Abstract:Phenolic acid is a family of nutrient and bioactive components in vegetable and fruits. Recently considerable studies found that phenolic acid contained many bioactivities, such as antioxidant, anticancer, antibacterial, anti-obesity, liver protection, improving immunity, mood modification, anti-inflammatory, antivirus and cardiovascular protection. The paper briefly introduced the structure, common form in plant and occurrence in food, emphasized especially the new researches of bioactivities.
Phenolic compounds as natural and multifunctional anti-obesity agents: A review

Abstract:Prevalence of obesity worldwide has reached pandemic proportions. Despite the increasing evidence in the implication of phenolic compounds in obesity management, the real effect is not completely understood. The available in vitro and in vivo studies have demonstrated the implication of phenolic compounds in: lowering food intake, decreasing lipogenesis, increasing lipolysis, stimulating fatty acids β-oxidation, inhibiting adipocyte differentiation and growth, attenuating inflammatory responses and suppress oxidative stress. This review encompasses the most recent evidence in the anti-obesity effect of phenolic compounds from plants to different nutraceuticals and functional foods based on the in vitro, in vivo and clinical studies. For that, this review has been focused on popular plant-based products highly consumed today such as cocoa, cinnamon, and olive oil, beverages such as red wine, tea (green, white and black tea) and Hibiscus sabdariffa L. tea, among others.

Abstract:Over the past two decades, obesity has been one of the major public health concerns in most countries. In the search for new molecules that could be used for the treatment of obesity, good perspectives have been opened up for polyphenols, a class of natural bioactive phytochemicals. Experimental and limited clinical trial evidence supports that some polyphenols such as quercetin, curcumin, and resveratrol have potential benefit functions on obesity treatment. This brief review focuses on the main functions of the above-named polyphenols on adipose tissue. These polyphenols may play beneficial effects on adipose tissue under obese condition by alleviating intracellular oxidative stress, reducing chronic low-grade inflammation, inhibiting adipogenesis and lipogenesis, and suppressing the differentiation of preadipocytes to mature adipocytes.

Abstract:Polyphenols, long-used components of medicinal plants, have drawn great interest in recent years as potential therapeutic agents because of their safety, efficacy, and wide range of biological effects. Approximately 75% of the world's population still use plant-based medicinal compounds, indicating the ongoing significance of phytochemicals for human health. This study emphasizes the growing body of research investigating the anti-adipogenic and anti-obesity functions of polyphenols. The functions of polyphenols, including phenylpropanoids, flavonoids, terpenoids, alkaloids, glycosides, and phenolic acids, are distinct due to changes in chemical diversity and structural characteristics. This review methodically investigates the mechanisms by which naturally occurring polyphenols mediate obesity and metabolic function in immunomodulation. To this end, hormonal control of hunger has the potential to inhibit pro-obesity enzymes such as pancreatic lipase, the promotion of energy expenditure, and the modulation of adipocytokine production. Specifically, polyphenols affect insulin, a hormone that is essential for regulating blood sugar, and they also play a role, in part, in a complex web of factors that affect the progression of obesity. This review also explores the immunomodulatory properties of polyphenols, providing insight into their ability to improve immune function and the effects of polyphenols on gut health, improving the number of commensal bacteria, cytokine production suppression, and immune cell mediation, including natural killer cells and macrophages. Taken together, continuous studies are required to understand the prudent and precise mechanisms underlying polyphenols' therapeutic potential in obesity and immunomodulation. In the interim, this review emphasizes a holistic approach to health and promotes the consumption of a wide range of foods and drinks high in polyphenols. This review lays the groundwork for future developments, indicating that the components of polyphenols and their derivatives may provide the answer to urgent worldwide health issues. This compilation of the body of knowledge paves the way for future discoveries in the global treatment of pressing health concerns in obesity and metabolic diseases.

