Abstract:Autophagy and apoptosis are two pivotal mechanisms in mediating cell survival and death. Cross-talk of autophagy and apoptosis has been documented in the tumorigenesis and progression of cancer, while the interplay between the two pathways in colorectal cancer (CRC) has not yet been comprehensively summarized. In this study, we outlined the basis of apoptosis and autophagy machinery firstly, and then reviewed the recent evidence in cellular settings or animal studies regarding the interplay between them in CRC. In addition, several key factors that modulate the cross-talk between autophagy and apoptosis as well as its significance in clinical practice were discussed. Understanding of the interplay between the cell death mechanisms may benefit the translation of CRC treatment from basic research to clinical use.

Abstract:Autophagy is a kind of cellular reaction,which is used to obtain nutrients and digest organelles.Cells can regulate autophagy to meet their needs.It plays an important role in maintaining protein metabolism balance and intracellular environment stability.It facilitates the cell growth,differentiation and maintenance of the organism's self-balance.Cells will induce autophagy in the situation of absence nutrition deficiency,degrading the cytoplasm and organelles to provide the energy for cell survival.However,if stimulating excessively,autophagy persistence will result in autophagy-like cell death.Abnormalities in autophagy regulation are directly related to tumorigenesis.Autophagy is a new hot spot in current tumor research,and it plays an important role in tumorigenesis and development.The induction of autophagy has a double-edged sword effect on the occurrence of tumors.On the one hand,it is a barrier that inhibits the growth of tumors,and on the other hand,it is an adaptive response of the relatively harsh environment,and promotes tumor progression.

Abstract:Autophagy is a highly conserved physiological phenomenon in most of eukaryotic cells and a cellular material recycling mechanism which is dependent on the lysosomes.Moderate autophagy allows cells to withstand the adverse environment,but excessive autophagy leads to cell death called autophagic cell death (ACD).Autophagic cell death is different from apoptosis,with the presence of a large number of autophagosomes wrapping cytoplasm and organelles.An increasing number of studies show that autophagy and autophagic cell death are closely related to the formation and development of some malignant tumors.In a variety of adverse circumstances,autophagy protects survival of tumour cells as an emergency mechanism,while initiation of autophagic cell death clears the tumour cells.In this paper,progress in the study on autophagy and tumor are reviewed.

Abstract:Autophagy has important functions in maintaining energy metabolism under conditions of starvation and to alleviate stress by removal of damaged and potentially harmful cellular components. Therefore, autophagy represents a pro-survival stress response in the majority of cases. However, the role of autophagy in cell survival and cell death decisions is highly dependent on its extent, duration, and on the respective cellular context. An alternative pro-death function of autophagy has been consistently observed in different settings, in particular, in developmental cell death of lower organisms and in drug-induced cancer cell death. This cell death is referred to as autophagic cell death (ACD) or autophagy-dependent cell death (ADCD), a type of cellular demise that may act as a backup cell death program in apoptosis-deficient tumors. This pro-death function of autophagy may be exerted either via non-selective bulk autophagy or excessive (lethal) removal of mitochondria via selective mitophagy, opening new avenues for the therapeutic exploitation of autophagy/mitophagy in cancer treatment.

Abstract:Abstract The balance between cell death and survival is a critical parameter in the regulation of cells and the maintenance of homeostasis in vivo. Three major mechanisms for cell death have been identified in mammalian cells: apoptosis (type I), autophagic cell death (type II), and necrosis (type III). These three mechanisms have been suggested to engage in cross talk with each other. Among them, autophagy was originally characterized as a cell survival mechanism for amino acid recycling during starvation. Whether autophagy functions primarily in cell survival or cell death is a critical question yet to be answered. Here, we present a comprehensive review of the cell death-related events that take place during autophagy and their underlying mechanisms in cancer and autoimmune disease development.

Abstract:Autophagy is an important catabolic process that delivers cytoplasmic material to the lysosome for degradation. Autophagy promotes cell survival by elimination of damaged organelles and proteins aggregates, as well as by facilitating bioenergetic homeostasis. Although autophagy has been considered a cell survival mechanism, recent studies have shown that autophagy can promote cell death. The core mechanisms that control autophagy are conserved between yeast and humans, but animals also possess genes that regulate autophagy that are not present in yeast. These regulatory differences may be explained by the need to control autophagy in a cell context-specific manner in multicellular animals, such as during cell survival and cell death. Autophagy was thought to be a bulk cytoplasmic degradation mechanism, but recent studies have shown that specific cargo is recruited for degradation. This suggests the possibility that either cell survival or death may be regulated by selective autophagic clearance of cytoplasmic material. Here we summarize the mechanisms that regulate autophagy and how they may contribute to cell survival and death.

Abstract:Autophagy is an evolutionarily conserved process responsible for protein degradation and organelle turnover. The literal translation of “autophagy” in Greek, “self-eating,” inclines that autophagy may be a phenomenon of cellular suicide. Indeed, cells undergoing cell death often display marked autophagic features, leading to the coining of the term “autophagic cell death.” However, linking autophagy to cell death is largely circumstantial, and whether it is the primary cause of cell death is intensively under debate. In contrast, autophagy, as a cellular recycling process, is an important protective mechanism against a range of various stress conditions including starvation. Therefore, the dialogue between autophagy and cell death programs influences the physiological homeostasis of cells and tissues. The imbalance between the two pathways is largely related to different human diseases, including cancer. Due to increasing interests in targeting autophagy pathway for different human diseases, molecular mechanisms of the cross talk between autophagy and cell death pathways have achieved major prominence. This chapter aims at summarizing the recent advances on the role of autophagy in cell death.

