Induction of Human Umbilical Cord Blood-Derived CD34~(+) Cells into Endothelial Cells in Vitro

Abstract:Objective To evaluate the feasibility of isolating, purifying, and expanding of umbilical cord blood-derived CD34~ + cells in vitro and the way to induce them into endothelial cells(ECs). Methods Mononuclear cells were isolated from human umbilical cord blood by density gradient centrifugation. CD34~ + mononuclear cells were selected by magnetic beads coated with antibody CD34. Cells were cultured in M-199 medium containing vascular endothelium growth factors and basic fibroblast growth factors. Morphology of the cells was observed and cell surface marker CD31 was assessed by fluorescence activated cell sorter(FACS) analysis.Expressions of ecNOS and flk-1/KDR proteins were detected by immunocytochemistry. Results After culturing CD34~ + cells for 3 days, multiple cell clusters appeared, and 7 days later, both isolated and linear cord-like spindle-shaped cells were observed. On the contrary, only few cultured CD34~ - cells could attach to the plate sporadically. In FACS assay, 70.03%±0.27% of cultured CD34~ + cells was positive-stained for CD31.In immunocytochemisty assay, most cultured CD34~ + cells expressed ecNOS and flk-1/KDR. Conclusion CD34~ + cells, which were isolated by magnetic bead separation method, could exhibit EC-like morphology such as spindle shape and cord-like structure during culturing. Most cells also expressed the EC lineage marks such as ecNOS, flk-1/KDR and CD31. Therefore, the method of isolating and inducing CD34~ + cells into EC in vitro is feasible.
