Abstract:The mechanism of the initial electron transfer step in the reaction center of photosynthetic bacteria has been the subject of intense study over the past 10 years. This initial step is ultrafast, occurring in about 3 ps at room temperature [1]. As the understanding of the reaction center improves the need arises for more precise kinetic data. In particular, questions arise to the exponentiality of the observed kinetic signals [2], the possibility of differing behavior at different wavelengths [3], the existence of oscillatory components [2], and of spectral shifts accompanying the excitation and subsequent electron transfer processes. We measured the spontaneous fluorescence decay of P* in Rb. Sphaeroidis R26, Rb. Capsulatus, and mutantsof Rb. Capsulatus using the upconversion technique. For all samples QA was chemically reduced with the exception of the Rb. Sphaeroidis R26 sample for which the quinone was removed. The Rb. Spaeroidis R26 quinone removed measurements with both direct excitation of P at 850 nm and indirect excitation through internal conversion from PQX and energy transfer from BChl excited at 608 nm. All other samples were excited @ 850 nm. Excitation at 608 nm was provided by an antiresonant ring dye laser amplified at 100 Khz by a YAG regen yielding 60 fs pulses [4]. Experiments using 850 nm excitation were performed with a Coherent Mira 900 F Ti sapphire laser operating at 76 MHz with 95 fs pulses [5]. The instrument response functions at 940 nm emission for the two apparatus is ~180 fs. A typical data set is shown in Figure 1. All the samples showed nonexponential decay (see Figure 1) which could be adequately fit by a sum of two exponentials. The shorter component compares very well with the single components determined by stimulated emission [6]. However, the improved dynamic range and absence of other components (excited state absorption and ground state bleaching) make the nonexponentiality very clear. With 850 nm excitation we were unable to detect a risetime for fluorescence at 940 nm. With 610 nm excitation a risetime of 200 fs is apparent (Figure 2). This risetime results from a combination of energy transfer from the accessory pigments and electronic relaxation with the special pair manifold.

Abstract:Restoration of positive mood, in addition to reducing negative mood, is an important treatment goal in the management of depression. The need to restore positive mood states in depressed patients is not adequately addressed by available treatments for major depressive disorder (MDD), suggesting that this mood dimension could be a useful target for drug development. However, for positive mood restoration to become a valid target for antidepressant drug development certain questions should be answered: are symptoms of decreased positive mood phenomenologically distinct from other symptoms of MDD? Should they be considered a distinct aspect of MDD in the diagnostic nomenclature? Is there evidence for differential responsiveness to treatment? Is the underlying pathophysiology understood and different from that of other MDD symptoms? Is low positive mood specific to depression or does it contribute to psychopathology in other disorders? Beyond these basic questions, this review identifies a number of design issues that need to be considered when conducting studies that target improving positive mood. These design issues include (1) what population to study, (2) what line of treatment to target, (3) the appropriateness and validation of methods and measures to evaluate positive mood and its restoration, (4) the role of functional outcome measures in determining success of the treatment, and (5) optimal designs for add-on therapy versus monotherapy agents.
Investigation of electrochromic band-shifts in the Soret region induced by the formation of Tyr<inf>D</inf> , Tyr<inf>Z</inf> , and Q<inf>A</inf> in Photosystem II

Abstract:The effects of Tyr<INF>D</INF> , Tyr<INF>Z</INF> , and QA radical formation on the absorption spectrum in the Soret region were studied in Mn-depleted Photosystem II at pH 8.6 (in order to be in the Tyr<inf>D</inf> state after dark adaptation). Flash-induced difference spectra were recorded in several PSII samples from: i) (formerly ), ii) sp. PCC 6803, iii) PCC 7203 grown under far-red light, and iv) . In the case of , mutants D1/H198Q, D1/T1789H, D2/I178H, and D2/Y160F, with PsbA1/Q130 instead of PsbA3/E130, were also studied for possible contributions from P<INF>D1</INF>, Chl<INF>D1</INF>, Chl<INF>D2</INF>, and Phe<INF>D1</INF>, respectively. For a possible contribution from P<INF>D2</INF>, the D2/H197A mutant was studied in . While P<INF>D1</INF> is clearly the species whose spectrum is blue-shifted by ~3nm in the presence of QA , as has already been well documented in the literature, the species whose spectra shift upon the formation of Tyr<INF>D</INF> and Tyr<INF>Z</INF> remain to be clearly identified, as they appear different from P<INF>D1</INF>, P<INF>D2</INF>, Phe<INF>D1</INF>, Chl<INF>D1</INF>, and Chl<INF>D2</INF>, as concluded by the lack of different light-induced difference spectra in the mutants listed above. Although we cannot rule out a weak effect, considering the accuracy of the experiments, it is proposed that other pigments, such as antenna Chl and/or Car, near the reaction center are involved. Additionally, it is shown that: i) there is no proton release into the bulk upon the oxidation of TyrD at pH 8.6, and ii) the rearrangement of the electrostatic environment of the pigments involved in the light-induced difference spectra in the samples studied, upon the formation of Tyr<INF>D</INF> , Tyr<INF>Z</INFD> , and QA , likely occurs differently from both a kinetic and structural perspective.

