Abstract:Methionine is one of the most hydrophobic, redox-sensitive, and one of the only two sulfur-containing amino acids on protein. Because of these biochemical properties, the methionine residue plays a central role in a variety of biological processes, such as metal coordination, antioxidant stress, and aging. However, studies on the molecular functions of methionine are much less common than the other primary sulfur-containing amino acid, cysteine. The limited number of publications on methionine-related studies is partially due to the lack of tools for methionine modification. Methionine bioconjugation offers a new strategy to decipher the biological function of methionine and expands the toolbox for protein functionalization in the context of the application, such as synthesizing proteins with novel properties and producing new biomaterials. The purpose of this Perspective is to highlight the biochemical properties and functions of methionine, list recent progress in the development of methionine bioconjugation reagents, and briefly demonstrate the application of these reagents on polypeptides, proteins, and proteomes.

Abstract:Cancer remains a global health challenge, demanding novel therapeutic options due to the debilitating side effects of conventional treatments on healthy tissues. The review highlights the potential of L-methioninase, a pyridoxal-5-phosphate (PLP)-dependent enzyme, as a promising avenue in alternative cancer therapy. L-methioninase offers a unique advantage, its ability to selectively target and inhibit the growth of cancer cells without harming healthy cells. This selectivity arises because tumor cells lack an essential enzyme called methionine synthase, which healthy cells use to make the vital amino acid L-methionine. Several sources harbor L-methioninase, including bacteria, fungi, plants, and protozoa. Future research efforts can explore and exploit this diverse range of sources to improve the therapeutic potential of L-methioninase in the fight against cancer. Despite challenges, research actively explores microbial L-methioninase for its anticancer potential. This review examines the enzyme's side effects, advancements in combination therapies, recombinant technologies, polymer conjugation and novel delivery methods like nanoparticles, while highlighting the success of oral administration in preclinical trials. Beyond its promising role in cancer therapy, L-methioninase holds potential applications in food science, antioxidants, and various health concerns like diabetes, cardiovascular issues, and neurodegenerative diseases. This review provides a piece of current knowledge and future prospects of L-methioninase, exploring its diverse therapeutic potential.

Abstract:Reprogramming of methionine metabolism is a conserved hallmark of tumorigenesis. Recent studies have revealed mechanisms regulating methionine metabolism within the tumor microenvironment (TME) that drive both cancer development and antitumor immunity evasion. In this review article we summarize advancements in our understanding of tumor regulation of methionine metabolism and therapies in development that target tumor methionine metabolism. We also delineate the challenges of methionine blockade therapies in cancer and discuss emerging strategies to address them.

Abstract:Abstract As one of the bicyclic metabolic pathways of one-carbon metabolism, methionine metabolism is the pivot linking the folate cycle to the transsulfuration pathway. In addition to being a precursor for glutathione synthesis, and the principal methyl donor for nucleic acid, phospholipid, histone, biogenic amine, and protein methylation, methionine metabolites can participate in polyamine synthesis. Methionine metabolism disorder can aggravate the damage in the pathological state of a disease. In the occurrence and development of chronic liver diseases (CLDs), changes in various components involved in methionine metabolism can affect the pathological state through various mechanisms. A methionine-deficient diet is commonly used for building CLD models. The conversion of key enzymes of methionine metabolism methionine adenosyltransferase (MAT) 1 A and MAT2A/MAT2B is closely related to fibrosis and hepatocellular carcinoma. In vivo and in vitro experiments have shown that by intervening related enzymes or downstream metabolites to interfere with methionine metabolism, the liver injuries could be reduced. Recently, methionine supplementation has gradually attracted the attention of many clinical researchers. Most researchers agree that adequate methionine supplementation can help reduce liver damage. Retrospective analysis of recently conducted relevant studies is of profound significance. This paper reviews the latest achievements related to methionine metabolism and CLD, from molecular mechanisms to clinical research, and provides some insights into the future direction of basic and clinical research.

Abstract:N6-methyladenosine (m6A) is the most abundant RNA modification; m6A modifications are installed by methyltransferases, removed by demethylases and recognized by reader proteins. M6A plays crucial roles in a variety of biological processes by regulating target RNA translation, splicing, nuclear export, and decay. Since the establishment of methylated RNA immunoprecipitation-sequencing methodology, over three hundred articles about m6A modulators, including "writers", "erasers" and "readers", have been reported in the last four years. In addition, an increasing number of molecular mechanisms underlying m6A RNA methylation in human cancers have been comprehensively clarified. The recently emerged molecular mechanisms of m6A modulators in cancer cell proliferation, cell cycle progression, migration and invasion, apoptosis, and autophagy remain to be summarized. Hence, this review specifically summarizes these recent advances in the understanding of m6A molecular mechanisms in tumorigenesis and cancer progression. In addition, we discuss the prospect of using an m6A methylation modulator as a new diagnostic biomarker and therapeutic target for human cancers.

