Abstract:Integrating synthetic biology into wearables could expand opportunities for noninvasive monitoring of physiological status, disease states and exposure to pathogens or toxins. However, the operation of synthetic circuits generally requires the presence of living, engineered bacteria, which has limited their application in wearables. Here we report lightweight, flexible substrates and textiles functionalized with freeze-dried, cell-free synthetic circuits, including CRISPR-based tools, that detect metabolites, chemicals and pathogen nucleic acid signatures. The wearable devices are activated upon rehydration from aqueous exposure events and report the presence of specific molecular targets by colorimetric changes or via an optical fiber network that detects fluorescent and luminescent outputs. The detection limits for nucleic acids rival current laboratory methods such as quantitative PCR. We demonstrate the development of a face mask with a lyophilized CRISPR sensor for wearable, noninvasive detection of SARS-CoV-2 at room temperature within 90 min, requiring no user intervention other than the press of a button.

Abstract:Fiber‐based biosensors enable a new approach in analytical diagnostic devices. The majority of textile‐based biosensors, however, rely on colorimetric detection. Here a woven biosensor that integrates microfluidics structures in combination with an electroanalytical readout based on a thiol‐self‐assembled monolayer (SAM) for Nucleic Acid Amplification Testing, NAATs is shown. Two types of fiber‐based electrodes are systematically characterized: pure gold microwires (bond wire) and off‐the‐shelf plasma gold‐coated polyester multifilament threads to evaluate their potential to form SAMs on their surface and their electrochemical performance in woven textile. A woven electrochemical DNA (E‐DNA) sensor using a SAM‐based stem‐loop probe‐modified gold microwire is fabricated. These sensors can specifically detect unpurified, isothermally amplified genomic DNA of Staphylococcus epidermidis (10 copies/µL) by recombinase polymerase amplification (RPA). This work demonstrates that textile‐based biosensors have the potential for integrating and being employed as automated, sample‐to‐answer analytical devices for point‐of‐care (POC) diagnostics.

Abstract:While the printed circuit board (PCB) has been widely considered as the building block of integrated electronics, the world is switching to pursue new ways of merging integrated electronic circuits with textiles to create flexible and wearable devices. Herein, as an alternative for PCB, we described a non-printed integrated-circuit textile (NIT) for biomedical and theranostic application via a weaving method. All the devices are built as fibers or interlaced nodes and woven into a deformable textile integrated circuit. Built on an electrochemical gating principle, the fiber-woven-type transistors exhibit superior bending or stretching robustness, and were woven as a textile logical computing module to distinguish different emergencies. A fiber-type sweat sensor was woven with strain and light sensors fibers for simultaneously monitoring body health and the environment. With a photo-rechargeable energy textile based on a detailed power consumption analysis, the woven circuit textile is completely self-powered and capable of both wireless biomedical monitoring and early warning. The NIT could be used as a 24/7 private AI "nurse" for routine healthcare, diabetes monitoring, or emergencies such as hypoglycemia, metabolic alkalosis, and even COVID-19 patient care, a potential future on-body AI hardware and possibly a forerunner to fabric-like computers.
An Electrochemiluminescence Sensing Platform Combining Entropy-Driven Amplification Strategy and Inverted DNA Tetrahedral Nanomachines for Detecting SARS-CoV-2 Specific Gene

Abstract:In this work, we propose a Ti3C2Tx-based electrochemiluminescence (ECL) sensing platform, combined with a programmable entropy-driven cycling strategy and inverted DNA tetrahedral nanomachines to detect the SARS-CoV-2 RdRp gene. The use of Ti3C2Tx in the ECL sensing platform provides unique advantages that improve the assay's sensitivity. Firstly, the SARS-CoV-2 RdRp gene triggers an entropy-driven cyclic amplification reaction to obtain a large number of Output strands. Subsequently, the Output strand reacts with an inverted DNA tetrahedral swing arm strand to start the self-driven process. Finally, the ferrocene (Fc)-labeled hairpin DNA strand of the inverted DNA tetrahedron is severed to achieve the signal change. Under optimal conditions, the ECL platform integrated with entropy-driven cycle amplification reaction and inverted DNA tetrahedral nano-machines obtained a wide range of 100 aM to 10 nM and a limit of detection (LOD) of 48.3 aM for the SARS-CoV-2 RdRp gene. Moreover, the recovery rates of 97% to 103.3% and 98.34% to 103% were obtained in real environments and human pharyngeal swabs, respectively. This ECL sensing platform, which combines an entropy-driven cycle amplification reaction and inverted DNA tetrahedral nanomachines, opens up new opportunities for the detection of other substances.

