Abstract:A regio- and stereoselective glycosylation of ribose tetraester with persilylated alloxazine to give either ‚-N1 or ‚-N3 nucleosides is described. The N3 product is potentially of interest as a fluorescent nucleoside and is predicted to have the hydrogen-bonding characteristics of thymidine.

Abstract:An asymmetric aldol strategy has been developed for the synthesis of L-daunosamine and D-ristosamine derivatives starting from noncarbohydrate precursors. Lithium and boron enolate mediated aldol reactions of 12 with O-TBS lactaldehyde gave non-Evans syn and Evans syn aldol products, respectively, with high selectivity. The chemical efficiency of the lithium enolate reactions were higher than the corresponding reactions with the boron enolates. Curtius rearrangement of lactone acids 23 and 26 gave the corresponding N-BOC amino lactones 30 and 32 in 64% and 62%, respectively, with complete retention of configuration. Lactone 30 was converted by a two-step sequence to N-benzoyldaunosamide 40. The overall yield for the amino sugar 40 was 18% over six steps. Similarly, lactone 32 was converted to N-benzoylristosamide 42 with an overall yield of 18% starting from 12.

Abstract:A concise approach for the synthesis of quinoxalin-2-one derivatives through substituted o-phenylenediamines and alpha-ketoester with high regioselectivity is developed. Interestingly, the two regioisomers can be selectively obtained and the regioselectivity can be regulated by acid and base. The typical regioisomer ratio is up to 15:1 in acidic conditions and it can be easily reversed as 1:4 in basic conditions.

Abstract:C-nucleosides, analogues of nucleosides by replacing a C-N glycosidic bond with a C-C bond, are potential anticancer or antiviral agents. Synthesis of C-nucleoside analogues enabled by photoredox-catalyzed radical coupling of glycosyl bromides with nonfunctionalized heteroarenes has been described. This strategy features readily available starting materials, mild conditions, good functional group tolerance, and site- and stereoselectivities (alpha-configuration). A variety of monosaccharides (D-lyxose, D-ribose, L-arabinose, D-glucose, D-mannose, D-glucuronide, and D-fucose), disaccharides (lactose, melibiose, and maltose), and polysaccharide (maltriose) can couple with a series of nonfunctionalized heteroarenes, such as purine, benzothiazole, thiazolopyridine, benzoxazole, benzimidazole, imidazopyridine, and phenanthridine. Biologically important molecules, such as theophylline, famciclovir, ribufuranoside, and adenine, can be glycosylated directly through a C-C bond using this method. Site- and stereoselectivities can be rationalized and predicted by density functional theory (DFT) calculations.

Abstract:Described here is a selective synthesis of 2,3-dihydropyrrole-fused N-glycosides through rhodium-catalyzed denitrogenative transannulation of N-sulfonyl-1,2,3-triazoles with glycals. A series of pyrroline-fused N-glycosides are afforded in moderate to excellent yields with exclusive regioselectivity and stereoselectivity. Functional application of such a resultant product by oxidative addition and epoxidation is also explored. Abstract:The design of novel 4'-thionucleoside analogues bearing a C2' stereogenic all-carbon quaternary center is described. The synthesis involves a highly diastereoselective Mukaiyama aldol reaction, and a diastereoselective radical-based vinyl group transfer to generate the all-carbon stereogenic C2' center, along with different approaches to control the selectivity of the N-glycosidic bond. Intramolecular SN2-like cyclization of a mixture of acyclic thioaminals provided analogues with a pyrimidine nucleobase. A kinetic bias favoring cyclization of the 1',2'-anti thioaminal furnished the desired β-D-4'-thionucleoside analogue in a 7:1 ratio. DFT calculations suggest that this kinetic resolution originates from additional steric clash in the SN2-like transition state for 1',4'-trans isomers, causing a significant decrease in their reaction rate relative to 1',4'-cis counterparts. N-glycosylation of cyclic glycosyl donors with a purine nucleobase enabled the formation of novel 2-chloroadenine 4'-thionucleoside analogues. These proprietary molecules and other derivatives are currently being evaluated both in vitro and in vivo to establish their biological profiles.

