Abstract:Biodegradable metals and their alloys have gained significant attention as promising candidates for biomedical applications. Notably, Magnesium, Zinc, and Iron stand out as the most prominent biodegradable materials, prompting extensive research into novel alloy development. These metals exhibit inherent biodegradability, acceptable biocompatibility, and commendable mechanical integrity, rendering them suitable for implant use. In this comprehensive review, we delve into recent advancements in biodegradable implants within the biomedical domain, with a primary focus on orthopedic and cardiovascular applications. The paper extensively explores the in vivo and in vitro degradation behavior, mechanical properties, and corrosion mechanism of these materials. Additionally, we discuss progress in surface modification and fabrication techniques, vital in optimizing implant performance. However, certain limitations associated with alloying materials, especially Zinc and Iron, have hindered their widespread implementation. We underscore the importance of overcoming these challenges to unlock the full potential of these materials. Furthermore, we present the promise of new metals, such as Molybdenum, which exhibit remarkable properties, opening doors to implants with superior mechanical characteristics. Overall, this review emphasizes the growing significance of biodegradable metallic implants, showcasing their potential to address diverse clinical needs. By addressing material limitations and harnessing innovative metals, we pave the way for the development of implants with superior mechanical properties and desirable degradation rates, thus revolutionizing the field of biomedical applications.

Abstract:Biodegradable metals (BMs) are metals and alloys expected to corrode gradually in vivo, with an appropriate host response elicited by released corrosion products, then dissolve completely upon fulfilling the mission to assist with tissue healing with no implant residues. In the present review article, three classes of BMs have been systematically reviewed, including Mg-based, Fe-based and Zn-based BMs. Among the three BM systems, Mg-based BMs, which now have several systems reported the successful of clinical trial results, are considered the vanguards and main force. Fe-based BMs, with pure iron and Fe–Mn based alloys as the most promising, are still on the animal test stage. Zn-based BMs, supposed to have the degradation rate between the fast Mg-based BMs and the slow Fe-based BMs, are a rising star with only several reports and need much further research. The future research and development direction for the BMs are proposed, based on the clinical requirements on controllable degradation rate, prolonged mechanical stability and excellent biocompatibility, by optimization of alloy composition design, regulation on microstructure and mechanical properties, and following surface modification.

Abstract:With attractive research and development of biomaterials, more and more opportunities have been brought to the treatments of human tissue repairs. The implant is usually no need to exist in the body accompanied with the recovery or regeneration of the tissue lesions, and the long-term effect of exotic substance to human body should be reduced as lower as possible. For this purpose, biodegradable materials, including polymers, magnesium alloys and ceramics, have attracted much attention for medical applications due to their biodegradable characters in body environment. This paper in turn introduces these three different types of widely studied biodegradable materials as well as their advantages as implants in applications for bone repairs. Relevant history and research progresses are summarized.

Abstract:Znic (Zn) alloys with good cytocompatibility and suitable degradation rate have been a kind of biodegradable metal with great potential for clinical applications. This paper summarizes the biological role of degradable Zn alloy as bone implant materials, discusses the mechanical properties of different Zn alloys and their advantages and disadvantages as bone implant materials, and analyzes the influence of different processing strategies (such as alloying and additive manufacturing) on the mechanical properties of Zn alloys. This paper provides systematic design approaches for biodegradable Zn alloys as bone implant materials in terms of the material selection, product processing, structural topology optimization, and assesses their application prospects with a view to better serve the clinic.

Abstract:Additive manufacturing (AM) can produce complicated structures accurately and freely, giving the implant a macro and micro geometry, which makes the implant match the patient's defect site and realize the needs for personalized clinical treatment. Thus, AM provides a new manufacturing method for biodegradable metals. Presently, biodegradable metals are the hotspot issues of metallic biomaterials research. Additively-manufactured biodegradable metals are new research field. In this paper, a comprehensive review on the AM of Mg-, Zn-, and Fe-based biodegradable metals, which focuses on their processes and influencing factors, mechanical properties, biodegradation behavior, and biocompatibility, is given. Finally, the future development trend of the AM biomedical metallic materials is explored.

