Abstract:Eleven previously undescribed sesquiterpenoids, arvestolides D-J, arvestonates A-C, and arvestonol were isolated from the aerial parts of Artemisia vestita W. Their structures were elucidated by extensive analysis of HRMS, UV, IR, and NMR spectroscopic data, and the absolute configurations were determined by single crystal X-ray diffraction, empirical rules, and comparison of calculated and experimental ECD data. Arvestolides H and I showed inhibitory effects on nitric oxide production in BV-2 cells induced by lipopolysaccharide with IC50 values of 43.2 and 39.9 mu M, respectively. (C) 2018 Elsevier Ltd. Abstract:Two new polyene polyketides, namely aurovertins T and U (1 and 2), were isolated from Calcarisporium arbuscula, together with aurovertins B (3), D and E (4 and 5), and M (6). The structures were elucidated by extensive spectroscopic methods (especially 2D‐NMR techniques). The cytotoxic activities of all isolates against human triple‐negative breast cancer cell line (MDA‐MB‐231) were evaluated. As a result, compounds 3, 4, and 6 exhibited more potent cytotoxic activities against MDA‐MB‐231 cell line than the positive control taxol. Also, discussion about the relationships between structure and activity of these aurovertins was presented.

Abstract:In this study, eight undescribed sesquiterpenoids (artemvulactone A-G and artemvulemdiol A), and two undescribed triterpenoids, (3S)-dammar-20,25-diene-3-hydroxy-24-one and (3S,23E)-dammar-20,23-diene-25- methoxy-3-ol were isolated from the leaves of Artemisia vulgaris L., together with ten known sesquiterpenoids and three known triterpenoids. The structures of these undescribed terpenoids were determined by extensive spectroscopy methods, including 1D and 2D-NMR, HRESIMS, IR, UV, X-ray diffraction, and ECD. The absolute configurations of artemvulactone A, artemvulactone D, and artemvulactone E were determined by X-ray diffraction (Cu Kα). All isolates were evaluated for their anti-inflammatory efficacy by detecting the expression of inflammatory mediator NO in LPS-induced RAW264.7 cells, and the results indicated that artemvulactone E exhibited significant anti-inflammatory effect with an IC50 value of 0.9 ± 0.2 μM. Furthermore, artemvulactone E could reduce LPS-induced COX-2 protein expression dose-dependently by western blotting experiments.

Abstract:The present study is to investigate the chemical constituents from the aerial parts of Artemisia myriantha. The chemical constituents were isolated by column chromatographies over silica gel, Sephadex LH-20, ODS and Semi-prep HPLC, and the structures were identified by NMR and MS data. Thirteen known compounds were isolated and identified as: blumenol A (1),(+)-dehydrovomifoliol (2),(+)-3-hydroxy-β-ionone (3),(3R, 6R, 7E)-3-hydroxy-4, 7-megastigmadien-9-one (4),(-)-10-oxo-isodauc-3-en-15-oic acid (5),isoerivanin (6),eudesmafraglaucolide (7), artanomalide A (8),13-acetoxy-3β-hydroxy-germacra-1(10) E,4E,7(11)-trien-12,6α-olide (9),13-acetoxy-3β-tigloyl-germacra-1(10) E, 4E, 7(11)-trien-12, 6α-olide (10),13-acetoxy-3β-(3-methylbutanoyl)-germacra-1(10)E, 4E, 7(11)-trien-12, 6α-olide (11),3,9-diacetoxy-13-hydroxy-1(10), 4, 7(11)-germacratrien-12,6α-olide (12), and 8α-angeloyloxycostunolide (13). Compounds 1-6 and 13 were obtained from the genus Artemisia for the first time, and 7-9 and 12 were isolated from this plant for the first time. Compound 8 exhibited selective cytotoxicity against human colon cancer (HCT-8) and human gastric cancer (BGC-823) with IC₅₀ values of 2.33 and 4.53 μmol•L ⁻¹, respectively.

Abstract:Two new ditetrahydrofuran lignans, named sieverlignans A and B (1 and 2), together with six known ones (3-8), were isolated from the aerial parts of Artemisia sieversiana. Their structures were established on the basis of spectroscopic analysis including HRMS, NMR spectra and circular dichroism experiments. All the compounds were evaluated for their anti-neuroinflammatory effects on the lipopolysaccharides (LPS)-induced nitric oxide production in BV-2 murine microglial cells. Compound 2 exhibited the significant activity with its IC50 value of 11.9 +/- 0.8 mu M, respectively, compared to a positive control quercetin with its IC50 value of 16.0 +/- 1.1 mu M.

