Abstract:We describe herein an intriguing method for the synthesis of biaryl aldehydes bearing both axial and central chirality through a desymmetric [3 + 2] cycloaddition reaction of activated isocyanides with prochiral biaryl dialdehydes under silver catalysis. This protocol features excellent enantioselectivity, 100% atom economy, good functional group compatibility, and operational simplicity.

Abstract:Herein, we report an unprecedented strategy for the simultaneous construction of a remote C-N stereogenic axis and three contiguous stereogenic carbon centers via silver-catalyzed desymmetrizative [3 + 2] cycloaddition of prochiral N-aryl maleimides with activated isocyanides. This method features operational simplicity, wide substrate scope, high efficiency, and good to excellent stereoselectivity. Notably, it represents the first example of catalytic enantioselective synthesis of C-N atropisomers with the use of activated isocyanides.

Abstract:We report here a Cu-catalyzed enantioselective acyloxylation of cyclic diaryliodonium salts. With readily available cyclic diaryliodonium salts and ubiquitous aliphatic or (hetero)aromatic carboxylic acids as the starting materials, various axially chiral acyloxylated 2-iodobiaryls were prepared in excellent yield and with excellent enantioselectivity (mostly 99% yield and 99% ee). Density functional theory calculations were conducted to reveal the stereo- and regioselectivities. This simple reaction protocol can be employed for the late-stage modification of some drug molecules. Finally, by diversity-oriented transformations, these acyloxylated 2-iodobiaryl products can be easily transformed into diverse valuable functionalized biaryls that could be used as chiral ligands or functional materials.

Abstract:The synthesis of axially chiral carbonyl compounds has attracted considerable interest in organic synthesis due to their prevalence in natural products and pharmaceuticals, as well as their utility in material science and catalysis. Transition‐metal‐catalyzed approaches have become powerful tools for their construction, offering high efficiency, selectivity, and versatility among the various methods developed. This concept aims to provide a comprehensive overview of recent advances in the transition‐metal‐catalyzed asymmetric synthesis of axially chiral carbonyl compounds, integrating scattered work with different catalytic systems. This feature concept is divided into four types of reactions based on the strategies employed: cross‐coupling reactions, cycloaddition reactions, desymmetrization reactions, and other C‒H activation reactions.

Abstract:By using diazonaphthoquinones and anilines as key reagents and through a point-to-axis chiral transfer strategy, the atroposelective synthesis via asymmetric C(sp2 )-H bond insertion reaction of arenes has been realized under rhodium catalysis, providing the resulting biaryl atropisomers in moderate to excellent yields with good enantiomeric ratios (up to 99:1). Further elaboration indicates this type of axially biaryl scaffold may have promising potentials in developing novel chiral ligands.

Abstract:C-N axially chiral skeletons are ubiquitous in bioactive natural products, pharmaceuticals, and chiral ligands. However, their atroposelective synthesis remains a formidable challenge because of their innate low configurational stability compared with that of well-developed C-C atropisomers. Herein, we report a general and efficient method for accessing C-N atropisomers through an axial-to-axial chirality transfer strategy based on palladium/chiral norbornene cooperative catalysis. The obtained C-N axial chirality originates from the preformed transient C-C axial chirality with high fidelity. A variety of C-N axially chiral phenanthridinones are obtained in excellent enantioselectivities (44 examples, up to > 99% ee). This method can be applied for the construction of two stereogenic axes via double atroposelective C-H arylation or further transformation of the products via axial-to-axial diastereoinduction. Additionally, the reaction mechanism and the chirality transfer process are elucidated by density functional theory calculations.

Abstract:Axially chiral tetrasubstituted alkenes are of increasing value and interest in chemistry‐related areas. However, their catalytic asymmetric synthesis remains elusive, due to the high steric repulsion and relatively low conformational stability. Herein, we disclose the straightforward construction of atropisomeric tetrasubstituted alkenes via an effective enantiocontrol of vinyl cations. This copper‐catalyzed enantioselective C(sp2)–H functionalization of sterically hindered (hetero)arenes with vinyl cations enables the efficient and atom‐economical preparation of axially chiral acyclic tetrasubstituted styrenes and pyrrolyl ethylenes with high atroposelectivities. Importantly, this reaction represents the first example for the assembly of axially chiral alkenes through vinyl cation approach. Computational mechanistic studies reveal the reaction mechanism, origin of regioselectivity, Z/E selectivity and enantioselectivity. The synthetic utility has been demonstrated by diverse product derivatizations, chiral organocatalyst synthesis, as well as further applications in asymmetric catalysis.

Abstract:Asymmetric de novo synthesis of benzocyclobutenes (BCBs) via catalytic intermolecular reaction is highly desired for efficient access to this important class of compounds, yet such a strategy remains unmet challenge. Here, we report a Pd/Pyrox-catalyzed asymmetric [2+2] annulation between arylboronic acids and functionalized alkenes, providing an unprecedented efficient protocol to access various enantio-enriched BCBs in a modular and versatile manner under mild conditions. A broad substrate scope with excellent enantioselectivity has been achieved under the current protocol. The isolation and characterization of the key chiral palladacycle intermediate, together with DFT calculations, provides strong evidence for the catalytic pathway including an enantiodetermining arylpalladation step.

