Abstract:Stem cells have fascinated the scientific and clinical communities for over a century. Despite the controversy that surrounds this field, it is clear that stem cells have the potential to revolutionize medicine. However, a number of significant hurdles still stand in the way of the realization of this potential. Chiefly among these are safety concerns, differentiation efficiency and overcoming immune rejection. Here we review current progress made in this field to optimize the safe use of stem cells with particular emphasis on prospective interventions to deal with challenges generated by immune rejection.

Abstract:Stem cells have the potential to advance therapy for many neurological diseases that are currently refractive to treatment. They are also key cellular players in homeostasis within several adult brain regions that host endogenous populations of neural stem cells. Investigations of the functions of stem cells in the adult CNS have historically approached these cells as sources of differentiated progeny, whether it be new neurons or new glial cells. Yet, as both basic research and pre-clinical efforts centered on stem cells in the brain push forward, it has become evident that this initial framework is incomplete. Emerging evidence indicates that stem and progenitor cells from a variety of tissues can regulate their microenvironment through production of secreted factors. This special issue highlights work investigating the role of the neural and non-neural stem cell secretome in regulating CNS function. These studies represent efforts both to more fully delineate the suite of factors secreted by stem cells and to evaluate its impact on CNS health and disease. Together, they demonstrate a broad potential for stem cell function through secreted proteins that urges continued basic and translational research in the years to come.

Abstract:Stem cells are undifferentiated cells capable of self-renewal and differentiation, giving rise to specialized functional cells. Stem cells are of pivotal importance for organ and tissue development, homeostasis, and injury and disease repair. Tissue-specific stem cells are a rare population residing in specific tissues and present powerful potential for regeneration when required. They are usually named based on the resident tissue, such as hematopoietic stem cells and germline stem cells. This review discusses the recent advances in stem cells of various tissues, including neural stem cells, muscle stem cells, liver progenitors, pancreatic islet stem/progenitor cells, intestinal stem cells, and prostate stem cells, and the future perspectives for tissue stem cell research.

Abstract:Over the past decade, stem cells and regenerative medicine have emerged as a very important field in natural sciences and medicine. We have witnessed remarkable progress in this thriving field. The activities of this field range from basic stem cell research and translational research to development of stem cell-based products and clinical applications. Some of the exciting scientific advances in this field by research groups who are members of the Asian Federation of Biotechnology (AFOB) are captured by this Special Issue of Biotechnology Journal. The current Special Issue contains reviews on recent progress in stem cell therapy for neuronal diseases, cell cryopreservation technology, cell sheet engineering for regenerative medicine, and antibody approaches to prepare clinically transplantable cells from human embryonic stem cells, together with research articles on technical advances in stem cell differentiation, myeloid cell behavior control, development of media and bioreactors for stem cell expansion, and perfusion culture of three-dimensional tissues. Several fundamental aspects of stem cells are reported in this Special Issue. Zhou et al. [1] investigate angiogenic/osteogenic response of bone marrow-derived mesenchymal stromal cells in bone-derived scaffolds. The effect of hypoxia and the role of PI3K/Akt-mediated VEGF-VEGFR pathway is also reported. Oh et al. [2] demonstrate the preparation of microspheres of multipotent adult rat hippocampal progenitor cells. The cellular behaviors of multipotent adult rat hippocampal progenitor cells in microspheres were compared with those cultured in monolayers, both in vitro and in vivo differences were examined. The outstanding work by Abdal Dayem and colleagues [3] deal with convergence of nano-sized materials and stem cells by using unique properties of silver nanoparticle to induce neuronal differentiation of cells. Choi et al. [4] identify human embryonic stem cell surface markers by monoclonal antibodies, and develop antibody approaches to prepare clinically transplantable cells. Kawahara and colleagues [5] apply the technique of single chain Fv (scFv)/receptor chimeras named “signalobodies” in a myeloid progenitor cell line, and examined the corresponding signaling pathways.