Abstract:Abstract The proper transfer of genetic information from DNA to RNA to protein is essential for cell-fate control, development, and health. Methylation of DNA, RNAs, histones, and non-histone proteins is a reversible post-synthesis modification that finetunes gene expression and function in diverse physiological processes. Aberrant methylation caused by genetic mutations or environmental stimuli promotes various diseases and accelerates aging, necessitating the development of therapies to correct the disease-driver methylation imbalance. In this Review, we summarize the operating system of methylation across the central dogma, which includes writers, erasers, readers, and reader-independent outputs. We then discuss how dysregulation of the system contributes to neurological disorders, cancer, and aging. Current small-molecule compounds that target the modifiers show modest success in certain cancers. The methylome-wide action and lack of specificity lead to undesirable biological effects and cytotoxicity, limiting their therapeutic application, especially for diseases with a monogenic cause or different directions of methylation changes. Emerging tools capable of site-specific methylation manipulation hold great promise to solve this dilemma. With the refinement of delivery vehicles, these new tools are well positioned to advance the basic research and clinical translation of the methylation field.

Abstract:For more than 70 years,it has been recognized that antifolates play a key role in treatment of malignant,microbial,parasitic and chronic inflammatory diseases.The discovery of new types of antifolates with improved properties and superior activities still remains an attractive strategy both in academia and in the pharmaceutical industry.Cobalamin-dependent methionine synthase is the pivotal enzyme in the human cells,which plays an important role in maintaining balance of folates.Inhibition of this enzyme can lead to kill cancer cells by blocking folate cycle.So cobalamin-dependent methionine synthase is an excellent target for anticancer drug research.This review mainly presents the development of methionine synthase inhibitors.

Abstract:Methyl group is not only an important building block of biologically active molecules and drug molecules, but also the important structural unit of complex molecules. Methylation of complex molecules is generally achieved through methylating reactions, which mainly include nucleophilic attack, electrophilic attack, and radical pathways. This review summarized the literatures of the past few decades and covered commonly used methylating agents used in methylation. In this review, the recently developed new methylating agents and methylation reactions are highlighted, and the reaction mechanisms are also detailed discussed. We expect that this review will inspire others to devote their research in the field of methylation.

Abstract:Methionine is the only nonpolar α-amino acid containing sulfur among the eight essential amino acids and is closely related to the metabolism of sulfur-containing compounds in the human body. Widely used in feed, medicine, food, and other fields, the market demand is increasing annually. However, low productivity and high cost largely limit the industrial production of methionine, and many novel production methods still have their own disadvantages. In this paper, the available methods for synthesizing methionine are reviewed and discussed. The latest strategies for improving methionine production are further introduced, including culture medium optimization, mutation technology, expression of key genes in the metabolic pathway, knockout and recombination, as well as the engineering of membrane transporters, the fermentation-enzymatic coupling route, and innovation of CO2 biotransformation.

Abstract:Methylation plays a vital role in biological systems. SAM (S-adenosyl-L-methionine), an abundant cofactor in life, acts as a methyl donor in most biological methylation reactions. SAM-dependent methyltransferases (MTase) transfer a methyl group from SAM to substrates, thereby altering their physicochemical properties or biological activities. In recent years, many SAM analogues with alternative methyl substituents have been synthesized and applied to methyltransferases that specifically transfer different groups to the substrates. These include functional groups for labeling experiments and novel alkyl modifications. This review summarizes the recent progress in the synthesis and application of SAM methyl analogues and prospects for future research directions in this field.

Abstract:DNA methylation abnormalities are regarded as critical event for cancer initiation and development. Tumor-associated genes encompassing aberrant DNA methylation alterations at specific locus are correlated with chromatin remodeling and dysregulation of gene expression in various malignancies. Thus, technologies designed to manipulate DNA methylation at specific loci of genome are necessary for the functional study and therapeutic application in the context of cancer management. Traditionally, the method for DNA methylation modifications demonstrates an unspecific feature, adversely causing global-genome epigenetic alterations and confusing the function of desired gene. Novel approaches for targeted DNA methylation regulation have a great advantage of manipulating gene epigenetic alterations in a more specific and efficient method. In this review, we described different targeting DNA methylation techniques, including both their advantages and limitations. Through a comprehensive understanding of these targeting tools, we hope to open a new perspective for cancer treatment.