Thermodynamics of the S2-to-S3 State Transition of the Oxygen-Evolving Complex of Photosystem II

Abstract:The room temperature pump-probe X-ray free electron laser (XFEL) measurements used for serial femtosecond crystallography provide remarkable information about the structures of the catalytic (S-state) intermediates of the oxygen-evolution reaction of photosystem II. However, mixed populations of these intermediates and moderate resolution limit the interpretation of the data from current experiments. The S3 XFEL structures show extra density near the OEC that may correspond to a water/hydroxide molecule. However, in the latest structure, this additional oxygen is 2.08 Å from the Oe2 of D1-E189, which is closer than the sum of the van der Waals radii of the two oxygens. Here, we use Boltzmann statistics and Monte Carlo sampling to provide a model for the S2-to-S3 state transition, allowing structural changes and the insertion of an additional water/hydroxide. Based on our model, water/hydroxide addition to the oxygen-evolving complex (OEC) is not thermodynamically favorable in the S2 g = 2 state, but it is in the S2 g = 4.1 redox isomer. Thus, formation of the S3 state starts by a transition from the S2 g = 2 to the S2 g = 4.1 structure. Then, electrostatic interactions support protonation of D1-H190 and deprotonation of the Ca2+-ligated water (W3) with proton loss to the lumen. The W3 hydroxide moves toward Mn4, completing the coordination shell of Mn4 and moving with its oxidation to Mn(IV) in the S3 state. In addition, binding additional hydroxide to Mn1 leads to a conformational change of D1-E189 in the S2 g = 4.1 and S3 structures. In the S3 state in the population of protonated D1-E189 increases.

Abstract:Accurate research of the photophysical processes is of great significance for the rational design of excellent thermally activated delayed fluorescence (TADF) materials. Herein, the interconversion and decay rates of the first excited singlet state (S1) and triplet states (T1) in the Cu(pop)(pz2BH2) complex are computed using the thermal vibration correlation function (TVCF) theory at different temperature. For consideration of the solid-state environment, a methodology that is based on the ONIOM model, combined with the optimally tuned range-separated hybrid functional (CAM-B3LYP*) method, was applied. Our calculated results are in excellent agreement with the experimentally available data. It has been found that the energy dissipation of the nonradiative processes from the S1 to ground state is promoted by low frequency vibrational modes in the solution phase, resulting in the high knr(S) = 1.68 × 108 s-1 at 300 K. However, for the crystal phase, they are easily hindered through intermolecular interactions, knr(S) is predicted to be decreased by about 5 orders of magnitude upon aggregation (2.98 × 103 s-1). With temperature increase, the reverse intersystem crossing (RISC) rate kRISC from T1 to S1 is drastically increased to 6.12 × 104 s-1 at 300 K, while the change of other rates is still small, which can compete with the radiative decay rate of kr(T) = 4.75 × 104 s-1 and nonradiative intersystem crossing rate of kISC(T1-S0) = 6.63 × 102 s-1 at the T1 state. This implies that the S1 state can be an efficient thermal population from the T1 state, leading to an occurrence of delayed fluorescence, and the complexes exhibit high emission quantum yields, 58.7%. But, at low temperature T < 100 K, the RISC rate is sharply change, kRISC ≪ kr(T) or kISC, which cannot induce an occurrence of delayed fluorescence. Our investigation would be helpful for designing novel, high-efficiency TADF materials.