Their structures were elucidated by a combination of extensive spectroscopic analysis, including 1D, 2D NMR spectroscopic and mass spectrometric data. Meanwhile, the stereochemistry of 1 and 2 was confirmed by single-crystal X-ray diffraction technique using Cu radiation. All the isolates were evaluated for their anti-neuroinflammatory effects on the lipopolysaccharide-induced nitric oxide production in BV-2 murine microglial cells. Compounds 2, 5, and 6 exhibited the significant activities with IC50 values of 6.5 +/- 0.5, 11.9 +/- 0.7, and 10.1 +/- 0.3 mu M, respectively, comparable to the positive control, quercetin, with an IC50 value of 16.3 +/- 0.4 mu M. Abstract:Aim of the study: Artemisia vestita is a common traditional Tibetan medicinal plant which has been used widely in China for treating various inflammatory diseases. Since little is known about its active components, the purpose of this study was to isolate and identify the immunosuppressive compounds from Artemisia vestita.Materials and methods: A bioassay-guided isolation was performed with picryl chloride-induced contact hypersensitivity in mice. MTT assay and Flow cytometric analysis were used for determining Con A-induced lymphocyte proliferation and CD25 expression in T cells, respectively.Results: The ethanol extract of the Artemisia vestita was found to possess significant inhibitory activity against the picryl chloride-induced contact hypersensitivity in mice. Then 4 fractions were isolated by macroporous adsorption resin and one of these fractions (AV3), which showed the highest activity in in vivo test, was further subjected to column chromatography. Nine known flavones were isolated and identified as pectolinarigenin (1), jaceosidin (2), cirsilineol (3), cirsimaritin (4), hispidulin (5), quercetin (6), 6-methoxytricin (7), acacetin (8), and apigenin (9). The structures of the 9 flavones were elucidated by spectral techniques. All the compounds were evaluated for their inhibitory activity on the proliferation and activation of T cells in vitro. Among the 9 flavones, cirsilineol (3), 6-methoxytricin (7) and apigenin (9) significantly inhibited T cell proliferation and activation in the bioassays.Conclusion: The result suggests that cirsilineol, 6-methoxytricin and apigenin are the major active components in Artemisia vestita. Abstract:A chemical investigation of the aerial parts of Artemisia vestita Wall. led to the isolation of 12 known sesquiterpenes, including2 furan-containing sesquiterpenoids and 10 eudesmane sesquiterpene lactones. Their structures were identified as negunfurol(1),schensianol A(2), artemine(3), erivanin(4), 1,5-diepi-artemin(5), acetylartemin(6), naphtho[1,2-b]furan-2(3H)-one, 6-(acetyloxy)decahydro-9a-hydroxy-3,5a-dimethyl-9-methylene-(3S,3aS,5aS,6S,9aS,9bS)(7), naphtho[1,2-b]furan-2(3H)-one, 6-(acetyloxy)-3a,4,5,5a,6,7,8,9b-octahydro-8-hydroxy-3,5a,9-trimethyl-(3S,3aS,5aR,6S,8S,9bS)(8), isoerivanin(9), barrelierin(10),(11S)-1-oxoeudesm-4(14)-eno-13,6α-lactone(11), 1-epi-dehydroisoeranin(12), respectively. All of these compounds were isolated from Artemisia vestita for the first time, and compounds 1 and 2 were isolated from the genus Artemisia for the first time.

Abstract:Six new anthracyclines (1-6) were obtained by the investigation of the solid fermented extract of a marine-derived Streptomyces sp. NB-A13. The structures were elucidated on the basis of spectroscopic analysis, including 1D and 2D NMR, HRESIMS, UV and IR. The absolute configurations of compounds 1-5 were established by calculated electronic circular dichroism (ECD) using time dependent density functional theory. Among the isolates, four compounds (2, 3, 5 and 6) showed moderate cytotoxic activity against PC3 cells (prostate cancer cells) with IC50 values of 7.0 +/- 0.3 mu M, 0.92 +/- 0.05 mu M, 2.1 +/- 0.7 mu M and 9.8 +/- 0.7 mu M, respectively.

Their structures were determined by analyses of MS and NMR spectroscopic data, and the absolute configuration of 1 was established by calculated ECD spectra using time-dependent density functional theory. Abstract:Random screening suggested that the EtOH extract of Artemisia myriantha (Asteraceae) and its EtOAc fraction had cytotoxicity against HepG2 cells with inhibitory ratios of 30.6% and 53.5% at 50.0 μg/mL. Bioassay-guided isolation of the most active fractions (Fr. C and Fr. D) afforded 19 new sesquiterpenolides, artemyrianolides A–S (1–19), involving 13 germacranolides (1–13), four guaianolides (14–17), and two eudesmanolides (18 and 19), together with 16 known sesquiterpenoids (20–35). The new compounds were characterized by physical data analyses (HRESIMS, IR, 1D and 2D NMR, ECD), and the absolute configurations of compounds 1, 2, and 11 were determined by X-ray crystallography. Structurally, compounds 2 and 11–13 maintain an uncommon cis-fused 10/5 bicyclic system and compound 12 possesses an unusual (7S) configuration. Twenty of the compounds exhibited cytotoxicity against HepG2, Huh7, and SMMC-7721 cell lines. Abstract:Thirteen new sesquiterpenoids, arteannoides F-R (1–13), along with 13 known analogues (14–26), were isolated from the dried aerial parts of Artemisia annua L. Their structures, including absolute configurations, were unambiguously determined by a combination of physical data analyses (HRESIMS, 1D and 2D NMR, and ECD) as well as the crystal structures of 1, 5, 6, 15, 19, and 23. Among the isolated compounds, 1 features an unusual 11-oxatricyclo[6.2.1.04,9]undecan-2-ene ring system, 5 possesses an uncommon 4,11-ether bridged tricyclic framework, whereas 6 is a new eudesmane-type sesquiterpenoid formed via rearrangement of its carbon backbone. The systemically anti-inflammatory activities of all isolates were evaluated by measuring their inhibitory effects on PGE2, NO, TNF-α, and IL-6 production in LPS-stimulated RAW 264.7 macrophages. Moreover, the structure activity relationships of some compounds are summarized, this study will provide new structural templates for discovering potential anti-inflammatory agents.

