Abstract:A direct route to branched N-allylpyrimidine analogues is herein reported via the highly regio- and enantioselective asymmetric allylation of pyrimidines with racemic allylic carbonates. With [Rh(COD)Cl]2/chiral diphosphine as the catalyst, a range of chiral pyrimidine acyclic nucleosides could be obtained under neutral conditions in good yields (up to 95% yield) with high levels of regio- and enantioselectivities (15:1 to >40:1 B/L and up to 99% ee). Furthermore, chiral pyrimidine acyclic nucleoside bearing two adjacent chiral centers has been successfully synthesized by asymmetric dihydroxylation.

Abstract:We herein describe a synergistic chiral primary amine/achiral palladium catalyzed enantioselective terminal addition to allenes with alpha-branched,beta-ketocarbonyls and aldehydes. The reactions afford allylic adducts bearing acyclic all-carbon quaternary centers with high regio- and enantioselectivity. A wide range of allenes including those aliphatic or 1,1'-disubstituted could be employed, thus expanding the scope of typical asymmetric allylic alkylation reactions.

Abstract:Chiral allene and N -heteroaryl motifs are present in an ever-growing list of biologically active natural products and synthetic drugs. Although significant progress has been made in asymmetric syntheses of chiral allenes, general and practical protocols for enantioselective syntheses of chiral N -heteroaryl-substituted allenes from readily available starting materials still remain rare. Here we report a highly enantioselective synthesis of quinolinyl-substituted chiral allenes through a copper-catalyzed asymmetric allenylation of quinoline N -oxides with readily available 1,3-enynes. A variety of 1,3-enynes react with quinoline N -oxides, affording the corresponding quinolinyl-substituted allenes in high yields (up to 95%) and high enantioselectivities (up to 99% ee). This transformation tolerates a variety of functional groups, such as chloro, bromo, trifluoromethyl ether, tertiary amine, siloxy, carboxylic ester, imide, pyridine, and thiophene moieties. DFT calculations suggest a pathway involving an intramolecular nucleophilic addition of an allenyl copper intermediate with a coordinated quinoline N -oxide through a five-membered, rather than seven-membered, transition state.

Abstract:Pyrazolones are a vital class of heterocycles possessing various biological properties and much attention is paid to the diversified synthesis of enantiopure pyrazolone derivatives. We describe here the development of diphenylphosphinoalkanoic acid based chiral bisphosphine ligands, which are successfully applied to the palladium-catalyzed asymmetric allenylation of racemic pyrazol-5-ones. The reaction affords C-allenylation products, optically active pyrazol-5-ones bearing an allene unit, in high chemo- and enantioselectivity, with DACH-ZYC-Phos-C1 as the best ligand. The synthetic potential of the C-allenylation products is demonstrated. Furthermore, the enantioselectivity observed with DACH-ZYC-Phos-C1 is rationalized by density functional theory studies.

Abstract:We report the development of a new class of multifunctional chiral guanidine/Pd(0) catalyst system for 1,4-addition/arylation tandem reaction. A variety of tetra-substituted allenes were readily accessible from three-component "one-pot" transformations of acyclic or cyclic 2-activated 1,3-enynes, malonates and halobenzenes under mild reaction conditions. High levels of yield and enantioselectivity were achieved in the construction of stereogenic center and axis using readily available acyclic guanidine-amides. The mechanistic studies suggest that the guanidine/Pd(0) collaboration has obvious synergism to both base-dominated conjugate addition, and Pd(0)-dominated Heck-type reaction.

Abstract:The first rhodium-catalyzed intermolecular [3+2] cycloaddition reaction of vinyl aziridines and allenes for the synthesis of enantioenriched functionalized pyrrolidines was realized. [3+2] cycloaddition with the proximal C=C bond of N-allenamides gave 3-methylene-pyrrolidines in high regio- and diastereoselectivity, whereas, 2-methylene-pyrrolidines were obtained as the major products by the cycloadditions of vinyl aziridines with the distal C=C bond of allenes. Use of readily available starting materials, a broad substrate scope, high selectivity, mild reaction conditions, as well as versatile functionalization of the cycloadducts make this approach very practical and attractive.

Abstract:CuI/bis(oxazoline)-catalyzed addition of propiolates and terminal ynones to 1-acylpyridinium salt (generated in situ from reaction of pyridine and methyl chloroformate) affords highly functionalized dihydropyridines with excellent enantioselectivity. It is found that the carbonyl group adjacent to the alkyne moiety is essential for the enantioselectivity of the addition. Short synthesis of indolizidines 167B and 223AB is achieved by employing two addition products.

