Abstract:Amino acid and primary amine/amide co-catalyzed acetone condensation was investigated. It was found that l-proline had overwhelming catalytic activity over other amino acids as well as the analogues with similar structures. Surprisingly, thiourea, a very cheap and stable chemical, was found to be the favorable co-catalyst. Co-catalyzed by the recyclable l-proline and thiourea, condensation of acetone led to the useful products mesityl oxide (MO), diacetone alcohol (DAA) and isophorone (IP) in the excellent 96.3% total selectivity.

Abstract:l-Proline amides were evaluated for catalyzing the direct aldol reaction of 4-nitrobenzaldehyde with chloroacetone. The presence of 30mol% (S)-pyrrolidine-2-carboxylic acid (2,4,6-trimethyl-phenyl)-amide catalyzed the direct aldol reactions of a range of aldehydes with chloroacetone to give anti-α-chloro-β-hydroxyketones with high regio-, diastereo- and enantioselectivity.

Abstract:Zinc-proline complex catalyzed Knoevenagel condensation in water was reported. Seventeen aldehydes and two ketones were investigated, which coupled with malononitrile smoothly affording desired Knoevenagel condensation products in 5 similar to 10 min at 80 degrees C in the yields of 84%similar to 99%. The catalyst can be recovered and reused for ten cycles without loss of activity.

Abstract:l-Prolinamides 2, prepared froml-proline and simple aliphatic and aromatic amines, have been found to be active catalysts for the direct aldol reaction of 4-nitrobenzaldehyde with neat acetone at room temperature. They give moderate enantioselectivities of up to 46% enantiomeric excess (ee). The enantioselectivity increases as the amide N—H becomes a better hydrogen bond donor.l-Prolinamides 3, derived from the reaction ofl-proline with α,β-hydroxyamines such that there is a terminal hydroxyl group, show more efficient catalysis and higher enantioselectivities. In particular, catalyst 3h, prepared froml-proline and (1S,2S)-diphenyl-2-aminoethanol, exhibits high enantioselectivities of up to 93% ee for aromatic aldehydes and up to >99% ee for aliphatic aldehydes under –25°C. Model reactions of benzaldehyde with three enamines derived from the condensation of prolinamides with acetone have been studied by quantum mechanics calculations. The calculations reveal that the amide N—H and the terminal hydroxyl groups form hydrogen bonds with the benzaldehyde substrate. These hydrogen bonds reduce the activation energy and cause high enantioselectivity. Our results suggest a new strategy in the design of new organic catalysts for direct asymmetric aldol reactions and related transformations.

Abstract:A series of poly(2-oxazoline) (POX) derivatives bearing prolinamide pendants were designed as organocatalysts and evaluated in the direct asymmetric aldol reaction between aromatic aldehydes and cyclic ketones. The structural variation of the alkyl spacer connecting the polymer backbone with the catalytic unit was applied so as to deduce structure–performance relationships combined with comparable experiments from model catalysts. Results showed that the POX-bound prolinamides can promote the aldol reaction more effectively as compared to their small-molecular and non-POX-bound analogs. The catalyst P3 containing the pyrrolidine moiety closer to the tertiary amide backbone exhibited the overall best catalytic efficiency, affording anti-products in 84% yield with 89% ee in the representative aldol addition of cyclohexanone to 4-nitrobenzaldehyde at a 10 mol.% catalyst loading. Furthermore, the influence of trifluoroacetic acid as an additive on the asymmetric transformation was investigated. Theoretical calculations revealed that the protonation of the aldehyde carbonyl group switched the activation mode of the aldol acceptor through hydrogen bond interactions, thereby changing the relative energy barrier of the enamine/aldehyde reaction transition states, which accounted well for the significant improvement in the enantioselectivity of the acidic additives observed experimentally.

Abstract:L-proline amides derived from various chiral beta-amino alcohols that bear substituents with various electron natures at their stereogenic centers are prepared and evaluated for catalyzing the direct Aldol reaction of 4-nitrobenzaldehyde with acetone. Catalysts with strong electron-withdrawing groups are found to exhibit higher catalytic activity and enantioselectivity than their analogues with electron-donating groups. The presence of 2 mol % catalyst 4g significantly catalyzes the direct Aldol reactions of a wide range of aldehydes with acetone and butanone, to give the beta-hydroxy ketones with very high enantioselectivities ranging from 96% to >99% ee. High diastereoselectivity of 95/5 was observed for the anti Aldol product from the reaction of cyclohexanone, and excellent enantioselectivity of 93% ee was provided for anti Aldol product from the reaction of cyclopentanone.

Abstract:A novel proline derivative, (4R)-4-(beta-Naphthalenyl)methoxy-(S)-proline, was conveniently prepared from the naturally occurring (4S)-hydroxy-(S)-proline; 5 mol % of this compound efficiently catalyzes the asymmetric aldol reactions of various benzaldehydes with acetone in excess of acetone as the solvent, giving the aldol adducts in good yields with ee up to 89.8%.

