Abstract:Recent advances in the sulfonylation of alkenes via the insertion of sulfur dioxide are summarized. The sulfur dioxide surrogate of DABCO<bold></bold>(SO2)(2) or inorganic sulfites is used in the transformation through a radical process. Two strategies for the sulfonylation of alkenes with the insertion of sulfur dioxide have been developed. In most cases, the vicinal difunctionalization of alkenes is initiated by the arylsulfonyl radical generated in situ. Although the sulfonylation of alkenes with the insertion of sulfur dioxide is efficient, the asymmetric versions are still challenging and less to be explored. It is expected that the asymmetric vicinal difunctionalization of alkenes with the insertion of sulfur dioxide will be developed rapidly in the near future.

Abstract:Organosulfur functionalities are ubiquitous in nature, pharmaceuticals, agrochemicals, materials and flavourants. Historically, these moieties were introduced almost exclusively using ionic chemistry; however, radical-based methods for the installation of sulfur-based functional groups have recently come to the fore. These radical methods have enabled their late-stage introduction into complex molecules, avoiding the need to preserve labile organosulfur moieties through multistep synthetic sequences. Here, we discuss homolytic C–S bond-forming processes, with a particular emphasis on radical substitution approaches to sulfide, disulfide and sulfinyl products, and the use of sulfur dioxide and its surrogates to build sulfonyl products. We also highlight the mechanistic considerations that we hope will guide further development of radical-based strategies compatible with the various organosulfur moieties that feature in modern chemistry.

Abstract:Sulfur oxides are widespread and gaining increasing attention in pharmaceutical and biotechnology discoveries owing to the heightened activities despite sulfur dioxide (SO2) is one of the most important gaseous air pollutants. Considerable research efforts have been devoted to deal with sulfur oxides in a greener and sustainable way. Herein, we report a visible-light-promoted four-component radical-polar crossover tandem cyclization involved twofold insertion of SO2, and the formation of S(IV) and S(IV) centers in one procedure. Various types of functionalized β-sulfonyl sultines were provided from the easy-to-access C(sp2)-H and C(sp3)-H building blocks with high group compatibility under mild and operationally simple conditions. Gram-scale preparation and late-stage transformations demonstrate opportunities to rapidly construct molecular complexity. Mechanistic studies supported a SO2 bi-inserted radical-polar crossover cyclization relay sequence, which allows for a more efficient utilization of SO2 and is also more environmentally friendly.

Abstract:Recent advances in the sulfonylation of C-H bonds with the insertion of sulfur dioxide are summarized. Usually, a sulfur dioxide surrogate, DABCO·(SO2)2, is used, which avoids the utilization of gaseous sulfur dioxide in the transformation. Inorganic sulfites such as sodium metabisulfite are also effective in C(sp3)-H bond sulfonylation. C-H bond sulfonylation under transition metal catalysis or through a radical process has been developed. In some cases, the sulfonylation can be performed under catalyst- and additive-free conditions, or can be facilitated by visible light irradiation. The efficiency is also investigated by merging the radical process and metal catalysis.

This unique capability has garnered significant interest across various domains. Many strategies have been developed for the synthesis of sulfonyl fluoride compounds. This review focuses on the latest advancements in synthesizing these compounds using the radical sulfur dioxide insertion and fluorination (RSIF) strategy, underscoring the versatility and efficacy of the RSIF strategy in constructing sulfonyl fluoride compounds.

Abstract:From the viewpoint of synthetic accessibility and functional group compatibility, photoredox-catalyzed sulfur dioxide insertion strategy enables in situ generation of functionalized sulfonyl radicals from easily accessible starting materials under mild conditions, thereby conferring broader application potential. Here we present two complementary photoinduced sulfur dioxide insertion systems to trigger radical asymmetric Truce-Smiles rearrangements for preparing a variety of chiral sulfones that bear a quaternary carbon stereocenter. This protocol features broad substrate scope and excellent stereospecificity. Aside from scalability, the introduction of a quaternary carbon stereocenter at position β to bioactive molecule-derived sulfones further demonstrates the practicality and potential of this methodology.

Abstract:The difunctionalization of alkenes based on the insertion of sulfur dioxide has been considered to be one of the most straightforward and powerful strategies for the construction of sulfones. Numerous strategies for activated alkenes have been developed in this area. However, progress in the SO2 insertion-mediated difunctionalization of unactivated alkenes, which encounters higher reactivity and shorter survival times of the alkyl radical intermediate, remains elusive and unresolved. Here, we report a strategy to access sulfonated quinazolinones from readily accessible aryl diazonium salts and Na2S2O5 via a catalyst-free photocatalytic system. The process is pro-posed to involve a photoinduced sulfur dioxide insertion/sulfonyl radical addition to an unactivated C=C double bond/Minisci-type cycliza-tion sequence. This approach features high functional group tolerance, broad substrate scope, and synthetic simplicity, and thus offers opportunities to rapidly build up sulfone-containing quinazolinones.

Abstract:β-Chiral sulfones are structural motifs widely found in natural products and bioactive molecules, while also serving as important intermediates for the synthesis of valuable chiral scaffolds. In contrast to the rapid growth of sulfur dioxide insertion chemistry over the last decade, enantioselective catalytic variants for accessing β-chiral sulfones, especially those bearing a quaternary carbon stereocenter, remain rare. Herein, we report an enantioselective copper/bisoxazoline catalyzed oxysulfonylation of alkenes to yield isoxazolinyl-containing β-chiral sulfones bearing quaternary carbon stereocenters with moderate to excellent yields and up to 96:4 er. The advantage and irreplaceability of this sulfur dioxide insertion catalytic system have been demonstrated by achieving transformations clearly unavailable with the previously reported sulfonyl chloride system. Additionally, the recoverability of the new-developed bisoxazoline ligand L9 and its reusability without any erosion of the catalytic activity and enantioselectivity further demonstrate the usefulness of the protocol.