Abstract:Excited-state intramolecular proton transfer (ESIPT) reactions are crucial in photoresponsive materials and fluorescent markers. The fluorescent compound 4-aminophthalimide (4-AP) has been reported to exhibit solvent-assisted ESIPT in protic solvents, such as methanol, wherein the solvent interacts with 4-AP to form a six-membered hydrogen-bonded ring that is strengthened upon excitation. Although the controversial observation of ESIPT in 4-AP has been extensively studied, the molecular mechanism has yet to be fully explored. In this study, femtosecond infrared spectroscopy was used to investigate the dynamics of 4-AP in methanol and acetonitrile after excitation at 350 and 300 nm, which promoted 4-AP to the S1 and S2 states, respectively. The excited 4-AP in the S1 state relaxed to the ground state, while 4-AP in the S2 state relaxed via the S1 state without the occurrence of ESIPT. The enol form of 4-AP (Enol 4-AP) in the S1 state was calculated to be ~10 kcal/mol higher in energy than the keto form in the S1 state, indicating that keto-to-enol tautomerization was endergonic, ultimately resulting in no observable ESIPT for 4-AP in the S1 state. Upon the excitation of 4-AP to the S2 state, the transition to Enol-4-AP in the S1 state was found to be exergonic; however, ESIPT must compete with an internal conversion from the S2 to the S1 state. The internal S2 → S1 conversion was significantly faster than the solvent-assisted ESIPT, resulting in a negligible ESIPT for the 4-AP excited to the S2 state. The detailed excitation dynamics of 4-AP clearly reveal the molecular mechanism underlying its negligible ESIPT, despite the fact that it forms a favorable structure for solvent-assisted ESIPT.

Abstract:In Photosystem II, light-induced water splitting occurs via the S state cycle of the CaMn 4 O 5 -cluster. To understand the role of various possible conformations of the CaMn 4 O 5 -cluster in this process, the temperature dependence of the S 1 → S 2 and S 2 → S 3 state transitions, induced by saturating laser flashes, was studied in spinach photosystem II membrane preparations under different conditions. The S 1 → S 2 transition temperature dependence was shown to be much dependent on the type of the cryoprotectant and presence of 3.5% methanol, resulting in the variation of transition half-inhibition temperature by 50 K. No similar effect was observed for the S 2 → S 3 state transition, for which we also show that both the low spin g = 2.0 multiline and high spin g = 4.1 EPR configurations of the S 2 state advance with similar efficiency to the S 3 state, both showing a transition half-inhibition temperature of 240 K. This was further confirmed by following the appearance of the Split S 3 EPR signal. The results are discussed in relevance to the functional and structural heterogeneity of the water oxidizing complex intermediates in photosystem II.

Chlorophyll triplet states in thylakoid membranes of Acaryochloris marina. Evidence for a triplet state sitting on the photosystem I primary donor populated by intersystem crossing

Abstract:Photo-induced triplet states in the thylakoid membranes isolated from the cyanobacterium Acaryocholoris marina, that harbours Chlorophyll (Chl) d as its main chromophore, have been investigated by Optically Detected Magnetic Resonance (ODMR) and time-resolved Electron Paramagnetic Resonance (TR-EPR). Thylakoids were subjected to treatments aimed at poising the redox state of the terminal electron transfer acceptors and donors of Photosystem II (PSII) and Photosystem I (PSI), respectively. Under ambient redox conditions, four Chl d triplet populations were detectable, identifiable by their characteristic zero field splitting parameters, after deconvolution of the Fluorescence Detected Magnetic Resonance (FDMR) spectra. Illumination in the presence of the redox mediator N,N,N',N'-Tetramethyl-p-phenylenediamine (TMPD) and sodium ascorbate at room temperature led to a redistribution of the triplet populations, with T3 (|D|= 0.0245 cm-1, |E|= 0.0042 cm-1) becoming dominant and increasing in intensity with respect to untreated samples. A second triplet population (T4, |D|= 0.0248 cm-1, |E|= 0.0040 cm-1) having an intensity ratio of about 1:4 with respect to T3 was also detectable after illumination in the presence of TMPD and ascorbate. The microwave-induced Triplet-minus-Singlet spectrum acquired at the maximum of the |D|-|E| transition (610 MHz) displays a broad minimum at 740 nm, accompanied by a set of complex spectral features that overall resemble, despite showing further fine spectral structure, the previously reported Triplet-minus-Singlet spectrum attributed to the recombination triplet of PSI reaction centre, 3 P 740 [Schenderlein M, Çetin M, Barber J, et al. Spectroscopic studies of the chlorophyll d containing photosystem I from the cyanobacterium Acaryochloris marina. Biochim Biophys Acta 1777:1400-1408]. However, TR-EPR experiments indicate that this triplet displays an eaeaea electron spin polarisation pattern which is characteristic of triplet sublevels populated by intersystem crossing rather than recombination, for which an aeeaae polarisation pattern is expected instead. It is proposed that the observed triplet, which leads to the bleaching of the P740 singlet state, sits on the PSI reaction centre.
Unusual Kinetics Of Thermal Decay Of Dim-Light Photoreceptors In Vertebrate Vision

