Abstract:Stimuli-responsive polymers exhibit properties that make them ideal candidates for biosensing and molecular diagnostics. Through rational design of polymer composition combined with new polymer functionalization and synthetic strategies, polymers with myriad responsivities,e.g., responses to temperature, pH, biomolecules, CO2, light, and electricity can be achieved. When these polymers are specifically designed to respond to biomarkers, stimuli-responsive devices/probes, capable of recognizing and transducing analyte signals, can be used to diagnose and treat disease. In this review, we highlight recent state-of-the-art examples of stimuli-responsive polymer-based systems for biosensing and bioimaging.

Abstract:This review focuses on smart nano-materials built of stimuli-responsive (SR) polymers and will discuss their numerous applications in the biomedical field. The authors will first provide an overview of different stimuli and their corresponding, responsive polymers. By introducing myriad functionalities, SR polymers present a wide range of possibilities in the design of stimuli-responsive devices, making use of virtually all types of polymer constructs, from self-assembled structures (micelles, vesicles) to surfaces (polymer brushes, films) as described in the second section of the review. In the last section of this review the authors report on some of the most promising applications of stimuli-responsive polymers in nanomedicine. In particular, we will discuss applications pertaining to diagnosis, where SR polymers are used to construct sensors capable of selective recognition and quantification of analytes and physical variables, as well as imaging devices. We will also highlight some examples of responsive systems used for therapeutic applications, including smart drug delivery systems (micelles, vesicles, dendrimers...) and surfaces for regenerative medicine.

Abstract:Polymeric stimuli‐responsive systems represent a group of materials that have experienced significant advancements in recent decades. These materials exhibit a physicochemical response based on various factors, such as temperature, pH, light, magnetism, insulin, and others. The promising future of biomedical sciences lies in the rapid and continuous development of these systems. Responsive polymer systems have the ability to change properties or behavior in response to external stimuli. The properties of responsive polymer systems can be fine‐tuned by adjusting the stimuli, enabling tailored responses for specific applications. These systems have applications in drug delivery, biosensors, tissue engineering, and more, as their ability to adapt and respond to dynamic environments leads to improved performance. However, challenges such as synthesis complexity, sensitivity limitations, and manufacturing issues need to be addressed for successful implementation. In our review, we provide a comprehensive summary on stimuli‐responsive polymer systems, delving into the intricacies of their mechanisms and actions. Future developments should focus on precision medicine, multifunctionality, reversibility, bioinspired designs, and integration with advanced technologies, driving the dynamic growth of sensitive polymer systems in biomedical applications.

Abstract:The past two decades have evidenced a tremendous growth in the field of responsive polymers, which can exhibit reversible or irreversible changes in physical properties and/or chemical structures to an external stimulus such as pH, temperature, ionic strength, light irradiation, mechanical forces, electric and magnetic fields, specific analytes, external additives (ions, bioactive molecules, etc.), or a combination of them. Responsive polymers can exist in the form of solutions, gels, self-assembled nanoparticles, (multilayer) films, and bulk solids. The field of responsive polymers has nowadays evolved well beyond the demonstration of novel and interesting properties. Currently, the exploitation of useful and advanced functions, drug or gene carriers with triggered release properties, catalysis, detection and imaging, environmentally adaptive coatings, and self-healing materials, has emerged to be a more relevant subject. In this Perspective, we focus on recent developments of responsive polymer-based chemo- and biosensors, highlighting this concept with selected literature reports. Such functional polymeric materials show prominent advantages such as tunable detection sensitivity, structural stability, aqueous dispersibility, biocompatibility, processability, and facile integration into detection devices, as compared to their small molecule analogues.

Abstract:Stimuli-responsive bio-based polymeric systems are gaining considerable attention as intelligent versatile tools that show great potential in various fields. In this review, an overview is given of recent developments of stimuli-responsive bio-based polymeric systems. The characteristics of bio-based polymers in different applications are discussed and the superiority of these advanced stimuli-responsive bio-based polymeric systems is highlighted. Furthermore, several emerging applications of these systems including intelligent drug delivery, responsive food packaging and smart water treatment are discussed and the section of intelligent drug delivery is emphasized in detail. Finally, the respective prospects and limitations inherent to these systems are addressed.

Abstract:Intelligent polymers or stimuli-responsive polymers may exhibit distinct transitions in physical-chemical properties, including conformation, polarity, phase structure and chemical composition in response to changes in environmental stimuli. Due to their unique ‘intelligent’ characteristics, stimuli-sensitive polymers have found a wide variety of applications in biomedical and nanotechnological fields. This review focuses on the recent developments in biomedical application of intelligent polymer systems, such as intelligent hydrogel systems, intelligent drug delivery systems and intelligent molecular recognition systems. Abstract:There has been growing interest in polymeric systems that break down or undergo property changes in response to stimuli. Such polymers can play important roles in biological systems, where they can be used to control the release of therapeutics, modulate imaging signals, actuate movement, or direct the growth of cells. In this Perspective, after discussing the most important stimuli relevant to biological applications, we will present a selection of recent exciting developments. The growing importance of stimuli-responsive polysaccharides will be discussed, followed by a variety of stimuli-responsive polymeric systems for the delivery of small molecule drugs and nucleic acids. Switchable polymers for the emerging area of therapeutic response measurement in theranostics will be described. Then, the diverse functions that can be achieved using hydrogels cross-linked covalently, as well as by various dynamic approaches will be presented. Finally, we will discuss some of the challenges and future perspectives for the field.

