Abstract:The main function region of the fusion (F) protein gene of 124 strains of Newcastle disease virus isolated from 2002 to 2004 in China was amplified and sequenced for further phylogenetic and residue substitutive analysis. Most of the isolates were classified into genotype VIIc, VIId, VIf, and VIb, while others into genotype IX, III, or II. The genotype IX, a unique genotype which includes strain F48, the first Chinese virulent NDV strain isolated in 1948, were still found inducing sporadic infections in certain areas. Subgenotype VIIc, VIId, and VIIe viruses, which were distributed in clusters in the phylogenetic tree distinct from members of subgenotypes VIIa and VIIb, were responsible for most outbreaks in China and circulated predominantly in China in recent years. Strain NDV03-026, an isolate of the genotype II which was normally lentogenic, was found carrying (112)RRQKRF(117) motif at the cleavage site of F protein as the virulent strain.
Genetic Diversity of the Genotype VII Newcastle Disease Virus: Identification of a Novel VIIj Sub-Genotype

Abstract:Newcastle disease (ND) is a highly contagious disease of poultry caused by Newcastle disease virus (NDV). Multiple genotypes of NDV have been circulating worldwide and NDV is continuously evolving, resulting into more diversity. Of multiple viral genotypes, VII is particularly important given that it had been associated with most recent ND outbreaks worldwide. In this study, an epidemiological investigation performed in northeastern China during 2014–2015 showed that 11 genotype VII isolates amounted to 55 percent in a total number of NDV isolates. Therefore, to evaluate the genetic diversity worldwide and epidemiological distribution in China of genotype VII NDV, a phylogenetic analysis based on the 1255 complete F gene sequences showed that VII is the most predominant genotype worldwide. A further detailed characterization on genotype VII was conducted based on the 477 complete F gene sequences from 11 isolates and 466 reference viruses available in GenBank. The results demonstrated that VII can be further divided into 8 sub-genotypes (VIIb, VIId–VIIj), indicating its complex genetic diversity. It is worthy of note that the isolation rate of VIIj is increasing recently. It emphasizes the necessity to pay close attention to the epidemiological dynamic of genotype VII NDV and highlights the importance of vaccination program.
Isolation and Molecular Identification of Two Virulent Newcastle Disease Virus Strains from Chicken Flocks

Abstract:Two strains of Newcastle disease virus(NDV) were isolated from infected chicken flocks in Shanghai.They were determined as virulent strains whose MDT was 51.2 h and 50.4 h,and ICPI was 1.850 and 1.875,respectively.The cDNA fragments of F gene of the 2 NDV strains were amplified by RT-PCR and then cloned into pGEM-T easy vector.The sequence analysis showed that the F gene fragments from the 2 NDV isolates consisted of l 654 bp,coding for 551 amino acids.The amino acid sequence homology between the 2 NDV isolates came to 100%,but that between the 2 isolates and common NDV vaccine strains(La Sota,B1,F48E9 and V4) was only 78.3%~83.7%.The sequence of cleavage site of F protein was 112R-R-Q-K-R-F117,belonging to the genotype Ⅶ of NDV.

Abstract:One virus isolate was obtained from a chicken flock with high morbidity and mortality.The isolate was confirmed as Newcastle disease virus.The biological tests proved it as a virulent strain,but the HA titers were as high as 28～10.The HN gene of the isolate was amplified by the Reverse Transcription-Polymerase Chain Reaction(RT-PCR)technique using a pair of specific primers.Then the whole HN gene was sequenced and the amino acid sequence was obtained from it.

Abstract:Ten Newcastle disease virus(NDV) strains were isolated from chickens in the field outbreaks in some regions of Hebei province during 2004-2008.Assessment of the virulence by MDT,ICPI and IVPI,RT-PCR and sequence analysis of fusion protein gene were used to compare the properties of NDV isolates.The results indicated that MDT,ICPI and IVPI of the isolates were 37.6-54.4 h,1.71-2.0 and 2.16-2.8,respectively,indicating that the all NDV isolates were highly virulent.The results of nucleotide sequencing and phylogentic analysis of fusion protein gene showed that the ten strains shared homology from 77.4% to 98.0% among themselves,from 87.0% to 98.9% and from 89.7% to 98.9% with NDV Lasota,F48E8,respectively.The putative amino acid sequences of fusion protein at the cleavage sites of eight isolates were 112 R-R-Q-K-R-F117,with the motif characteristics of the virulent NDV strain;two isolates were 112 G-R-Q-G-R-L117,with the motif characteristics of the Lentongenic NDV strain.Phylogenetic analysis showed that the NDV epidemic belonged to 3 genotypes(II,Ⅶand Ⅸ) and the genotype VII(70%)was the major pathogeny causing the epizootic of ND in Hebei.

