Abstract:Continuous monoculture can increase plant morbidity and reduce crop yields. However, the Ganoderma lucidum mushroom possesses unique nutrient acquisition methods and displays an altered response to the generation of reactive oxygen species (ROS). The effects of continuous cropping on G. lucidum remain unclear. Here, we investigated the effects of consecutive monoculture soil on the yield, bioactive components, antioxidant properties, nutrient metabolism, and enzyme activity of G. lucidum. The results showed no significant differences in terms of yield between the control check and treatment groups after 2 years. However, the content of polysaccharides and triterpenoids after 2 years of treatment and the content of total phenols and flavonoids after 3 years of treatment significantly increased. Moreover, the antioxidant activity (AOA) of the polysaccharides under consecutive monoculture soil treatments also significantly increased. These differences were caused by changes in nutrient metabolism and the activities of antioxidant and ligninolytic enzymes. Under continuous monoculture soil treatment, the content of reducing sugars, water-soluble sugars, free amino acids, and water-soluble proteins significantly increased. The activities of superoxide dismutase, manganese peroxidase, and lignin peroxidase increased with the time of continuous monoculture, while the activities of peroxidase, catalase, and laccase decreased. The increase in ligninolytic enzymes was postulated to mediate the ROS scavenging ability of G. lucidum. In conclusion, continuous cropping soil could increase the levels of bioactive components and AOAs of G. lucidum.

Abstract:Saccharomyces cerevisiae FL 1 is selected as the mode yeast to study the dynamics of the bioprocess of yeast growth on the solid media and the influences of different osmolytes on the cellular growth under various physiological phases. By simplifying the colony to a sphere segment and theoretical deduction, one concise expression was built up, which can depict the bio-process and fit the observation well. Cell culture trial indicates that sodium chlorine significantly inhibits cell proliferation in the exponential and stationary growth phase via decreasing cellular specific growth rate and biomass. Glycerol can promote the cell proliferation in the stationary growth phase by enhancing cellular specific growth rate. Furthermore, there exists conjunct effect between sodium chlorine glycerol.

Abstract:In recent decades, an enormous potential of fungal-based products with characteristics equal to, or even outperforming, classic petroleum-derived products has been acknowledged. The production of these new materials uses mycelium, a root-like structure of fungi consisting of a mass of branching, thread-like hyphae. Optimizing the production of mycelium-based materials and fungal growth under technical conditions needs to be further investigated. The main objective of this study was to select fast-growing fungi and identify optimized incubation conditions to obtain a dense mycelium mat in a short time. Further, the influence of the initial substrate characteristics on hyphae expansion was determined. Fungal isolates of Ganoderma lucidum , Pleurotus ostreatus , and Trametes versicolor were cultivated for seven days on substrate mixtures consisting of various proportions of pine bark and cotton fibers. Furthermore, the substrates were mixed with 0, 2, and 5 wt.% calcium carbonate (CaCO 3 ), and the incubator was flushed with 0, 5, and 10 vol.% carbon dioxide (CO 2 ). All samples grew in the dark at 26 °C and a relative humidity of 80%. Evaluation of growth rate shows that cotton fiber-rich substrates performed best for all investigated fungi. Although Pleurotus ostreatus and Trametes versicolor showed comparatively high growth rates of up to 5.4 and 5.3 mm d −1 , respectively, mycelium density was thin and transparent. Ganoderma lucidum showed a significantly denser mycelium at a maximum growth rate of 3.3 mm d −1 on a cotton fiber-rich substrate (75 wt.%) without CaCO 3 but flushed with 5 vol.% CO 2 during incubation.

Abstract:The growth and sporulation of Mucor bacilliformis on an inert solid support (polyurethane foam) were studied at different conditions of water content and nutrient concentration. Maximum biomass (0.59g/ginertsupport) and spore production (6×108spores/ginertsupport) were obtained for the highest water content (90%, w/w) and the most concentrated medium studied. The growth, CO2 production and carbon source consumption of M. bacilliformis were modeled using the logistic model and the linear growth model of Pirt, and estimates of the kinetic parameters were obtained. Maximum specific growth rates (μmax) were determined from the biomass and CO2 data using the exponential and logistic model. The exponential model gave higher μmax values than the logistic model. Yield coefficients (YX/CO2), maintenance (mCO2/X) and endogenous decay coefficients (ke) were also determined.