Anti-diabetic and anti-obesity: Efficacy evaluation and exploitation of polyphenols in fruits and vegetables

Abstract:Polyphenols are a group of secondary plant metabolites widely present in diets and have antagonistic effects on some chronic metabolic diseases, like type 2 diabetes (T2D) and obesity. We attempt to investigate the effects of polyphenols in fruits and vegetables on reducing the risk of T2D and obesity by collecting epidemiological evidence, including cross-sectional survey (CSS), prospective cohort study (PCS), and randomized controlled trial (RCT). Further, we provide possible mechanisms for the anti-diabetic effects including protecting pancreatic β-cells, affecting glucose digestion, absorption, and uptake, and activating glucose/lipid metabolism pathways, while improving obesity by reducing lipid accumulation, regulating intestinal microflora, alleviating inflammation, and reducing food intake. Polyphenols also play an important role in the relationship between T2D and obesity. On the one hand, obesity is a low-grade chronic inflammation causing insulin resistance, so polyphenols can reduce T2D risk by improving obesity. On the other hand, obesity decreases the polyphenols bioavailability by disturbing gastrointestinal microflora, thus increasing T2D risk. These are instructive for diets and bring considerable development value. Therefore, we discussed the hotspots of polyphenols exploitation in the food industry, including masking bitter and astringent taste, ensuring stability, and improving the bioavailability, which provides ideas for polyphenols application in anti-diabetics and anti-obesity.
Anthropometric and cardiometabolic effects of polyphenols in people with overweight and obesity: an umbrella review

Abstract:Polyphenols are plant-based compounds with potential anti-inflammatory, antioxidant, and anti-obesogenic properties. However, their effects on health outcomes remain unclear. To evaluate the effects of polyphenols on anthropometric and cardiometabolic markers. Six electronic databases—namely, EMBASE, CINAHL, PubMed, Scopus, The Cochrane Library (reviews only), and Web of Science—were searched for relevant systematic reviews with meta-analyses (SRMAs). Three reviewers performed the data extraction via a data-extraction Microsoft Excel spreadsheet. An umbrella review and meta-analysis of existing SRMAs was conducted. Eighteen SRMAs published from 2015 to 2023, representing 445 primary studies and 838 unique effect sizes, were identified. Meta-analyses were conducted using random-effects models with general inverse variance. Polyphenol-containing foods were found to significantly improve weight (-0.36 kg; 95% confidence interval [CI]: −0.62, 0.77 kg; P < 0.01, I 2 = 64.9%), body mass index (−0.25 kg/m 2 ; 95% CI: −0.34, −0.17 kg/m 2 ; P < 0.001, I 2 = 82.4%), waist circumference (−0.74 cm; 95% CI: −1.34, −0.15 cm; P < 0.01, I 2 = 99.3%), low-density-lipoprotein cholesterol (−1.75 mg/dL; 95% CI: −2.56, −0.94; P < 0.001, I 2 = 98.6%), total cholesterol (−1.23 mg/dL; 95% CI: −2.00, −0.46; P = 0.002, I 2 = 94.6%), systolic blood pressure (−1.77 mmHg; 95% CI: −1.77, −0.93 mmHg; P < 0.001, I 2 = 72.4%), diastolic blood pressure (−1.45 mmHg; 95% CI: −2.09, −0.80 mmHg; P < 0.001, I 2 = 61.0%), fat percentage (−0.70%; 95% CI: −1.03, −0.36%; P < 0.001, I 2 = 52.6%), fasting blood glucose (−0.18 mg/dL; 95% CI: −0.35, −0.01 mg/dL; P = 0.04, I 2 = 62.0%), and C-reactive protein (CRP; including high-sensitivity-CRP [hs-CRP]) (−0.2972 mg/dL; 95% CI: −0.52, −0.08 mg/dL; P = 0.01, I 2 = 87.9%). No significant changes were found for high-density-lipoprotein cholesterol (−0.12 mg/dL; 95% CI: −1.44, 0.69; P = 0.67, I 2 = 89.4%) and triglycerides (−1.29 mg/dL; 95% CI: −2.74, 0.16; P = 0.08, I 2 = 85.4%). Between-study heterogeneity could be explained by polyphenol subclass differences. The findings of this umbrella review support the beneficial effects of polyphenols on anthropometric and metabolic markers, but discretion is warranted to determine the clinical significance of the magnitude of the biomarker improvements. Abstract:Phenolic compounds constitute a large and diverse group of secondary metabolites derived from phenylalanine and tyrosine and are widely distributed throughout the plant kingdom. They could be divided into different classes such as simple phenol, phenyl acetic acid, hydroxybenzoic acid, hydroxycinnamic acid, and other phenylpropanoids as well as condensed tannins (proanthocyanidins) and hydrolysable tannins, among others, depending on their basic carbon skeleton structure. Phenolic compounds in plant-based foods have been suggested to have a number of beneficial health effects including prevention of cancer, cardiovascular disease, diabetes, immune disorders, neurogenerative disease and others. These properties are largely attributable to the antioxidant activity of the phenolic compounds as well as other mechanisms of action. Therefore, nutraceuticals of plant origin may evolve to be considered a vital aspect of dietary-disease preventive food components. Agri-food industries generate substantial quantities of phenolic rich by-products, which could serve as an attractive and commercially viable source of nutraceuticals. This contribution mainly summarizes the occurrence of phenolic compounds and some other bioactives in various Agri-food by-products, their bioavailability and health benefits.