Abstract:Autophagy is a self-degradative process that is important for balancing sources of energy at critical times in development and in response to nutrient stress. Autophagy also plays a housekeeping role in removing misfolded or aggregated proteins, clearing damaged organelles, such as mitochondria, endoplasmic reticulum and peroxisomes, as well as eliminating intracellular pathogens. Thus, autophagy is generally thought of as a survival mechanism, although its deregulation has been linked to non-apoptotic cell death. Autophagy can be either non-selective or selective in the removal of specific organelles, ribosomes and protein aggregates, although the mechanisms regulating aspects of selective autophagy are not fully worked out. In addition to elimination of intracellular aggregates and damaged organelles, autophagy promotes cellular senescence and cell surface antigen presentation, protects against genome instability and prevents necrosis, giving it a key role in preventing diseases such as cancer, neurodegeneration, cardiomyopathy, diabetes, liver disease, autoimmune diseases and infections. This review summarizes the most up-to-date findings on how autophagy is executed and regulated at the molecular level and how its disruption can lead to disease.

Abstract:Autophagy is a lysosomal degradation pathway that is essential for survival, differentiation, development, and homeostasis. Autophagy principally serves an adaptive role to protect organisms against diverse pathologies, including infections, cancer, neurodegeneration, aging, and heart disease. However, in certain experimental disease settings, the self-cannibalistic or, paradoxically, even the prosurvival functions of autophagy may be deleterious. This Review summarizes recent advances in understanding the physiological functions of autophagy and its possible roles in the causation and prevention of human diseases.

Abstract:Autophagy, a cellular process of self-eating by which intracellular components are degraded within the lysosome, is an evolutionarily conserved response to various stresses. Autophagy is associated with numerous patho-physiological conditions, and dysregulation of autophagy contributes to the pathogenesis of a variety of human diseases including cancer. Depending on context, activation of autophagy may promote either cell survival or death, two major events that determine pathological process of many illnesses. Importantly, the activity of autophagy is often associated with apoptosis, another critical cellular process determining cellular fate. A better understanding of biology of autophagy and its implication in human health and disorder, as well as the relationship between autophagy and apoptosis, has the potential of facilitating the development of autophagy -based therapeutic interventions for human diseases such as cancer.

Abstract:Autophagy is an evolutionarily conserved intracellular self-digestion process, which mediates homeostasis in response to various stresses via degradation of damaged organelles or unnecessary proteins. It has been demonstrated that autophagy involves in tumorigenesis and progression. Autophagy serves either as tumor suppressor or promotor in a context-dependent way. It has been revealed in multiple studies that autophagy plays a pro-survival role upon treatment of anticancer drugs. Thus, combination of autophagy inhibitors with anticancer drugs may provide a desirable strategy to improve therapeutic efficacy. In this review, we summarize recent progress in the process and regulation of autophagy with a highlight in advances in the role of autophagy in cancer treatment. We also summarize some recent clinical outcomes of combinatorial use of autophagy inhibitors and anticancer drugs, and introduce latest discovered selective autophagy inhibitors. Some issues which should be paid attention to during the research to improve the clinical outcomes are discussed.

Abstract:Macroautophagy (hereafter referred to as autophagy) is a lysosomal catabolic pathway whereby cells recycle macromolecules and organelles. The capacity of autophagy to maintain cellular metabolism under starvation conditions and to remove damaged organelles under stress conditions improves the survival of cells. Yet, autophagy appears to suppress tumorigenesis. In this review we discuss recent data that begin to elucidate the molecular basis for this apparent controversy. First, we summarize our current knowledge on the autophagy-mediated control of both cell survival and cell death in general. Then, we highlight the common cancer-associated changes in autophagy induction, regulation and execution. And finally we discuss the potential of pro- as well as anti-autophagic signaling pathways as targets for future cancer therapy.

Beside macroautophagy, there are several forms of selective autophagy, including chaperone-mediated autophagy (CMA), cytoplasm to vacuole targeting (Cvt), pexophagy, and mitophagy. In this review, we summarize what is currently known about selective autophagy, and discuss its role in cell death and survival. We also discuss possible mechanisms underlying the selectivity of macroautophagy.

Abstract:Autophagy plays a complex impact role in tumor initiation and development. It serves as a double-edged sword by supporting cell survival in certain situations while also triggering autophagic cell death in specific cellular contexts. Understanding the intricate functions and mechanisms of autophagy in tumors is crucial for guiding clinical approaches to cancer treatment. Recent studies highlight its significance in various aspects of cancer biology. Autophagy enables cancer cells to adapt to and survive unfavorable conditions by recycling cellular components. However, excessive or prolonged autophagy can lead to the self-destruction of cancer cells via a process known as autophagic cell death. Unraveling the molecular mechanisms underlying autophagy regulation in cancer is crucial for the development of targeted therapeutic interventions. In this review, we seek to present a comprehensive summary of current knowledge regarding autophagy, its impact on cancer cell survival and death, and the molecular mechanisms involved in the modulation of autophagy for cancer therapy.
