Abstract:MicroRNAs (miRNAs) are a family of endogenous, small (approximately 22 nucleotides in length), noncoding, functional RNAs. With the development of molecular biology, the research of miRNA biological function has attracted significant interest, as abnormal miRNA expression is identified to contribute to serious human diseases such as cancers. Traditional methods for miRNA detection do not meet current demands. In particular, nanomaterial-based methods, nucleic acid amplification-based methods such as rolling circle amplification (RCA), loop-mediated isothermal amplification (LAMP), strand-displacement amplification (SDA) and some enzyme-free amplifications have been employed widely for the highly sensitive detection of miRNA. MiRNA functional research and clinical diagnostics have been accelerated by these new techniques. Herein, we summarize and discuss the recent progress in the development of miRNA detection methods and new applications. This review will provide guidelines for the development of follow-up miRNA detection methods with high sensitivity and specificity, and applicability to disease diagnosis and therapy.

Abstract:MicroRNAs(miRNAs) are a kind of small non-coding RNA molecules with about 22 nt long,which negatively regulate specific target genes by mRNA degradation or translational repression.Over 10,000 miRNAs have been discovered in animals by different detect methods.Northern blotting,as the standard of miRNA detection,was substituted by various new methods due to its low sensitivity.Many miRNA detection methods are based on hybridization,including bioluminescent,electrochemical,surface-enhanced Raman spectroscopy,BRET-based,Fluorescence correlation spectroscopy,and bioluminescent protein reassembly;while some methods rely on profiling techniques,such as microarray,deep sequencing,bead-based assay,miRAGE,and nanostring technology.In this review,we summarized the current methods for identifying miRNAs,discussed the advantages and the disadvantages of the methods and prospected the miRNA detection trends.

Abstract:MicroRNAs (miRNAs) are small, highly conserved, non-coding RNAs that regulate gene expression of target mRNAs through cleavage or translational inhibition. Computer-based approaches for miRNA gene identification are being considered as indispensable in miRNAs research. Similarly, experimental approaches for detection of miRNAs are crucial to the testing and validating of computational algorithms. The detection of miRNAs in tissues or cells can supply valuable information for investigating the biological function of these molecules. Selective and highly sensitive detection methods will pave the way for extended understanding of miRNA function within organisms. In this review, we summarize the various computational methods for identification of miRNAs as well as the methodologies that have been developed to detection miRNAs.

Abstract:MicroRNAs (miRNAs) are a class of endogenous non-coding RNAs that play an important role in the control of developmental process of different cells by negative regulation of protein-coding gene expression. Analyzing miRNA expression in tissues or cells can supply valuable information for investigating the biological function of these molecules. Recently, researchers had proposed a number of approaches for analyzing the differences of miRNA expression among different physiological or pathological conditions, and found that aberrant expression of miRNA was related to cancers, neurological disorders and heart diseases, etc. This review focuses on newly developed strategies for miRNA quantification, and elucidates in detail the probe-hybridization based methods including Northern blotting, microarray, gold nanoparticle labelling, and splinted ligation with radioactive labels. The amplification-based methods including quantitative PCR, rolling cycle amplification, invader assay, and the next generation sequencing methods were also discussed. The advantages and disadvantages of these methods were compared.

Abstract:MicroRNAs (miRNA) are a kind of endogenous non-coding small molecule RNA, which are widely found in plants, animals, and viruses. miRNA can regulate the post-transcriptional cleavage of target message RNA or inhibit its translation process, and plays an important regulatory role in gene expression. Further studies have shown that miRNAs also play an important role in tumorigenesis and host-pathogen interactions. Given the important contribution of miRNAs in gene regulation and biological functions, rapid and accurate detection of miRNAs has become increasingly significant. Lateral flow assay is a detection method derived to achieve rapid detection in the field. It overcomes the problems of cumbersome, time-consuming, and high cost in traditional detection methods, and provides a good platform for accurate, sensitive, and immediate detection of target analytes. With the advantages convenient, cost-saving, and repaid result readout, the lateral flow test strips are now a recognized point-of-care testing (POCT) device and has been widely used in medical diagnosis, agriculture, food, and environmental safety. For the first time, we summarized the research progress of miRNA detection based on lateral flow methods, focusing on the strategies used in the lateral flow assays for miRNA detection and introducing some main experimental operation details, as well as providing an outlook on the future development direction of this field, aiming to provide a general guidance for colleagues preparing to conduct related research.

