Abstract:Alfalfa (Medicago sativa L.), an important forage crop with high nutritional value and good palatability, plays a vital role in the development of animal husbandry in China. In Northeast China, there are vast areas of saline–alkali land that remain undeveloped. Given that alfalfa is a highly adaptable forage crop, exploring its salt tolerance at the molecular transcriptional level and identifying salt-tolerant genes has great significance for breeding salt-resistant alfalfa varieties. This also provides valuable genetic resources for better utilization of saline–alkali land. In this study, we conducted two rounds of screening on 41 alfalfa varieties and identified WL168 as a salt-sensitive variety and Longmu801 as a salt-tolerant variety. After 7 days of 300 mM salt stress, both varieties showed a decreasing trend in plant height, fresh weight, and dry weight over time, but Longmu801 demonstrated better water retention ability compared to WL168. Chlorophyll content also declined, but chlorophyll a and total chlorophyll levels in Longmu801 were higher than in WL168. Hydrogen peroxide and malondialdehyde levels increased overall, but Longmu801 had significantly lower levels than WL168 under prolonged stress. Both varieties showed increasing trends in soluble sugars, proline, and antioxidant enzymes (SOD, POD, CAT), with Longmu801 significantly outperforming WL168. This suggests that the two varieties share similar growth and physiological response mechanisms, with their differences primarily arising from variations in indicator levels. In the above, comparisons between varieties were conducted based on the relative values of the indicators in relation to their controls. Transcriptomic analysis revealed that under salt stress, Longmu801 had 16,485 differentially expressed genes (DEGs) relative to its control, while WL168 had 18,726 DEGs compared to its control. Among these, 2164 DEGs shared the same expression trend, with GO functions enriched in response to oxidative stress, nucleus, plasma membrane, and others. The KEGG pathways were enriched in phenylpropanoid biosynthesis, protein processing in the endoplasmic reticulum, starch and sucrose metabolism, and others. This suggests that alfalfa's transcriptional response mechanism to salt stress involves these pathways. Additionally, the variety-specific DEGs were also enriched in the same KEGG pathways and GO functions, indicating that the differences between the two varieties stem from their unique stress-responsive DEGs, while their overall mechanisms for coping with stress remain similar. To further identify salt stress-related genes, this study conducted WGCNA analysis using 32,683 genes and physiological indicators. Six modules closely related to physiological traits were identified, and the top five genes ranked by degree in each module were selected as hub genes. Further analysis of these hub genes identified five genes directly related to salt stress: Msa085011, Msa0605650, Msa0397400, Msa1258740, and Msa0958830. Mantel test analysis revealed that these genes showed strong correlations with physiological indicators. This study will provide important insights for breeding salt-tolerant alfalfa varieties.

Abstract:Alfalfa growth and production in China are negatively impacted by high salt concentrations in soils, especially in regions with limited water supplies. Few reliable genetic markers are currently available for salt tolerance selection. As a result, molecular breeding strategies targeting alfalfa are hindered. Therefore, with the continuous increase in soil salinity in agricultural lands, it is indispensable that a salt-tolerant variety of alfalfa is produced. We collected 220 alfalfa varieties around the world for resequencing and performed genome-wide association studies (GWASs). Alfalfa seeds were germinated in saline water with different concentrations of NaCl, and the phenotypic differences in several key root traits were recorded. In the phenotypic analysis, the breeding status and geographical origin strongly affected the salt tolerance of alfalfa. Forty-nine markers were significantly associated with salt tolerance, and 103 candidate genes were identified based on linkage disequilibrium. A total of 2712 differentially expressed genes were upregulated and 3570 were downregulated based on transcriptomic analyses. Some candidate genes that affected root development in the seed germination stage were identified through the combination of GWASs and transcriptome analyses. These genes could be used for molecular breeding strategies to increase alfalfa's salt tolerance and for further research on salt tolerance in general.

Abstract:Alfalfa (Medicago sativa L.) is an important legume forage crop. However, its genetic improvement for salt tolerance is challenging, as alfalfa’s response to salt stress is genetically and physiologically complex. A review was made to update the knowledge of morphological, physiological, biochemical, and genetic responses of alfalfa plants to salt stress, and to discuss the potential of applying modern plant technologies to enhance alfalfa salt-resistant breeding, including genomic selection, RNA-Seq analysis, and cutting-edge Synchrotron beamlines. It is clear that alfalfa salt tolerance can be better characterized, genes conditioning salt tolerance be identified, and new marker-based tools be developed to accelerate alfalfa breeding for salt tolerance.