Abstract:This article was about the growth experiment of Agaricus bisporus,which the growth parameters of the mycelial growth vigor,the extracellular protein content,intracellular polysaccharide,peroxidase activity were influenced by different density of the leached liquor of Ginkgo biloba leaves,Bambusoideae leaves,Castanea mollissima shell,Allium sativum L.,Zingiber offcinale Rose.and Citrus reticulate peel with plate culture and shaking fermentation.Compared with the control,the results showed the mycelial growth vigor,colony diameter and peroxidase activity of Agaricus bisporus were promoted by adding the above-mentioned 6 kind of 1-3g/L water leaching solutions.The most obvious effect on the mycelium growth was Ginkgo biloba leaves extract,the dry weight of mycelium reached the highest value 432mg/100mL.Intracellular polysaccharide and peroxidase activity reached the highest values 12.38mg/mL and 40.9U respectively,namely 3 times and 40 times compared the control by adding the 1g/L extract of Zingiber offcinale Rose.Considered the producing cost and economy benefit,water leaching solutions were better than ethanol extract.2g/L Ginkgo biloba leaves water extract added in general culture medium would in favor of the Agaricus bisporus growth.

Abstract:The studies on the growth of Cryptomanas ovata were first conducted in five different media. The medium BBM and FPW selected among them are better for the growth of C.ovata. Culture conditions including temperature, light intensity, light period and PH were also studied by gradient or orthogonal experiments. The result showed that temperature 25 ℃, light intensity 6000 xL,light period 18L∶[KG-*2]16D was the optimum culture condition for C.ovata. The medium BBM is advantageous for C.ovata to endure much higher temperature; the medium FPW is more useful, especially when used for large-scale culture of C.ovata, because it is more simple in composition and preparation with pond water.

Abstract:The dimorphic fungus Mucor indicus was grown in different forms classified as purely filamentous, mostly filamentous, mostly yeast-like and purely yeast-like, and the relationship between morphology and metabolite production, inhibitor tolerance and the cell wall composition was investigated. Low concentrations of spores in the inoculum with subsequent aeration promoted filamentous growth, whereas higher spore concentrations and anaerobic conditions promoted yeast-like growth. Ethanol was the main metabolite with glycerol next under all conditions tested. The yields of ethanol from glucose were between 0.39 and 0.42 g g(-1) with productivities of 3.2-5.0 g l(-1) h(-1). The ethanol productivity of mostly filamentous cells was increased from 3.9 to 5.0 g l(-1) h(-1) by the presence of oxygen, whereas aeration of purely yeast-like cells showed no such effect. All growth forms were able to tolerate 4.6 g l(-1) furfural and 10 g l(-1) acetic acid and assimilate the sugars, although with different consumption rates. The cell wall content of the fungus measured as alkali insoluble materials (AIM) of the purely yeast-like cells was 26% of the biomass, compared to 8% of the pure filaments. However, the chitosan concentration of the filaments was 29% of the AIM, compared to 6% of the yeast-like cells.

Microbial inoculants using spent mushroom substrates as carriers improve soil multifunctionality and plant growth by changing soil microbial community structure

Abstract:Peat is typically used as a carrier for microbial inoculants; however, due to its non-renewable nature alternatives need to be identified as reliable and renewable carriers for mineral-solubilizing inoculants. In pot experiments, solid microbial inoculants were comprised of peat (P), biochar (BC), and spent mushroom substrates (SMS) using Medicago sativa L. as experimental materials, and the purpose of this study is to assess the effect of solid microbial inoculants on soil multifunctionality and plant growth. The results revealed that the SMS microbial inoculant had the greatest positive impact on plant biomass and significantly stimulated soil multifunctionality which is typically managed or assessed based on various soil functions or processes that are crucial for sustaining productivity, in contrast to the peat microbial inoculant, particularly at a supply level of 100 g/pot. There was no significant correlation between soil multifunctionality and bacterial/fungal microbial diversity. However, according to the co-occurrence network of bacteria and fungi, soil multifunctionality was intimately correlated with the biodiversity of the main ecological clusters (modules) of bacteria and fungi, rather than to the entire soil microbial community structure. The keystone species of module hubs and connectors play critical roles in maintaining the stability of ecological clusters of microbial co-occurrence networks and linkages between ecological clusters. Soil pH is a major predictor of changes in plant biomass, and leads to changes therein by affecting the major ecological clusters of bacterial and fungal co-occurrence networks. These results suggested that SMS may serve as a good alternative to peat as a carrier of mineral-solubilizing microorganisms to maintain soil multifunctionality and promote plant growth.