Abstract:Utilizing RT-PCR,the cDNA fragment of NDV V4 strain was successfully amplified,which was about 4. 2 kb in length,and was cloned and sequenced. The cDNA fragment covers the open reading frame (ORF),the intergenic regions in NP and P gene coding region junction and the whole nucleotide sequence of Phosphoprotein (P) gene. According to the comparative analysis between the nucleotide and amino sequence of NP and P gene in NDV V4 and the counterpart in 8 other NDV strains,the homology was highest with Queenland V4. Comparison of nucleotide sequence was carried out between intergenic region in NP and P gene coding region junction of NDV V4 strain and the counterpart of other 13 NDV strains respectively. The results showed that the mesogenic or velogenic strains,such as F48E9 and ZJ1, had the extra 6 nucleosides insertion between 1 647 nt and 1 648 nt of lentogenic strains,such as La Sota and D26, indicating the homology was different (63. 9%-100%),and presented the homology between lentogenic strains was higher than that between velogenic strains,and than that between lentogenic strains and velogenic strains.

Abstract:This study was conducted to evaluate the virulence and evolution of genotype IX Newcastle disease virus (NDV) isolates obtained from wild birds in the northern Qinling Mountains of China. Five isolates were obtained from 374 larynx and cloacae swabs, which were collected from multiple asymptomatic wild bird species from August 2008 to July 2011, and were subsequently characterized by pathotype and genotype. Deduced amino acid sequences revealed that all five NDV isolates exhibited velogenic fusion protein cleavage sites motif 112 R-R-Q-R-R-F 117 , shared as high as 99.8–99.9 % homology with each other, and varied in pathotype by intracerebral pathogenicity indices (ICPI) of 0.425–1.638. Phylogenetic analysis showed that all five isolates were clustered to genotype IX NDV. This is the first study to confirm multiple asymptomatic wild bird species as natural carriers of virulent genotype IX NDV. A novel NDV isolate from the Spotted-necked Dove (family Columbidae) exhibited discordance between its lentogenic ICPI and its virulent proteolytic cleavage site motif 112 R-R-Q-R-R-F 117 . Although the five isolates underwent several amino acid mutations in the fusion protein, evidence of continuous evolutionary divergence did exist in the genotype IX NDV, which was always regarded as a conservative genotype.
Pathotypical Characterization and Molecular Epidemiology of Newcastle Disease Virus Isolates from Different Hosts in China from 1996 to 2005

Abstract:Thirty Newcastle disease virus (NDV) strains isolated from outbreaks in China during 1996 to 2005 were characterized pathotypically and genotypically. All strains except one were velogenic. An analysis of the variable region (nucleotides 47 to 420) of the F gene indicated that 6 isolates belonged to genotype II, 3 to genotype III, 1 (isolated from a pigeon) to genotype VI, and 20 to genotype VII. Isolates belonging to genotype VII were further divided into five subtypes, VIIa, VIIb, VIIc, VIId, and VIIe, and subtype VIId was made up of VIId1 to VIId5. These results showed that genotype VII isolates might have been the most prevalent in China during the past two decades. Genotype VII isolates shared high homology, but the homology was less than that between genotype VII viruses and the vaccine virus LaSota. Among these NDV isolates, 25 isolates had the velogenic motif (112)R/K-R-Q-K/R-R-F(117) that is consistent with results of the biological tests. However, four of five LaSota-type isolates that contained the lentogenic motif (112)G-R-Q-G-R-L(117) were velogenic, except SY/03, in the view of the biological test. The majority of genotype VII isolates had lost one or two N-glycosylation sites. Finally, a cross-protection experiment in which specific-pathogen-free chickens vaccinated with LaSota were challenged by six NDV isolates showed that more than three isolates were antigenic variants that could be responsible for recent outbreaks of Newcastle disease.

Abstract:Twenty four field strains of newcastle disease viruses(NDV) isolated from different geographical locations in China in the last 10 years were characterized genetically.A pair of primers specific to NDV gene F 3′ end was designed according to the known sequences from GenBank,and was used to amplify the NDV cDNA gene F 3′ end by RT-PCR from total RNA,which was 450 bp in length.Analysis of the F gene sequence revealed that 20 strains belonged to the genetype VII,whose amino acid sequences of the cleavage site region(112-117) were similar to that of all other virulent strains.The results confirmed that the majority of epidemic strains of NDV in China recently are virulent.

