Abstract:Solid-state nanopores have been widely employed in sensing applications from Coulter counters to DNA sequencing devices. The analytical signal in such experiments is the change in ionic current flowing through the orifice caused by the large molecule or nanoparticle translocation through the pore. Conceptually similar nanopipette-based sensors can offer several advantages including the ease of fabrication and small physical size essential for local measurements and experiments in small spaces. This paper describes the first evaluation of nanopipettes with well characterized geometry for resistive-pulse sensing of Au nanoparticles (AuNP), nanoparticles coated with an allergen epitope peptide layer, and AuNP-peptide particles with bound antipeanut antibodies (IgY) on the peptide layer. The label-free signal produced by IgY-conjugated particles was strikingly different from those obtained with other analytes, thus suggesting the possibility of selective and sensitive resistive-pulse sensing of antibodies.

Abstract:Developing a rapid and ultrasensitive immunosensor is significant for detecting allergenic contamination in food safety fields. This study reports a microfluidic biosensing device consisting of two top-bottom opposite electrode chips and a channel slab for constructing peanut allergen-Ara h 1 immunosensors. Under optimal fluidic manipulation (OFM), transporting an 80 μL aliquot of Ara h 1-containing phosphate buffer solution spiked by 2.5 mM Fe(CN) 6 3−/4− mediator with a 50 μL/min flow rate for 2.5 min, the device can obtain better immunoreaction. The electron transfer resistance (R et ) of the label-free immunosensors was analyzed by electrochemical impedance spectroscopy (EIS) to quantify Ara h 1 immunoreaction. After performing OFM once, the linear range and the limit of detection were 1 pg/mL− 10 ng/mL and 3.9 fg/mL, respectively. Furthermore, the R et response of the immunosensors increased with OFM times. The sensing device can detect the Ara h 1 concentration with good recovery (95 −103%) for the diluted supernatant extracted from standard peanut butter. The microfluidic device integrating EIS-based immunosensors exhibits rapid and ultrasensitive sensing properties with promising potential for label-free detection of allergens in food products.

Abstract:The rise in antibiotic-resistant pathogens, highly infectious viruses, and chronic diseases has prompted the search for rapid and versatile medical tests that can be performed by the patient. Field-effect transistor (FET)-based electronic biosensing platforms are particularly attractive due to their sensitivity, fast turn-around time, potential for parallel detection of multiple pathogens, and compatibility with semiconductor manufacturing. However, an unmet critical need is a scalable, site-selective multiplexed biofunctionalization method with nanoscale precision for immobilizing different types of pathogen-specific bioreceptors on individual FETs, preventing parallel detection of multiple targets. Here, we propose a paradigm shift in FET biofunctionalization using thermal scanning probe lithography (tSPL) with a thermochemically sensitive polymer. This polymer can be spin-coated on fully-fabricated FET chips, making this approach applicable to any FET sensor material and technology. Crucially, we demonstrate the spatially selective multiplexed functionalization capability of this method by immobilizing different types of bioreceptors at prescribed locations on a chip with sub-20 nm resolution, paving the way for massively parallel FET detection of multiple pathogens. Antibody- and aptamer-modified graphene FET sensors are then realized, achieving ultra-sensitive detection of a minimum measured concentrations of 3 aM of SARS-CoV-2 spike proteins and 10 human SARS-CoV-2 infectious live virus particles per ml, and selectivity against human influenza A (H1N1) live virus.

Abstract:We report the fabrication of a highly sensitive field-effect transistor (FET) biosensor using thermally-reduced graphene oxide (TRGO) sheets functionalized with gold nanoparticle (NP)-antibody conjugates. Probe antibody was labeled on the surface of TRGO sheets through Au NPs and electrical detection of protein binding (Immunoglobulin G/IgG and anti-Immunoglobulin G/anti-IgG) was accomplished by FET and direct current (dc) measurements. The protein binding events induced significant changes in the resistance of the TRGO sheet, which is referred to as the sensor response. The dependence of the sensor response on the TRGO base resistance in the sensor and the antibody areal density on the TRGO sheet was systematically studied, from which a correlation of the sensor response with sensor parameters was found: the sensor response was more significant with larger TRGO base resistance and higher antibody areal density. The detection limit of the novel biosensor was around the 0.2 ng/mL level, which is among the best of reported carbon nanomaterial-based protein sensors and can be further optimized by tuning the sensor structure.