Abstract:Lentinus edodes (shiitake mushroom) is a well-known medicinal and edible mushroom throughout the world. Statistical optimization was employed to optimize the culture medium for maximum mycelial growth and exo-polysaccharide (EPS) production in submerged fermentation: Plackett–Burman design (PBD) was applied to determine significant factors, followed by the paths of steepest ascent to move to the general vicinity of the optimum and Box–Behnken design (BBD) to obtain the final optimum culture medium composition. Glucose, yeast-powder and pH were significant for fermentation. 15.4g glucose and 5.32g yeast-powder per liter and pH 4.61 were optimum for the biomass accumulation from 2.75g/l to 6.88g/l, while 15.78g glucose and 5.86g yeast-powder per liter and pH 4.48 were optimum for EPS production from 0.214g/l to 0.751g/l. Under the optimized cultivation condition, cultivation kinetic models were studied in flask (pellet growth form) and airlift (filamentous growth form) reactors. The two morphologies resulted in different culture processes and mathematical models.

Abstract:The experiment objective was to study the effect of organic acid and amino acid on the mycelial growth of Agaricus Bisporus 2796;In the article,the effects of three kinds of organic acid from citric acid, tartaric acid and succinic acid and three kinds of amino acid from alanine,tyrosine and arginine in different concentration on the growth of Agaricus bisporus were studied by the solid and liquid culture methods.The growth parameters of the clony diameter,extracellular protein content,mycelium dry weight,intracellular polysaccharide content,peroxidase activity and reducing sugar content were analyzed in treat groups.The results showed that the growth of Agaricus bisporus could be mostly promoted by adding 0.04%citric acid.It would produce higher intracellular polysaccharide content,peroxidase activity and extracellular protein content.The mycelial growth could be also promoted by adding succinic acid,arginine and tyrosine.Compared to the control in the medium,the mycelium dry weight was highest by adding succinic acid,it increased 131%; the extracellular protein content was highest by adding arginine,it increased 151%;intracellular polysaccharide content and peroxidase activity of Agaricus bisporus were the highest value by adding citric acid,they increased 21%and 146%respectively.The reducing sugar content was declined by adding acids. But higher concentration organic acid and amino acid would inhibit the growth of Agaricus bisporus.
Analysis of growth dynamics in five different media and metabolic phenotypic characteristics of Piriformospora indica

Abstract:Piriformospora indica is an important endophytic fungus with broad potential for alleviating biotic and abiotic stress on host plants. This study monitored the growth dynamics of P. indica on five commonly used artificial media for microorganisms and analyzed its metabolic characteristics using Biolog Phenotype Microarray (PM) technology. The results showed that P. indica grew fastest on Potato Dextrose Agar (PDA), followed by Kidney Bean Agar (KBA), Alkyl Ester Agar (AEA), Oatmeal Agar (OA), and Luria-Bertani Agar (LB), and the most suitable medium for spore production was OA. Using Biolog PM1-10, 950 metabolic phenotypes of P. indica were obtained. P. indica could metabolize 87.89% of the tested carbon sources, 87.63% of the tested nitrogen sources, 96.61% of the tested phosphorus sources, and 100% of the tested sulfur sources. P. indica displayed 92 kinds of tested biosynthetic pathways, and it could grow under 92 kinds of tested osmotic pressures and 88 kinds of tested pH conditions. PM plates 1-2 revealed 43 efficient carbon sources, including M-Hydroxyphenyl acid, N-Acetyl-D-Glucosamine, Tyramine, Maltotrios, α-D-Glucosine, I-Erythritol, L-Valine, D-Melezitose, D-Tagatose, and Turanose. PM plates 3,6-8 indicated 170 efficient nitrogen sources, including Adenosine, Inosine Allantoin, D, L-Lactamide, Arg-Met, lle-Trp, Ala-Arg, Thr-Arg, Trp-Tyr, Val-Asn, Gly-Gly-D-Leu, Gly-Gly-Phe, and Leu-Leu-Leu. This study demonstrates that P. indica can metabolize a variety of substrates, such as carbon and nitrogen sources, and has a wide range of environmental adaptability. The growth dynamics on artificial culture media and metabolic phenotypes of P. indica can be used to investigate its biological characteristics, screen for more suitable growth and sporulation conditions, and elucidate the physiological mechanisms that enhance the stress resistance of host plants. This study provides a theoretical basis for its better application in agriculture.
Cultural characteristics of mycelia of Ganoderma gibbosum