Abstract:Cultivating salt-tolerant crops is a feasible way to effectively utilize saline-alkali land and solve the problem of underutilization of saline soils. Quinoa, a protein-comprehensive cereal in the plant kingdom, is an exceptional crop in terms of salt stress tolerance level. It seems an excellent model for the exploration of salt-tolerance mechanisms and cultivation of salt-tolerant germplasms. In this study, the seeds and seedlings of the quinoa cul-tivar Shelly were treated with different concentrations of NaCl solution. The physiological, biochemical charac-teristics and agronomic traits were investigated, and the response patterns of three salt stress-responsive genes (SSRGs) in quinoa were determined by real-time PCR. The optimum level of stress tolerance of quinoa cultivar Shelly was found in the range of 250-350 mM concentration of NaCl. Salt stress significantly induced expression of superoxide dismutase (SOD), peroxidase (POD), and particularly betaine aldehyde dehydrogenase (BADH). BADH was discovered to be more sensitive to salt stress and played an important role in the salt stress tolerance of quinoa seedlings, particularly at high NaCl concentrations, as it displayed upregulation until 24 h under 100 mM salt treatment. Moreover, it showed upregulation until 12 h under 250 mM salt stress. Taken together, these results suggest that BADH played an essential role in the salt-tolerance mechanism of quinoa. Based on the expression level and prompt response induced by NaCl, we suggest that the BADH can be considered as a mole-cular marker for screening salt-tolerant quinoa germplasm at the early stages of crop development. Salt treatment at different plant ontogeny or at different concentrations had a significant impact on quinoa growth. Therefore, an appropriate treatment approach needs to be chosen rationally in the process of screening salt-tolerant quinoa germplasm, which is useful to the utilization of saline soils. Our study provides a fundamental information to deepen knowledge of the salt tolerance mechanism of quinoa for the development of salt-tolerant germplasm in crop breeding programs.

Abstract:Alfalfa (Medicago sativa L.) is an important forage crop worldwide. However, little is known about the effects of breeding status and different geographical populations on alfalfa improvement. Here, we sequenced 220 alfalfa core germplasms and determined that Chinese alfalfa cultivars form an independent group, as evidenced by comparisons of FST values between different subgroups, suggesting that the geographical origin plays an important role in group differentiation. By tracing the influence of geographical regions on the genetic diversity of alfalfa varieties in China, we identified 350 common candidate genetic regions and 686 genes under selection. We also defined 165 loci associated with 24 important traits from genome-wide association studies (GWASs). Of those, 17 genomic regions closely associated with a given phenotype were under selection, with the underlying haplotypes showing significant differences between subgroups of distinct geographical origins. Based on results from expression analysis and association mapping, we propose that 6-phosphogluconolactonase (MsPGL) and a gene encoding a protein with NHL domains (MsNHL) are critical candidate genes for root growth. Abstract:Alfalfa ( Medicago sativa L.), a perennial forage plant, is a rich source of nutrients such as vitamins, minerals, and proteins. Salt stress, however, impedes its growth. The plant-specific transcription factor abscisic acid insensitive 3 (ABI3) has a critical contribution to the control of abscisic acid (ABA) signaling pathway and abiotic stress response. The gene ScABI3 from Syntrichia caninervis , a moss species tolerant to desiccation, could be considered a potential candidate gene to modify alfalfa's nutritional and growth aspects. However, it remains unclear how ScABI3 affects the salt stress response of transgenic alfalfa. Therefore, we elucidated the role and molecular mechanism of ScABI3 from S. caninervis as an ABA signaling factor in transgenic alfalfa. Our findings demonstrate that ScABI3 overexpression in transgenic alfalfa improves salt tolerance by promoting relative water content, antioxidant enzyme activity, and photosynthetic parameters. Furthermore, the key genes of plant hormone signaling and the classical salt tolerance pathway were activated in ScABI3 transgenic lines under salt stress. Based on these results, ScABI3 could be considered a potentially critical candidate gene to alleviate salt stress in alfalfa. The present study provides valuable insights for developing transgenic crop breeding strategies for saline-alkaline soils.

