Over the past few decades, the efficient synthesis of axially chiral biaryls has been a research topic of great interest. This feature article will provide an overview of recent advances in the synthesis of these chiral skeletons via transition metal-catalysed asymmetric C–H functionalization.

Abstract:We report here a Cu-catalyzed enantioselective acyloxylation of cyclic diaryliodonium salts. With readily available cyclic diaryliodonium salts and ubiquitous aliphatic or (hetero)aromatic carboxylic acids as the starting materials, various axially chiral acyloxylated 2-iodobiaryls were prepared in excellent yield and with excellent enantioselectivity (mostly 99% yield and 99% ee). Density functional theory calculations were conducted to reveal the stereo- and regioselectivities. This simple reaction protocol can be employed for the late-stage modification of some drug molecules. Finally, by diversity-oriented transformations, these acyloxylated 2-iodobiaryl products can be easily transformed into diverse valuable functionalized biaryls that could be used as chiral ligands or functional materials.

Abstract:We describe herein an intriguing method for the synthesis of biaryl aldehydes bearing both axial and central chirality through a desymmetric [3 + 2] cycloaddition reaction of activated isocyanides with prochiral biaryl dialdehydes under silver catalysis. This protocol features excellent enantioselectivity, 100% atom economy, good functional group compatibility, and operational simplicity.

In this feature article, recent progress made in this field will be overviewed. Attention will be given to the background, substrate scopes, applications, advances and mechanistic investigations of various asymmetric sequential double hydrofunctionalizations of alkynes.

Abstract:Here, we report a chiral copper(II)-bisoxazoline-catalyzed enantioselective ring opening of cyclic diaryliodonium salts with heteroaryl thiols. The readily available 2-mercaptoben-zoxazole and 2-mercaptobenzothiazole derivatives reacted efficiently with cyclic diaryliodonium salts and afforded various axially chiral biaryls bearing iodine and sulfur functional groups in excellent yields and enantioselectivities. The products were further transformed into diverse enantiopure alkyl biaryl sulfides, which can be employed as chiral ligands.

Abstract:The synthesis of axially chiral carbonyl compounds has attracted considerable interest in organic synthesis due to their prevalence in natural products and pharmaceuticals, as well as their utility in material science and catalysis. Transition‐metal‐catalyzed approaches have become powerful tools for their construction, offering high efficiency, selectivity, and versatility among the various methods developed. This concept aims to provide a comprehensive overview of recent advances in the transition‐metal‐catalyzed asymmetric synthesis of axially chiral carbonyl compounds, integrating scattered work with different catalytic systems. This feature concept is divided into four types of reactions based on the strategies employed: cross‐coupling reactions, cycloaddition reactions, desymmetrization reactions, and other C‒H activation reactions.

Abstract:Axially chiral tetrasubstituted alkenes are of increasing value and interest in chemistry‐related areas. However, their catalytic asymmetric synthesis remains elusive, due to the high steric repulsion and relatively low conformational stability. Herein, we disclose the straightforward construction of atropisomeric tetrasubstituted alkenes via an effective enantiocontrol of vinyl cations. This copper‐catalyzed enantioselective C(sp2)–H functionalization of sterically hindered (hetero)arenes with vinyl cations enables the efficient and atom‐economical preparation of axially chiral acyclic tetrasubstituted styrenes and pyrrolyl ethylenes with high atroposelectivities. Importantly, this reaction represents the first example for the assembly of axially chiral alkenes through vinyl cation approach. Computational mechanistic studies reveal the reaction mechanism, origin of regioselectivity, Z/E selectivity and enantioselectivity. The synthetic utility has been demonstrated by diverse product derivatizations, chiral organocatalyst synthesis, as well as further applications in asymmetric catalysis.

Abstract:Axially chiral styrenes, a type of atropisomer analogous to biaryls, have attracted great interests because of their unique presence in natural products and asymmetric catalysis. Since 2016, a number of methodologies have been devloped for the atroposelective construction of these chiral skeletons, involving both transition metal catalysis and organocatalysis. In this feature article, we aim to provide a comprehensive understanding of recent advances in the asymmetric synthesis of axially chiral styrenes catalyzed by transition metals, integrating scattered work with different catalytic systems together. This article is cataloged into five sections according to the strategies, including asymmetric coupling, enantioselective C–H activation, central-to-axial chirality transfer, asymmetric alkyne functionalization, and atroposelective [2+2+2] cycloaddition.

