Abstract:The electrons extracted from the CaMn(4) cluster during water oxidation in photosystem II are transferred to P(680)(+) via the redox-active tyrosine D1-Tyr161 (Y(Z)). Upon Y(Z) oxidation a proton moves in a hydrogen bond toward D1-His190 (His(Z)). The deprotonation and reprotonation mechanism of Y(Z)-OH/Y(Z)-O is of key importance for the catalytic turnover of photosystem II. By light illumination at liquid helium temperatures (∼5 K) Y(Z) can be oxidized to its neutral radical, Y(Z)(•). This can be followed by the induction of a split EPR signal from Y(Z)(•) in a magnetic interaction with the CaMn(4) cluster, offering a way to probe for Y(Z) oxidation in active photosystem II. In the S(3) state, light in the near-infrared region induces the split S(3) EPR signal, S(2)'Y(Z)(•). Here we report on the pH dependence for the induction of S(2)'Y(Z)(•) between pH 4.0 and pH 8.7. At acidic pH the split S(3) EPR signal decreases with the apparent pK(a) (pK(app)) ∼ 4.1. This can be correlated to a titration event that disrupts the essential H-bond in the Y(Z)-His(Z) motif. At alkaline pH, the split S(3) EPR signal decreases with the pK(app) ∼ 7.5. The analysis of this pH dependence is complicated by the presence of an alkaline-induced split EPR signal (pK(app) ∼ 8.3) promoted by a change in the redox potential of Y(Z). Our results allow dissection of the proton-coupled electron transfer reactions in the S(3) state and provide further evidence that the radical involved in the split EPR signals is indeed Y(Z)(•).

Abstract:The photosystem II (PSII) reaction center contains two redox active tyrosines, YZ and YD, situated on the D1 and D2 proteins, respectively. By illumination at 5 K, oxidation of YZ in oxygen-evolving PSII can be observed as induction of the Split S1 EPR signal from YZ* in magnetic interaction with the CaMn4 cluster, whereas oxidation of YD can be observed as the formation of the free radical EPR signal from YD*. We have followed the light induced induction at 5 K of the Split S1 signal between pH 4-8.5. The formation of the signal, that is, the oxidation of YZ, is pH independent and efficient between pH 5.5 and 8.5. At low pH, the split signal formation decreases with pKa approximately 4.7-4.9. In samples with chemically pre-reduced YD, the pH dependent competition between YZ and YD was studied. Only YZ was oxidized below pH 7.2, but at pH above 7.2, the oxidation of YD became possible, and the formation of the Split S1 signal diminished. The onset of YD oxidation occurred with pKa approximately 8.0, while the Split S1 signal decreased with pKa approximately 7.9 demonstrating that the two tyrosines compete in this pH interval. The results reflect the formation and breaking of hydrogen bonds between YZ and D1-His190 (HisZ) and YD and D2-His190 (HisD), respectively. The oxidation of respective tyrosine at 5 K demands that the hydrogen bond is well-defined; otherwise, the low-temperature oxidation is not possible. The results are discussed in the framework of recent literature data and with respect to the different oxidation kinetics of YZ and YD.
Light-adapted charge-separated state of photosystem II: structural and functional dynamics of the closed reaction center

Abstract:Photosystem II (PSII) uses solar energy to oxidize water and delivers electrons for life on Earth. The photochemical reaction center of PSII is known to possess two stationary states. In the open state (PSIIO), the absorption of a single photon triggers electron-transfer steps, which convert PSII into the charge-separated closed state (PSIIC). Here, by using steady-state and time-resolved spectroscopic techniques on Spinacia oleracea and Thermosynechococcus vulcanus preparations, we show that additional illumination gradually transforms PSIIC into a light-adapted charge-separated state (PSIIL). The PSIIC-to-PSIIL transition, observed at all temperatures between 80 and 308 K, is responsible for a large part of the variable chlorophyll-a fluorescence (Fv) and is associated with subtle, dark-reversible reorganizations in the core complexes, protein conformational changes at noncryogenic temperatures, and marked variations in the rates of photochemical and photophysical reactions. The build-up of PSIIL requires a series of light-induced events generating rapidly recombining primary radical pairs, spaced by sufficient waiting times between these events-pointing to the roles of local electric-field transients and dielectric relaxation processes. We show that the maximum fluorescence level, Fm, is associated with PSIIL rather than with PSIIC, and thus the Fv/Fm parameter cannot be equated with the quantum efficiency of PSII photochemistry. Our findings resolve the controversies and explain the peculiar features of chlorophyll-a fluorescence kinetics, a tool to monitor the functional activity and the structural-functional plasticity of PSII in different wild-types and mutant organisms and under stress conditions.
The S1 to S2 and S2 to S3 state transitions in plant photosystem II: relevance to the functional and structural heterogeneity of the water oxidizing complex