Abstract:The need for more efficient health services and the trend of a healthy lifestyle pushes the development of smart textiles. Since textiles have always been an object of everyday life, smart textiles promise, and extensive user acceptance. Thereby, the manufacture of electrical components based on textile materials is of great interest for applications as biosensors. Organic electrochemical transistors (OECTs) are often used as biosensors for the detection of saline content, adrenaline, glucose, etc., in diverse body fluids. Textile-based OECTs are mostly prepared by combining a liquid electrolyte solution with two separate electro-active yarns that must be precisely arranged in a textile structure. Herein, on the other hand, a biosensor based on a textile single-component organic electrochemical transistor with a hardened electrolyte was developed by common textile technologies such as impregnation and laminating. Its working principle was demonstrated by showing that the herein-produced transistor functions similarly to a switch or an amplifier and that it is able to detect ionic analytes of a saline solution. These findings support the idea of using this new device layout of textile-based OECTs as biosensors in near-body applications, though future work must be carried out to ensure reproducibility and selectivity, and to achieve an increased level of textile integration.

Abstract:Unpredictable pandemics are likely to pose a significant global threat in the future, and biologically protective textiles will play critical roles in controlling the spread of pathogens during outbreaks. Herein, we present a novel metal–organic framework (MOF) composed of repeating units of a Cu(II)/(L-Cys)2 complex formed through coordination bonds between Cu(II) and L-Cys, while being interconnected by ionic bonds involving Cu(II) and the carboxylate group of L-Cys. After covalently embedding the MOF nanofibers onto cotton fiber surfaces, the resulting fabrics exhibit remarkable virucidal and antibacterial capabilities. Abstract:Textile-based microfluidic biosensors represent an innovative fusion of various multidisciplinary fields, including bioelectronics, material sciences, and microfluidics. Their potential in biomedicine is significant as they leverage textiles to achieve high demands of biocompatibility with the human body and conform to the irregular surfaces of the body. In the field of microfluidics, fabric coated with hydrophobic materials serves as channels through which liquids are transferred in precise amounts to the sensing element, which in this case is a biosensor. This paper presents a condensed overview of the current developments in textile-based microfluidics and biosensors in biomedical applications over the past 20 years (2005-2024). A literature search was performed using the Scopus database. The fabrication techniques and materials used are discussed in this paper, as these will be key in various modifications and advancements in textile-based microfluidics. Furthermore, we also address the gaps in the application of textile-based microfluidic analytical devices in biomedicine and discuss the potential solutions. Advances in textile-based microfluidics are enabled by various printing and fabric manufacturing techniques, such as screen printing, embroidery, and weaving. Integration of these devices into everyday clothing holds promise for future vital sign monitoring, such as glucose, albumin, lactate, and ion levels, as well as early detection of hereditary diseases through gene detection. Although most testing currently takes place in a laboratory or controlled environment, this field is rapidly evolving and pushing the boundaries of biomedicine, improving the quality of human life.