Abstract:Newcastle disease virus (NDV) V protein is crucial for viral interferon (IFN) antagonism and virulence, determining its host range restriction. However, little information is available on the B cell epitopes of V protein and the subcellular movement of V protein in the process of NDV infection. In this study, the monoclonal antibody (mAb) clone 3D7 against genotype VII NDV V protein was generated by immunizing mice with a purified recombinant His-tagged carboxyl-terminal domain (CTD) region of V protein. Fine epitope mapping analysis and B-cell epitope prediction indicated that mAb 3D7 recognized a linear epitope 152RGPAELWK159, which is located in the V protein CTD region. Sequence alignment showed that the mAb clone 3D7-recognized epitope is highly conserved among Class II genotype VII NDV strains, but not among other genotypes, suggesting it could serve as a genetic marker to differentiate NDV genotypes. Furthermore, the movement of V protein during NDV replication in infected cells were determined by using this mAb. It was found that V protein localized around the nucleus during virus replication. The establishment of V protein-specific mAb and identification of its epitope extend our understanding of the antigenic characteristics of V protein and provide a basis for the development of epitope-based diagnostic assays.

Abstract:Twenty Newcastle disease virus (NDV) strains were isolated from chickens and geese in the field outbreaks during 2005 and 2006 in some regions of Jiangsu and Guangxi province. Assessment of the virulence by MDT and ICPI, RT-PCR and sequence analysis of fusion protein gene were used to compare the properties of NDV isolates. The results indicated that MDT and ICPI of the isolates were 45.3h - 58.2h and 1.61 - 2.00 respectively, which confirmed that the all NDV isolates were highly virulent. And their hemagglutinin were not resistant to heat and belonged to fast pattern of elution. The results of nucleotide sequencing and phylogentic analysis of fusion protein gene showed that the twenty strains shared homology from 79.7% to 100% among themselves, from 78.1% to 83.4% and from 80.2% to 90.1% with NDV LaSota, F48E8, respectively. The putative amino acid sequences of fusion protein at the cleavage sites of all the isolates were 112R-R-Q-R/K-R-F117, with the motif characteristics of the virulent NDV strain, which was in accordant with the results of assessment of the pathogenicity. The phylogentic tree based on sequences of fusion protein gene variable regions (47-420nt) revealed that the 18 strains belonged to sub-genotype VIId and the others belonged to an old genotype III of NDV, revealing that subgenotype VIId virus was responsible for the NDV outbreaks in some regions of Jiangsu and Guangxi promince recently.
Development of Real-time Fluorescent Quantitative RT-PCR Assay for Detection of Genotype VII Newcastle Disease Virus (NDV)

Abstract:In order to establish a differential diagnosis Real-time RT-PCR with highly sensitivity,specificity and repeatability,a pair of primers and TaqMan probe were designed according to characteristics of cleavage site sequences of F gene of genotype Ⅶ Newcastle disease virus strains in this study.The standard curve was established by using cRNA standard prepared in vitro transcription as positive template,the related indexes were tested.The results showed that this assay has a good effect of differential detection of genotype Ⅶ NDV;102 copies·μL-1 of template RNA could be detected at least.The sensitive of this assay was 100 times higher than conventional RT-PCR.The real-time fluorescent quantitative RT-PCR for detection of genotype Ⅶ Newcastle disease virus was established.This assay could be used as differential diagnosis method of genotype Ⅶ NDV strains,which couldn’t be identified by conventional methods.Our results indicated that the method established in this study can be used as rapid identification assay for the detection of genotype Ⅶ NDVs.