Abstract:In Photosystem II, light-induced water splitting occurs via the S state cycle of the CaMn 4 O 5 -cluster. To understand the role of various possible conformations of the CaMn 4 O 5 -cluster in this process, the temperature dependence of the S 1 → S 2 and S 2 → S 3 state transitions, induced by saturating laser flashes, was studied in spinach photosystem II membrane preparations under different conditions. The S 1 → S 2 transition temperature dependence was shown to be much dependent on the type of the cryoprotectant and presence of 3.5% methanol, resulting in the variation of transition half-inhibition temperature by 50 K. No similar effect was observed for the S 2 → S 3 state transition, for which we also show that both the low spin g = 2.0 multiline and high spin g = 4.1 EPR configurations of the S 2 state advance with similar efficiency to the S 3 state, both showing a transition half-inhibition temperature of 240 K. This was further confirmed by following the appearance of the Split S 3 EPR signal. The results are discussed in relevance to the functional and structural heterogeneity of the water oxidizing complex intermediates in photosystem II.

Abstract:The effects of Tyr<INF>D</INF> , Tyr<INF>Z</INF> , and QA radical formation on the absorption spectrum in the Soret region were studied in Mn-depleted Photosystem II at pH 8.6 (in order to be in the Tyr<inf>D</inf> state after dark adaptation). Flash-induced difference spectra were recorded in several PSII samples from: i) (formerly ), ii) sp. PCC 6803, iii) PCC 7203 grown under far-red light, and iv) . In the case of , mutants D1/H198Q, D1/T1789H, D2/I178H, and D2/Y160F, with PsbA1/Q130 instead of PsbA3/E130, were also studied for possible contributions from P<INF>D1</INF>, Chl<INF>D1</INF>, Chl<INF>D2</INF>, and Phe<INF>D1</INF>, respectively. For a possible contribution from P<INF>D2</INF>, the D2/H197A mutant was studied in . While P<INF>D1</INF> is clearly the species whose spectrum is blue-shifted by ~3nm in the presence of QA , as has already been well documented in the literature, the species whose spectra shift upon the formation of Tyr<INF>D</INF> and Tyr<INF>Z</INF> remain to be clearly identified, as they appear different from P<INF>D1</INF>, P<INF>D2</INF>, Phe<INF>D1</INF>, Chl<INF>D1</INF>, and Chl<INF>D2</INF>, as concluded by the lack of different light-induced difference spectra in the mutants listed above. Although we cannot rule out a weak effect, considering the accuracy of the experiments, it is proposed that other pigments, such as antenna Chl and/or Car, near the reaction center are involved. Additionally, it is shown that: i) there is no proton release into the bulk upon the oxidation of TyrD at pH 8.6, and ii) the rearrangement of the electrostatic environment of the pigments involved in the light-induced difference spectra in the samples studied, upon the formation of Tyr<INF>D</INF> , Tyr<INF>Z</INFD> , and QA , likely occurs differently from both a kinetic and structural perspective.

Formation and decay of the S3 EPR signal species in acetate-inhibited photosystem II