Abstract:A systematic study on the enzymatic transglycosylation of 3’‐amino nucleosides has been reported. Using the commercially available 3’‐amino‐3’‐deoxythymidine as the sugar donor, 13 pyrimidine and purine nucleobases were shown as the viable sugar acceptors in the nucleoside phosphorylase EcPPNP‐ and/or nucleoside 2’‐deoxyribosyltransferase LlNDT‐catalyzed reactions, furnishing the corresponding 3’‐amino containing nucleosides in 23‐73% NMR yields at an analytical scale. The synthetic utility of this diversification approach was further showcased by the successful synthesis of 3’‐amino‐2’,3’‐dideoxy‐5‐bromouridine in 29% isolated yield at a preparative scale. The current work demonstrates the transglycosylation activity of EcPPNP for the first time, expands the substrate scope of LlNDT, and provides an efficient, direct access to various 3’‐amino nucleosides of high value.

Abstract:A series of novel 1'-O-alpha-methyl isonucleosides were efficiently and stereoselectively synthesized with 1,2,3,5-tetra-O-acetyl-D-ribofuranose as the starting material. The key steps include regioselective and stereoselective deprotection of the 2,4-dichlorobenzyl group at C2, triflation, and substitution with appropriate nucleobases using cesium carbonate as the base. Removal of the residual 2,4-dichlorobenzyl groups and subsequent transformation afforded the title compounds in 30-37% overall yield. The products represent a new type of isonucleosides with 1'-O-substitution.

Abstract:Nucleosides find applications in medicinal chemistry, chemical biology and diagnostics. Among the nucleosides, C‐nucleosides have attracted particular attention because their carbon‐carbon bond between nucleobase and ribose provides stability against degradation. While several well‐established methods exist for the synthesis of 2'‐deoxy‐C‐nucleosides, the preparation of their ribofuranosyl counterparts is more challenging. Established methods for their synthesis give mixtures of anomers and require harsh reagents or conditions. Here we report a diastereoselective glycosylation method involving a Heck reaction between a glycal and an aryl iodide, followed by oxidation of the silyl enol ether with methyl(trifluoromethyl)dioxirane (TFDO). The resulting ketone is then diastereoselectively reduced to the ribonucleoside. The new route has been applied to pyridones and is expected to also yield other ribo‐C‐nucleosides in diastereoselective fashion.

Abstract:A direct route to branched N-allylpyrimidine analogues is herein reported via the highly regio- and enantioselective asymmetric allylation of pyrimidines with racemic allylic carbonates. With [Rh(COD)Cl]2/chiral diphosphine as the catalyst, a range of chiral pyrimidine acyclic nucleosides could be obtained under neutral conditions in good yields (up to 95% yield) with high levels of regio- and enantioselectivities (15:1 to >40:1 B/L and up to 99% ee). Furthermore, chiral pyrimidine acyclic nucleoside bearing two adjacent chiral centers has been successfully synthesized by asymmetric dihydroxylation.

Abstract:The ribonucleosides of pyrazolo (5,4-e)-1,2,4-triazine derivatives were synthesized. The ��,��-isomes were distinguished with 1H NMR. ��-isomer was the major product in acetyl protected ribonucleosides. In benzoyl protected ribonucleosides, no ��-isomer was formed. The J H1H2 of cis isomer was bigger than that of trans in five membered ring.

Abstract:The (nitrosocarbonyl)mesitylene intermediate undergoes an ene reaction with crotyl alcohol, affording two regioisomeric adducts in fair yields. The sterically demanding (nitrosocarbonyl)mesitylene slightly shifts the C2/Markovnikov orientation towards a C3/anti-Markovnikov pathway, affording a 5-hydroxyisoxazolidine that serves as a synthon for the preparation of N,O-nucleoside analogues through the Vorbrüggen protocol. The selectivity of the ene reaction is discussed in the light of C=C bond polarization and steric effects. The structures of the N,O-nucleosides are assigned and discussed on the basis of spectroscopic observations and X-ray analysis.

Abstract:A general synthetic approach to 2,3-unsaturated glycosides connecting with nucleosides involving Ferrier rearrangements of glycals is discussed. The new compounds were identified by NMR and MS (HRFAB+). The hydroxylation of the resulting 2,3-unsaturated glycosides was completed using OsO4 to give 5′-O-glycosylnucleosides in good yield.