Abstract:Rapid detection of bacterial pathogens and toxic contaminants in the environment is essential for providing real-time results to mitigate potential safety hazards. This study prepared a wearable fabric sensing material based on fluorescent nanomaterials. The nano silver/europium complex is assembled layer by layer with sodium alginate to obtain nanoparticle aggregates with strong fluorescence, and then these are covalently bonded onto the cotton fabric surface to obtain the fluorescence cotton fabric (FCF). Via fluorescence quenching, FCF can sensitively sense Escherichia coli and Staphylococcus aureus and show good antibacterial activity. Besides, the selected transition metal ions can be detected qualitatively through the functional relationship between the fluorescence intensity and metal ion dose. The viability test of NIH3T3 cells showed that FCF had good biocompatibility. This fluorescent fabric sensing material has the characteristics of being wearable, antibacterial, and biocompatible. It realizes the real-time detection of bacteria and toxic metal ions and has great application prospects in environmental and biological monitoring.

Abstract:Infectious diseases severely threaten public health and global biosafety. In addition to transmission through the air, pathogenic microorganisms have also been detected in environmental liquid samples, such as sewage water. Conventional biochemical detection methodologies are time-consuming and cost-ineffective, and their detection limits hinder early diagnosis. In the present study, ultrafine plasmonic fiber probes with a diameter of 125 μm are fabricated for clustered regularly interspaced short palindromic repeats/CRISPR-associated protein (CRISPR/Cas)-12a-mediated sensing of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Single-stranded DNA exposed on the fiber surface is trans-cleaved by the Cas12a enzyme to release gold nanoparticles that are immobilized onto the fiber surface, causing a sharp reduction in the surface plasmon resonance (SPR) wavelength. The proposed fiber probe is virus-specific with the limit of detection of ~2,300 copies/ml, and genomic copy numbers can be reflected as shifts in wavelengths. A total of 21 sewage water samples have been examined, and the data obtained are consistent with those of quantitative polymerase chain reaction (qPCR). In addition, the Omicron variant and its mutation sites have been fast detected using S gene-specific Cas12a. This study provides an accurate and convenient approach for the real-time surveillance of microbial contamination in sewage water.

Abstract:The rampaging COVID-19 needs bioassaying methods of low cost and high robustness for those living in the poorly developed regions. Here, we propose such a method that does not need expensive and complicated equipment. Only a set of hand-held small devices is sufficient. A section along an optic fiber cable is stripped, so that laser light travelling through it will leak outside, while biosensing process taken place on this stripped section can form a new cladding layer of hydrogel, restoring the laser output of the fiber. A short peptide probe immobilized on the stripped section of the fiber can covalently capture a biomarker protein of SARS-Cov-2 from the serum sample. Through the cross-linking of the target protein with the interfering proteins in the serum sample, a hydrogel is covalently immobilized around the stripped section, highly resistant to detergent rinsing that is indispensable for removing nonspecific interference from the clinical sample. Using this "covalent biosensing" strategy, only one peptide probe is sufficient to simultaneously achieve ultrahigh affinity toward the biomarker protein of SARS-Cov-2 and effective signal amplification.This article has not yet been cited by other publications.

Abstract:<p>Accessible and adaptable nucleic acid diagnostics remains a critical challenge in managing the evolving COVID-19 pandemic. Here, an integrated molecular nanotechnology that enables direct and programmable detection of SARS-CoV-2 RNA targets in native patient specimens is reported. Termed synergistic coupling of responsive equilibrium in enzymatic network (SCREEN), the technology leverages tunable, catalytic molecular nanostructures to establish an interconnected, collaborative architecture. SCREEN mimics the extraordinary organization and functionality of cellular signaling cascades. Through programmable enzyme–DNA nanostructures, SCREEN activates upon interaction with different RNA targets to initiate multi-enzyme catalysis; through system-wide favorable equilibrium shifting, SCREEN directly transduces a single target binding into an amplified electrical signal. To establish collaborative equilibrium coupling in the architecture, a computational model that simulates all reactions to predict overall performance and optimize assay configuration is developed. The developed platform achieves direct and sensitive RNA detection (approaching single-copy detection), fast response (assay reaction is completed within 30 min at room temperature), and robust programmability (across different genetic loci of SARS-CoV-2). When clinically evaluated, the technology demonstrates robust and direct detection in clinical swab lysates to accurately diagnose COVID-19 patients.</p>