Abstract:Despite the frequent use of ND vaccines in Egypt, Newcastle Disease Virus still causes outbreaks in the commercial, backyard, and wild captive poultry. Avian Orthoavula virus 1 (AOaV-1) belongs to the genus Orthoavulavirus and causes ND in a variety of avian species around the world. To explore the evolution of the NDV and evaluate the efficacy of the vaccine regimens currently being used in Egypt's commercial poultry, spleen, brain, ileocecal tonsil, and tracheal tissue specimens were collected from twenty-three vaccinated broiler flocks from four Egyptian governorates (Kafr-Elsheikh, Alexandria, Matrouh, and El-Behera) between 2017 and 2020 for molecular analysis. qRT-PCR was used to characterize the NDV that was propagated in specific pathogen-free embryonated chicken eggs (SPF ECEs). The isolates were screened for other endemic avian respiratory viruses including avian influenza and infectious bronchitis viruses. Five isolates were selected for further genetic characterization, based on the motif of the cleavage site (112RRQKR ↓F117) in the fusion (F) protein. All five isolates were identified as velogenic NDV strains based on partial-length F gene sequences. These isolates are clustered into subgenotype VII.1.1 of the class II of ND viruses, which is the main genotype in the Egyptian poultry industry and in those of many Middle East countries.
Identification of Newcastle Disease Virus(ndv)isolates by RT-PCR and Its Clinical Application

Abstract:RT-PCR assays have been conducted to differentiate NDV isolates by using 3 pairs of primers,which were designed according to the gene sequence at the cleavage site of F protein.PA+PB primers directed at all NDV strains;PA+PC,PA+PD primers were specific to virulent and non-virulent strains respectively.The results showed a fragment of 359 bp was amplified by PA+PB primers from 15 strains of NDV isolates.The RT-PCR by using PA+PC primers resulted in a fragment of 254 bp from F48E8,ND98,Fw,H2,P3,BY,L2,P1,P2 strain and no band was found by using PA+PD primers.Meanwhile ND89,Lasota strains produced a fragment of 254 bp by using PA+PD primers and manifested negative reaction by using PA+PC primers.The brain,spleen,pharyngeal and cloacal swabs were sounpled from naturally infected cockfights and detected with RT-PCR assays by using the 3 pairs of primers.The sick cockfights were definitely diagnosed as virulent NDV infection.It is proved to be in accordance with the traditional methods of virus isolation and virulence identification.
Establishment of Reverse Genetics for Genotype VII Newcastle Disease Virus and Altering the Cell Tropism by Inserting TMPRSS2 into the Viral Genome

Abstract:Newcastle disease (ND) is the most important infectious disease in poultry, which is caused by avian orthoavulavirus type 1 (AOAV-1), previously known as Newcastle disease virus (NDV). In this study, an NDV strain SD19 (GenBank accession number OP797800) was isolated, and phylogenetic analysis suggested the virus belongs to the class II genotype VII. After generating wild-type rescued SD19 (rSD19), the attenuating strain (raSD19) was generated by mutating the F protein cleavage site. To explore the potential role of the transmembrane protease, serine S1 member 2 (TMPRSS2), the TMPRSS2 gene was inserted into the region between the P and M genes of raSD19 to generate raSD19-TMPRSS2. Besides, the coding sequence of the enhanced green fluorescent protein (EGFP) gene was inserted in the same region as a control (rSD19-EGFP and raSD19-EGFP). The Western blot, indirect immunofluorescence assay (IFA), and real-time quantitative PCR were employed to determine the replication activity of these constructs. The results reveal that all the rescued viruses can replicate in chicken embryo fibroblast (DF-1) cells; however, the proliferation of raSD19 and raSD19-EGFP needs additional trypsin. We next evaluated the virulence of these constructs, and our results reveal that the SD19, rSD19, and rSD19-EGFP are velogenic; the raSD19 and raSD19-EGFP are lentogenic; and the raSD19-TMPRSS2 are mesogenic. Moreover, due to the enzymatic hydrolysis of serine protease, the raSD19-TMPRSS2 can support itself to proliferate in the DF-1 cells without adding exogenous trypsin. These results may provide a new method for the NDV cell culture and contribute to ND’s vaccine development.
Molecular Characteristics of Newcastle Disease Virus Isolates from Guizhou Province

Abstract:The fusion protein gene(F gene) of eight isolates of Newcastle disease virus(NDV),which is isolated from different avian in the different years in Guizhou province,were amplified by polymerase chain reaction(PCR) technique,then were cloned respectively into the pMD18-T vector and sequenced,and these F gene were analyzed with reference NDV stains in GenBank.The results showed that F gene of all isolates were 1 662 bp and encoding 553 amino acids,and the sequence identity matrix indicated that the homology of nucleotide sequence of these isolates were 84.0%～99.7% and that of amino acid sequence were 87.5%～99.3%,and were respectively 84.2%～98.9%and 87.2%～98.6% compared with that of Lasota,B1,F48E9,CH2000 and TW2000.The phylogenetic analysis showed that strain DQ,N98,P1 and H2 belonged to Ⅶ genotype,strain BY and FW to Ⅸ genotype,and strain GZ and TN to Ⅱ genotype,and revealed that the same genotype of NDV could induce outbreaks in different avian and the genotype of NDV had transformed in different years,and Ⅶ genotype is the main genotype of NDV occurred in Guizhou province in recent years.