Abstract:The reagent system based on the combined use of Et3SiH/I2 acts as an efficient N-glycosidation promoter for the synthesis of natural and sugar-modified nucleosides. An analysis of reaction stereoselectivity in the absence of C2-positioned stereodirecting groups revealed high selectivity with six-membered substrates, depending on the nucleophilic character of the nucleobase or based on anomerization reactions. The synthetic utility of the Et3SiH/I2-mediated N-glycosidation reaction was highlighted by its use in the synthesis of the investigational drug apricitabine.

Abstract:Abstract β-Nucleosides and their analogs are dominant clinically-used antiviral and antitumor drugs. α-Nucleosides, the anomers of β-nucleosides, exist in nature and have significant potential as drugs or drug carriers. Currently, the most widely used methods for synthesizing β- and α-nucleosides are via N -glycosylation and pentose aminooxazoline, respectively. However, the stereoselectivities of both methods highly depend on the assisting group at the C2’ position. Herein, we report an additive-controlled stereodivergent iodocyclization method for the selective synthesis of α- or β-nucleosides. The stereoselectivity at the anomeric carbon is controlled by the additive (NaI for β-nucleosides; PPh 3 S for α-nucleosides). A series of β- and α-nucleosides are prepared in high yields (up to 95%) and stereoselectivities (β:α up to 66:1, α:β up to 70:1). Notably, the introduced iodine at the C2’ position of the nucleoside is readily functionalized, leading to multiple structurally diverse nucleoside analogs, including stavudine, an FDA-approved anti-HIV agent, and molnupiravir, an FDA-approved anti-SARS-CoV-2 agent.

Abstract:A regioselective synthesis of isoxazoles with propargyl alcohols, N‐iodosuccinimide (NIS), and N‐tert‐butyl hydroxylamine hydrochloride (t‐BuNHOH•HCl) as the reaction substrates is reported. Reaction scope investigations indicated that primary and secondary propargyl alcohols underwent this transformation smoothly to afford 5‐, 3‐, and 3,5‐substituted isoxazoles in moderate to good yields. The reaction most likely involves an electrophilic‐intercepted Meyer–Schuster rearrangement of the propargyl alcohol, followed by intermolecular cyclocondensation of N‐tert‐butyl hydroxylamine and in‐situ‐formed α‐iodo enones/enals. This one‐pot method is regioselective and easy to perform, making it a valuable method for the construction of isoxazoles. Finally, its application was explored through the synthesis of valdecoxib.

Abstract:Abstract Modern, highly evolved nucleoside-processing enzymes are known to exhibit perfect regioselectivity over the glycosylation of purine nucleobases at N 9. We herein report an exception to this paradigm. Wild-type nucleoside phosphorylases also furnish N 7-xanthosine, a “non-native” ribosylation regioisomer of xanthosine. This unusual nucleoside possesses several atypical physicochemical properties such as redshifted absorption spectra, a high equilibrium constant of phosphorolysis and low acidity. Ultimately, the biosynthesis of this previously unknown natural product illustrates how even highly evolved, essential enzymes from primary metabolism are imperfect catalysts.

Abstract:1,3,4,6-Tetra-O-acetyl-2-deoxy-2-phthalimido-β-d-glucopyranose and 1,3,4,6-tetra-O-acetyl-2-deoxy-2-phthalimido-β-d-galactopyranose reacted with protected nucleosides in the presence of BF3 as promoter at room temperature to give selectively 2-amino-2-deoxy-β-glycosyl (1→5)nucleosides in good yields. CD spectra and thermal melting studies showed that 2-amino-2-deoxy-β-d-glucopyranosyl-(1→5)-nucleosides could interact with RNA in solution and 2-deoxy-2-amino-β-d-galactopyranosyl-(1→5)-nucleosides (17–19) exhibit higher affinity to RNA than 2-deoxy-2-amino-β-d-glucopyranosyl-(1→5)-nucleosides (14–16). It indicated that the majority of interactions are established between the polar group of glycosylnucleosides and the sugar-phosphate backbone of RNA helices and weak stacking interaction is observed. The different configuration of hydroxyl group on the glycosyl moiety may affect the glycosyl-nucleoside binding to RNA by induced fit.
