Abstract:It has long been proposed that emotionally “prepared” (i.e., fear-related) stimuli are privileged in the unconscious acquisition of conditioned fear. However, as fear processing is suggested to highly depend on the coarse, low-spatial-frequency (LSF) components of the fear-related stimuli, it is plausible that LSF may play a unique role in the unconscious fear conditioning even with emotionally neutral stimuli. Here, we provided empirical evidence that, following classical fear conditioning, an invisible, emotionally neutral conditioned stimulus (CS+) with LSF, but not with high spatial frequency (HSF), can rapidly elicit stronger skin conductance responses (SCRs) and larger pupil diameters than its CS− counterpart. In comparison, consciously perceived emotionally neutral CS+ with LSF and HSF elicited comparable SCRs. Taken together, these results support that the unconscious fear conditioning does not necessarily entail emotionally prepared stimuli but prioritizes LSF information processing and highlight the crucial distinctions between the unconscious and the conscious fear learning. These findings not only coincide with the postulation that a rapid, spatial-frequency-dependent subcortical route is engaged in unconscious fear processing but also suggest the existence of multiple routes for conscious fear processing.

Rapid Synaptic Potentiation Within the Anterior Cingulate Cortex Mediates Trace Fear Learning

Abstract:Although the cortex has been extensively studied in long-term memory storage, less emphasis has been placed on immediate cortical contributions to fear memory formation. AMPA receptor plasticity is strongly implicated in learning and memory, and studies have identified calcium permeable AMPA receptors (CP-AMPARs) as mediators of synaptic strengthening. Trace fear learning engages the anterior cingulate cortex (ACC), but whether plastic events occur within the ACC in response to trace fear learning, and whether GluN2B subunits are required remains unknown. Here we show that the ACC is necessary for trace fear learning, and shows a rapid 20% upregulation of membrane AMPA receptor GluA1 subunits that is evident immediately after conditioning. Inhibition of NMDA receptor GluN2B subunits during training prevented the upregulation, and disrupted trace fear memory retrieval 48 h later. Furthermore, intra-ACC injections of the CP-AMPAR channel antagonist, 1-naphthylacetyl spermine (NASPM) immediately following trace fear conditioning blocked 24 h fear memory retrieval. Accordingly, whole cell patch clamp recordings from c-fos positive and c-fos negative neurons within the ACC in response to trace fear learning revealed an increased sensitivity to NASPM in recently activated neurons that was reversed by reconsolidation update extinction. Our results suggest that trace fear learning is mediated through rapid GluN2B dependent trafficking of CP-AMPARs, and present in vivo evidence that CP-AMPAR activity within the ACC immediately after conditioning is necessary for subsequent memory consolidation processes.
Rapid Processing of Invisible Fearful Faces in the Human Amygdala

Abstract:Rapid detection of a threat or its symbol (e.g., fearful face), whether visible or invisible, is critical for human survival. This function is suggested to be enabled by a subcortical pathway to the amygdala independent of the cortex. However, conclusive electrophysiological evidence in humans is scarce. Here, we explored whether the amygdala can rapidly encode invisible fearful faces. We recorded intracranial electroencephalogram (iEEG) responses in the human (both sexes) amygdala to faces with fearful, happy, and neutral emotions rendered invisible by backward masking. We found that a short-latency intracranial event-related potential (iERP) in the amygdala, beginning 88 ms poststimulus onset, was preferentially evoked by invisible fearful faces relative to invisible happy or neutral faces. The rapid iERP exhibited selectivity to the low spatial frequency (LSF) component of the fearful faces. Time-frequency iEEG analyses further identified a rapid amygdala response preferentially for LSF fearful faces at the low gamma frequency band, beginning 45 ms poststimulus onset. In contrast, these rapid responses to invisible fearful faces were absent in cortical regions, including early visual areas, the fusiform gyrus, and the parahippocampal gyrus. These findings provide direct evidence for the existence of a subcortical pathway specific for rapid fear detection in the amygdala and demonstrate that the subcortical pathway can function without conscious awareness and under minimal influence from cortical areas. SIGNIFICANCE STATEMENT Automatic detection of biologically relevant stimuli, such as threats or dangers, has remarkable survival value. Here, we provide direct intracranial electrophysiological evidence that the human amygdala preferentially responds to fearful faces at a rapid speed, despite the faces being invisible. This rapid, fear-selective response is restricted to faces containing low spatial frequency information transmitted by magnocellular neurons and does not appear in cortical regions. These results support the existence of a rapid subcortical pathway independent of cortical pathways to the human amygdala.