GO annotations and KEGG pathway enrichments of the DEGs based on RNA-seq data indicated that DEGs were involved in (1) ion and membrane homeostasis, including ABC transporter, CLC , NCX , and NHX ; (2) Ca 2+ sensing and transduction, including BK channel, EF-hand domain, and calmodulin binding protein; (3) phytohormone signaling and regulation, including TPR, FBP, LRR , and PP2C ; (4) transcription factors, including zinc finger proteins, YABBY , and SBP-box; (5) antioxidation process, including GST , PYROX, and ALDH ; (6) post-translational modification, including UCH , ubiquitin family, GT, MT and SOT . Abstract:Background: With soil salinity levels rising at an alarming rate, accelerated by climate change and human interventions, there is a growing need for crop varieties that can grow on saline soils. Alfalfa (Medicago sativa) is a cool-season perennial leguminous crop, commonly grown as forage, biofuel feedstock, and soil conditioner. It demonstrates significant potential for agricultural circularity and sustainability, for example by fixing nitrogen, sequestering carbon, and improving soil structures. Although alfalfa is traditionally regarded as moderately salt-tolerant species, modern alfalfa varieties display specific salt-tolerance mechanisms, which could be used to pave alfalfa's role as a leading crop able to grow on saline soils. Scope: Alfalfa's salt tolerance underlies a large variety of cascading biochemical and physiological mechanisms. These are partly enabled by alfalfa's complex genome structure and out-crossing nature, which on the other hand entail impediments for molecular and genetic studies. This review first summarizes the general effects of salinity on plants and the broad-ranging mechanisms for dealing with salt-induced osmotic stress, ion toxicity, and secondary stress. Secondly, we address defensive and adaptive strategies that have been described for alfalfa, such as the plasticity of alfalfa's root system, hormonal crosstalk for maintaining ion homeostasis, spatiotemporal specialized metabolite profiles, and the protection of alfalfa-rhizobia associations. Finally, bottlenecks for research of the physiological and molecular salt-stress responses as well as biotechnology-driven improvements of salt tolerance are identified and discussed. Conclusion: Understanding morpho-anatomical, physiological, and molecular responses to salinity is essential for the improvement of alfalfa and other crops in saline land reclamation. This review identifies potential breeding targets for enhancing alfalfa performance stability and general crop robustness for rising salt levels as well as to promote alfalfa applications in saline land management.

Abstract:Alfalfa is an important forage crop known for its perennial nature, high biomass yield, high nutritive value, and high water demand. Global reduction in the availability of clean water in arid and semiarid regions is forcing farmers to irrigate with low-quality degraded/recycled waters, which are generally higher in salts than freshwater. High salt concentrations in soils not only affect soil properties but also result in reduced plant growth and tissue ion toxicities. Plants employ a wide variety of morphological, anatomical, physiological, biochemical, and molecular mechanisms to cope with salinity stress. The self-incompatibility and polyploidy of alfalfa makes the genetic dissection of these complex mechanisms challenging. Understanding these mechanisms is critical to effectively manipulate underlying genetic determinants needed to develop salt-tolerant alfalfa genotypes. This chapter focuses on the effects of salinity on alfalfa growth and development, mechanisms of salinity tolerance in plants, and the current status of the knowledge related to salinity tolerance in alfalfa. Recent studies on the effects of salinity on growth, biomass yield, photosynthesis, water and ion relationships, nutritive value, and antioxidant capacity provide a clear picture of different component traits involved in the salinity response of alfalfa. Based on these studies and the molecular information generated on model plants, tremendous progress has been made to fill the major knowledge gaps in different pathways regulating salt stress responses in alfalfa. This knowledge will facilitate the manipulation of genetic components that would allow alfalfa to thrive when irrigated with recycled/highly saline waters and expand alfalfa cultivation to marginal lands.
Transcriptome Analysis for Salt-Responsive Genes in Two Different Alfalfa (Medicago sativa L.) Cultivars and Functional Analysis of MsHPCA1

