, respectively, compared to a positive control quercetin with its IC50 value of 16.0 +/- 1.1 mu M.

Abstract:Eleven previously undescribed sesquiterpenoids (8-18), one undescribed jasmonic acid derivative (35) and 28 known compounds were isolated from the leaves of Artemisia stolonifera. Undescribed compounds with their absolute configurations were determined by extensive spectroscopic analysis, single-crystal X-ray diffraction and ECD calculation. Compound 8 was identified as a rare sesquiterpenoid featuring a rearranged 5/8 bicyclic ring system, whereas compound 17 was found to be an unprecedented monocyclic sesquiterpenoid with methyl rearrangement. Evaluation of biological activity showed that compounds 1-5 and 7 displayed cytotoxicity against six tumor cells. In the meantime, compounds 11, 12, 18 and 35 exhibited inhibitory effects against LPS-stimulated NO production in RAW 264.7 macrophage cells and reduced the transcription of IL-6 and IL-1β in a dose-dependent manner at 25, 50 and 100 μM. Moreover, the anti-inflammatory-based network pharmacology and molecular docking analyses revealed potential target proteins of 11, 12, 18 and 35.

Abstract:A new sesquiterpene, rupestrisin A (1), and three new thiophene derivatives, rupestrienes A–C (2–4), were isolated from Artemisia rupestris. Their structures were determined by analyses of MS and NMR spectroscopic data, and the absolute configuration of 1 was established by calculated ECD spectra using time-dependent density functional theory. In in vitro bioassays, compounds 1–4 showed inhibitory effects on LPS-stimulated NO production in BV-2 microglial cells with IC50 values of 24.3, 20.3, 8.5, and 5.3 μM, respectively.

Abstract:Twelve undescribed and 13 known eudesmane-type sesquiterpenoids were obtained from Artemisia leucophylla , and structurally elucidated based on comprehensive analyses of spectral data, including HRESIMS, IR, 1D and 2D NMR, and ECD calculation. The absolute configuration of compound 1 was determined by a single X-ray single crystal diffraction. Chemically, compounds 1–5 featured unprecedented 1,2- seco -1- nor- eudesmane-type skeleton with a cis -fused 6/5 bicyclic system. Antihepatoma evaluation against three human hepatoma cell lines (HepG2, Huh7, and SK-Hep-1) for all compounds demonstrated that compound 7 displayed the most active cytotoxicity with IC 50 values of 35.1, 35.0, and 32.7 μ Μ.

Abstract:Chemical fractionation of the n-BuOH partition, which was generated from the EtOH extract of the flower buds of Tussilago farfara, afforded a series of polar constituents including four new sesquiterpenoids (1-4), one new sesquiterpenoid glucoside (5) and one known analogue (6) of the eudesmane type, as well as five known quinic acid derivatives (7-11). Structures of the new compounds were unambiguously characterized by detailed spectroscopic analyses, with their absolute configurations being established by X-ray crystallography, electronic circular dichroism (ECD) calculation and induced ECD experiments. The inhibitory effect of all the isolates against LPS-induced NO production in murine RAW264.7 macrophages was evaluated, with isochlorogenic acid A (7) showing significant inhibitory activity.

Structure elucidation of the new compounds was unambiguously performed based on HRESIMS, 1D and 2D NMR spectroscopic analyses. The two new compounds exhibited in vitro strong cytotoxic activities against human cancer cell lines HCT-8 with IC50 values of 0.76 and 0.28 μM, respectively.

Abstract:Random screening suggested that the EtOH extract of Artemisia myriantha (Asteraceae) and its EtOAc fraction had cytotoxicity against HepG2 cells with inhibitory ratios of 30.6% and 53.5% at 50.0 μg/mL. Bioassay-guided isolation of the most active fractions (Fr. C and Fr. D) afforded 19 new sesquiterpenolides, artemyrianolides A–S (1–19), involving 13 germacranolides (1–13), four guaianolides (14–17), and two eudesmanolides (18 and 19), together with 16 known sesquiterpenoids (20–35). The new compounds were characterized by physical data analyses (HRESIMS, IR, 1D and 2D NMR, ECD), and the absolute configurations of compounds 1, 2, and 11 were determined by X-ray crystallography. Structurally, compounds 2 and 11–13 maintain an uncommon cis-fused 10/5 bicyclic system and compound 12 possesses an unusual (7S) configuration. Twenty of the compounds exhibited cytotoxicity against HepG2, Huh7, and SMMC-7721 cell lines.
