Changes in Platelet-Derived Growth Factor Receptor-Mediated Signal Transduction Pathway in the Aorta of Spontaneously Hypertensive Rats

Abstract:Objective: To investigate the changes of platelet derived growth factor (PDGF) receptor mediated signal transduction pathway of aorta of spontaneously hypertensive rats (SHR). Methods: Cellular membrane protein and cytoplasmic protein of rat aorta were prepared from SHR and WKY rats. Western blot analysis was used to measure the changes of PDGF receptor β subunit , extracellular signal regulated kinase 1/2 (ERK 1/2) and phosphorylated ERK 1/2 (ppERK 1/2) of aorta. Results: Arterial blood pressure was normal in 4 week old SHR, and increased significantly and stably in 12 week old SHR. A progressive increase with age in aorta protein levels of the PDGF receptor β subunit was observed in SHR and WKY strains. But the levels of PDGF receptor β subunit of aorta in 4 and 12 week old SHR were significantly increased by 28.0% and 28.6% ( P 0.05), respectively, compared with their age matched WKY groups. The expression of ERK 1/2, the downstream signal molecule of PDGF receptor mediated signal transduction pathway, was increased significantly in 4 and 12 week old SHR compared with their age matched WKY rats ( P 0.05). The content of ppERK 1/2 of aorta from 12 week old SHR was also higher than that of age matched WKY rats by 28.4% ( P 0.05). Conclusion: The up regulations of PDGF receptor β subunit and ERK 1/2 of aorta were present before the appearence of hypertension, and maintained in higher level during hypertension. These data suggest that the activation of PDGF receptor β subunit mediated signal transduction pathway of aorta may contribute to the initiation and maintenance of spontaneously hypertension.