Abstract:Alfalfa (Medicago sativa L.) is an important forage legume and soil salinization seriously affects its growth and yield. In a previous study, we identified a salt-tolerant variety 'Gongnong NO.1' and a salt-sensitive variety 'Sibeide'. To unravel the molecular mechanism involved in salt stress, we conducted transcriptomic analysis on these two cultivars grown under 0 and 250 mM NaCl treatments for 0, 12, and 24 h. Totals of 336, and 548 differentially expressed genes (DEGs) in response to NaCl were, respectively, identified in the 'Gongnong NO.1' and 'Sibeide' varieties. The Kyoto Encyclopedia of Genes and Genomes (KEGG) and Gene Ontology (GO) pathway enrichment analysis showed that the DEGs were classified in carbohydrate metabolism, energy production, transcription factor, and stress-associated pathway. Expression of MsHPCA1, encoding a putative H2O2 receptor, was responsive to both NaCl and H2O2 treatment. MsHPCA1 was localized in cell membrane and overexpression of MsHPCA1 in alfalfa increased salt tolerance and H2O2 content. This study will provide new gene resources for the improvement in salt tolerance in alfalfa and legume crops, which has important theoretical significance and potential application value.

Abstract:Alfalfa ( Medicago sativa L.), an important perennial legume forage crop with high nutritional value and forage yield, is widely used in animal husbandry. However, it is very sensitive to aluminum, which severely limits its growth in acidic soils. In this study, we analyzed the genotype variation of each agronomic trait in 44 alfalfa varieties in two acidic soil environments. Then, analysis of variance (ANOVA) of the variance components was performed using the Residual Maximum Likelihood (REML). The best linear unbiased predictor analysis was used to obtain the mean trait of each variety, and the mean values were used to construct the mean matrix of varieties × traits and interaction analysis of varieties × years. The results showed that there was significant ( P < 0.05) genotypic variation for each trait of the 44 varieties and the genetic diversity was abundant. The average repeatability ( R value) of interannual plant height (PH), stem thickness (ST), number of branches (NS), fresh weight (FW), total fresh weight (TFW), and total dry weight (TDW) was high (0.21–0.34), whereas the genetics were relatively stable. PH, NS, FW, TFW, and dry weight (DW) were positively correlated ( P < 0.01) with TDW. Six alfalfa varieties (Algonquin, Xinjiang daye, Trifecta, Vernal, WL354HQ, and Boja) with excellent TDW and TFW were identified in different years, environmental regions, and climatic altitudes. Our research results can provide suggestions and critical information regarding the future improvement and development of new alfalfa strains and varieties that are resistant to acidic soil conditions.

Abstract:Alfalfa is the most important forage internationally due to its high yield and nutritional value, but its production is severely limited by soil salinization. A better understanding of alfalfa's salt response mechanism is needed to provide guidance for breeding salt tolerant varieties. Lipidomic profiling of the salt-tolerant alfalfa cultivar Medicago sativa cv. Zhongmu-4 was performed after salt treatment. Of the 275 molecular species of lipid identified, 171 had altered levels after salt treatment. The levels of phospholipids, glycolipids, fatty acids, and sphingolipids decreased whereas lysophospholipids and glycerides accumulated in the salt treated plants. Specifically, higher PC/PE and DGDG/MGDG ratios and increased levels of FA 18:3 were observed, suggesting that changes in bilayer membrane structure integrity and fluidity maintenance occur in alfalfa leaves during salt stress. An accumulation of DAG (36:6, 36:5) and TAG (54:8, 54:9, 54:7, 54:6, 52:5, 52:6) was found, consistent with their potential role as storage substrates for membrane regeneration under salt stress. Changes in PI, LPC (16:0, 18:3, 18:2, 18:0), LPE (16:0, 18:2), and Cer (42:1;4 O, 34:1;4 O, 40:1;4 O) suggest these compounds may play a role in signal transduction under salt stress. The levels of PE, DGDG, and TAG were highly correlated (r>0.8) with changes in salt-related physiological parameters. In addition, the apparent remodeling of lipid profiles was supported by complementary changes in gene expression, including DGD2 , SQD2 , FAD2 , DGK , DGAT , and TAGL , that can be important candidate genes for alfalfa salt tolerance improvement. The detailed description of the changes occurring in lipid profiles and gene expression in alfalfa under salt stress provided by this study implicate lipid remodeling as an important feature of the salt-stress response, and provides a basis for the genetic improvement of alfalfa salt tolerance.

