Abstract:Rh(III)-catalyzed C-H functionalization of 7-arylpyrazolo[1,5-a]pyrimidines was developed wherein the pyrazolo[1,5-a]pyrimidine moiety is reported for the first time to direct the C-H bond activation. Various 7-arylpyrazolo[1,5-a]pyrimidines underwent smooth C-H amidation with alkyl-, aryl-, and heteroaryl-substituted dioxazolones to afford the products in moderate to good yields. Mechanistic studies suggest that a six-membered rhodacycle intermediate involving N1 might play a key role in the regioselective catalytic cycle.

Abstract:Abstract A Rh-catalyzed electrochemical vinylic C–H annulation of acrylamides with alkynes has been developed in an undivided cell, affording cyclic products in good to excellent yield. Divergent syntheses of α-pyridones and cyclic imidates are accomplished by employing N-phenyl acrylamides and N-tosyl acrylamides as substrates, respectively. Additionally, excellent regioselectivities are achieved when using unsymmetrical alkynes. This electrochemical process is environmentally benign compared to traditional transition metal-catalyzed C–H annulations because it avoids the use of stoichiometric metal oxidants. DFT calculations elucidated the reaction mechanism and origins of substituent-controlled chemoselectivity. The sequential C–H activation and alkyne insertion under rhodium catalysis leads to the seven-membered ring vinyl-rhodium intermediate. This intermediate undergoes either the classic neutral concerted reductive elimination to produce α-pyridones, or the ionic stepwise pathway to produce cyclic imidates.

Abstract:A Cp*Rh-III-catalyzed annulative carbooxygenation of challenging 1,1-disubstituted alkenes triggered by C-H activation of N-aryloxyacetamides has been established, which affords 2,3-dihydrobenzofuran derivatives with a quaternary carbon center in good to excellent yields under mild redox-neutral conditions. An amide group on the alkenes is essential for the process, and may inhibit the beta-H elimination from C(sp3)-Rh species by saturating the rhodium center through coordination. Furthermore, mechanistic insights obtained from control experiments suggest a mechanism involving a Rh-III-Rh-V-Rh-III catalytic cycle.

Abstract:Cp*Rh(III)‐catalyzed regioselective C−H annulation and alkenylation of 2‐pyridones with terminal alkynes have been developed. The cationic Cp*Rh(III) catalytic system containing FeCl3 additive enables annulation of 1‐(2‐pyridyl)‐2‐pyridones with terminal alkynes, providing efficient access to 5,7‐diarylated 2‐quinolinones. The reaction pathway can be switched to alkenylation with [Cp*RhCl2]2 as the catalyst, NaOAc as the additive and HOAc as the solvent, affording C6‐alkenylated 1‐(2‐pyridyl)‐2‐pyridones in high yields. These protocols accommodate a wide range of substrates with good functional group compatibility.

Abstract:In this paper, rhodium‐catalyzed sequential twofold ortho ‐C‐H functionalization of N ‐phenylbenzimidamide with internal alkyne is studied. The double C‐H activations proved viable in a one‐pot fashion with the assistance of C=N and C‐N, providing a serial of benzimidazoisoquinolines with high levels of positional selectivity control. The operationally simple transformation showed high functional group compatibility and featured with cleavage of the C‐H bonds located on different moiety of the N ‐phenylbenzimidamide substrates. Detailed mechanistic studies provided strong support for C−H bond cleavage on N ‐phenyl ring is preferential than C‐H bond on C‐phenyl ring. As a multifunctional catalytic platform, the rhodium catalyst conducted two independent and compatible catalytic cycles in one pot.

Abstract:We disclose a mediated electrochemical [2+2+2] annulation of alkynes with nitriles, forming substituted pyridines in a single step from low-cost, readily available starting materials. The combination of electrochemistry and a triarylamine redox mediator obviates the requirements of transition metals and additional oxidants. Besides the formation of diarylpyridine moieties via the homocoupling of two identical alkynes, the heterocoupling of two different alkynes depending on their electronic nature is possible, highlighting the unprecedented control of chemoselectivity in this catalytic [2+2+2] process. Mechanistic investigations like cyclic voltammetry and crossover experiments combined with DFT calculations indicate the initial oxidation of an alkyne as the key step leading to the formation of a vinyl radical cation intermediate. The utilization of continuous flow technology proved instrumental for an efficient process scale-up. The utility of the products is exemplified by the synthesis of π-extended molecules, being relevant for material or drug synthesis.