Abstract:In recent years, the increase in high-calorie diets and sedentary lifestyles has made obesity a global public health problem. An unbalanced diet promotes the production of proinflammatory cytokines and causes redox imbalance in the body. Phenolics have potent antioxidant activity and cytoprotective ability. They can scavenge free radicals and reactive oxygen species, and enhance the activity of antioxidant enzymes, thus combating the body's oxidative stress. They can also improve the body's inflammatory response, enhance the enzyme activity of lipid metabolism, and reduce the contents of cholesterol and triglyceride. Most phenolics are biotransformed and absorbed into the blood after the action by gut microbiota, then these metabolites undergo phase I and II metabolism and regulate oxidative stress by scavenging free radicals and increasing expression of antioxidant enzymes. Phenolics induce the expression of genes encoding anti-oxidant enzymes and phase II detoxification enzymes by stimulating Nrf2 to enter the nucleus and bind to the ARE after uncoupling from Keap1, thereby promoting the production of antioxidant enzymes and phase II detoxification enzymes. The absorption rate of phenolics in the small intestine is extremely low. Most phenolics reach the colon, where they interact with the microbiota and undergo a series of metabolism. Their metabolites will reach the liver via the portal vein and undergo conjugation reactions. Afterward, the metabolites reach the whole body to exert biological activity by traveling with the systemic circulation. Phenolics can promote the growth of probiotics, reduce the ratio of Firmicutes/Bacteroidetes (F/B), and improve intestinal microecological imbalance. This paper reviewed the nutritional value, bioactivity, and antioxidant mechanism of phenolics in the body, aiming to provide a scientific basis for the development and utilization of natural antioxidants and provide a reference for elucidating the mechanism of action of phenolics for regulating oxidative stress in the body. Abstract:Phenolic acids have been reported to have many biological activities, but daily intake information is scarce. In this study, the phenolic acid contents of 116 commonly consumed food in five regions of China (Beijing, Hangzhou, Wuhan, Chongqing and Guangzhou) were analyzed by high performance liquid chromatographymass spectrometry (HPLC-MS) and dietary intakes estimated. Tea had the highest total phenolic acids (TPA) content in all regions investigated. Phenolic acids were in esterified and bound forms, and hydroxycinnamic acids accounted for more than half of TPA content, except in tea and legumes. Average TPA intake was 193.50 mg/ day at the national level but ranged from 157.09 to 263.01 mg/ day among the regions. Rice, tea, and fruits were the main contributors to dietary intakes of TPA. Also, TPA intake in the period 2009-2013 increased 31.65 mg/ day compared with 2002, largely due to increased intakes of fruits, vegetables, and legumes.

Abstract:Background: Diets rich in phenolics are associated with multiple health benefits. The majority of related studies have focused nearly exclusively on soluble free and conjugated phenolics in plant-based foods, whereas a large amount of insoluble phenolics bound to food matrix in the remaining solid residues are neglected. The fate of bound phenolics in human digestive tract and health implications are not well understood. Scope and approach: In this review, we provide a thorough literature review regarding the occurrence, chemistry and potential health implications of the insoluble-bound phenolics in food matrix. Research gaps were identified and future perspectives on insoluble-bound phenolics have been proposed. Key findings and conclusions: Significant amount of phenolic compounds occurs in insoluble forms through covalently binding to cell wall structural components in plant food matrix. These bound phenolics must be released by various pretreatments such as hydrolysis by enzyme, alkali and acid, or other assisted technologies before they can be identified and characterized, and there is no standard method currently. The health benefits of insoluble-bound phenolics are not only largely governed by their intrinsic content in food matrix, but their bioaccessibility and bioavailability in human body. Insoluble-bound phenolics obtained via hydrolysis possess antioxidant and anti-inflammatory activities in vitro, and have been implicated in different health benefits. Recent studies have suggested their involvement in modulating gut microbiota and intestinal immune response as a dietary component; however, the roles of bound phenolics and phenolic metabolites in intestinal inflammation and gut health and the interplay mechanisms are still poorly understood. This contribution provides an update on the insoluble-bound phenolics in food matrix and new perspectives for future studies.