Abstract:Methionine is an essential amino acid in animals and it is also a limited amino acid in the diet. Methionine can be used as a substrate to participate in protein synthesis. It can also be used as a methyl donor involved in the methylation of DNA, RNA and protein, or through the transfer of sulfhydryl pathways to play anti-oxidative roles and participate in the formation of polyamines. These metabolic pathways have important regulatory effects on animal performance, antioxidant capacity, immune function, and intestinal health. This article reviews the mechanism of methionine modulating physiological functions in recent years in order to provide reference for the study and scientific application of methionine metabolism mechanism.

Abstract:Metallenes, atomically thin-layered materials composed of coordination-deficient metal atoms, have emerged as a new category of two-dimensional materials. Metallenes exhibit exciting properties with a fusion of atom economy, ultrathin structure, photonic properties, and catalytic activity, which make them intriguing for a wide range of applications in biomedicine. The development of biomedical applications of metallenes is in its infancy yet fast-growing. In this review, after a brief introduction of the definition, structures, properties, and classification of metallenes, we outline two common synthesis strategies and identify their shortcomings. Then, we comprehensively discuss the biological effects of metallenes, such as nano-biointeractions and signaling pathway regulation. We also highlight their recent advances in biomedical applications, including antitumor, biosensing, bioimaging, antibacterial, and anti-inflammation. Finally, we provide personal perspectives on remaining challenges and future opportunities for the biomedical applications of metallenes.

Abstract:Cysteine can be specifically functionalized by a myriad of acid-base conjugation strategies for applications ranging from probing protein function to antibody-drug conjugates and proteomics. In contrast, selective ligation to the other sulfur-containing amino acid, methionine, has been precluded by its intrinsically weaker nucleophilicity. Here, we report a strategy for chemoselective methionine bioconjugation through redox reactivity, using oxaziridine-based reagents to achieve highly selective, rapid, and robust methionine labeling under a range of biocompatible reaction conditions. We highlight the broad utility of this conjugation method to enable precise addition of payloads to proteins, synthesis of antibody-drug conjugates, and identification of hyperreactive methionine residues in whole proteomes.

Abstract:Advances in mass spectrometry allow for broader applications of metabolomics in research and clinical applications. In a recent issue of Nature Metabolism, Vande Voorde and colleagues utilized metabolite profiling to investigate the metabolism of colorectal cancer in mouse models, organoids, and patients. This study underscores the utility of metabolomics in distinguishing colorectal cancer, offering potential for its use in precision medicine. It also revealed a pivotal role for adenosylhomocysteinase in the methionine cycle and highlighted its potential as a therapeutic target.

Abstract:Peptide-based drugs have garnered considerable attention in recent years owing to their increasingly crucial role in the treatment of diverse diseases. However, the limited pharmacokinetic properties of peptides have hindered their full potential. One prominent strategy for enhancing the druggability of peptides is N-methylation, which involves the addition of a methyl group to the nitrogen atom of the peptide backbone. This modification significantly improves the stability, bioavailability, receptor binding affinity and selectivity of peptide drug candidates. In this review, we provide a comprehensive overview of the advancements in synthetic methods for N-methylated peptide synthesis, as well as the associated limitations. Moreover, we explore the versatile effects of N-methylation on various aspects of peptide properties. Furthermore, we emphasize the efforts dedicated to N-methylated peptide pharmaceuticals that have successfully obtained marketing approval.

Abstract:As the predominant epigenetic markers, methyl-modifying enzymes-catalyzed methylation modification widely participates in the regulation of cellular functions, and the dysregulation of methyl-modifying enzyme activity is closely related with various malignant tumors, autoimmune disorders, and diabetes. Consequently, methylation modification and methyl-modifying enzymes have become valuable biomarkers for clinical disease diagnosis. Single-molecule fluorescent detection has distinct advantages of minimum sample consumption, excellent sensitivity, and visualization capability. However, there is no review about single-molecule detection of methylation modification and methyl-modifying enzymes so far. Herein, we summarize the single-molecule detection-based strategies for the measurement of methylation modification and methyl-modifying enzymes in the last five years (2019-2023), and focus on five categories of methylation modification and methyl-modifying enzymes including DNA/RNA methylation modifications, DNA methyltransferases, DNA demethylases, RNA demethylases, and RNA methyltransferases. We summarize the working principles and applications of these strategies, and discuss the challenges and prospects in this field.

Abstract:Methionine (Met) offers a valuable handle to achieve peptide chemical modification owing to its unique thioether functional group. In contrast with cysteine, the site-selective functionalization of the hydrophobic and redox-sensitive thioether motif on peptides is still challenging, and strategies for diversification on the Met residue are rarely disclosed. Herein we report a transition-metal-free and redox-neutral approach for Met diversification with substrate diversity, which could be applied to synthesize cyclic peptides.