Abstract:Due to the ubiquity and essential of the proteins in all living organisms, the identification and quantification of disease-specific proteins are particularly important. Because the conformational change of aptamer upon its target or probe/target/probe sandwich often is the primary prerequisite for the design of an electrochemical aptameric assay system, it is extremely difficult to construct the electrochemical aptasensor for protein assay because the corresponding aptamers cannot often meet the requirement. To circumvent the obstacles mentioned, an electronic channel switching-based (ECS) aptasensor for ultrasensitive protein detection is developed. The essential achievement made is that an innovative sensing concept is proposed: the hairpin structure of aptamer is designed to pull electroactive species toward electrode surface and makes the surface-immobilized IgE serve as a barrier that separates enzyme from its substrate. It seemingly ensures that the ECS aptasensor exhibits most excellent assay features, such as, a detection limit of 4.44×10(-6)μg mL(-1) (22.7fM, 220zmol in 10-μL sample) (demonstrating a 5 orders of magnitude improvement in detection sensitivity compared with classical electronic aptasensors) and dynamic response range from 4.44×10(-6) to 4.44×10(-1)μg mL(-1). We believe that the described sensing concept here might open a new avenue for the detection of proteins and other biomacromolecules.

Abstract:Allergy is a prevalent disease, and the potential allergic population is expanding with industrialization and changes in people's living standards. Serum immunoglobulin E (IgE) level is one of the critical indicators for determining allergy. Here, we proposed a simple, real-time monitoring, low chip cost, label-free aptamer biosensing strategy based on weak value amplification (WVA) for the quantitative detection of IgE in serum samples, enabling early and accurate diagnosis of allergic or hypersensitive patients. The aptasensor combined an imaging weak measurement system with the high specificity of the aptamer for the marker IgE. By modifying the amino group at the 3-terminal end, the anti-IgE aptamers can attach to a dopamine-modified prism's surface and selectively recognize IgE in human serum. In the presence of IgE, a specific binding reaction occurred, resulting in a change in the refractive index of the reactive region's surface, manifested as a change in the light intensity of the camera acquired experimental images. As the concentration of IgE increased, the relative light intensity advanced sequentially. The WVA-aptasensing strategy achieved a wide detection range of 0.01 ng/mL to 2 mu g/mL in phosphate buffered saline buffer, with the resolution as low as 4.3 pg/mL. IgE testing experiments in human serum have proved the feasibility of our methods in detecting complex samples. In addition, the method specifically recognized IgE without interference from other proteins. We believe that our proposed sensing strategy opens up new possibilities for ultrahigh sensitivity screening of IgE and can be expanded to detecting other biomolecules.

Abstract:It is crucial for early stage medical diagnostics to identify disease biomarkers at ultralow concentrations. A wide range of analytes can be identified using low-dimensional materials to build highly sensitive, targeted, label-free, field-effect transistor (FET) biosensors. Two-dimensional (2D) materials are preferable for high-performance biosensing because of their dramatic change in resistivity upon analyte adsorption or biomarker detection, tunable electronic properties, high surface activities, adequate stability, and layer-dependent semiconducting properties. We give a succinct overview of interesting applications for protein sensing with various architectural styles, such as 2D transition metal dichalcogenides (TMDs)-based FETs that include carbon nanotubes (CNTs), graphene (Gr), reduced graphene oxide (rGr), 2D transition-metal carbides (MXene), and Gr/MXene heterostructures. Because it might enable individuals to perform better, this review will be an important contribution to the field of medical science. These achievements demonstrate point-of-care diagnostics' abilities to detect biomarkers at ultrahigh performance levels. A summary of the present opportunities and challenges appears in the conclusion.
Microfluidic Origami Nano-Aptasensor for Peanut Allergen Ara H1 Detection.

