The Effect of Vascular Endothelial Growth Factor on Invasive Capability and Homotypic Adhesion of Hepatocellular Carcinoma Cells

Abstract:Objective To observe the effect of Vascular Endothelial Growth Factor ( VEGF ) on metastasis and homotypic adhesion of hepatocellular carcinoma cells. Methods The functions of homotypic adhesion and metastasis in hepatocellular cancinoma cells ( HepG 2 ) were measured with 3 H-TdR infiltration and Boyden chamber respectively. Results After 60、90、120 minutes, the values ( dpm/min ) of 3 H-TdR infiltration in HepG 2 cells induced by 1ng/ml VEGF were 1758.67±289.46、1380.03±328.55 and 2657.43±310.31 respectively, by 5ng/ml were 3124.3±2262.14、2245.6±273.24 and 2091.52±213.84, and that by 10ng/ml VEGF were 1232.32±201.04、2337.5±333.04 and 2236.99±237.07 . The 10ng/ml group was dramatically lower than that in control group, ( P 0.05 or 0.01). The cells in lower chamber were respectively (5.75±1.00)×10 4 /ml, (17.17±2.38)×10 4 /ml and (10.33±0.88)×10 4 /ml after cultured for 5 hours by 1ng/ml、5ng/ml and 10ng/ml VEGF, which were remarkably higher than that (1.58±0.38)×10 4 /ml in control group, P 0.05 or 0.01. Conclusion VEGF can decrease homotypic adhesion of hepatocellular carcinoma cell HepG 2 , and is one of the cause of metastasis.