Abstract:Fear is defined as a fundamental emotion promptly arising in the context of threat and when danger is perceived. Fear can be innate or learned. Examples of innate fear include fears that are triggered by predators, pain, heights, rapidly approaching objects, and ancestral threats such as snakes and spiders. Animals and humans detect and respond more rapidly to threatening stimuli than to nonthreatening stimuli in the natural world. The threatening stimuli for most animals are predators, and most predators are themselves prey to other animals. Predatory avoidance is of crucial importance for survival of animals. Although humans are rarely affected by predators, we are constantly challenged by social threats such as a fearful or angry facial expression. This chapter will summarize the current knowledge on brain circuits processing innate fear responses to visual stimuli derived from studies conducted in mice and humans.

Abstract:A subcortical pathway is thought to have evolved to facilitate fear information transmission, but direct evidence for its existence in humans is lacking. In recent years, rapid, preattentive, and preconscious fear processing has been demonstrated, providing indirect support for the existence of the subcortical pathway by challenging the necessity of canonical cortical pathways in fear processing. However, direct support also requires evidence for the involvement of subcortical regions in fear processing. To address this issue, here we investigate whether fear processing reflects the characteristics of the subcortical structures in the hypothesized subcortical pathway. Using a monocular/dichoptic paradigm, Experiment 1 demonstrated a same-eye advantage for fearful but not neutral face processing, suggesting that fear processing relied on monocular neurons existing mainly in the subcortex. Experiments 2 and 3 further showed insensitivity to short-wavelength stimuli and a nasal–temporal hemifield asymmetry in fear processing, both of which were functional characteristics of the superior colliculus, a key hub of the subcortical pathway. Furthermore, all three experiments revealed a low spatial frequency selectivity of fear processing, consistent with magnocellular input via subcortical neurons. These results suggest a selective involvement of subcortical structures in fear processing, which, together with the indirect evidence for automatic fear processing, provides a more complete picture of the existence of a subcortical pathway for fear processing in humans. Abstract:In daily social situations, faces rarely appear in isolation and are often contextual. This study investigated individuals' behavioral and neural responses to facial expressions with different intensities embedded in emotional scenes. Participants were presented with neutral, low-intensity, and prototypical fearful expressions embedded in positive and negative scenes and instructed to categorize the facial expressions as neutral or fearful. The behavioral results showed that neutral and low-intensity fearful expressions embedded in negative scenes were rated as fearful significantly more frequently than the same faces embedded in positive scenes. Event-related potential analyses revealed the time course of the integration of contextual information when processing facial expressions. N170 modulation by scene valence was found for low-intensity and prototypical fearful expressions, showing greater N170 elicited by negative than positive scenes. The results suggest that affective information extracted from contextual scenes influences the processing of facial emotions in the early perceptual encoding stage. Furthermore, the pattern of effect varies for different intensities of expressions.

