Abstract:The use of nitroarenes as amino sources in synthesis is challenging. Herein is reported an unusual, straightforward, and transition metal-free method for the net [3 + 2]-cycloaddition reaction of 2-azaallyl anions with nitroarenes. The products of this reaction are diverse 2,5-dihydro-1,2,4-oxadiazoles (>40 examples, up to 95% yield). This method does not require an external reductant to reduce nitroarenes, nor does it employ nitrosoarenes, which are often used in N–O cycloadditions. Instead, it is proposed that the 2-azaallyl anions, which behave as super electron donors (SEDs), deliver an electron to the nitroarene to generate a nitroarene radical anion. A downstream 2-azaallyl radical coupling with a newly formed nitrosoarene is followed by ring closure to afford the observed products. This proposed reaction pathway is supported by computational studies and experimental evidence. Overall, this method uses readily available materials, is green, and exhibits a broad scope.

Abstract:A HFIP-promoted highly selective hydroxyalkylation of aniline derivatives with arylglyoxal hydrates has been realized. The reaction produces various N,N-dialkylanilines and their derivatives with α-hydroxy carbonyl units in good to excellent yields under mild conditions. Furthermore, the synthetic potential of this method has been demonstrated by the facile synthesis of several structurally interesting molecules such as benzil, 1,2,4-triazine, quinoxaline, hydantoin, and 2-thiohydantoin with aromatic amine units.

Abstract:The chemoselective catalytic reduction of nitro scaffolds to value-added amine functions is desirable but challenging. We developed a combined KOtBu/BEt3 catalyst for deoxygenative reduction of nitroarenes, nitro heteroarenes and even notoriously challenging nitroalkanes using pinacolborane under mild reaction conditions. The novel transition-metal-free strategy is readily applicable to the preparation of commercially available pharmaceuticals.

Abstract:We present an efficient approach for chemoselective synthesis of various functionalized arylamines from nitroarenes over an oxygen-implanted MoS2 catalyst (O-MoS2). The HRTEM, XRD, XPS, Raman, EXAFS and NH3-TPD characterizations show the existence of (MoOx)-O-Iv structure and abundant coordinative unsaturated (CUS) Mo sites in the 2D-layer structure of O-MoS2. In the transfer hydrogenation of nitroarenes with hydrazine hydrate, the (MoOx)-O-Iv structure works as the catalytic active center. The N2H4 selectively decomposes to the active hydrogen species in polar electronic states (H delta- and He delta+), which show high chemoselectivity toward the nitro reduction over C=C, C=C, and C N groups. The O-MoS2 catalyst can be recovered in a facile manner from the reaction mixture and recycled four times without any significant loss of activity. (C) 2016 Elsevier B.V. All rights reserved.

Abstract:The highly enantioselective totally N-selective hydroxyamination reaction of -aryl--cyanoacetates with 2-nitrosopyridines was realized by using a chiral N,N-dioxide/Mg(OTf)(2) complex as catalyst, which enriches the nitroso chemistry. A variety of 2-cyano-2-[hydroxyl(pyrydin-2-yl)amino]acetates with quaternary stereocenters and potential antibacterial activities were obtained in excellent yields with good to excellent ee values under as low as 0.05mol% catalyst loading. The products could be easily transformed to useful -amino amides and 1,2-diamines. Besides, a possible transition state model was proposed to elucidate the origin of the chirality induction.
Mechanistic study of NHC-catalyzed oxidative N vs O chemoselective acylation of aldehydes with alkanolamines

Abstract:The possible catalytic mechanism was proposed and studied comprehensively at the M062X/6-31G(d,p) level of theory for a reported chemoselective intermolecular S N 2 nucleophilic substitution of aldehydes with alkanoamine. The calculated results show that the catalytic cycle occurs through six stages including the nucleophilic attack of active N -heterocyclic carbene (NHC) on the substrate, generation of the Breslow intermediate, oxidative addition of NBS and alkanolamine, S N 2 nucleophilic substitution, Et 3 N-assisted debrominationand finally regeneration of the NHC along with the release of the product. The results show that TSs of the C−N or C−O bond formation is the key transition state that determines the chemoselectivity of the title reaction. Moreover, our results demonstrated that the N -bromoamine intermediate is more favorable both thermodynamically and kinetically than hypobromite by 14.2 kcal/mol, which is consistent with the fact that only N -acylation product was observed in the experiment. In addition, we found that alkanolamine not only acts as a reactant but also as a proton-shuttle and stabilizer to stabilize the structure of chemoselectivity favored transition state by multiple N-H···N, N-H···O and C-H···Br noncovalent interactions. Et 3 N firstly acts as a base to promote the formation of active catalyst NHC, then the protonated Et 3 N as proton-shuttle to accelerate Breslow intermediate formation and decreasing the activation barrier. The Parr functions and frontier molecular orbital analyses of key species indicate that NHC enhances remarkably the nucleophilicity of substrate aldehyde, leading to a significant increase of the HOMO energy and a moderate increase of the LUMO energy. We hope this work could provide deeper insight into the fundamental mechanisms of the Et 3 N assisted NHC-catalyzed S N 2 substitution reactions.