Abstract:A novel smart biochromic textile sensor was developed by immobilizing anthocyanin extract into electrospun cellulose acetate nanofibers to detect bacteria for numerous potential uses, such as healthcare monitoring. Red-cabbage was employed to extract anthocyanin, which was then applied to cellulose acetate nanofibers treated with potassium aluminum sulfate as a mordant. Thus, nanoparticles (NPs) of mordant/anthocyanin (65-115 nm) were generated in situ on the surface of cellulose acetate nanofibrous film. The pH of a growing bacterial culture medium is known to change when bacteria multiply. The absorbance spectra revealed a bluish shift from 595 nm (purple) to 448 nm (green) during the growth of Gram-negative bacteria (E. coli) owing to the discharge of total volatile basic amines as secretion metabolites. On the other hand, the absorption spectra of a growing bacterial culture containing Gram-positive bacteria (L. acidophilus) showed a blue shift from 595 nm (purplish) to 478 nm (pink) as a result of releasing lactic acid as a secretion metabolite. Both absorbance spectra and CIE Lab parameters were used to determine the color shifts. Various analytical techniques were utilized to study the morphology of the anthocyanin-encapsulated electrospun cellulose nanofibers. The cytotoxic effects of the colored cellulose acetate nanofibers were tested.

Abstract:In recent years, the application of cell-free protein synthesis systems in biosensing has been developing rapidly. Cell-free synthetic biology, with its advantages of high biosafety, fast material transport, and high sensitivity, has overcome many defects of cell-based biosensors and provided an abiotic substitute for biosensors. In addition, the application of freeze-drying technology has improved the stability of such systems, making it possible to realize point-of-care application of field detection and broadening the application prospects of cell-free biosensors. However, despite these advancements, challenges such as the risk of sample interference due to the lack of physical barriers, maintenance of activity during storage, and poor robustness still need to be addressed before the full potential of cell-free biosensors can be realized on a larger scale. In this review, current strategies and research results for improving the performance of cell-free biosensors are summarized, including a comprehensive discussion of the existing challenges, future trends, and potential investments needed for improvement.

Abstract:With the development of flexible advanced materials and microfluidic technology, wearable biosensors provide a new strategy for the continuous monitoring of health. In this study, a novel bandage-like wearable flexible microfluidic recombinase polymerase amplification (RPA) sensor was constructed for the rapid and visual detection of nucleic acids. This wearable sensor is triggered by human body heat (30°C-37 °C) and allows for visual nucleic acid (a conserved nucleic acid fragments of zika virus) detection within 10 min. The sensor displays good sensitivity and selectivity, with a detection limit of 10 copies/μL. The wearable sensor has exhibited well-defined accuracy when applied to testing clinical serum samples. In addition, the wearable RPA sensor was proved to be feasible by human trials under different daily activities. This wearable sensor of nucleic acids will probably be of great significance in the field of online pathogen detection for wounds, for tumour biomarker diagnosis, and for the detection of epidermal cell molecular lesions.

Abstract:The timely control of infectious diseases can prevent the spread of infections and mitigate the significant socio-economic damage witnessed during recent pandemics. Diagnostic methods play a significant role in detecting highly contagious agents, such as viruses, to prevent further transmission. The emergence of advanced point-of-care techniques offers several advantages over conventional approaches for detecting infectious agents. These techniques are highly sensitive, rapid, can be miniaturized, and are cost-effective. Recently, MXene-based 2D nanocomposites have proven beneficial for fabricating electrochemical biosensors due to their suitable electrical, optical, and mechanical properties. This article covers electrochemical biosensors based on MXene nanocomposite for the detection of viruses, along with the associated challenges and future possibilities. Additionally, we highlight various conventional techniques for the detection of infectious agents, discussing their pros and cons. We delve into the challenges faced during the fabrication of MXene-based biosensors and explore future endeavors. It is anticipated that the information presented in this work will pave the way for the development of Point-of-Care (POC) devices capable of sensitive and selective virus detection, enhancing preparedness for ongoing and future pandemics.