Predicting Aversive Events And Terminating Fear In The Mouse Anterior Cingulate Cortex During Trace Fear Conditioning

Abstract:A variety of studies have implicated the anterior cingulate cortex (ACC) in fear, including permanent storage of fear memory. Recent pharmacological and genetic studies indicate that early synaptic plasticity in the ACC may also contribute to certain forms of fear memory at early time points. However, no study has directly examined the possible changes in neuronal activity of ACC neurons in freely behaving mice during early learning. In the present study, we examined the neural responses of the ACC during trace fear conditioning. We found that ACC putative pyramidal and nonpyramidal neurons were involved in the termination of fear behavior ("un-freezing"), and the spike activity of these neurons was reduced during freezing. Some of the neurons were also found to acquire un-freezing locked activity and change their tuning. The results implicate the ACC neurons in fear learning and controlling the abolition of fear behavior. We also show that the ACC is important for making cue-related fear memory associations in the trace fear paradigm as measured with tone-evoked potentials and single-unit activity. Collectively, our findings indicate that the ACC is involved in predicting future aversive events and terminating fear during trace fear.

Abstract:Human adaptive behaviour to potential threats involves specialized brain responses allowing rapid and reflexive processing of the sensory input and a more directed processing for later evaluation of the nature of the threat. The amygdalae are known to play a key role in emotion processing. It is suggested that the amygdalae process threat-related information through a fast subcortical route and slower cortical feedback. Evidence from human data supporting this hypothesis is lacking. The present study investigated event-related neural responses during processing of facial emotions in the unattended hemifield using magnetoencephalography (MEG) and found activations of the amygdala and anterior cingulate cortex to fear as early as 100 ms. The right amygdala exhibited temporally dissociated activations to input from different visual fields, suggesting early subcortical versus later cortical processing of fear. We also observed asymmetrical fusiform activity related to lateralized feed-forward processing of the faces in the visual-ventral stream. Results demonstrate fast, automatic, and parallel processing of unattended emotional faces, providing important insights into the specific and dissociated neural pathways in emotion and face perception.

Abstract:It has long been suggested that emotion, especially threatening emotion, facilitates early visual perception to promote adaptive responses to potential threats in the environment. Here, we tested whether and how fearful emotion affects the basic visual ability of visual acuity. An adapted Posner’s spatial cueing task was employed, with fearful and neutral faces as cues and a Vernier discrimination task as the probe. The time course of the emotional attention effect was examined by varying the stimulus onset asynchrony (SOA) of the cue and probe. Two independent experiments (Experiments 1 and 3) consistently demonstrated that the brief presentation of a fearful face increased visual acuity at its location. The facilitation of perceptual sensitivity was detected at an SOA around 300 ms when the face cues were presented for both 250 ms (Experiment 1) and 150 ms (Experiment 3). This effect cannot be explained by physical differences between the fearful and neutral faces because no improvement was found when the faces were presented inverted (Experiment 2). In the last experiment (Experiment 4), the face cues were flashed very briefly (17 ms), and we did not find any improvement induced by the fearful face. Overall, we provide evidence that emotion interacts with attention to affect basic visual functions.

Abstract:Fear is closely related to the evolution of the basic emotions,it is biologically adaptive responses to environmental threat and it can stimulate a series of defense mechanisms.A subcortical pathway through the superior colliculus and pulvinar to the amygdala is assumed to mediate the unconscious processing of affective visual stimuli.Instead,the researchers propose that the primary role of the amygdala in visual processing,like that of the pulvinar,is to coordinate the function of cortical networks during the evaluation of the biological significance of affective visual stimuli.So studying the neural mechanisms of the fear not only has high scientific value,but also has important clinical value.Classical fear conditioning is regarded as an important animal model to study emotional learning and memory.Previous evidence from functional magnetic resonance imaging(fMRI) studies has shown that the amygdala,doral Anterior cingulate cortex(dACC),orbitofrontal cortex(OFC) play an important role in the formation and expression of fear.Hippocampus is the core limbic regions in the encoding and consolidation of fear memories.In fear of emotional regulation in the process,the anterior cingulate(ACC) and prefrontal cortex(PFC) play an important role in the extinction of fear;The prefrontal cortex(PFC) also plays an important role in the extinction of the fear and affects the amygdala,hippocampus and other brain-related activities.Future research should focus on the following areas: such as the interaction of various brain regions,metabolism clinical research and so on.Finally,the author made on the future prospects of research on the existing paradigm for improvement and innovation and intends to integrate resting state paradigm to study the process of fear,so the neural mechanisms in consolidation and reconsolidation can be well understand.
Early Influence of Emotional Scenes on the Encoding of Fearful Expressions with Different Intensities: an Event-Related Potential Study

