Abstract:Methyl group is not only an important building block of biologically active molecules and drug molecules, but also the important structural unit of complex molecules. Methylation of complex molecules is generally achieved through methylating reactions, which mainly include nucleophilic attack, electrophilic attack, and radical pathways. This review summarized the literatures of the past few decades and covered commonly used methylating agents used in methylation. In this review, the recently developed new methylating agents and methylation reactions are highlighted, and the reaction mechanisms are also detailed discussed. We expect that this review will inspire others to devote their research in the field of methylation.
