The Use of Vascular Endothelial Growth Factor with Silk Fibroin Scaffolds and Bladder Acellular Matrix Grafts to Support Bladder Reconstruction in Rabbit Model

Abstract:Background: Bladder tissue engineering (BTE) plays vital role in bladder repair and construction in urology. Biomaterials with excellent biocompatibility and good compliance in association with volume and contractility variations will be suitable for BTE. Materials and Methods: VEGF-SF electrospinning dope was prepared by mixing VEGF and SF together. Coaxial electrospinning technique was employed to develop VEGF-SF-BAGM. Two groups of rabbits underwent partial bladder wall cystectomy and reconstruction of the defects with an equally sized BAMG. Results: VEGF release profile exhibited a sustained release manner which was suitable for drug delivery and controlled release. At 24 weeks after implantation, the capacity of experimental group (34.1 +/- 1.98 ml) was significantly smaller than that of control group (48.9 +/- 1.44 ml). At 4 weeks, the silk fibroin partly degraded and the silk fibroin which covered by organization uniformly and become fragments. Futhermore, well-differentiated and arrangement of smooth muscle cells and the formation of the angiogenesis were observed after implantation. Conclusions: VEGF-SF-BAMG can effectively trigger angiogenesis by stimulating the growth of urethra epitheliums and smooth muscle cells after implantation and promote the urethral defect repair.
