Alterations in gene expression of calcium release channel in spontaneously hypertensive rats using RNA array

Abstract:Objective To evaluate the mRNA expression of calcium release channel proteins and relationship between the alteration in gene expression and the experimental hypertension in spontaneously hypertensive rats ( SHR) . Methods 294 total RNA were obtained from the tissue of ventriculum, aortic smooth muscle, liver and kidney in SHR and normotensive rats ( Wistar-kyoto rats,WKY). RNA array was used to determine the mRNA levels of ryanodine receptor 2 ( RyR2 ) and inositol 1 , 4, 5-triphosphate receptors 1 (IP3R1 ) in the two groups. Results Compared with age-matched WKY, the systolic blood pressure increased significantly in SHR at 6-week-old(P 0. 01) , 8-week-old(P 0. 01 ) , 10-week-old(P 0. 01) and 12-week-old( P 0. 01) , and the cardiosomatic ratio was significant high at 10-week-old (P 0. 01) and 12-week-old ( P 0. 01 ) . In cardiac sarcoplasmic reticulum, the mRNA levels of RyR2 were significantly increased at 4-week-old ( P 0. 05 ) , 6-week-old ( P 0. 01 ) , 8-week-old ( P 0. 05 ) , 10-week-old(P 0. 01 ) and 12-week-old (P 0. 01 ) and the expression of IP3R1 was also significantly increased at 6-week-old( P 0. 05 ) , 8-week-old ( P 0. 01) , 10-week-old ( P 0. 01) and 12-week-old ( P 0. 01) separately. In the aortic sarcoplasmic reticulum , the mRNA levels of RyR2 were significantly increased at 4-week-old( P 0. 05 ) , 6-week-old ( P 0. 05 ) , 8-week-old ( P 0. 05 ) , 10-week-old ( P 0. 05) and 12-week-old (P 0. 01) , and the expression of IP3 R, was also significantly increased at 4-week-old(P0.05) , 6-week-old(P0.05) , 8-week-old(P =0. 01) , 10-week-old (P0.05)and 12-week-old ( P 0. 05 ) . No significant signals were detected in the liver and the kidney. Conclusion Our results provided the evidence of the altered gene expression of Ca2 + release channel in SHR, which suggested the progressive nature of experimental hypertension.
