Study of the Biocompatibility on Bioactive Glass Ceramics with Cultured Periosteal-derived Osteoblasts in Vitro

Abstract:Objective:The purpose of this study was to investigate the biocompatibility of the bioactive glass ceramics (BGC) combined with cultured periosteal-derived osteoblasts(POBs) in vitro, and to select a favorable scaffold for bone tissue engineering. Methods: POBs harvested from a new born rabbit were cultured with the supernate of BGC, at different time points, cell proliferation ability and alkaline phosphatase(ALP) activity were examined by MTT method and enzyme assay. The morphological characters of POBs seeded into the porous BGC were observed by scanning electronic microscope (SEM). Results: The supernate of BGC had a little negative effect on the proliferation of POBs. BGC could stimulate the POBs to secrete more ALPs. Most of the seeded cells grew well on the surface and could produce some extracellular matrix (ECM). Conclusion: BGC had an excellent biocompatibility and osteoconduction with cultured POBs in vitro, and could be used as scaffolds in bone tissue engineering.