Abstract:We present measurements of rate constants for thermal-induced reactions of the 11-cis retinyl chromophore in vertebrate visual pigment rhodopsin, a process that produces noise and limits the sensitivity of vision in dim light. At temperatures of 52.0-64.6 degrees C, the rate constants fit well to an Arrhenius straight line with, however, an unexpectedly large activation energy of 114 +/- 8 kcal/mol, which is much larger than the 60-kcal/mol photoactivation energy at 500 nm. Moreover, we obtain an unprecedentedly large prefactor of 10(72 +/- 5) s(-1), which is roughly 60 orders of magnitude larger than typical frequencies of molecular motions! At lower temperatures, the measured Arrhenius parameters become more normal: E-a = 22 +/- 2 kcal/mol and A(pref) = 10(9 +/- 1) s(-1) in the range of 37.0-44.5 degrees C. We present a theoretical framework and supporting calculations that attribute this unusual temperature-dependent kinetics of rhodopsin to a lowering of the reaction barrier at higher temperatures due to entropy-driven partial breakup of the rigid hydrogen-bonding network that hinders the reaction at lower temperatures.
Femtosecond Spontaneous-Emission Studies of Reaction Centers from Photosynthetic Bacteria

Abstract:Spontaneous emission from reaction centers of photosynthetic bacteria has been recorded with a time resolution of 50 fs. Excitation was made directly into both the special-pair band (850 nm) and the Q(x) band of bacteriochlorophylls (608 nm). Rhodobacter sphaeroides R26, Rhodobacter capsulatus wild type, and four mutants of Rb. capsulatus were studied. In all cases the fluorescence decay was not single exponential and was well fit as a sum of two exponential decay components. The short components are in excellent agreement with the single component detected by measurements of stimulated emission. The origin of the nonexponential decay is discussed in terms of heterogeneity, the kinetic scheme, and the possibility of slow vibrational relaxation.
Ethanol Production Kinetics by a Thermo-Tolerant Mutant of Saccharomyces Cerevisiae from Starch Industry Waste (Hydrol)

Abstract:A thermo-tolerant and deoxyglucose-resistant mutant of Saccharomyces cerevisiae was developed and employed to convert them to fuel ethanol in a 150 litre fermenter. Maximum ethanol production was achieved when fermentation of dextrozyme- treated hydrol was carried out for about 36 hours under optimized fermenting conditions. The maximum specific ethanol production rate (qP), and overall ethanol yield (YP/S) were found to be 2.82 g L -1 h -1 and 0.49 g/g respectively. Determination of activation energy for cell growth (Ea= 20.8 kJ/mol) and death (Ed = 19.1 kJ/mol) and product formation and inactivation (EP=35.8 kJ/mol and Edp = 33.5 kJ/mol) revealed the thermo-stability of the organism for up to 47°C.

Abstract:Flash-induced Fourier transform infrared (FTIR) difference spectra for the four-step S-state cycle and the effects of global (15)N- and (13)C-isotope labeling on the difference spectra were examined for the first time in the mid- to low-frequency (1200-800 cm(-1)) as well as the mid-frequency (1700-1200 cm(-1)) regions using photosystem (PS) II core particles from cyanobacterium Synechocystis sp. PCC 6803. The difference spectra clearly exhibited the characteristic vibrational features for each transition during the S-state cycling. It is likely that the bands that change their sign and intensity with the S-state advances reflect the changes of the amino acid residues and protein matrices that have functional and/or structural roles within the oxygen-evolving complex (OEC). Except for some minor differences, the trends of S-state dependence in the 1700-1200 cm(-1) frequency spectra of the PS II cores from Synechocystis were comparable to that of spinach, indicating that the structural changes of the polypeptide backbones and amino acid side chains that occur during the oxygen evolution are inherently identical between cyanobacteria and higher plants. Upon (13)C-labeling, most of the bands, including amide I and II modes and carboxylate stretching modes, showed downward shifts; in contrast, (15)N-labeling induced isotopic shifts that were predominantly observed in the amide II region. In the mid- to low-frequency region, several bands in the 1200-1140 cm(-1) region were attributable to the nitrogen- and/or carbon-containing group(s) that are closely related to the oxygen evolution process. Specifically, the putative histidine ligand exhibited a band at 1113 cm(-1) which was affected by both (15)N- and (13)C-labeling and showed distinct S-state dependency. The light-induced bands in the 900-800 cm(-1) region were downshifted only by (13)C-labeling, whereas the bands in the 1000-900 cm(-1) region were affected by both (15)N- and (13)C-labeling. Several modes in the mid- to low-frequency spectra were induced by the change in protonation state of the buffer molecules accompanied by S-state transitions. Our studies on the light-induced spectrum showed that contributions from the redox changes of Q(A) and the non-heme iron at the acceptor side and Y(D) were minimal. It was, therefore, suggested that the observed bands in the 1000-800 cm(-1) region include the modes of the amino acid side chains that are coupled to the oxidation of the Mn cluster. S-state-dependent changes were observed in some of the bands.