Abstract:Contextual affective information influences the processing of facial expressions at the relatively early stages of face processing, but the effect of the context on the processing of facial expressions with varying intensities remains unclear. In this study, we investigated the influence of emotional scenes (fearful, happy, and neutral) on the processing of fear expressions at different levels of intensity (high, medium, and low) during the early stages of facial recognition using event-related potential (ERP) technology. EEG data were collected while participants performed a fearful facial expression recognition task. The results showed that (1) the recognition of high-intensity fear expression was higher than that of medium- and low-intensity fear expressions. Facial expression recognition was the highest when faces appeared in fearful scenes. (2) Emotional scenes modulated the amplitudes of N170 for fear expressions with different intensities. Specifically, the N170 amplitude, induced by high-intensity fear expressions, was significantly higher than that induced by low-intensity fear expressions when faces appeared in both neutral and fearful scenes. No significant differences were found between the N170 amplitudes induced by high-, medium-, and low-intensity fear expressions when faces appeared in happy scenes. These results suggest that individuals may tend to allocate their attention resources to the processing of face information when the valence between emotional context and expression conflicts i.e., when the conflict is absent (fear scene and fearful faces) or is low (neutral scene and fearful faces).

Abstract:Rapid detection of evolutionarily relevant threats (e.g., fearful faces) is important for human survival. The ability to rapidly detect fearful faces exhibits high variability across individuals. The present study aimed to investigate the relationship between behavioral detection ability and brain activity, using both event-related potential (ERP) and event-related oscillation (ERO) measurements. Faces with fearful or neutral facial expressions were presented for 17 ms or 200 ms in a backward masking paradigm. Forty-two participants were required to discriminate facial expressions of the masked faces. The behavioral sensitivity index d' showed that the detection ability to rapidly presented and masked fearful faces varied across participants. The ANOVA analyses showed that the facial expression, hemisphere, and presentation duration affected the grand-mean ERP (N1, P1, and N170) and ERO (below 20 Hz and lasted from 100 ms to 250 ms post-stimulus, mainly in theta band) brain activity. More importantly, the overall detection ability of 42 subjects was significantly correlated with the emotion effect (i.e., fearful vs. neutral) on ERP (r = 0.403) and ERO (r = 0.552) measurements. A higher d' value was corresponding to a larger size of the emotional effect (i.e., fearful -neutral) of N170 amplitude and a larger size of the emotional effect of the specific ERO spectral power at the right hemisphere. The present results suggested a close link between behavioral detection ability and the N170 amplitude as well as the ERO spectral power below 20 Hz in individuals. The emotional effect size between fearful and neutral faces in brain activity may reflect the level of conscious awareness of fearful faces.
[Neural mechanisms of fear responses to emotional stimuli: a preliminary study combining early posterior negativity and electroencephalogram source network analysis]

Abstract:Fear emotion is a typical negative emotion that is commonly present in daily life and significantly influences human behavior. A deeper understanding of the mechanisms underlying negative emotions contributes to the improvement of diagnosing and treating disorders related to negative emotions. However, the neural mechanisms of the brain when faced with fearful emotional stimuli remain unclear. To this end, this study further combined electroencephalogram (EEG) source analysis and cortical brain network construction based on early posterior negativity (EPN) analysis to explore the differences in brain information processing mechanisms under fearful and neutral emotional picture stimuli from a spatiotemporal perspective. The results revealed that neutral emotional stimuli could elicit higher EPN amplitudes compared to fearful stimuli. Further source analysis of EEG data containing EPN components revealed significant differences in brain cortical activation areas between fearful and neutral emotional stimuli. Subsequently, more functional connections were observed in the brain network in the alpha frequency band for fearful emotions compared to neutral emotions. By quantifying brain network properties, we found that the average node degree and average clustering coefficient under fearful emotional stimuli were significantly larger compared to neutral emotions. These results indicate that combining EPN analysis with EEG source component and brain network analysis helps to explore brain functional modulation in the processing of fearful emotions with higher spatiotemporal resolution, providing a new perspective on the neural mechanisms of negative emotions.
Time Course of Influence on the Allocation of Attentional Resources Caused by Unconscious Fearful Faces.