Abstract:To study the cultural characteristics of mycelia of Ganoderma gibbosum, a medicinal fungus used in China. The growth rate and biomass of G. gibbosum mycelia were measured under different temperature, lightning carbon and nitrogen sources conditions. It showed that the optimal growth temperature for mycelia was 25 degrees C. Darkness was beneficial for mycelium growth. The initial pH 5.5 was suitable. The sucrose was the best carbon source and yeast extract the best nitrogen source, the optimal carbon-nitrogen ratio 60:2. These conclusions will offer references for further artificial cultivation and liquid fermentation.
Effects of Carbon and Nitrogen Nutrient on Submerged Fermentation in Agaricus Bisporus

Abstract:Carbon and nitrogen sources were the essential nutrients of edible fungus growth.Under the liquid culture conditions,the effects of four carbon sources and four nitrogen sources on mycelium biomass,intracellular polysaccharides,extracellular protein content and extracellular enzyme activity of Agaricus bisporus were studied by single factor tests method in this paper.The results showed that the optimum carbon and nitrogen sources were soluble starch and cornstarch to mycelial dry weight.In terms of extracellular protein,glucose and peptone were the most suitable carbon and nitrogen sources,sucrose and yeast powder were the best carbon and nitrogen sources respectively to inducing peroxydase,amylase and polyphenol oxidase.The experiments fixed a basic to research on the nutrition of Agaricus bisporus,also provided a science evidence to select good medium in the produce process and utilize the metabolize outcome in deep liquid culture.

Abstract:A growth experiment was conducted at Madhav Science College , Ujjain , India to find out the optimum growth requirements of flower decomposing fungi. In this experiment, Selected test fungi i.e. Penecillium sp., Aspergillus sp., Mucor sp.,Rhizopus sp. , Alterneria sp.etc, were taken and allow to grow in various kinds of media viz. semi defined media with floral extract , chemical defined media, semi defined media with yeast extract and semi defined media with floral extract & yeast extract. After incubation, observations indicate that semi defined media with yeast extract and floral extract is more suitable for fungal growth and absence of yeast extract slightly affect the fungal growth.

Abstract:Background: Certain microorganisms during the preparation of substrate for growing Agaricus bisporus are of great interest for the conversion of organic material and for the excretion of a wide range of metabolites with growth regulation activities that may affect the mycelial growth processes and mushroom yield. Adding beneficial microorganisms in the substrate may be a biotechnological alternative to optimize A. bisporus cultivation. Objective: Isolate and evaluate thermophilic microorganisms from self-heating pasteurized substrate with growth-promoting effects on A. bisporus cultivation. Methods: Different microorganisms were isolated and selected at 45 and 55 °C. They were tested for siderophore production, 1-octen-3-ol consumption, and phosphate solubilization in coculture with A. bisporus to determine their growth effects on potato dextrose agar (PDA) and on sterile pangola grass (Digitaria eriantha). Results: Of the 106 microorganisms isolated, 88 % were able to grow in the presence of 1-octen-3-ol, while 1 % had the capacity to produce siderophores, and 55 % had the ability to solubilize phosphate. The strains Bacillus hisashii ECS-B-65, B. licheniformis ECS-B-78, Rhizomucor pusillus ECS-710 and ECS-712, Aspergillus fumigatus ECS-709, and Thermomyces sp. ECS-711 were found to have a positive effect on A. bisporus mycelial growth.

Abstract:The effects of carbon and nitrogen sources, carbon-to-nitrogen ratio (C:N) and initial pH value on the growth and sporulation of the nematophagous fungus Pochonia chlamydosporia in liquid culture were examined. Among the 21 carbon sources and 15 nitrogen compounds tested, the optimal carbon and nitrogen sources for mycelial growth were sweet potato and L-tyrosine, and for sporulation were sweet potato and casein peptone. A C:N ratio of 10:1 at pH 3.7 gave the maximum yield of conidia and a C:N ratio of 40:1 at pH 6.8 gave the maximum biomass. The initial pH value had a significant effect on mycelial growth and conidial production, with the optimal ranges being 3.5–4.5 for sporulation and 5–6 for growth. Maximum conidial production was obtained at an initial pH of 4.0 and the maximum biomass at pH 6.0. The results also showed that the final pH after 7 days cultivation was always higher than the initial value. The variability in growth and sporulation of seven strains of P. chlamydosporia in liquid culture was also compared and discussed.