Abstract:Mg-based alloys as revolutionary implantable biomaterials have increasingly attracted considerable attention, owing to their biodegradability in vivo and beneficial effects on biological systems. The degradation process and products of Mg-based alloys have been reported to exhibit significant biological effects on host-tissue responses. However, these effects have not yet been fully understood. This review systemically summarizes and analyses the current understandings and recent research progress in this area. The primary focal points are the biological effects and related mechanisms associated with the degradation behaviour of Mg-based alloys. The biological impacts of the degradation products are elucidated and the arguable or controversial issues are also discussed, providing a pathway toward a greater understanding of the biological implications of Mg-based alloys. Furthermore, based on these biological implications, the restorative potential of Mg-based alloys for applications in tissue repair and regeneration is summarized. Finally, outlooks on biosafety evaluation and design strategies for Mg-based alloy implants are briefly discussed.

Abstract:Degradable metal scaffolds, including multifunctional scaffolds doped with metal elements, enhance bone regeneration through mechanical and biological properties. This review highlights the materials, techniques, and potential for designing multifunctional scaffolds doped with metal elements. The clinical application of degradable metal‐based bone tissue engineering scaffolds in human bone repair and the research on different types of metal‐based tissue engineering scaffolds are discussed. The increasing prevalence of orthopedic‐related diseases necessitates the development of effective orthopedic implants. Conventional metal scaffolds used in orthopedic devices have limitations such as poor biocompatibility and the need for a second surgery to remove the scaffold. Degradable metal‐based scaffolds, including metal‐based scaffolds and multifunctional scaffolds doped with metal elements, are a rapidly developing tissue engineering strategy aimed at utilizing the mechanical and biological properties of metal elements to create a support structure that matches the complex bone regeneration environment. Repairing bone defects involves the regeneration of various tissues, and incomplete repair can negatively affect bone function and overall recovery. Therefore, combining metal‐based degradable materials with other active components has great potential for enhancing the versatility and clinical applications of scaffolds. Multifunctional scaffolds doped with metal elements have better biocompatibility, osteoinductivity, biodegradability, matching mechanical, and microenvironmental adjustment capabilities. Moreover, these metal‐doped scaffolds possess the advantages of controlled release of metal ions, multifunctionality, and faster degradation. This review focuses on the materials and techniques used for constructing degradable metal‐based scaffolds. Furthermore, this study discussed the potential for designing and constructing multifunctional biodegradable metal‐based scaffolds doped with metal elements by integrating multiple strategies.

Abstract:This review discusses and summarizes the recent developments and advances in the use of biodegradable materials for bone repair purposes. The choice between using degradable and non-degradable devices for orthopedic and maxillofacial applications must be carefully weighed. Traditional biodegradable devices for osteosynthesis have been successful in low or mild load bearing applications. However, continuing research and recent developments in the field of material science has resulted in development of biomaterials with improved strength and mechanical properties. For this purpose, biodegradable materials, including polymers, ceramics and magnesium alloys have attracted much attention for osteologic repair and applications. The next generation of biodegradable materials would benefit from recent knowledge gained regarding cell material interactions, with better control of interfacing between the material and the surrounding bone tissue. The next generations of biodegradable materials for bone repair and regeneration applications require better control of interfacing between the material and the surrounding bone tissue. Also, the mechanical properties and degradation/resorption profiles of these materials require further improvement to broaden their use and achieve better clinical results.

Abstract:The first systematic review & meta-analysis on biodegradable metals.The quality of evidence of biodegradable metals for bone defect repair is very low.Evidence-based study is an effective approach for biomaterials research.Biodegradable metals are promising candidates for bone defect repair. With an evidence-based approach, this study investigated and analyzed the performance and degradation properties of biodegradable metals in animal models for bone defect repair to explore their potential clinical translation. Animal studies on bone defect repair with biodegradable metals in comparison with other traditional biomaterials were reviewed. Data was carefully collected after identification of population, intervention, comparison, outcome, and study design (PICOS), and following the inclusion criteria of biodegradable metals in animal studies. 30 publications on pure Mg, Mg alloys, pure Zn and Zn alloys were finally included after extraction from a collected database of 2543 publications. A qualitative systematic review and a quantitative meta-analysis were performed. Given the heterogeneity in animal model, anatomical site and critical size defect (CSD), biodegradable metals exhibited mixed effects on bone defect repair and degradation in animal studies in comparison with traditional non-degradable metals, biodegradable polymers, bioceramics, and autogenous bone grafts. The results indicated that there were limitations in the experimental design of the included studies, and quality of the evidence presented by the studies was very low. To enhance clinical translation of biodegradable metals, evidence-based research with data validity is needed. Abstract:Zinc (Zn)-based biodegradable materials show moderate degradation rates in comparison with other biodegradable materials (Fe and Mg). Biocompatibility and non-toxicity also make them a viable option for implant applications. Furthermore, Pure Zn has poor mechanical behavior, with a tensile strength of around 100-150 MPa and an elongation of 0.3-2%, which is far from reaching the strength required as an orthopedic implant material (tensile strength is more than 300 MPa, elongation more than 15%). Alloy and composite fabrication have proven to be excellent ways to improve the mechanical performance of Zn. Therefore, their alloys and composites have emerged as an innovative category of biodegradable materials. This paper summarizes the most important recent research results on the mechanical and biological characteristics of biodegradable Zn-based implants for orthopedic applications and the most commonly added components in Zn alloys and composites.