Abstract:Chemoselective control over N/O selectivity is an intriguing issue in nitroso chemistry. Recently, we reported an unprecedented asymmetric α-amination reaction of β-ketocarbonyl compounds that proceeded through the catalytic coupling of enamine carbonyl groups with in-situ-generated carbonyl nitroso moieties. This process was facilitated by a simple chiral primary and tertiary diamine that was derived from tert-leucine. This reaction featured high chemoselectivity and excellent enantioselectivity for a broad range of substrates. Herein, a computational study was performed to elucidate the origins of the enantioselectivity and N/O regioselectivity. We found that a bidentate hydrogen-bonding interaction between the tertiary N(+)-H and nitrosocarbonyl groups accounted for the high N selectivity, whilst the enantioselectivity was determined by Si-facial attack on the (E)- and (Z)-enamines in a Curtin-Hammett-type manner. The bidentate hydrogen-bonding interaction with the nitrosocarbonyl moieties reinforced the facial selectivity in this process.

Abstract:Nickel-catalyzed aminoxylation of an unactivated C(sp3)–H bond with a stable nitroxyl radical has been accomplished for the first time to offer various N-alkoxyamine derivatives, which further enables a one-pot approach to α-formyl acid derivatives. The aminoxylation process reported also provides direct evidence for the oxidative addition of a cyclometallic intermediate with a free radical, which is helpful for the reaction-mechanism study in transition-metal-catalyzed functionalization of inert C(sp3)–H bonds.

Abstract:AbstractCompared to the α‐functionalization of aldehydes, ketones, even esters, the direct α‐modification of amides is still a challenge because of the low acidity of α‐CH groups. The α‐functionalization of N−H (primary and secondary) amides, containing both an unactived α‐C−H bond and a competitively active N−H bond, remains elusive. Shown herein is the general and efficient oxidative α‐oxyamination and hydroxylation of aliphatic amides including secondary N−H amides. This transition‐metal‐free chemistry with high chemoselectivity provides an efficient approach to α‐hydroxy amides. This oxidative protocol significantly enables the selective functionalization of inert α‐C−H bonds with the complete preservation of active N−H bond.

Abstract:Asymmetric functionalization of inert 0-C-H is one of the most powerful strategies in organic synthesis, but of great challenge in the efficient 0-C-H activation and the enantioselective C-heteroatom formation. Here, this work proposes an elegant strategy for one-pot asymmetric 0-C-H functionalization of aldehyde under O2 catalyzed by hydroxide-layered Fe(III) sites synergistic with confined interlayer amine through a cascade oxidation-addition process on L-proline intercalated Fe-containing layered double hydroxides (LDHs), in which the direct 0-H elimination of aldehyde on hydroxide-layered Fe(III) sites with the two -electron abstraction promoted by the Fe(III):N coordination as the key step to efficiently oxidize the enamine. The major 0-functionalized diastereomer in the asymmetric nitromethylation of 3-phenylpropanal has been afforded in a regioselectivity of up to >99%, a stereoselectivity of 90%, and a yield of 65% on L-proline intercalated Mg3.00Al0.47Fe0.45-LDHs.(c) 2022 Elsevier Inc. Abstract:The transition metal-catalyzed C-H functionalization with hydroxylamine derivatives serving as both reactants and internal oxidants has attracted a lot of interest. These reactions obviate the need for external oxidants and therefore result in high reactivity and selectivity, as well as excellent functional group tolerance under mild reaction conditions, and moreover, water, methanol or carboxylic acid is generally released as the by-product, thus leading to reduced waste. This review focuses on the transition metal-catalyzed oxidative C-H functionalization of N-oxyenamine internal oxidants, with an emphasis on the scope and limitations, as well as the mechanisms of these reactions.

Abstract:Transition-metal-free-catalyzed nucleophilic hydrofunctionalization of both terminal and internal unactivated aliphatic alkenes has been described for the first time. Most topical classes of carbon, nitrogen and oxygen nucleophiles are well-compatible. The highly chemoselective unprotected dinucleophiles are also presented in the atom-economical approach. More than 80 structurally complex beta-hetero-substituted aliphatic amides were rapidly synthesized in good yield with exclusive Markovnikov selectivity, which are difficult to be achieved efficiently by the traditional Michael addition of conjugated amides due to their poor intrinsic electrophilicity.

Abstract:The utilization of unprotected NH2OH, which is not only an oxygen nucleophile but also a nitrogen nucleophile, in iridium catalyzed allylic substitution is realized under mild conditions. The chemoselectivity, stereoselectivity, and multiple allylation are controlled by adjusting the reaction conditions. This method produces the N-(1-allyl)-hydroxylamines in good to high yields with high level of chemoselectivities, regioselectivities, and enantioselectivities. The application of allylated hydroxylamine (R)-3a in the synthesis of diallylated hydroxylamine 6 is achieved, along with an excellent diastereomeric ratio.

Abstract:An unprecedented allylation reaction of protected hydrazines with enantioenriched allylic amines has been developed in a stereospecific manner with net stereoretention. A wide variety of protected hydrazines underwent palladium/acid-catalyzed allylation with highly enantioenriched primary allylic amines to give structurally diverse N-allylhydrazines in moderate to excellent yields with extremely high regioselectivity and complete retention of the configuration. Importantly, the reaction exhibits high atom-economy by yielding ammonia as the sole byproduct.