Abstract:Herein, we report a new approach for the light‐mediated generation of sulfamoyl radicals using sulfur dioxide as key building block and the direct application of these radicals in the synthesis of sulfonamides. In the presence of different photoredox catalysts, sulfamoyl radicals can be generated directly from SO2 or the SO2 surrogate DABSO (1,4‐diazabicyclo[2.2.2]octane·bis (sulfur dioxide) adduct) and N‐aminopyridinium salts as nitrogen radical precursors. Trapping of the in situ generated sulfamoyl radicals with selected electron‐rich olefins affords different sulfonamides in up to 86% yield in a three‐component procedure. This transformation provides an attractive and complementary approach for the in situ generation of sulfamoyl radicals as synthetic intermediates for the assembly of the sulfonamide functionality, a motif in active pharmaceutical ingredients.

Abstract:Sodium sulfinates have been widely utilized as sulfonyl radical precursors for preparing a diverse array of value-added sulfur-containing compounds (sulfones, sulfonamides, sulfonates, thiosulfonates, etc.) through S-C, S-N, S-O and S-S bonds formation reactions. Organic electrosynthesis has become an attractive alternative to conventional methods for redox reactions because it utilizes electric current instead of chemical redox agents. As such, the electrochemical generation of sulfonyl radicals from sodium sulfinates and their applications in organic electrosynthesis have attracted much attention. In this review, the recent advances in the electrochemical sulfonylation of organic compounds involving sodium sulfinates as sulfonyl radical precursors since 2015 were reviewed, along with related reaction mechanisms.

Abstract:Katritzky salts are used as the alkyl radical precursors with the insertion of sulfur dioxide under photoredox catalysis. This transformation first enables direct generation of various alkylsulfonyl radicals by photoinduced single electron reduction, leading to diverse dialkyl sulfones in good to excellent yields. A radical pathway is proposed under visible-light induced conditions with the insertion of sulfur dioxide.

Abstract:Sulfur-containing heterocycles, where the S(R) moiety is located within the cyclic structure, have found tremendous applications in the fields of chemical, pharmaceutical, and materials sciences due to their unique chemical, biological and pharmaceutical activities. Recent years have witnessed increasing interest in sulfur-containing heterocycles, and new methods for their synthesis have been reported by adopting modern methodologies and technologies through insertion of sulfur dioxide. Abstract:A three-component reaction of 2-ethynylbenzenesulfonamides, DABCO-bis(sulfur dioxide), and aryldiazonium tetrafluoroborates affords sulfonated benzosultams in moderate to good yields. This reaction proceeds through a tandem radical process. DABCO acts as a carrier for single electron transfer, as well as a base to promote the C–N bond formation.

Abstract:Sulfur(VI) fluoride exchange (SuFEx) chemistry that relies on the unique reactivity–stability balance of high valent organosulfur has emerged as a promising topic for the next-generation click reaction. Sulfonyl fluorides are the most widely used connective hubs of the SuFEx click reaction with widespread applications in the fields of chemical biology, drug discovery, and materials science. Compared with current methods, direct fluorosulfonylation with fluorosulfonyl radicals has emerged as a concise and efficient approach for the production of sulfonyl fluorides. The highly active SO 2 F radical was an unstable and inaccessible precursor until it was observed in the decomposition of fluorosulfonyl azide, which inspired efforts towards the production of fluorosulfonyl radical precursors for direct radical fluorosulfonylation processes. Abstract:Sulfonyl fluorides are key components in the fields of chemical biology, materials science and drug discovery. In this line, the highly active SO2F radical has been employed for the construction of sulfonyl fluorides, but the utilization of gaseous ClSO2F as radical precursor is limited due to the tedious and hazardous preparation. Meanwhile, the synthesis of sulfonyl fluorides from inert SO2F2 gas through a fluorosulfonyl radical (·SO2F) process has met with inevitable difficulties due to the high homolytic bond dissociation energy of the S(VI)-F bond. Here we report a radical fluorosulfonylation strategy for the stereoselective synthesis of alkenyl sulfonyl fluorides and functional alkyl sulfonyl fluorides with an air-stable crystalline benzimidazolium fluorosulfonate cationic salt reagent. This bench-stable redox-active reagent offers a useful and operational protocol for the radical fluorosulfonylation of unsaturated hydrocarbons with good yield and high stereoselectivity, which can be further transformed into valuable functional SO2F moieties.

Abstract:A palladium-catalyzed direct sulfonylation of C-H bonds with the insertion of sulfur dioxide under mild conditions is reported. The sulfonylative couplings with the insertion of sulfur dioxide into C-H bonds are effective, and two classes of sulfonylative products are formed in moderate to good yields by the combination of radical chemistry and palladium-catalyzed C-H activation.

Abstract:An organocatalytic enantioselective radical reaction of potassium alkyltrifluoroborates, DABCO·(SO2)2 and α,β-unsaturated carbonyl compounds under photoinduced conditions is developed, which provides an efficient pathway for the synthesis of chiral β-sulfonyl carbonyl compounds in good yields with excellent enantioselectivity (up to 96% ee). Aside from α,β-unsaturated carbonyl compounds with auxiliary groups, common chalcone substrates are also well compatible with this organocatalytic system. This method proceeds through an organocatalytic enantioselective radical sulfonylation under photoinduced conditions, and represents a rare example of asymmetric transformation involving sulfur dioxide insertion.