Abstract:In this study, an origami microfluidic electrochemical nano-aptasensor was developed for the rapid detection of the peanut allergen Ara h1. Specifically, the microfluidic aptasensor was fabricated through sequential folding of a piece of chromatography paper substrate patterned with microchannel and screen-printed electrodes. Aptamer-decorated black phosphorus nanosheets (BPNSs) were electrodeposited onto the paper-based electrode surface as sensing probes for enhanced electrochemical detection and high specificity and selectivity. Critical design parameters (the concentration of probe, time for self-assembly of aptamer and reaction time) were investigated to optimize the aptasensor performance. The prepared aptasensor was able to complete detection within 20 min and demonstrated a linear range from 50 similar to 1000 ng/mL with a detection limit of 21.6 ng/mL. The aptasensor was successfully used to detect the Ara h1 spiked cookie dough sample. The proposed method reduces the gap between complex lab testing and food allergen analysis at the point of need.

Abstract:Miniaturized biosensor design is widely preferred for its application in integrated platform to provide rapid and sensitive diagnosis. However, there are only few reports which employ extended gate field effect transistors (EG-FET) for immunosensing application. Herein, we designed EG-FET using four level lithography techniques. Further, the gate layer (Au/Ti) of device has been modified with Tin disulfide nanoflowers (SnS 2 ) and nitrogen doped graphene oxide (N-GO) hybrid composite for enhanced electrical sensing. The as-prepared SnS 2 and N-GO based nanocomposite (SnS 2 /N-GO) was characterized by various techniques. The designed SnS 2 /N-GO based EG-FET was employed as an immunosensor to detect HIgG using anti-HIgG antibody. Under optimum conditions, our miniaturized immunosensor demonstrated the detection of HIgG as low as 4.81 ng/mL. The proposed immunosensor exhibited excellent sensitivity, selectivity and reproducibility towards HIgG detection, thereby suitable for a reliable integrated biosensing platform which can detect protein concentration in real samples for clinical diagnostics.

Abstract:In the present study, a back-gate thin-film transistor (TFT) based on a single-walled carbon nanotubes (SWCNTs)/poly(9,9-dioctylfluorene-co-bithiophene) (F8T2) composite semiconductor is proposed as electronic label-free immunosensor for the detection of the C-reactive protein (CRP). The nano-composite semiconducting ultra-thin film is deposited on the channel area of the TFT and is functionalized with a monoclonal anti-CRP antibody coating acting as biorecognition layer. The immunesensor was then used for the selective detection of the CRP both in phosphate buffered saline (PBS) solution and in human serum samples. The proposed immunosensor exhibits high current (10 - 20 μA) and good field-effect carriers mobility. Furthermore, the CRP could be revealed at a detection limit as low as 1 mg/L and in a wide dynamic range (from 0.4 nM to 2.2 μM).

Abstract:Antibody-antigen interactions are shaped by the solution pH level, ionic strength, and electric fields, if present. In biological field-effect transistors (BioFETs) the interactions take place at the sensing area in which the pH level, the ionic strength and the electric fields are determined by the Poisson-Boltzmann equation and the boundary conditions at the solid-solution interface and the potential applied at the solution electrode. The present study demonstrates how a BioFET solution electrode potential affects the sensing area double layer pH level, ionic strength, and electric fields and in this way shapes the biological interactions at the sensing area. We refer to this as 'active sensing'. To this end, we employ the meta-nano-channel (MNC) BioFET and demonstrate how the solution electrode can determine the antibody-antigen equilibrium constant and allows the control and tuning of the sensing performance in terms of dynamic range and limit-of-detection. In the current work we employ this method to demonstrate specific and label-free sensing of Alpha-Fetoprotein (AFP) molecule from 0.5 μL drops of 1:100 diluted serum. AFP is measured during pregnancy as part of the prenatal screening program for fetal anomalies, chromosomal abnormalities, and abnormal placentation. We demonstrate AFP sensing with a limit-of-detection of 10.5 aM and a dynamic range of 6 orders of magnitude in concentration. Extensive control measurements are reported.