Abstract:MicroRNA( miRNA ) is a class of endogenous noncoding RNA molecule with the length of about 22 nucleotides. MicroRNA may regulate more than 50% of human protein. coding genes, and plays important roles in regulating cell differentiation and other biological processes. The abnormal expression of miRNA is closely related to the occurrence of many diseases including cancers. Therefore, miRNA may function as a potential biomarker for molecular diagnostics. The sensitive detection of miRNA is essential to the early clinical diagnosis and the development of anticancer drugs. Because of the short size, low abundance in total RNA samples and the homologous similarities in miRNA family, the ultrasensitive detection of miRNA remains a great challenge. Recently, many new methods have been developed to improve the detection sensitivity. This review focuses on the advance in miRNA detection methods including colorimetric assay, fluorescent assay, chemiluminescent assay, surface enhanced Raman scattering, single molecules detection and electrochemical detection. In addition, we give a new insight into future development of miRNA assay.

Abstract:With the recent development of investigating biological functions of microRNA( miRNA),the importance of miRNA in the biological processes including developmental biology, metabolic regulation, disease progression and treatment has been well recognized. Due to its the instability, low level of expression and minor difference in sequence,more sophisticated analytical methods for rapid and easy quantitative detection of miRNA with strong specificity and high sensitivity play an important role in the study of the biological functions of miRNA. This review analyzes and compares recent quantitative methods to detect miRNA, with an attempt to provide a foundation for choosing an appropriate method to quantitatively analyze miRNA.

Abstract:MicroRNAs (miRNAs) are a series of promising molecules that could regulate gene expression, while its expression level and type are strictly related to the early diagnosis, targeted therapy, and prognosis of diseases. Improved detection accuracy of miRNAs would lead to unprecedented progress in early treatment. Numerous methods to improve sensitivity have been proposed and confirmed valuable in miRNAs detection recently. However, the research which especially targets at improving detection specificity remains rare. This Feature gives a retrospective summary of the most common detection methods including Northern blot, reverse transcription quantitative polymerase chain reaction, next-generation sequencing, and microarray. Common methods play an important role in many aspects because of maturity and stability. However, the problem of specificity still exists. Then, this Feature summarizes breakthroughs in traditional techniques which are closely related to progressive methods to improve specificity. In addition, considerable studies focusing on improving accuracy are described in detail. Most of them involve the reasonable combination of common methods, and the strategies that contribute to improving accuracy are classified. This Feature aims to illustrate the importance of detection accuracy and explore how to improve specificity on detecting miRNAs, especially homologous families.

Abstract:Drug resistance is a significant factor that hinders the success of cancer chemotherapy. The widely recognized mechanisms of drug resistance include changes to cell proliferation, cycle/apoptosis, drug metabolism/transport, DNA damage and the epithelial to mesenchymal transition. MicroRNAs (miRNAs), short non-coding RNAs with lengths of approximately 19-25 nucleotides, are related to cancer drug resistance, which is regulated by the aforementioned mechanisms. Based on the importance of miRNAs in regulating drug resistance, it is also necessary to take appropriate miRNA detection methods into consideration. To date, a number of advanced miRNA detection methods with high specificity and sensitivity have been developed, such as isothermal amplification-based methods, nanomaterial-based methods, chromatography-based methods, mass spectrometry-based methods and so on. Herein, biogenesis of miRNAs, the relationship between miRNAs and cancer drug resistance, and miRNA detection methods are introduced and discussed to facilitate the development of non-invasive diagnosis and inhibition of cancer drug resistance.

Abstract:microRNAs(miRNAs),a class of small,endogenous and non-coding RNAs,play critical regulatory functions in many cell processes such as cell development,fat metabolism,organs generation,cell differentiation and apoptosis,etc. Researchers have discovered that the dysregulation of miRNA expression is closely associated with various diseases including human cancers and diabetes. Therefore,the prompt and accurate detection of miRNA expression is the prerequisite for the further researches on it. Many assays for miRNA detection have recently been developed,but the detection techniques for polymorphism and mutation of miRNA are not so mature. Furthermore,the sensitivity for detection technology of microscale miRNA in biological samples also needs further development and breakthroughs. The new developed miRNA detection techniques with high selectivity and high sensitivity are reviewed in this paper,and then this may provide important assistance for further study on the related detection technologies.

