Abstract:Through amplification and sequence analysis of F genes segments of 3 pigeon NDV strains to discuss the genetic relationship among the three NDV strains and the vaccine strains.Collect samples,inoculat samples into chicken embryos to isolate virus.They were identified NDV strains by HA and HI;a pair of primers were designed and synthetized,in which were used to amplify the important fragment of F gene(1395 bp) by RT-PCR.Nucleotide sequences and deduced amino acids sequences analysis showed : homologous rate of F gene nucleotides sequences was 98.0%～99.3%,amino acid sequences was 98.5%～99.1%.The results showed that the deduced amino acid sequence at the cleavage site of the F protein was 112 R-R-Q-K-R-F117,which is the same as virulent strains of NDV.The phylogenetic tree showed that there was a close genetic relationship between the 3 isolates and strain isolated from Guizhou which all belonged to the genotype Ⅵ.The results indicated that the 3 strains were highly homologous with each other,and pathological changes and nucleotide sequences and deduced amino acids sequences analysis showed the 3 strains were virulent NDV strains which were lower homologies with traditional La Sota and B1.