Abstract:AbstractA Co‐catalyzed enantioselective desymmetric [2+2+2] cycloaddition for synthesis of pyridines with all‐carbon quaternary carbon centers has been developed. The regio‐ and enantioselectivities are controlled by the inherent nature of terminal alkynes and the substituents on the bisoxazolinephosphine ligands. Pyridines with 5‐substitutents could be obtained with >20:1 regioselectivity and up to 94 % ee when terminal alkyl, alkenyl or silyl alkynes and DTBM/Ph‐based NPN* ligand L6 were used. Terminal aryl alkynes and Ph/Bn‐based ligand L4 leads to formation of pyridines with 6‐substitutents in up to 99 % ee.

Abstract:A new asymmetric catalytic protocol for the synthesis of enantioenriched N-allyl 2-pyrodones has been developed via the first Pd-catalyzed regio- and enantioselective aminoarylation of allenols with aryl iodides and 2-pyridones. By using a palladium complex generated in situ from Pd2(dba)3·CHCl3 and (S,S,S)-SKP as a catalyst, the three-component aminoarylation proceeded smoothly to afford a variety of functionalized N-allylic 2-pyridones in high yields with good regioselectivities and excellent enantioselectivities.

Abstract:Chiral phosphines were found to catalyze the enantioselective asymmetric gamma-addition of heteroaromatic compounds to allenoates in good yields with high enantiomeric ratios and regioselectivity in the presence of (S)-BINOL. Both pyrazole and imidazole could be employed in this process. The synthetic value of these gamma-addition products was demonstrated by the preparation of biologically relevant molecules and structural scaffolds. Remarkably, the synthetic utility of this strategy was demonstrated through a two-step synthesis of a Janus kinase (JAK) inhibitor.

Abstract:Abstract Amines are a class of compounds of essential importance in organic synthesis, pharmaceuticals and agrochemicals. Due to the importance of chirality in many practical applications of amines, enantioselective syntheses of amines are of high current interest. Here, we wish to report the development of ( R , R a )- N -Nap-Pyrinap and ( R , S a )- N -Nap-Pyrinap ligands working with CuBr to catalyze the enantioselective A 3 -coupling of terminal alkynes, aldehydes, and amines affording optically active propargylic amines, which are platform molecules for the effective derivatization to different chiral amines. With a catalyst loading as low as 0.1 mol% even in gram scale reactions, this protocol is applied to the late stage modification of some drug molecules with highly sensitive functionalities and the asymmetric synthesis of the tubulin polymerization inhibitor ( S )-(-)- N -acetylcolchinol in four steps. Mechanistic studies reveal that, unlike reported catalysts, a monomeric copper(I) complex bearing a single chiral ligand is involved in the enantioselectivity-determining step.

Abstract:The iridium-catalysed enantioselective intramolecular allylic substitution of pyrimidine-tethered allylic carbonates was developed. A wide range of chiral pyrimidine-fused diazepinone derivatives were successfully constructed in 88–96% yields with 85–99% ees. This work further highlights the power of chiral-bridged biphenyl phosphoramidites in asymmetric synthesis.

Abstract:A novel and efficient palladium-catalyzed highly regioselective reaction of 1-[2-(2,2-dibromoethenyl)phenyl]-1H-pyrrole with allenes was realized to synthesize pyrrolo[1,2-a]quinolones. The tandem process involves intermolecular cyclization and intramolecular direct arylation, leading to the formation three new C-C bonds and two new rings. Notably, this transformation exhibits broad substrate scope and high functional group tolerance.

Abstract:We herein describe an asymmetric α-allylic allenylation of β-ketocarbonyls and aldehydes with 1,3-enynes. A synergistic chiral primary amine/Pd catalyst was identified to facilitate the utilization of 1,3-enynes as atom-economic and achiral allene precursors. The synergistic catalysis enables the construction of all-carbon quaternary centers-tethered allenes bearing non-adjacent 1,3-axial central stereogenic centers in high level of diastereo- and enantio-selectivity. By switching the configurations of ligands and aminocatalysts, diastereodivergence can be achieved and any of the four diastereoisomers can be accessed in high diastereo- and enantio- selectivity.

Abstract:A strategy for the synthesis of iodo-substituted allylic pyrazoles through NIS-mediated regioselective additions of allenamides with pyrazoles is presented. This reaction proceeds rapidly, is scalable up to a gram scale, and tolerates a broad scope of substrates. Moreover, using 1,2,4-triazole as the nucleophile, the corresponding 1,2-addition product was obtained in good yield. Significantly, the products of this reaction, iodo-substituted allylic pyrazoles could undergo coupling with phenylacetylene under Sonogashira Cross-Coupling conditions.