Abstract:The asymmetric cross-coupling reaction is developed as a straightforward strategy toward 1,1-diaryl alkanes, which are a key skeleton in a series of natural products and bioactive molecules in recent years. Here we report an enantioselective benzylic C(sp(3))-H bond arylation via photoredox/nickel dual catalysis. Sterically hindered chiral biimidazoline ligands are designed for this asymmetric cross-coupling reaction. Readily available alkyl benzenes and aryl bromides with various functional groups tolerance can be easily and directly transferred to useful chiral 1,1-diaryl alkanes including pharmaceutical intermediates and bioactive molecules. This reaction proceeds smoothly under mild conditions without the use of external redox reagents.

Abstract:Axially chiral molecular scaffolds are widely found in pharmaceutical molecules, functionalized materials, and chiral ligands. The synthesis of these compounds has garnered considerable interest from both academia and industry. The construction of such molecules, enabled by transition metal catalysis and organocatalysis under thermodynamic conditions, has been extensively studied and well-reviewed. In recent years, photoinduced enantioselective reactions have emerged as powerful methods for the catalytic construction of axial chirality. In this review, we provide an overview of various synthetic strategies for the photoinduced construction of axial chirality, with a specific focus on reaction design and catalytic mechanisms. Additionally, we discuss the limitations of current methods and highlight future directions in this field.

Abstract:Due to the significance of chiral benzoheterocycles, the synthesis of these skeletons draws considerable attention from chemists and remarkable progress has been made in the last few decades. However, most of methods employ the noble transition metals as the catalysts. Herein, this review mainly focuses on the facile assembly of enantio-enriched benzoheterocylces via earth-abundant metal catalysis. To date, numerous methods have been well-developed, such as the Ullmann reaction, hydroarylation, borylation or cycloetherification of alkenes, O-H insertion of α-diazo esters, tandem arylation of heteroatom-linked olefins, kinetic resolution, hydrofunctionalization of dihydroquinolines, and other kinds of methods, which are discussed in detail in this review. Furthermore, we also discuss the existed limitations of the previously reported methods and the perspective in this field.

Abstract:Abstract The simultaneous construction of two different chiralities via a simple operation poses considerable challenge. Herein a cationic gold-catalyzed asymmetric hydroarylation of ortho-alkynylaryl ferrocenes derivatives is developed, which enable the simultaneous construction of axial and planar chirality. The here identified TY-Phos derived gold complex is responsible for the high yield, good diastereoselectivity (>20:1 dr), high enantioselectivities (up to 99% ee) and mild conditions. The catalyst system also shows potential application in the synthesis of chiral biaryl compounds. The cause of high enantioselectivity of this hydroarylation is investigated with density functional theory caculation.

Abstract:Axially chiral compounds have widespread applications in many fields. While significant progress has been made for the asymmetric synthesis of atropisomers consisting of C-C bonds, an efficient approach for the construction of C-B atropisomers is still lacking. Herein, we report a practical method for atroposelective synthesis of axially chiral arylboron compounds through Rh-catalyzed [2 + 2 + 2] cycloaddition. The reaction exhibits a broad substrate scope and good functional group tolerance, thus providing an efficient protocol to access a variety of C-B atropisomers in high yields and good enantiocontrol. The utility of these C-B atropisomers has been adequately demonstrated by versatile transformations into highly valuable compounds such as chiral phosphine ligands and fluorescent molecules. Mechanistic experiments and DFT calculations have provided insights into the reaction mechanism as well as the origin of enantiocontrol.

Abstract:Catalytic dynamic kinetic asymmetric transformation (DyKAT) provides a powerful tool to access chiral stereoisomers from racemic substrates. Such transformation has been widely employed on the construction of central chirality, however, the application in axial chirality remains underexplored because its equilibrium of substrate enantiomers is limited to five-membered metalacyclic intermediate. Here we report a tetracoordinate boron-directed dynamic kinetic asymmetric cross-coupling of racemic, configurationally stable 3-bromo-2,1-azaborines with boronic acid derivatives. A series of challenging C-B axially chiral compounds were prepared with generally good to excellent enantioselectivities. Moreover, this transformation can also be extended to prepare atropisomers bearing adjacent C-B and C-C diaxes with excellent diastereo- and enantio-control. The key to the success relies on the rational design of a reversible tetracoordinate boron intermediate, which is supported by theoretical calculations that dramatically reduces the rotational barrier of the original C-B axis and achieves the goal of DyKAT.

Abstract:Atropisomers possessing additional stereogenic centres have found wide applications in various fields, however, the development of catalytic asymmetric reactions towards their preparation through a single transformation has been much less studied. In particular, no example of catalytic diastereodivergent and enantioselective simultaneous construction of axial and point chirality has been disclosed and stands for an unmet synthetic challenge. Herein, we report the realization of such a goal for the first time. An unprecedented silver-catalyzed desymmetric [3+2] cycloaddition reaction of N-quinazolinone maleimides with isocyanoacetates was developed, producing a series of bicyclic N-heterocycles bearing a remote chiral N-N axis and three contiguous stereogenic carbon centres in high efficiencies with high-to-excellent stereoselectivities. Most strikingly, an interesting diastereodivergent synthesis was also achieved allowing facile access to both endo- and exo-cycloadducts. DFT calculations revealed that the stereoselectivity is mainly controlled by the coordination type of ligand, ligand-substrate non-bond interaction, and distortion of the annulation transition state.