Abstract:Emotionally affective stimuli have priority in our visual processing even in the absence of conscious processing. However, the influence of unconscious emotional stimuli on our attentional resources remains unclear. Using the continuous flash suppression (CFS) paradigm, we concurrently recorded and analyzed visual event-related potential (ERP) components evoked by the images of suppressed fearful and neutral faces, and the steady-state visual evoked potential (SSVEP) elicited by dynamic Mondrian pictures. Fearful faces, relative to neutral faces, elicited larger late ERP components on parietal electrodes, indicating emotional expression processing without consciousness. More importantly, the presentation of a suppressed fearful face in the CFS resulted in a significantly greater decrease in SSVEP amplitude which started about 1-1.2 s after the face images first appeared. This suggests that the time course of the attentional bias occurs at about 1 s after the appearance of the fearful face and demonstrates that unconscious fearful faces may influence attentional resource allocation. Moreover, we proposed a new method that could eliminate the interaction of ERPs and SSVEPs when recorded concurrently.
Interaction Between the Prefrontal and Visual Cortices Supports Subjective Fear

Abstract:It has been reported that threatening and non-threatening visual stimuli can be distinguished based on the multi-voxel patterns of hemodynamic activity in the human ventral visual stream. Do these findings mean that there may be evolutionarily hardwired mechanisms within early perception, for the fast and automatic detection of threat, and maybe even for the generation of the subjective experience of fear? In this human neuroimaging study, we presented participants (Fear group: N=30; No Fear group: N = 30) with 2700 images of animals that could trigger subjective fear or not as a function of individual’s idiosyncratic “fear profiles” (i.e., fear ratings of animals reported by a given participant). We provide evidence that the ventral visual stream may represent affectively neutral visual features that are statistically associated with fear ratings of participants, without representing the subjective experience of fear itself. More specifically, we show that patterns of hemodynamic activity predictive of a specific “fear profile” can be observed in the ventral visual stream whether a participant reports being afraid of the stimuli or not. Further, we found that the multivariate information synchronization between ventral visual areas and prefrontal regions distinguished participants who reported being subjectively afraid of the stimuli from those who did not. Together, these findings support the view that the subjective experience of fear may depend on the relevant visual information triggering implicit metacognitive mechanisms in the prefrontal cortex.

Early Sensory Pathways for Detection of Fearful Conditioned Stimuli: Tectal and Thalamic Relays

Abstract:Sensory stimuli acquire significance through learning. A neutral sensory stimulus can become a fearful conditioned stimulus (CS) through conditioning. Here we report that the sensory pathways used to detect the CS depend on the conditioning paradigm. Animals trained to detect an electrical somatosensory stimulus delivered to the whisker pad in an active avoidance task were able to detect this CS and perform the task when a reversible or irreversible lesion was placed in either the somatosensory thalamus or the superior colliculus contralateral to the CS. However, simultaneous lesions of the somatosensory thalamus and superior colliculus contralateral to the CS blocked performance in the active avoidance task. In contrast, a lesion only of the somatosensory thalamus contralateral to the same CS, but not of the superior colliculus, blocked performance in a pavlovian fear conditioning task. In conclusion, during pavlovian fear conditioning, which is a situation in which the aversive outcome is not contingent on the behavior of the animal, the sensory thalamus is a critical relay for the detection of the CS. During active avoidance conditioning, a situation in which the aversive outcome is contingent on the behavior of the animal (i.e., the animal can avoid the aversive event), the sensory thalamus and the superior colliculus function as alternative routes for CS detection. Thus, even from early stages of sensory processing, the neural signals representing a CS are highly distributed in parallel and redundant sensory circuits, each of which can accomplish CS detection effectively depending on the conditioned behavior.