Abstract:Stimuli-responsive polymers have received tremendous attention from scientists and engineers for several decades due to the wide applications of these smart materials in biotechnology and nanotechnology. Driven by the complex functions of living systems, multi-stimuli-responsive polymer materials have been designed and developed in recent years. Compared with conventional single- or dual-stimuli-based polymer materials, multi-stimuli-responsive polymer materials would be more intriguing since more functions and finer modulations can be achieved through more parameters. This critical review highlights the recent advances in this area and focuses on three types of multi-stimuli-responsive polymer materials, namely, multi-stimuli-responsive particles (micelles, micro/nanogels, vesicles, and hybrid particles), multi-stimuli-responsive films (polymer brushes, layer-by-layer polymer films, and porous membranes), and multi-stimuli-responsive bulk gels (hydrogels, organogels, and metallogels) from recent publications. Various stimuli, such as light, temperature, pH, reduction/oxidation, enzymes, ions, glucose, ultrasound, magnetic fields, mechanical stress, solvent, voltage, and electrochemistry, have been combined to switch the functions of polymers. The polymer design, preparation, and function of multi-stimuli-responsive particles, films, and bulk gels are comprehensively discussed here.

Abstract:Stimuli-responsive polymers can respond to internal stimuli, such as reactive oxygen species (ROS), glutathione (GSH), and pH, biological stimuli, such as enzymes, and external stimuli, such as lasers and ultrasound, etc., by changing their hydrophobicity/hydrophilicity, degradability, ionizability, etc., and thus have been widely used in biomedical applications. Due to the characteristics of the tumor microenvironment (TME), stimuli-responsive polymers that cater specifically to the TME have been extensively used to prepare smart nanovehicles for the targeted delivery of therapeutic and diagnostic agents to tumor tissues. Compared to conventional drug delivery nanosystems, TME-responsive nanosystems have many advantages, such as high sensitivity, broad applicability among different tumors, functional versatility, and improved biosafety. In recent years, a great deal of research has been devoted to engineering efficient stimuli-responsive polymeric nanosystems, and significant improvement has been made to both cancer diagnosis and therapy. In this review, we summarize some recent research advances involving the use of stimuli-responsive polymer nanocarriers in drug delivery, tumor imaging, therapy, and theranostics. Various chemical stimuli will be described in the context of stimuli-responsive nanosystems. Accordingly, the functional chemical groups responsible for the responsiveness and the strategies to incorporate these groups into the polymer will be discussed in detail. With the research on this topic expending at a fast pace, some innovative concepts, such as sequential and cascade drug release, NIR-II imaging, and multifunctional formulations, have emerged as popular strategies for enhanced performance, which will also be included here with up-to-date illustrations. We hope that this review will offer valuable insights for the selection and optimization of stimuli-responsive polymers to help accelerate their future applications in cancer diagnosis and treatment.

Abstract:<p>As we enter the age of artificial intelligence (AI), new technologies will be needed to "sample" environmental conditions to provide data to AI systems that will yield a specific response. This review focuses on stimuli-responsive polymers (SRPs), and their ability to react to changes in external environmental conditions by undergoing a physical and/or chemical change. We first describe various methods for SRP synthesis, which leads into a discussion of the mechanism of SRPs response to external conditions, and finally we highlight examples of their use for sensing, biosensing, and actuation. The aim of this review is to familiarize readers with some of the most recent developments in SRP design and application, which we hope will encourage further research in this field that can help overcome some of the current challenges and inspire new and creative applications for these materials.</p>

Abstract:Stimuli-responsive supramolecular polymers are ordered nanosized materials that are held together by non-covalent interactions (hydrogen-bonding, metal-ligand coordination, π-stacking and, host–guest interactions) and can reversibly undergo self-assembly. Their non-covalent nature endows supramolecular polymers with the ability to respond to external stimuli (temperature, light, ultrasound, electric/magnetic field) or environmental changes (temperature, pH, redox potential, enzyme activity), making them attractive candidates for a variety of biomedical applications. To date, supramolecular research has largely evolved in the development of smart water-soluble self-assemblies with the aim of mimicking the biological function of natural supramolecular systems. Indeed, there is a wide variety of synthetic biomaterials formulated with responsiveness to control and trigger, or not to trigger, aqueous self-assembly. The design of responsive supramolecular polymers ranges from the use of hydrophobic cores (i.e., benzene-1,3,5-tricarboxamide) to the introduction of macrocyclic hosts (i.e., cyclodextrins). In this review, we summarize the most relevant advances achieved in the design of stimuli-responsive supramolecular systems used to control transport and release of both diagnosis agents and therapeutic drugs in order to prevent, diagnose, and treat human diseases.

Abstract:Stimuli-responsive polymers (SRPs) are capable of changing their solubility, conformation, and volume in response to external stimuli. By understanding the nature of the responsivity and the synthetic challenges, new SRPs can be synthesized with enhanced responsivities at scales that will allow their use in the real world. In this review, many fundamental concepts and recent advances in SRPs for sensing and actuation are presented. A variety of sensors that can respond to physiologically relevant species and stimuli are reviewed, including temperature/pH, proteins, glucose and ions. The review also includes examples of the use of SRPs for artificial muscles, soft robots, cell culture, and drug delivery.
