Expressions and Clinicopathological Significance of VEGF-C,VEGF-D and VEGFR-3 in Gastric Carcinoma

Abstract:OBJECTIVE: To explore the significance of VEGF-C, VEGF-D and vascular endothelial growth factor receptor-3 (VEGFR-3) in lymphangiogenesis and lymphatic metastasis in gastric carcinoma. METHODS: The expressions of VEGF-C, VEGF-D and VEGFR-3 in paraffin-embedded specimens obtained from 113 patients with primary gastric carcinoma were examined by immunohistochemistry. The expressions of VEGF-C, VEGF-D and VEGFR-3 in frozen specimens from 28 patients with primary gastric carcinoma were detected by RT-PCR. Lymphatic and blood vessels were quantitated immunohistochemically by using anti-VEGFR-3 antibody and anti-CD34 antibody. RESULTS: The positive rates of VEGF-C/D in specimens with lymph node metastasis were significantly higher than those in specimens without lymph node metastasis, P=0.018, P=0.032. The numbers of lymphatic vessels in the VEGF-C/D-positive group were higher than those in the VEGF-C/D-negative group, P=0.001, P=0.007. The survival time of patients with VEGF-C-positive tumors was significantly shorter than that of patients with VEGF-C-negative tumors. CONCLUSIONS: The expressions of VEGF-C and VEGF-D are significantly related with lymph node metastasis in gastric carcinoma and the number of lymphtic vessels in stroma of gastric carcinoma. VEGF-C may be an independent prognostic factor.