Amygdala Adaptation and Temporal Dynamics of the Salience Network in Conditioned Fear: A Single-Trial fMRI Study

Abstract:Research in rodents has established the role of the amygdaloid complex in defensive responses to conditioned threat. In human imaging studies, however, activation of the amygdala by conditioned threat cues is often not observed. One hypothesis states that this finding reflects adaptation of amygdaloid responses over time. We tested this hypothesis by estimating single-trial neural responses over a large number of conditioning trials. Functional MRI (fMRI) was recorded from 18 participants during classical differential fear conditioning: Participants viewed oriented grayscale grating stimuli (45° or 135°) presented centrally in random order. In the acquisition block, one grating (the CS+) was paired with a noxious noise, the unconditioned stimulus (US), on 25% of trials. The other grating, denoted CS-, was never paired with the US. Consistent with previous reports, BOLD in dorsal anterior cingulate cortex (dACC) and insula, but not the amygdala, was heightened when viewing CS+ stimuli that were not paired with US compared to CS- stimuli. Trial-by-trial analysis showed that over the course of acquisition, activity in the amygdala attenuated. Interestingly, activity in the dACC and insula also declined. Representational similarity analysis (RSA) corroborated these results, indicating that the voxel patterns evoked by CS+ and CS- in these brain regions became less distinguishable over time. Together, the present findings support the hypothesis that the lack of BOLD differences in the amygdaloid complex in many studies of classical conditioning is due to adaptation, and the adaptation effects may reflect changes in large-scale networks mediating aversive conditioning, particularly the salience network.
Conscious perception of fear in faces: Insights from high-density EEG and perceptual awareness scale with threshold stimuli

Abstract:Contrary to the extensive research on processing subliminal and/or unattended emotional facial expressions, only a minority of studies have investigated the neural correlates of consciousness (NCCs) of emotions conveyed by faces. In the present high-density electroencephalography (EEG) study, we first employed a staircase procedure to identify each participant's perceptual threshold of the emotion expressed by the face and then compared the EEG signals elicited in trials where the participants were aware with the activity elicited in trials where participants were unaware of the emotions expressed by these, otherwise identical, faces. Drawing on existing knowledge of the neural mechanisms of face processing and NCCs, we hypothesized that activity in frontal electrodes would be modulated in relation to participants' awareness of facial emotional content. More specifically, we hypothesized that the NCC of fear seen on someone else's face could be detected as a modulation of a later and more anterior (i.e., at frontal sites) event-related potential (ERP) than the face-sensitive N170. By adopting a data-driven approach and cluster-based statistics to the analysis of EEG signals, the results were clear-cut in showing that visual awareness of fear was associated with the modulation of a frontal ERP component in a 150–300 msec interval. These insights are dissected and contextualized in relation to prevailing theories of visual consciousness and their proposed NCC benchmarks.

Abstract:Appropriate processing of human voices with different threat-related emotions is of evolutionarily adaptive value for the survival of individuals. Nevertheless, it is still not clear whether the sensitivity to threat-related information is present at birth. Using an odd-ball paradigm, the current study investigated the neural correlates underlying automatic processing of emotional voices of fear and anger in sleeping neonates. Event-related potential data showed that the fronto-central scalp distribution of the neonatal brain could discriminate fearful voices from angry voices; the mismatch response (MMR) was larger in response to the deviant stimuli of anger, compared with the standard stimuli of fear. Furthermore, this fear-anger MMR discrimination was observed only when neonates were in active sleep state. Although the neonates' sensitivity to threat-related voices is not likely associated with a conceptual understanding of fearful and angry emotions, this special discrimination in early life may provide a foundation for later emotion and social cognition development.