Abstract:Over the past decade, electrical detection of chemical and biological species using novel nanostructure-based devices has attracted significant attention for chemical, genomics, biomedical diagnostics, and drug discovery applications. The use of nanostructured devices in chemical/biological sensors in place of conventional sensing technologies has advantages of high sensitivity, low decreased energy consumption and potentially highly miniaturized integration. Owing to their particular structure, excellent electrical properties and high chemical stability, carbon nanotube and graphene based electrical devices have been widely developed for high performance label-free chemical/biological sensors. Here, we review the latest developments of carbon nanostructure-based transistor sensors in ultrasensitive detection of chemical/biological entities, such as poisonous gases, nucleic acids, proteins and cells.

Abstract:Peanut allergy has garnered worldwide attention due to its high incidence rate and severe symptoms, stimulating the demand for the ultrasensitive detection method of peanut allergen. Herein, we successfully developed a novel electrochemical aptasensor for ultrasensitive detection Ara h1, a major allergenic protein present in peanuts. A conductive nickel atoms Anchored Hydrogen-Bonded Organic Frameworks (PFC-73-Ni) were utilized as excellent electrocatalysts toward hydroquinone (HQ) oxidation to generate a readable current signal. The developed electrochemical aptasensor offers wide linear range (1-120 nM) and low detection limit (0.26 nM) for Ara h1. This method demonstrated a recovery rate ranging from 95.00% to 107.42% in standard addition detection of non-peanut food samples. Additionally, the developed electrochemical method was validated with actual samples and demonstrated good consistency with the results obtained from a commercial ELISA kit. This indicates that the established Ara h1 detection method is a promising tool for peanut allergy prevention.

Abstract:Flexible sensors are essential for advancing implantable and wearable bioelectronics toward monitoring chemical signals within and on the body. Developing biosensors for monitoring multiple neurotransmitters in real time represents a key in vivo application that will increase understanding of information encoded in brain neurochemical fluxes. Here, arrays of devices having multiple In2O3 nanoribbon field-effect transistors (FETs) were fabricated on 1.4-μm-thick polyethylene terephthalate (PET) substrates using shadow mask patterning techniques. Thin PET-FET devices withstood crumpling and bending such that stable transistor performance with high mobility was maintained over >100 bending cycles. Real-time detection of the small-molecule neurotransmitters serotonin and dopamine was achieved by immobilizing recently identified high-affinity nucleic-acid aptamers on individual In2O3 nanoribbon devices. Limits of detection were 10 fM for serotonin and dopamine with detection ranges spanning eight orders of magnitude. Simultaneous sensing of temperature, pH, serotonin, and dopamine enabled integration of physiological and neurochemical data from individual bioelectronic devices.

Abstract:Molecular imprinting is a technique for preparing polymer scaffolds that function as synthetic receptors 1 , 2 , 3 . Imprinted polymers that can selectively bind organic compounds have proven useful in sensor development 2 , 3 , 4 , 5 , 6 , 7 . Although creating synthetic molecular-imprinting polymers that recognize proteins remains challenging 8 , 9 , 10 , 11 , nanodevices and nanomaterials show promise in this area 12 , 13 , 14 . Here, we show that arrays of carbon-nanotube tips with an imprinted non-conducting polymer coating can recognize proteins with subpicogram per litre sensitivity using electrochemical impedance spectroscopy. We have developed molecular-imprinting sensors specific for human ferritin and human papillomavirus derived E7 protein. The molecular-imprinting-based nanosensor can also discriminate between Ca 2+ -induced conformational changes in calmodulin. This ultrasensitive, label-free electrochemical detection of proteins offers an alternative to biosensors based on biomolecule recognition.

Abstract:Low-abundance biomolecule detection is very crucial in many biological and medical applications. In this paper, we present a novel electrolyte-gated graphene field-effect transistor (EGFET) biosensor consisting of acoustic tweezers to increase the sensitivity. The acoustic tweezers are based on a high-frequency bulk acoustic resonator with thousands of MHz, which has excellent ability to concentrate nanoparticles. The operating principle of the acoustic tweezers to concentrate biomolecules is analyzed and verified by experiments. After the actuation of acoustic tweezers for 10 min, the IgG molecules are accumulated onto the graphene. The sensitivities of the EGFET biosensor with accumulation and without accumulation are compared. As a result, the sensitivity of the graphene-based biosensor is remarkably increased using SMR as the biomolecule concentrator. Since the device has advantages such as miniaturized size, low reagent consumption, high sensitivity, and rapid detection, we expect it to be readily applied to many biological and medical applications.