Identification of Molecular Marker Linked to Salt Tolerance Gene in Alfalfa

Abstract:The study has established the F2 offspring obtained by crossing salt-tolerant with salt-sensitive alfalfa, and appraised the salt-tolerant F2 offspring seedling was evaluated in pot culture. With the F2 segregated population, the research has obtained a molecular marker linked with salt-tolerant genes of alfalfa using the improved BSA combined with RAPD. The RAPD PCR products were excised from the agarose gel and purified using a kit, then were mixed with pMD-18T vector and sequenced. Sequencing result indicated the RAPD marker was 1 438 bp in length. Similarity researches using blast in Genbank indicated that the nucleotide sequence of the RAPD marker showed 93% and 91% similarity with mth2-6el8 gene fragment (347 bp) and mth2-33122 gene fragment (334 bp) of Medicago truncatula respectively. Medicago truncatula is a close relative of alfalfa and Mth2-6e18 is a molecular marker of the gene coding for a cysteine protease which was salt inducible in some plants. These results indicated the RAPD marker was possibly related to cysteine protease genes in alfalfa.

Abstract:Soil salinization is a widespread hindrance that endangers agricultural production and ecological security. High salt concentrations in saline soils are primarily caused by osmotic stress, ionic toxicity and oxidative stress, which have a negative impact on plant growth and development. In order to withstand salt stress, plants have developed a series of complicated physiological and molecular mechanisms, encompassing adaptive changes in the structure and function of various plant organs, as well as the intricate signal transduction networks enabling plants to survive in high-salinity environments. This review summarizes the recent advances in salt perception under different tissues, physiological responses and signaling regulations of plant tolerance to salt stress. We also examine the current knowledge of strategies for breeding salt-tolerant plants, including the applications of omics technologies and transgenic approaches, aiming to provide the basis for the cultivation of salt-tolerant crops through molecular breeding. Finally, future research on the application of wild germplasm resources and muti-omics technologies to discover new tolerant genes as well as investigation of crosstalk among plant hormone signaling pathways to uncover plant salt tolerance mechanisms are also discussed in this review.

Abstract:Soil salinization and a degraded ecological environment are challenging agricultural productivity and food security. Rice (Oryza sativa), the staple food of much of the world’s population, is categorized as a salt-susceptible crop. Improving the salt tolerance of rice would increase the potential of saline-alkali land and ensure food security. Salt tolerance is a complex quantitative trait. Biotechnological efforts to improve the salt tolerance of rice hinge on a detailed understanding of the molecular mechanisms underlying salt stress tolerance. In this review, we summarize progress in the breeding of salt-tolerant rice and in the mapping and cloning of genes and quantitative trait loci (QTLs) associated with salt tolerance in rice. Furthermore, we describe biotechnological tools that can be used to cultivate salt-tolerant rice, providing a reference for efforts aimed at rapidly and precisely cultivating salt-tolerance rice varieties.

Abstract:The physiological and biochemical indexes of different salt-tolerant wheat cultivars under salt stress are affected to different degrees. The changes in physiological and biochemical indexes in salt-tolerant wheat varieties are moderate, while salt-sensitive wheat varieties have bigger changes. In this article, through comprehensive utilization of germination and seedling indoor test morphological indexes, physiological and biochemical index, and output index, combined with the evaluation mechanism of wheat's salt resistance, different salt tolerance types in the middle and lower reaches of the Yangtze River in Shandong province were studied; a collection of 100 wheat varieties and 11 wheat varieties planted over a large area were classified into three groups: strong salt resistance, medium salt resistance, and weak salt resistance. Comparative analysis of different wheat varieties' salt resistance evaluation mechanism was performed, as well as the analysis of the germination rate of wheat varieties, coleoptile growth situation, emergence rate, protect wheat seeding rate, tillering rate, seedling height, root length, seedling dry weight, wet weight, number of leaves, plant growth situation, agronomic characters of the Na+/K+ ratio, and other physiological and biochemical indexes such as salt resistance index. The evaluation mechanism of salt tolerance and the relationship of salt tolerance of different wheat varieties were obtained. The results showed that there were significant differences in salt tolerance among the 100 wheat varieties at the germination stage. Among them, three varieties, YM (Yangmai) 25, YM (Yangmai) 24, and EM (Emai) 25, had the strongest salt tolerance at the germination stage, reaching the level of the salt tolerance variety DK 961, and the salt tolerance was in the first level, while NM 17, NM 23, and other 21 varieties reached the level of the salt tolerance. These 24 varieties with strong salt tolerance at the germination stage can be used to screen the salt tolerance of wheat planted in the saline soil of Jiangsu coastal beach.
