METHODS Bacterial Cellulose/Gelatin composite Scaffold was fabricated with a nanofibrous layer and 3D microporous structure. The macro and micro-morphology observaton for Gelatin/BC composites and urethral acellular matrix were characterized using SEM. Then we cultured autologus lingual keratinocytes on the nanofibrous BC layer and muscle cells on the nano-microporous layer to obtain a new kind of 3D urethral graft for treating 5 cm urethral defect. 36 female beagle dogs were divided into three groups of 12 dogs each. Group A was given Pure BC scaffolds, group B was given final Gel/BC scaffolds, and group C was given Gel/BC scaffolds seeded with keratinocytes and muscle cells. The entire urethra was removed at 1,3, and 6 months postoperatively, then prepared by H&E/Masson staining and assessed by viewing sections of tissue in the mucosal membrane. RESULTS Our results showed that the composites could support keratincocytes and muscle cells adhesion and growth in vitro for its microporous and nanofibrous property. Gelatin/BC composites can withstand a higher mechanical load than gelatin alone. Our study also suggested that Gelatin/BC bilayer scaffolds with keratincocytes and muscle cells enhanced the repair in dog urethral defect models, resulting in patent urethra. Improved organized muscle bundles and epithelial layer were observed in animals treated with Gelatin/BC scaffold seeded by keratincocytes and muscle cells. CONCLUSIONS This study demonstrate the feasibility of developing 3-D bilayer scaffolds with both microporous structures and nanotextured surfaces by stationary cultivation Gluconacetobacte xylinus using gelatin microporous scaffold as a template. This study provides preclinical evidence that cell-seeded scaffolds can be used in the reconstruction of urethral defects.