Abstract:Newcastle disease (ND) is one of the most important diseases of poultry, and causes severe economic losses in the global poultry industry. Although all Newcastle disease virus (NDV) isolates belong to a single serotype, significant genetic diversity has been described between different NDV isolates. Here, we report the molecular characterization of 23 virulent genotype VIId NDV isolates of class II circulating in China. Phylogenetic construction and analysis revealed the existence of distinctly genomic and amino acid differences that clearly distinguished these isolates from other typical NDV genotypes and vaccine strains. We also report a new 582-amino-acid hemagglutinin-neuraminidase in genotype VII NDV strains. This is believed to be the first study to investigate systematically the most predominant NDV strains, and provides more information on the genetic nature of genotype VIId NDV of class II circulating in China.

Abstract:The NP and P genes of a Newcastle disease virus(NDV) isolate NDV09-056,obtained from domestic duck in China in 2009,were amplified by RT-PCR and sequenced.The complete NP gene of NDV09-056 isolate was composed of 1746 nucleotides encoding 489 amino acids.Homology analysis of NP gene of the isolate revealed that the homology of nucleotides among class Ⅰ NDV isolates were between 93.5%～98.5%,and showed relative lower homology between this isolate and the class Ⅱ NDV strains,77.0%～79.2%.The full length of P gene of the isolate was composed of 1463 nucleotides,encoding 399 amino acids.Homology analysis of the P gene of the isolate revealed that the homology of nucleotides among class Ⅰ NDV isolates were between 90.4%～95.3%,and showed relatively lower homology between the isolate and the class Ⅱ NDV strains,68.3%～72.5%.Phylogenetic analysis based on the complete NP gene and P gene showed that the NDV09-056 isolate was grouped into genotype 3 in class I.The evolutionary rates of NP gene and P gene of class Ⅰ was similar based on the homology analysis and phylogenetic analysis.

Abstract:A strain NDV named DQ was isolated successfully in a case of ND.The entire function fragment of F gene of the isolate DQ was amplified by RT-PCR,and then cloned into pMD18-T vector.The sequence analysis showed that the nucleotide sequences of the fragment were 1 662 bp,coding for 553 amino acid.The isolate(DQ) is closer to two strains(CH2000,TW2000) in genetic relationship,neucleitide and amino acid sequence homologies were found to be 95.7%-96.6% and 96.6%-96.8% respectively,but the neucleitide and the amino acid sequence homologies to other NDV strains such as LaSota、B1 and F48E9 were only from 84.0%-86.5% and 87.2%-89.9% respectively.The deduced amino acid sequence of the cleavage site of F protein of the NDV isolates was112R-R-Q-K-R-F117 matching to the virulent NDV strains,the isolate possess K and V,which was similar to the characteristic of genotype Ⅶ,the NDV systemic evolution tree was depicted by MegAlign software and showed that the isolate was gene type Ⅶd.

Abstract:A pair of primers were designed.Utilizing RT-PCR,4 overlapping cDNA fragments of NDV V4 strain were successfully amplified using above NDV specific primers.The 4 PCR fragments length is about 4.2,4.1,4.1,3.5 kb respectively,which were cloned and sequenced.The nucleotide sequence of 15 061 bp in length gained by spliced the nucleotide sequence of the 4 fragments, which covers the part of nucleocappsid protein(NP), the whole nucleotide sequence of Phosphoprotein(P),Matrix protein(M),Fusion protein(F),Haemagglutinin-Neuraminidase(HN),Large protein(L),the intergenic regions in NP-P,P-M,M-F,F-HN,HN-L junctions,and the 108 bp of 5′ trailer region of NDV V4 strain.But the gained NP nucleotide sequence have spanned its open reading frame(ORF) and 3′ non-coding region.According to the length of complete genome sequence known at present,we found there are at least two types of NDV strains:15 186 bp and 15 192 bp.Equaled to the 15 186 bp strain,the NDV V4 strain isn′t the extra 6 nucleosides insertion between 1 647 nt and 1 652 nt as compared with the 15 192 bp strain.The amino acid sequence of F protein cleavage site is 112GKQGRL117,and the amino acid sequence of HN protein is 616 in length,which conformed to the trait of lentogenic strain.