Abstract:Direct asymmetric aldol addition of methyl ketones to 2,2,2-trifluoro-1-phenylethanone and its ring-substituted derivatives was achieved using L-proline as a chiral promoter. Various optically active beta-trifluoromethyl-beta-hydroxy ketones were obtained in almost quantitative yields with moderate enantioselectivities up to 64 % ee.

Abstract:A photoredox-catalyzed sequential decarboxylative/defluorinative aminoalkylation of CF3-alkenes with N-arylglycines is described. This metal-free and redox-neutral protocol provided efficient access to the monofluoroalkenyl-1,5-diamines in good yields with excellent functional group compatibility. Mechanistic studies revealed that the reaction proceeds via a radical pathway with the gem-difluoroalkenyl amine as an intermediate.

Abstract:A HFIP-promoted highly selective hydroxyalkylation of aniline derivatives with arylglyoxal hydrates has been realized. The reaction produces various N,N-dialkylanilines and their derivatives with α-hydroxy carbonyl units in good to excellent yields under mild conditions. Furthermore, the synthetic potential of this method has been demonstrated by the facile synthesis of several structurally interesting molecules such as benzil, 1,2,4-triazine, quinoxaline, hydantoin, and 2-thiohydantoin with aromatic amine units.

Abstract:Asymmetric functionalization of inert 0-C-H is one of the most powerful strategies in organic synthesis, but of great challenge in the efficient 0-C-H activation and the enantioselective C-heteroatom formation. Here, this work proposes an elegant strategy for one-pot asymmetric 0-C-H functionalization of aldehyde under O2 catalyzed by hydroxide-layered Fe(III) sites synergistic with confined interlayer amine through a cascade oxidation-addition process on L-proline intercalated Fe-containing layered double hydroxides (LDHs), in which the direct 0-H elimination of aldehyde on hydroxide-layered Fe(III) sites with the two -electron abstraction promoted by the Fe(III):N coordination as the key step to efficiently oxidize the enamine. The major 0-functionalized diastereomer in the asymmetric nitromethylation of 3-phenylpropanal has been afforded in a regioselectivity of up to >99%, a stereoselectivity of 90%, and a yield of 65% on L-proline intercalated Mg3.00Al0.47Fe0.45-LDHs.(c) 2022 Elsevier Inc. Abstract:A novel hyperbranched polyethylene (HBPE)-supported L-proline has been developed via a bottom-up copolymerization strategy. Copolymerization of ethylene and a protected proline acrylate comonomer with cationic Pd-diimine catalysts was conducted, followed by de-protection of the proline groups. Well-defined HBPE copolymers having molecular weights (MWs) of 10.3-50.3 kDa and 3.2-15.6 L-proline molecules per HBPE polymer chain were synthesized. The effects of the L-proline amount and HBPE MW on the catalyst performance were studied. The HBPE catalysts were efficient in asymmetric aldol reactions of p-nitrobenzaldehyde (p-NBA) or benzaldehyde derivatives with cyclohexanone. High p-NBA conversions of up to 98% and excellent product selectivities with anti/syn = 98/2 and ee > 99% were achieved. Moreover, the HBPE catalysts could be easily recovered by adding water into the product. The recovered catalysts could be reused multiple times with only a slight decline in reactivity and selectivity.

Abstract:Direct asymmetric aldol additions of acetone/and butanone to alpha-ketoesters were achieved using L-proline as a chiral catalyst. Various optically active alpha-hydroxyesters were obtained in the yields of 43 % - 93 % with enantioselectivities up to 81 % e.e. The steric effect seems to be an important factor which influences the activity and the selectivity of the reaction. Acetone showed higher reactivity than butanone while the latter provided better enantioselectivity in some cases.

Abstract:Aldol condensation reactions are very important C–C coupling reactions in organic chemistry. In this study, the catalytic performance of layered double hydroxides (LDHs) in the aldol condensation reaction of formaldehyde (FA) and propionaldehyde (PA) was investigated. The MxAl-LDHs (denoted as re-MxAl–LDHs; M = Ca and Mg; X = 2–4), as heterogeneous basic catalysts toward the aldol condensation reaction, were prepared via a two-step procedure. The catalyst exhibited a high PA conversion (82.59%), but the methacrolein (MAL) selectivity was only 36.01% due to the limitation of the alkali-catalyzed mechanism. On this basis, the direct intercalation of L-proline into LDHs also has been investigated. The influences of several operating conditions, including the temperature, reaction time, and substrate content, on the reaction results were systematically studied, and the optimized reaction conditions were obtained. The optimized Mg3Al–Pro-LDHs catalyst exhibited a much higher MAL selectivity than those of re-MgxAl–LDHs.