Abstract:In recent years, asymmetric catalysis of ynamides has attracted much attention, but these reactions mostly constructed central chirality, except for a few examples on the synthesis of axially chiral compounds which exclusively relied on noble-metal catalysis. Herein, a facile access to axially chiral N-heterocycles enabled by chiral Bronsted acid-catalyzed 5-endo-dig cyclization of ynamides is disclosed, which represents the first metal-free protocol for the construction of axially chiral compounds from ynamides. This method allows the practical and atom-economical synthesis of valuable N-arylindoles in excellent yields with generally excellent enantioselectivities. Moreover, organocatalysts and ligands based on such axially chiral N-arylindole skeletons are demonstrated to be applicable to asymmetric catalysis.

Abstract:Catalytic enantioselective preparation of axially chiral molecules has gained considerable interest over the past decades, due to their numerous applications in bioactive molecules, natural products, pharmaceuticals, materials, ligands, and catalysts. Compared with the well-established synthetic approaches for six-membered axially chiral skeletons, methodologies directed towards five-membered axially chiral compounds are relatively rare. Among these, axially chiral arylpyrroles are especially important structural motifs with wide utility, and the atroposelective synthesis of them is highly desirable. In recent years, novel strategies have been developed based on transition-metal catalysis and organocatalysis. This review summarizes the recent achievements in atroposelective preparation of arylpyrroles, by emphasizing the synthetic methods for each axially chiral framework, reaction mechanisms, and applications.

Abstract:Direct C–H functionalization has recently emerged as one of the most efficient strategies to access structurally complex molecules from readily accessible feedstocks in an atom- and step-economic manner. In particular, enantioselective C–H activation has garnered increasing attention by enabling chemists to efficiently assemble valuable chiral compounds by asymmetrically manipulating C–H bonds into useful functionalities. Apart from the extensively studied C–C bond formation, very few endeavors have been focused on the C–X formation analogs. Motivated by the utility of the latter approach in constructing academically and industrially important heteroatom-containing chiral compounds, we provide herein an overview on C–X forming asymmetric C–H activation reactions proceeding through C–H metalation. The advancements are organized according to the employed catalytic systems, which include Pd(II) catalysis, group-9 CpxM(III) catalysis, monovalent group-9 metal catalysis, and multi-boryl/silyl Ir(III) catalysis, with emphasis on the design philosophy, mechanism, and mode of enantiocontrol.

Abstract:A novel method by a one-step introduction of axial chirality and sterically hindered group has been developed for facile synthesis of axially chiral styrene-type carboxylic acids. With the palladium-catalyzed C-H arylation and olefination of readily available cinnamic acid established, this transformation demonstrated excellent yield, excellent stereocontrol (up to 99% yield and 99% ee), and broad substrate scope under mild conditions. The axially chiral styrene-type carboxylic acids produced have been successfully applied to Cp*Co-III-catalyzed asymmetric C-H activation reactions, indicating their potential as chiral ligands or catalysts in asymmetric synthesis.

Abstract:Atropisomeric compounds have been discovered in pharmaceutical and material sciences, and their enantioselective syntheses have gained tremendous attention. Among strategies for catalytic atroposelective synthesis, desymmetrization provides robust and straightforward approaches to axially chiral biaryls. Due to the relative ease of substrate design compared to other atroposelective strategies, desymmetrization has emerged as a pivotal stage for converting fascinating chemistry into its atroposelective counterpart, despite challenges such as the formation of achiral products by subsequent catalytic reactions and the long distance between the reaction site and the stereogenic axis. This review offers a comprehensive overview of recent advancements in atroposelective desymmetrization using chiral organo‐ and metal catalysts, addressing challenges and solutions, and aims to provide insights into future developments in this field.

Abstract:Atropisomers featuring restricted rotation around a single bond have emerged as important structural motifs due to their prevalence in drug discovery, materials science and catalyst design. In recent years, the catalytic asymmetric synthesis of atropisomers has hold significant fascination from chemists. While traditional six-membered axially chiral skeletons are commonly studied, methodologies directed towards less abundant five-membered heterobiaryls are relatively rare. In particular, axially chiral azole-based skeletons are essential components of five-membered heterocyclic framework and have received considerable interest. This is the focus of this review, which summarizes the recent achievements in atroposelective preparation of azole derivatives, by emphasizing mechanistic insights and synthetic applications. We hope that this review will encourage chemists worldwide to promote a continued development of the field in the future.