Abstract:A 230-G-wide EPR signal is induced in acetate-treated photosystem II by 30 s of illumination at 277 K followed by freezing under illumination to 77 K [MacLachlan, D. J., & Nugent, J. H. A. (1993) Biochemistry 32, 9772-9780]. This signal, referred to as the S3 EPR signal, has been interpreted to arise from an S2X+ species where X+ is an amino acid radical. Investigation of the factors responsible for the formation and decay of the S3 EPR signal reveals that the yield of the S3 EPR signal is strongly temperature-dependent and depends on the rate of oxidation of QA-. Quantitation of the number of centers contributing to the S3 EPR signal produced by the optimal continuous illumination times of 3 min at 250 K, 30 s at 273 K, and 5 s at 294 K gave values of 13, 38, and 49 +/- 3%, respectively. By using 5 s of illumination at 294 K to induce the S3 EPR signal, and then illumination at 200 K to reduce QA, both the S3 and QA(-)Fe EPR signals were induced in high yield. This result indicates that the S3 EPR signal does not arise from an acceptor-side species. When saturating laser flashes were used to induce the S3 EPR signal in a dark-prepared, dark-adapted, acetate-treated sample, the yield was small after one flash and close to maximal after two flashes. An EPR signal at g = 4.1 was observed to be formed at intermediate times during the decay of the S3 EPR signal in the dark; the rates of decay of the S3 EPR signal at 273 and 294 K corresponded to the rates of formation of the g = 4.1 EPR signal. These results, together with the flash results, indicate that two steps are involved in both the generation and decay of the S3 EPR signal. The rates of formation and decay of both the S3 and QA(-)Fe EPR signals were measured at 250, 273, and 294 K. A kinetic model is presented that accounts for these kinetic data and the yield of the S3 EPR signal.

Light-Induced Charge Separation in Photosystem I from Different Biological Species Characterized by Multifrequency Electron Paramagnetic Resonance Spectroscopy

Abstract:Photosystem I (PSI) serves as a model system for studying fundamental processes such as electron transfer (ET) and energy conversion, which are not only central to photosynthesis but also have broader implications for bioenergy production and biomimetic device design. In this study, we employed electron paramagnetic resonance (EPR) spectroscopy to investigate key light-induced charge separation steps in PSI isolated from several green algal and cyanobacterial species. Following photoexcitation, rapid sequential ET occurs through either of two quasi-symmetric branches of donor/acceptor cofactors embedded within the protein core, termed the A and B branches. Using high-frequency (130 GHz) time-resolved EPR (TR-EPR) and deuteration techniques to enhance spectral resolution, we observed that at low temperatures prokaryotic PSI exhibits reversible ET in the A branch and irreversible ET in the B branch, while PSI from eukaryotic counterparts displays either reversible ET in both branches or exclusively in the B branch. Furthermore, we observed a notable correlation between low-temperature charge separation to the terminal [4Fe-4S] clusters of PSI, termed FA and FB, as reflected in the measured FA/FB ratio. These findings enhance our understanding of the mechanistic diversity of PSI's ET across different species and underscore the importance of experimental design in resolving these differences. Though further research is necessary to elucidate the underlying mechanisms and the evolutionary significance of these variations in PSI charge separation, this study sets the stage for future investigations into the complex interplay between protein structure, ET pathways, and the environmental adaptations of photosynthetic organisms.

Abstract:In oxygenic photosynthesis,photosystem II(PSII) catalyzes the water splitting/oxidation and O2 release into the ambient atmosphere upon light absorption.The catalytic active site therein is an inorganic cluster consisted of 4 Mn,1 Ca and 5 O.Electron paramagnetic resonance(EPR) spectroscopy plays a key role in the endeavor to elucidate the mechanism of water splitting.In this short review,we emphasized the interpretation of EPR spectra originating from different oxidized Mn4O5Ca clusters and from different magnetic interactions between the Mn4O5Ca cluster and a nearby tyrosine radical,combined with the methanol effects.The overall oxidation of two substrate water molecules is a stepwise and proton-coupled electron transfer.

Abstract:Photochemistry plays a significant role in shaping the chemical reaction network in the solar nebula and interstellar clouds. However, even in a simple triatomic molecule photodissociation, determination of all fragmentation processes is yet to be achieved. In this work, we present a comprehensive study of the photochemistry of H2S, derived from cutting-edge translational spectroscopy measurements of the H, S(1D) and S(1S) atom products formed by photolysis at wavelengths across the range 155-120 nm. The results provide detailed insights into the energy disposal in the SH(X), SH(A) and H2 co-fragments, and the atomisation routes leading to two H atoms along with S(3P) and S(1D) atoms. Theoretical calculations allow the dynamics of all fragmentation processes, especially the bimodal internal energy distributions in the diatomic products, to be rationalised in terms of non-adiabatic transitions between potential energy surfaces of both 1A' and 1A'' symmetry. The comprehensive picture of the wavelength-dependent (or vibronic state-dependent) photofragmentation behaviour of H2S will serve as a text-book example illustrating the importance of non-Born-Oppenheimer effects in molecular photochemistry, and the findings should be incorporated in future astrochemical modelling.