Abstract:After decades of developing strategies to minimize the corrosion of metallic biomaterials, there is now an increasing interest to use corrodible metals in a number of medical device applications. Abstract:Compared with non-degradable materials, biodegradable biomaterials play an increasingly important role in the repairing of severe bone defects, and have attracted extensive attention from researchers. In the treatment of bone defects, scaffolds made of biodegradable materials can provide a crawling bridge for new bone tissue in the gap and a platform for cells and growth factors to play a physiological role, which will eventually be degraded and absorbed in the body and be replaced by the new bone tissue. Traditional biodegradable materials include polymers, ceramics and metals, which have been used in bone defect repairing for many years. Although these materials have more or fewer shortcomings, they are still the cornerstone of our development of a new generation of degradable materials. With the rapid development of modern science and technology, in the twenty-first century, more and more kinds of new biodegradable materials emerge in endlessly, such as new intelligent micro-nano materials and cell-based products. At the same time, there are many new fabrication technologies of improving biodegradable materials, such as modular fabrication, 3D and 4D printing, interface reinforcement and nanotechnology. This review will introduce various kinds of biodegradable materials commonly used in bone defect repairing, especially the newly emerging materials and their fabrication technology in recent years, and look forward to the future research direction, hoping to provide researchers in the field with some inspiration and reference.

Abstract:Degradable metallic biomaterials represent a new concept of bioactive biomaterials used for implants with temporary function. They should support the tissue healing process for a certain period and should progressively degrade thereafter. Degradable metallic materials could potentially replace the corrosion-resistant metals currently used for orthopaedic, cardiovascular, and paediatric implants. The interest in the study of degradable metallic biomaterials has dramatically increased in the last decade. This article reviews the current achievements in the design of biodegradable iron-based materials for orthopaedic load-bearing applications. It introduces a broad overview of the different alloying elements, coating materials, and processing methods used to improve the corrosion behaviour, mechanical properties, and biocompatibility of iron implant materials for temporary hard tissue scaffolds. An emphasis is set on Mn and Zn as the most promising alloying elements for Fe as well as on innovative calcium phosphate-based ceramic and polymeric coatings. In addition, the novel iron–ceramic composite biomaterials for orthopaedic implants are mentioned. Finally, recent challenges and future development direction for iron-based materials are proposed.

Abstract:Recently, zinc and its alloys have been proposed as promising candidates for biodegradable metals (BMs), owning to their preferable corrosion behavior and acceptable biocompatibility in cardiovascular, bone and gastrointestinal environments, together with Mg-based and Fe-based BMs. However, there is the desire for surface treatment for Zn-based BMs to better control their biodegradation behavior. Firstly, the implantation of some Zn-based BMs in cardiovascular environment exhibited intimal activation with mild inflammation. Secondly, for orthopedic applications, the biodegradation rates of Zn-based BMs are relatively slow, resulting in a long-term retention after fulfilling their mission. Meanwhile, excessive Zn2+ release during degradation will cause in vitro cytotoxicity and in vivo delayed osseointegration. Abstract:Repair of large bone defects caused by trauma or disease poses significant clinical challenges. Extensive research has focused on metallic materials for bone repair because of their favorable mechanical properties, biocompatibility, and manufacturing processes. Traditional metallic materials, such as stainless steel and titanium alloys, are widely used in clinics. Biodegradable metallic materials, such as iron, magnesium, and zinc alloys, are promising candidates for bone repair because of their ability to degrade over time. Emerging metallic materials, such as porous tantalum and bismuth alloys, have gained attention as bone implants owing to their bone affinity and multifunctionality. However, these metallic materials encounter many practical difficulties that require urgent improvement. In this article, we systematically reviewed and analyzed the metallic materials used for bone repair, providing a comprehensive overview of their morphology, mechanical properties, biocompatibility, and in vivo implantation. Furthermore, we summarized the strategies and efforts made to address the short‐comings of metallic materials.