Abstract:Arteannoide A (1), an unusual cadinane dimer featuring a rare fused 6,8-dioxabicyclo[3.2.l]octan-7-one ring system, arteannoides B and C (2 and 3), two novel heterodimers incorporating a rearranged cadinene sesquiterpenoid and a phenylpropanoid, together with two new rearranged cadinene sesquiterpenoids 4 and 5, were isolated from Artemisia annua L. Their structures were determined by a combination of NMR spectroscopy, electronic circular dichroism calculations, and X-ray diffraction analyses. Compounds 2 and 3 showed inhibition of nitric oxide production in lipopolysaccharide-induced RAW 264.7 mouse macrophage cell lines with IC50 values of 4.5 and 2.9 μM, respectively.

Abstract:Introduction: Natural plants are valuable resources for exploring new bioactive compounds. Artemisia vulgaris L. is a traditional Chinese medicinal herb that has been historically used for treating multiple diseases. Active compounds isolated and extracted from A. vulgaris L. typically possess immunomodulatory and anti-inflammatory properties. Artemvulactone E (AE) is a new sesquiterpene lactone isolated and extracted from A. vulgaris L. with unclear biological activities. Methods: The immunoregulatory effects of AE on macrophages were assessed by ELISA, RT-qPCR, immunofluorescence, and western blot assay. The effect of AE on lipopolysaccharide (LPS) -relates signaling pathways was examined by western blot assay. In zebrafish models, the larvae were yolk-microinjected with LPS to establish inflammation model and the effect of AE was evaluated by determining the survival rate, heart rate, yolk sac edema size, neutrophils and macrophages infiltration of zebrafish. The interaction between AE and Toll-like receptor 4 (TLR4) was examined by molecular docking and dynamic stimulation. Results: AE reduced the expression and secretion of pro-inflammatory cytokines (TNF-α and IL-6), inflammatory mediators iNOS and COX-2, as well as decreases the production of intracellular NO and ROS in LPS-stimulated macrophages. In addition, AE exerted its anti-inflammatory effect synergistically by inhibiting MAPK/JAK/STAT3-NF-κB signaling pathways. Furthermore, AE enhanced the survival rate and attenuated inflammatory response in zebrafish embryos treated with LPS. Finally, the molecular dynamics results indicate that AE forms stable complexes with LPS receptor TLR4 through the Ser127 residue, thus completely impairing the subsequent activation of MAPK-NF-κB signaling. Conclusion: AE exhibits notable anti-inflammatory activity and represents as a potential agent for treating inflammation-associated diseases.

Abstract:In addition to beta-sitosterol and alpha-amyrin detected in all the investigated species, the extract of the aerial parts of Artemisia giraldii var. giraldii gave stigmasterol, daucosterol, sesamine, luteolin, eupafolin, hispidulin, eupatilin, belamcanidin, pinitol, artemin, ridentin, and a new antifungal monoterpene (named santolinylol) while that of the aerial parts of A. mongolica afforded sesamine, eupafolin, eupatilin, matricarin, and a new germacranolide (3-oxo-11 alpha H-germacra-1(10)E,4Z-dien-12,6 alpha-olide), and that of the aerial parts of A. vestita yielded stigmasterol, daucosterol, umbelliferone, scopolin, scoparone, and isoscopoletin-O-glucoside. Pinitol, first reisolated from Artemisia genus, was shown to inhibit the growth of the human pathogenic fungi Candida albicans, Aspergillus flavus, A. niger, Geotrichun candidum, Trichophyton rubrum, and Epidermophyton floccosum. Umbelliferone was also active against Candida tropicalis, A. flavus, G. candidum, T. rubrum, and E. floccosum. The flavones hispidulin and belamcanidin were almost equally inhibitory to the growth of A. flavus, G. candidum, T. rubrum, and E. floccosum, and santolinylol to C. albicans, A. flavus, A. niger, G. candidum, T. rubrum, and E. floccosum. In addition, ridentin was active against the growth of the plant pathogenic fungus Cladosporium cucumerinum.
