Metastasis of Hepatocellular Carcinoma Induced by Vascular Endothelial Growth Factor

Abstract:Objective To investigate the relationship between vascular endothelial growth factor (VEGF) and metastasis of hepatocellular carcinoma (HCC). Methods We cultured the HCC cell line HepG2 induced by VEGF by using Bodyen Chamber Membrane Invasion Culture System (MICS). At the end of the culture, the amnion was stained by Mayer's hematoxillin. We counted the number of cells going through the membrane from upper part to lower one of the chamber and that of cells staying inside the membrane. Results The cell number in the lower part of chamber was 5.8±1.0, 17.2±2.4 and (10.3±0.9)×10 4/ml after the cells were cultured for 5 hours by 1, 5 and 10 ng/ml VEGF, which was remarkably higher than that (1 6±0.4)×10 4/ml in the control (P0.05 or 0.01). After the cells were cultured by 5 ng/ml VEGF, the number of cells in the membrane was significantly higher in the experimental group than in the control (42 vs. 4,P0.01). Conclusion VEGF can induce metastasis of HCC. The most suitable concentration is 5 ng/ml and the most suitable duration is about 2~5 hours. The metastasis of HCC caused by VEGF is closely related to enhancement of the migration ability of HCC cells.