Abstract:The present study focuses on the first electrochemical immunosensor built with polymeric mats enriched with nanomaterials, targeting the dust mite protein, for the detection of allergens in flour sources with important implications in allergic reactions. As a proof-of-concept, we have used the Blo t 5 allergen from Blomia tropicalis (target) and established electrochemical parameters to recognize and detect the specific allergen using a polyclonal immunoglobulin (Ig) Y (probe). Detection was performed in a portable potentiostat (EmStat) using a graphite screen-printed electrode. For that, the nanofibrous mats of poly(lactic acid)/poly(ethylene glycol), and carbon nanotubes were placed in contact with the surface of the electrodes. The functionalization that occurs through the deposition of the polymer was induced through the passage of current. Subsequently, the modification was validated by cyclic voltammetry (CV) readings and ferri/ferrocyanide was used as a redox indicator in CV analyses. The novel immunosensor was able to discriminate between allergen-contaminated and uncontaminated farinaceous samples. The immunosensor displays high sensitivity detecting up to 50 μg/ml in the calibration curve with a linear response between 5 and 500 μg/ml. The new biosensor shows great potential in the detection of farinaceous contaminants in laboratories, food factories and in the field.

Abstract:The ability to sense proteins and protein‐related interactions at the single‐molecule level is becoming of increasing importance to understand biological processes and diseases better. Single‐molecule sensors, such as nanopores have shown substantial promise for the label‐free detection of proteins; however, challenges remain due to the lack of selectivity and the need for relatively high analyte concentrations. An aptamer‐functionalized nanopore extended field‐effect transistor (nexFET) sensor is reported here, where protein transport can be controlled via the gate voltage that in turn improves single‐molecule sensitivity and analyte capture rates. Importantly, these sensors allow for selective detection, based on the choice of aptamer chemistry, and can provide a valuable addition to the existing methods for the analysis of proteins and biomarkers in biological fluids.

Abstract:Reusable point-of-care biosensors offer a cost-effective solution for serial biomarker monitoring, addressing the critical demand for tumour treatments and recurrence diagnosis. However, their realization has been limited by the contradictory requirements of robust reusability and high sensing capability to multiple interactions among transducer surface, sensing probes and target analytes. Here we propose a drug-mediated organic electrochemical transistor as a robust, reusable epidermal growth factor receptor sensor with striking sensitivity and selectivity. By electrostatically adsorbing protonated gefitinib onto poly(3,4-ethylenedioxythiophene):polystyrene sulfonate and leveraging its strong binding to the epidermal growth factor receptor target, the device operates with a unique refresh-in-sensing mechanism. It not only yields an ultralow limit-of-detection concentration down to 5.74 fg ml-1 for epidermal growth factor receptor but, more importantly, also produces an unprecedented regeneration cycle exceeding 200. We further validate the potential of our devices for easy-to-use biomedical applications by creating an 8 × 12 diagnostic drug-mediated organic electrochemical transistor array with excellent uniformity to clinical blood samples.

Abstract:There has been a significant drive to deliver nanotechnological solutions to biosensing, yet there remains an unmet need in the development of biosensors that are affordable, integrated, fast, capable of multiplexed detection, and offer high selectivity for trace analyte detection in biological fluids. Herein, some of these challenges are addressed by designing a new class of nanoscale sensors dubbed nanopore extended field-effect transistor (nexFET) that combine the advantages of nanopore single-molecule sensing, field-effect transistors, and recognition chemistry. We report on a polypyrrole functionalized nexFET, with controllable gate voltage that can be used to switch on/off, and slow down single-molecule DNA transport through a nanopore. This strategy enables higher molecular throughput, enhanced signal-to-noise, and even heightened selectivity via functionalization with an embedded receptor. This is shown for selective sensing of an anti-insulin antibody in the presence of its IgG isotype.