Abstract:Quantifying the photoinduced electron transfer properties of a single entity is of paramount importance for darifying the link between the photoelectrochemical performance and the specific properties of an individual. Here, we successfully monitored the photoelectrochemical behavior of a single dye sensitized ZnO entity on a Au ultramicroelectrode with different TiO2 film thicknesses. Due to a trap-limited electron diffusion in TiO2 film, a sub-millisecond photocurrent transient was observed for an individual N719@ZnO associated with single-particle photo catalytic water splitting. Furthermore, a Monte Carlo random walk numerical simulation model was developed to simulate the photoinjected electron transport dynamics and recombination in a nanoparticulate TiO2 film. Our approach allowed the photocatalytic properties of N719 at the single-molecule to be quantified, and electron diffusivity and electron collection efficiency as a function of the film thickness were estimated by simulation analyses. Excellent agreement was obtained between the experimental results and theoretical simulations, indicating that the underlying photoinduced electron transfer processes can be reliably explored.

Abstract:Excitation energy transfer (EET) and electron transfer (ET) are crucially involved in photosynthetic processes. In reality, the photosynthetic reaction center constitutes an open quantum system of EET and ET, which manifests an interplay of pigments, solar light and phonon baths. So far theoretical studies have been mainly based on master equation approaches in the Markovian condition. The non-Markovian environmental effect, which may play a crucial role, has not been sufficiently considered. In this work, we propose a mixed dynamic approach to investigate this open system. The influence of phonon bath is treated via the exact dissipaton equation of motion (DEOM) while that of photon bath is via the Lindblad master equation. Specifically, we explore the effect of non-Markovian quantum phonon bath on the coherent transfer dynamics and its manipulation on the current--voltage behavior. Distinguished from the results of completely Markovian Lindblad equation and those adopting classical environment description, the mixed DEOM--Lindblad simulations exhibittransfer coherence up to a few hundreds femtosecondsand the related environmental manipulation effect on current.These non-Markovian quantum coherent effects may be extended tomore complex and realistic systems and be helpful to thedesign of organic photovoltaic devices.

Abstract:Zero-field splitting (ZFS) parameters are fundamentally tied to the geometries of metal ion complexes. Despite their critical importance for understanding the magnetism and spectroscopy of metal complexes, they are not routinely available through general laboratory-based techniques, and are often inferred from magnetism data. Here we demonstrate a simple tabletop experimental approach that enables direct and reliable determination of ZFS parameters in the terahertz (THz) regime. We report time-domain measurements of electron paramagnetic resonance (EPR) signals associated with THz-frequency ZFSs in molecular complexes containing high-spin transition-metal ions. We measure the temporal profiles of the free-induction decays of spin resonances in the complexes at zero and nonzero external magnetic fields, and we derive the EPR spectra via numerical Fourier transformation of the time-domain signals. In most cases, absolute values of the ZFS parameters are extracted from the measured zero-field EPR frequencies, and the signs can be determined by zero-field measurements at two different temperatures. Field-dependent EPR measurements further allow refined determination of the ZFS parameters and access to the g-factor. The results show good agreement with those obtained by other methods. The simplicity of the method portends wide applicability in chemistry, biology and material science.
A quantum picture of light-suppressed photosynthetic charge transfer: Photo-blockade

Abstract:We propose a dynamic mechanism for the reversible regulation of photochemistry in plants under varying light environments. We employ a three-level quantum model to take into account the correlations between charge donors and charge acceptors immediately before photoexcitation, and show that under steady and coherent driving of light, the efficiency of charge transfer is inversely proportional to the intensity of incident light, which can be suppressed so severely that it becomes a limiting factor on photosynthetic electron transport. These results are analyzed to gain insight in the light responses of photosynthetic parameters. We discuss the implications of thermal fluctuation in the light source used in photochemical experiments, and argue that in high light conditions, the quantum yields measured with an incandescent lamp may be higher than those measured with a laser, a manifestation of thermal fluctuation in lamp illumination. Our new picture renders a consistent interpretation of a wide range of experiments, including plastocyanin-dependent electron transport in photosystem I, biphasic redox kinetics of P700 and wavelength-dependent quantum yields, and provides a donor-side scheme for the onset of irreversible damage to photosystem II.