[Cultural characteristics of Armillaria mellea on solid media]

Abstract:Objective: To study the cultural characteristics of Armillaria mellea (A. mellea ) on solid media. Methods: A. mellea was cultured on semi-solid agar medium in dark conditions. Effects of different media, carbon sources, nitrogen sources, and temperature on growth and morphology of A. mellea were observed. The contents of polysaccharide, mannitol, glucose, and reducing sugars in A. mellea during different stages of development were determined. Results: The biomass and morphology of A. mellea were different in various media. Sugars were more effective carbon sources than the relevant sugar alcohols. Little molecular carbon sources such as alcohol and glycerol could be utilized by A. mellea, but starch only could be utilized slowly. Either organic or inorganic nitrogen sources could be uptaken and utilized effectively by A. mellea. No evidence was found that VitB1 affects the growth of A. mellea. The growth cycle on wort medium at 30 degrees C was shorter than that at 25 degrees C for 7 days. In logarithmic growth phase and stable phase, the polysaccharide contents of A. mellea were 9.24% and 4.70% respectively, while the mannitol contents were 10.08% and 10.58% respectively; glucose and reducing sugar contents remained low level in the whole growth stage. Conclusions: Carbon sources have a more remarked effect on the growth of A. mellea than the nitrogen sources do. Optimal temperature for the growth of A. mellea ranges 20-30 degrees C. Mannitol accumulates more than other little molecular carbohydrates in A. mellea.

Abstract:Conventional growth media for microbes contains lots of non-edible components that are harmful to consume when the organism is ready to intake—like probiotics growing in chemical media ( Lactobacillus grown in MRS media). The 04 isolates of Lactobacillus ( L. plantarum, L. rhamnosus, L. ferciminis and L. bifarmentans) and 02 Bifidobacterium (B. infantis and B. Bifidum) were used for the evaluation of medium efficacy. To formulate the edible culture media, 04 vegetables and 02 pulses were used. The media was formulated in different formulations. For Lactobacillus, maximum growth was observed at MM-2 media formulation that was about 15.62 log CFU/ml for L. plantarum and compared to MRS media that was 11.78 log CFU/ml. Bifidobaterium showed the highest viability at MW-1 edible media formulation which was about 12.72 log CFU/ml whereas in Bifidobacteria selective media the cell viability was 12.48 log CFU/ml. The edible media has no toxic or unhealthy effect on mice while trailing in an animal model and shows excellent results in encapsulation with alginate. In comparison with the performance of traditional chemical media, the formulated media were found to be cost-effective and safe for human consumption.
Influence of Culturing Mediums and Conditions on Characterization of Growth and Enzyme Production of White Rot Fungi

Abstract:The characterizations of growth and enzyme production of white rot fungi(Phanerochaete chrysosporium) in the natural culturing medium were compared with that in the synthetical culturing medium.The effects of bamboo lixivium addition,pH,support and antibiotics on the growth and enzyme production of Phanerochaete chrysosporium were studied.The results indicated that the biomass,shape of mycelia and the lignin peroxidase(LiP) activity presented higher values in the natural culturing medium than that in the synthetical medium.The lixivium of bamboo promoted the growth and enhanced the LiP activity of white rot fungi.The shape of mycelia was affected by the pH and carriers.The mycelia were small under low pH(4.5) and grew in the form of adhesion with the addition of carrier.Further studies with antibiotics showed that amphotericin B could inhibit the growth and enzyme production of white rot fungi at the mass concentration of 50(mg/L).

Abstract:This paper is concerned with the optimization of effect factors for mycelial growth and exopolysaccharide production by Schizophyllum commune by one-factor-at-a-time and orthogonal methods. The one-factor-at-a-time method was adopted to investigate the effects of six different compounds (sodium carboxymethylcellulose, l -glutamic acid, V B1 , naphthalene acetic acid, oleic acid, and Tween 80) on mycelial growth and exopolysaccharide production. Among these factors, oleic acid, V B1 and Tween 80 were identified to be the most important factors. Subsequently, the concentration of oleic acid, V B1 and Tween 80 were optimized using the orthogonal matrix method. The effects of the factors on the mycelial growth of S. commune were in the order of oleic acid > V B1 > Tween 80, and those on exopolysaccharide production were in the same order. The optimal concentration for mycelia and exopolysaccharide were determined as oleic acid 0.1% ( v / v ), V B1 0.5 mg/L, and Tween 80 6 mg/L. The subsequent verification experiments confirmed the validity of the models. Under this optimized conditions in shake flask culture, the mycelial yield and exo-biopolymer production were 25.93 and 2.79 g/L, respectively, which were considerably higher than those obtained in the preliminary studies. The result was further confirmed in a 7-L fermentor experiments.