Abstract:This study demonstrates a novel methodology for developing a textile-based adsorption sensor via mixed solvent dyeing with aggregation-induced emission (AIE) dyes on recycled fabrics. AIE dyes were incorporated into the fabrics using a mixed solvent dyeing method with a co-solvent mixture of H2O and organic solvents. This method imparted unique fluorescence properties to fabrics, altering fluorescence intensity or wavelength based on whether the AIE dye molecules were in an isolated or aggregated state on the fabrics. The precise control of the H2O fraction to organic solvent during dyeing was crucial for influencing fluorescence intensity and sensing characteristics. These dyed fabrics exhibited reactive thermochromic and vaporchromic properties, with changes in fluorescence intensity corresponding to variations in temperature and exposure to volatile organic solvents (VOCs). Their superior characteristics, including a repetitive fluorescence switching property and resistance to photo-bleaching, enhance their practicality across various applications. Consequently, the smart fabrics dyed with AIE dye not only find applications in clothing and fashion design but demonstrate versatility in various fields, extending to sensing temperature, humidity, and hazardous chemicals.

Smart Textiles with Self-Disinfection and Photothermochromic Effects

Abstract:Self-disinfecting textile materials employing combined photodynamic/photothermal effects enable the prevention of microbial infections, a property that has great potential in healthcare applications. However, smart textiles with stimulus responses to ambient temperature are marvelous materials for enhancing their photothermal applications with additional functions. It is still challenging to realize vivid and contrasting color changes as temperature indicators. Herein, through the in situ growth of PCN-224 metal-organic frameworks (MOFs), the electrospraying of a Ti3C2 MXene colloid, and the screen printing of a thermochromic dye, a smart photothermochromic self-disinfecting textile has been fabricated. An antibacterial inactivation study revealed 99.9999% inactivation toward gram-negative (Escherichia coli ATCC 8099) and gram-positive (Staphylococcus aureus ATCC 6538) bacteria in 30 min. A mechanism study revealed that light-driven singlet oxygen and heat are the main reasons for bacterial inactivation. Interestingly, the fabrics presented photothermal effects not only under a handheld 780 nm NIR laser but also under visible Xe lamp (λ ≥ 420 nm) illumination. The color of the fabrics (S-CF@PCN0.08) changed completely from dark green to dark red when the temperature exceeded 45 °C under Xe lamp illumination. Furthermore, the photothermochromic effect occurred in just 1 s under a 780 nm laser. Taken together, this smart photothermochromic self-disinfecting textile permits a new way to feedback the timely signal of temperature by color change and provides novel insights into the development of self-disinfecting textiles.
A smartphone-based visual biosensor for CRISPR-Cas powered SARS-CoV-2 diagnostics

Abstract:The pandemic of coronavirus disease 2019 (COVID-19) resulted from novel severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has become a worldwide concern. It is imperative to develop rapid, sensitive, and specific biosensing methods. Herein, we developed a CRISPR-Cas12a powered visual biosensor with a smartphone readout for ultrasensitive and selective detection of SARS-CoV-2. Simply, the SARS-CoV-2 derived nucleic acids triggered CRISPR-Cas12a based indiscriminate degradation of a single-stranded DNA that was supposed to link two gold nanoparticles, inducing the dis-aggregation of gold nanoparticles and thus generating observable color changes. This change can be readily distinguished by naked eyes as well as a smartphone with a Color Picker App. The proposed biosensor was successfully applied to detect SARS-CoV-2 gene in synthetic vectors, transcribed RNA and SARS-CoV-2 pseudoviruses. It rendered "single copy resolution" as evidenced by the 1 copy/μL limit of detection of pseudoviruses with no cross-reactivity. When the developed biosensor was challenged with SARS-CoV-2 clinical bio-samples, it provided 100% agreement (both positive and negative) with qPCR results. The sample-to-result time was roughly 90 min. Our work provides a novel and robust technology for ultrasensitive detection of SARS-CoV-2 that could be used clinically.