The following observations were made: (1) The free energy state of P+IA− was pH independent below pH 10 (–370 ± 10 meV relative to that of the excited dimer P*) and showed a remarkable decrease (about 20 meV/pH unit) above pH 10. A part of the dielectric relaxation of the P+IA− charge pair that is not insignificant (about 120 meV) should come from protonation-related changes. (2) The single exponential decay character of the kinetics proves that the protonated/unprotonated P+IA− and P+QA− states are in equilibria and the rate constants of protonation konH +koffH are much larger than those of the charge back reaction kback ~103 s−1. (3) Highly similar pH profiles were measured to determine the free energy states of P+QA− and P+IA−, indicating that the same acidic cluster at around QB should respond to both anionic species. This was supported by model calculations based on anticooperative proton distribution in the cluster with key residues of GluL212, AspL213, AspM17, and GluH173, and the effect of the polarization of the aqueous phase on electrostatic interactions. The larger distance of IA− from the cluster (25.2 Å) compared to that of QA− (14.5 Å) is compensated by a smaller effective dielectric constant (6.5 ± 0.5 and 10.0 ± 0.5, respectively). (4) The P* → P+QA− and IA−QA → IAQA− electron transfers are enthalpy-driven reactions with the exemption of very large (>60%) or negligible entropic contributions in cases of substitution by 2,3-dimethyl-AQ or 1-chloro-AQ, respectively. The possible structural consequences are discussed.

Abstract:Transient absorption and emission spectra of IR-140 dye (5,5'-dichloro-11-diphenylamine-3,3'-diethyl- 10,12-ethylene-thiatricarbocyanine-perchlorate) in ethanol have been investigated by using the pump supercontinuum-probing technique of 130 fs time resolution. Excited at a wavelength of 807 nm, two peaks at 820 and 840 nm in the emission spectra were observed. The decay of stimulated emission (SE) at 840 nm can be fitted by a dual-exponential function. Meanwhile, a new transient absorption band ranging from 500 to 650 nm was observed and attributed to the transition between the excited singlet states. Its relaxation process could also be fitted by a dual -exponential function. Both relaxation processes of SE and excited-state absorption were probing wavelength-dependent and included a fast relaxation with a decay time of about several picoseconds and a slow relaxation with decay time longer than 0.1 ns. The prime energy structure was proposed according to the relaxation process and transient spectra of IR-140.

Abstract:Biological research in agriculture needs a lot of specialized electronic sensors in order to fulfill different goals, like as: climate monitoring, soil and fruit assessment, control of insects and diseases, chemical pollutants, identification and control of weeds, crop tracking, and so on. That research must be supported by consistent biological models able to simulate diverse environmental conditions, in order to predict the right human actions before risky biological damage could be irreversible. In this paper an experimental distributed network based on climatic and biological wireless sensors is described, for providing real measurements in order to validate different biological models used for viticulture applications.

Abstract:Salicylidenaniline (SA) is an archetypal system for excited-state intramolecular proton transfer (ESIPT) in non-planar systems. Multiple channels for relaxation involving both the keto and enol forms have been proposed after excitation to S$_1$ with near-UV light. Here we present transient absorption measurements of hot gas-phase SA, jet-cooled SA, and SA in Ar clusters using cavity-enhanced transient absorption spectroscopy (CE-TAS). Assignment of the spectra is aided by simulated TAS spectra, computed by applying time-dependent complete active space configuration interaction (TD-CASCI) to structures drawn from nonadiabatic molecular dynamics simulations. We find prompt ESIPT in all conditions followed by the rapid parallel generation of the trans-keto metastable photochrome state and fluorescent keto state in parallel. Increasing the internal energy increases the photochrome yield and decreases the fluorescent yield and fluorescent state lifetime observed in TAS. In Ar clusters, internal conversion of SA is severely hindered but the photochrome yield is unchanged. Taken together, these results are consistent with the photochrome being produced via the vibrationally excited keto population after ESIPT.