Abstract:This work represents the first demonstration of a light-induced pH-switchable process monitored by in situ rapid-scan (RS) electron paramagnetic resonance (EPR) spectroscopy on the millisecond timescale. Here, we investigate the protonation state of an imidazoline type raical as pH sensor under visible light irradiation of a merocyanine photoacid in bulk solution. The results highlight the utility of photoacids in combination with pH-sensitive spin probes as an effective tool for real-time investigation of biochemical mechanisms regulated by changes of the pH value.

Space Quantization of Light Transmission by Strong Coupling of Plasmonic Cavity Modes with Photosynthetic Complexes

Abstract:The interaction between molecules and surface plasmons in defined geometries can lead to new light mater hybrid states where light propagation is strongly influenced by molecular photon absorption. Their application range from lasing LEDs to controlling chemical reactions and are relevant in light harvesting. The coupling between the electromagnetic field and molecular excitations may also lead to macroscopic extended coherent states characterized by an increase in temporal and spatial coherency. In this respect, it is intriguing to explore the coherency of the hybrid system for molecules that possess highly efficient exciton energy transfer. Such a molecule, is the photosynthetic light harvesting complex photosystem I which has an extended antenna system dedicated for efficient light harvesting. In this work, we demonstrate space quantization of light transmission through a single slit in free standing Au film coated with several layers of PS I. A self assembly technique for multilayer fabrication is used, enabling fabrication of multilayers which leaves most of the hole vacant with only the surface and slit walls coated with molecules. When a broad band, non coherent white light source excites the cavity, a strong SQOL is observed which is attributed to molecular photon absorption that induces coherency in the plasmon cavity modes. The SQOL is accompanied by a 13 fold enhancement in the extraordinary optical transmission through the cavity. This work demonstrate the emergence of spatial coherency in a cavity strongly coupled to one of the most efficient energy transfer photosynthetic protein in nature and provides the path for engineering quantum electrodynamics by the vast diversity of electronic properties of biological macromolecules.

Abstract:Upon the addition of imidazole (Im) to the artificial photosynthetic systems composed by ZnTPP/BQ and p-H2TPP-NH-CO-CH2-BQ, half-lives of ESR signals of semibenzoquinone radical BQ(.-) and -CH2-BQ(.-) are significantly prolonged by the secondary electron transfer pathway from imidazole to porphyrin cation in polar solvents. The imidazole cation radicals (Im can simultaneously be identified by PBN(spin trap) both in BQ/ZnTPP/Im and p-H2TPP-NH-CO-CH2-BQ systems when laser-flashing. On the basis of our ESR observation, a secondary electron transfer mechanism can be proposed as follows: (1) TPP+hv-->TPP*; (2) TPP*+BQ-->TPP.++BQ(.-); (3) TPP.++Im-->TPP+Im(.+).
Differentiation of correlated fluctuations in site energy on excitation energy transfer in photosynthetic light-harvesting complexes

Abstract:One of the promising approaches to revealing the photosynthetic efficiency of close to one unit is to investigate the quantum regime of excitation energy transfer (EET). The majority of studies, however, have concluded that different pigment molecules contribute equally to EET, rather than differently. We investigate the roles of different site-energies in EET by evaluating the correlated fluctuations of site-energies in two adjacent pigment molecules (namely Site 1 and Site 2), and we attempt to demonstrate different site roles in EET with the j-V characteristics and power via a photosynthetic quantum heat engine (QHE) model rather than an actual photosynthetic protein. The results show that fluctuations at Site 1 (the pigment molecule absorbing solar photons) provide ascending and then descending EET. At Site 2, the EET is reduced through the use of correlated fluctuation increments (the pigment molecule acting as the charge-transfer excited state). Furthermore, when investigating the correlated fluctuations at Site 2, the different gap differences of the output terminal play a positive role in EET, but a sharply decreasing EET process is also achieved with less correlated fluctuations at Site 2 compared to those at Site 1.The findings show that different pigment molecules contribute differently to EET. The significance of this work is that it not only clarifies the roles of different pigment molecules in EET, but it also deepens our understanding of the fundamental physics of EET as it transports through the molecular chain in photosynthetic light-harvesting complexes. Furthermore, the results are appropriate to the EET in organic semiconductors, photovoltaic devices, and quantum networks, when these systems couple to the environment of photons via the vibrational motion of sites in the molecular chain.
