IN VITRO STUDY ON THE CHARACTERISTICS OF PLANT GUM FERMENTED BY RECTAL MICROFLORA

Abstract:Objective To investigate the characteristics of plant gum fermentation by rectal microflora.Method Guar gum and pectin were taken respectively as the only carbon source fermented by rectal microflora in vitro,and then sampled at different fermentation periods,measured pH,counted colonies with selective media,measured the amount of short chain fatty acids(SCFAs) with gas chromatography.Results During the fermentation process in vitro,the pH value of the two cultures declined gradually.Apart from Lactobacterium,other bacteria in rectum showed clear downward trend in two media with different carbon sources,and the colonies count of Bacteroides declined dramatically after mid-term fermentation especially.Moreover,the amount of total anaerobic bacteria kept stable.Both of the two carbon sources could be utilized to produce SCFAs by microflora in colon..For guar gum,the amount of isobutyric acid was the most,then that of propionic acid.Butyric acid production was the least.Lactic acid was promptly utilized after short time of accumulation during fermentation.While for pectin,the amount of propionic acid was the most,and acetic acid was inferior to it.The least one was butyric acid.Lactic acid was also accumulated little in short period and could not be detected after 8h.Conclusion Plant gum contained in food can be fermented by human rectal microflora.Bacterial growth varies between species during fermentation and the two plant gums both inhibit the growth of bacteriodes.
