N-Cycle Genes Abundance Determined N Mineralization Rate Following Afforestation in the Loess Plateau of China

Abstract:Exploring the mechanism of soil nitrogen mineralization is of great significance of assessing forest ecosystem function. However, it is unclear how soil N-cycling genes drive soil nitrogen mineralization during afforestation. In this study, soil samples were collected from areas with a chronosequence of 14, 20, 30 and 45 years, of Robinia pseudoacacia L (RP14, RP20, RP30, and RP45) and a sloped farmland (FL) as a control. Analysis of soil N-cycling microbial diversity and composition by metagenomic sequencing, we found that soil microbial diversity (alpha and beta diversity) involved in N-cycling significantly changed along the afforestation chronosequence, and afforestation effectively increased the diversity of soil microbial communities. Through indoor culture experiments and correlation analysis, we found that both soil nitrification rate (Rn) and soil nitrogen mineralization rate (Rm) increased significantly with stand age, and soil Rm was significantly correlated with soil microbial community and soil N-cycling gene abundance. PLS-PM analysis showed that nitrification (narH,narY,nxrB, narG,narZ,nxrA, hao, pmoC-amoC) and denitrification(norB, nosZ, nirK) genes had a greater direct effect on soil Rm than soil microbial communities. The results revealed the relationship between soil nitrogen mineralization rate and soil microbial communities and functional genes involved in nitrogen cycling under the background of Robinia pseudoacacia L restoration on the Loess Plateau. This study enriches the understanding of the effects of microorganisms on soil nitrogen mineralization rate during afforestation and provides a new theoretical basis for evaluating soil nitrogen mineralization mechanisms during forest succession.