Abstract:microRNAs (miRNAs) are an extensive class of endogenous, non-conding, short (21 similar to 25 nt) RNA molecules directly involved in signal transduction and play an important role in gene expression regulation. The detection of miRNA expression is a very important first step in miRNA exploration. With the discovery of new plant microRNAs and the functional research of miRNA thoroughly, new demand has been put forward to the plant miRNA detecting methods. Recently, plant miRNA expression detecting methods is mostly based on the hybridization and PCR techniques. In order to provide some suggestions for the plant miRNA detection research, this review summarizes the progress of several kinds of plant miRNA expression detecting methods, discusses the advantages and the disadvantages of several kinds of methods and prospects the plant miRNA detecting methods.

Abstract:MicroRNAs (miRNAs) are small, non-coding, endogenous RNA molecules that play important roles in a variety of normal and diseased biological processes by post-transcriptionally regulating the expression of target genes. They can bind to target messenger RNA (mRNA) transcripts of protein-coding genes and negatively control their translation or cause mRNA degradation. miRNAs have been found to actively regulate a variety of cellular processes, including cell proliferation, death, and metabolism. Therefore, their study is crucial for the better understanding of cellular functions in eukaryotes. To better understand the mechanisms of miRNA: mRNA interaction and their cellular functions, it is important to identify the miRNA targets accurately. In this paper, we provide a brief review for the advances in the animal miRNA target prediction methods and available resources to facilitate further study of miRNAs and their functions.

Abstract:Over the decades, the biological role of microRNAs (miRNAs) in the post-transcriptional regulation of gene expression has been discovered in many cancer types, thus initiating the tremendous expectation of their application as biomarkers in the diagnosis, prognosis, and treatment of cancer. Hence, the development of efficient miRNA detection methods in vitro is in high demand. Extensive efforts have been made based on the intrinsic properties of miRNAs, such as low expression levels, high sequence homology, and short length, to develop novel in vitro miRNA detection methods with high accuracy, low cost, practicality, and multiplexity at point-of-care settings. In this review, we mainly summarized the newly developed in vitro miRNA detection methods classified by three key elements, including biological recognition elements, additional micro-/nano-materials and signal transduction/readout elements, their current challenges and further applications are also discussed. Abstract:MicroRNAs (miRNAs) are small RNAs of 18-25 nucleotides in length that are widely distributed in eukaryotes and are produced by DNA transcription. As regulators of post-transcriptional gene expression, it plays an important role in the physiological processes of cells. As some miRNAs in the body are abnormally expressed at different and earlier stages of diseases, this phenomenon suggests that accurate, sensitive and specifical detection of them can be helpful for early and differential diagnosis. To expound the technological progress of miRNA detection, we reviewed all the related articles in PubMed database published before May 6, 2019, with the following keywords: "miRNA", "real-time fluorescent quantitative PCR", "electrochemical detection", "next-generation sequencing", "digital PCR technology". Original articles and reviews on the topics were selected. The present methods established for quantitative detection of miRNAs mainly relies on various probe design and labeling techniques, and the improvement of the sensitivity and specificity of detection is often through combination of microarray chips, real-time fluorescent quantitative PCR, high-throughput sequencing and other techniques. This paper combines the existing microRNA detection methods to provide a reference for researchers to choose the best detection method.

Abstract:MicroRNAs (miRNAs), as the small, non-coding, evolutionary conserved, and post-transcriptional gene regulators of the genome, have been highly associated with various diseases such as cancers, viral infections, and cardiovascular diseases. Several techniques have been established to detect miRNAs, including northern blotting, real-time polymerase chain reaction (RT-PCR), and fluorescent microarray platform. However, it remains a significant challenge to develop sensitive, accurate, rapid, and cost-effective methods to detect miRNAs due to their short size, high similarity, and low abundance. The electrochemical biosensors exhibit tremendous potential in miRNA detection because they satisfy feature integration, portability, mass production, short response time, and minimal sample consumption. This article reviewed the working principles and signal amplification strategies of electrochemical DNA biosensors summarized the recent improvements. With the development of DNA nanotechnology, nanomaterials and biotechnology, electrochemical DNA biosensors of high sensitivity and specificity for microRNA detection will shortly be commercially accessible.
