Expressions of VEGF-D and MMP-7 in Gastric Cancer and Their Relationship with Prognosis

Abstract:Background: Lymphatic metastasis is the most common metastatic pattern of gastric cancer. Vascular endothelial growth factor D (VEGF-D) is a lymphangiogenic factor playing an important role in lymph node metastasis (LNM). Matrix metalloproteinases (MMPs) are capable of degrading all kinds of extraeellular matrix, and their roles in invasion and metastasis of malignancies have attracted much attention in recent years. Aims: To investigate the expressions of VEGF-D and MMP-7 in gastric cancer, and to study their relationship with invasiveness, LNM and prognosis of gastric cancer. Methods: Gastric cancer specimens of 108 patients who had undergone radical gastrectomy (54 patients with LNM, and 54 patients without LNM) and 20 normal gastric tissues were collected. The expressions of VEGF-D and MMP-7 were determined by immunohistochemistry. Results: VEGF-D and MMP-7 were generally localized in cytoplasm of gastric cancer cells with a moderate intensity. The positivity rates were 77.8% and 65.7%, respectively, which were significantly higher than those in normal gastric tissues (0% and 25.0%, respectively, P0.05). There were significant correlations between the expressions of VEGF-D and MMP-7, and the tumor size, differentiation, depth of invasion, LNM, TNM staging and survival less than five years in gastric cancer patients. Conclusions: Overexpressions of VEGFD and MMP-7 are significantly correlated with the invasiveness and LNM of gastric cancer, and might be the important prognostic factors.