Abstract:Phenolic acids, a class of secondary metabolites containing phenolic hydroxyl and carboxyl groups, are widely distributed in higher plants. Phenolic acids are bound to structural components of the cell wall through ester and ether linkages in plant food materials, and have multiple bioactivities, such as antioxidant, antimicrobial and anticancer activity. Phenolic compounds have gained wide interest in recent years and become one of the hotspots in the field of food research. Through a series of biological regulation techniques, various phenolic acids can be enriched in plant food materials, thereby effectively improving their nutritional value. In this paper, the types, structures, biosynthesis, metabolic regulation and bioactivities of phenolic acids in plant food materials are summarized. In the end, the future directions are also discussed.
Novel insights of dietary polyphenols and obesity

Abstract:The prevalence of obesity has steadily increased over the past three decades both in the United States and worldwide. Recent studies have shown the role of dietary polyphenols in the prevention of obesity and obesity-related chronic diseases. Here, we evaluated the impact of commonly consumed polyphenols, including green tea catechins, especially epigallocatechin gallates, resveratrol and curcumin, on obesity and obesity-related inflammation. Cellular studies demonstrated that these dietary polyphenols reduce viability of adipocytes and proliferation of preadipocytes, suppress adipocyte differentiation and triglyceride accumulation, stimulate lipolysis and fatty acid β-oxidation, and reduce inflammation. Concomitantly, the polyphenols modulate signaling pathways including the adenosine-monophosphate-activated protein kinase, peroxisome proliferator activated receptor γ, CCAAT/enhancer binding protein α, peroxisome proliferator activator receptor gamma activator 1-alpha, sirtuin 1, sterol regulatory element binding protein-1c, uncoupling proteins 1 and 2, and nuclear factor-κB that regulate adipogenesis, antioxidant and anti-inflammatory responses. Animal studies strongly suggest that commonly consumed polyphenols described in this review have a pronounced effect on obesity as shown by lower body weight, fat mass and triglycerides through enhancing energy expenditure and fat utilization, and modulating glucose hemostasis. Limited human studies have been conducted in this area and are inconsistent about the antiobesity impact of dietary polyphenols probably due to the various study designs and lengths, variation among subjects (age, gender, ethnicity), chemical forms of the dietary polyphenols used and confounding factors such as other weight-reducing agents. Future randomized controlled trials are warranted to reconcile the discrepancies between preclinical efficacies and inconclusive clinic outcomes of these polyphenols.

Abstract:Polyphenols are widely known for their putative antioxidant and anti‐inflammatory effects and their potential protective role in several diseases such as type 2 diabetes, cardiovascular disease and cancer. They have also attracted significant interest as 'anti‐obesity' agents, although with mechanisms of action that have been exclusively demonstrated in animal and in vitro studies. This umbrella review aims to evaluate current evidence surrounding the role of polyphenols in obesity and weight management and to establish the usefulness of these agents in combatting obesity. A search of systematic reviews and meta‐analyses of randomised controlled trials was carried out. Nine systematic reviews (of which eight included a meta‐analysis) were included. Evidence of polyphenols' effects on reducing bodyweight is mixed, and where the effects are significant, they are numerically small and unlikely to be of help in reducing bodyweight or preventing weight gain. Future research should focus on establishing the anti‐inflammatory and antioxidant effects of polyphenols through well‐designed randomised controlled trials. Such research could be more valuable and cost‐effective since it has shown potential to improve human health.

Abstract:Objective: Phenolic compounds are essential for defense reactions, including antioxidant, anti-inflammatory, antidiabetic, anti-proliferative activity, and anti-aging activity. Coming from the class of phytomedicine, they are widespread in the plant kingdom and are commonly taken as substances in the daily diet. They are mainly found in various types of edible plants, especially fruits, vegetables, and tea. Phenolic compounds have been investigated for their effects on human health due to their beneficial effects against oxidative stress activities. The proposed main mechanism of this protective effect against harmful oxidation processes is related to the radical scavenging activity of the phenolic compounds due to reactive oxygen and nitrogen substances. The review uses up-to-date data via manual screening of the titles and abstracts of retrieved articles using string pharmaceutical and medical benefits of phenolic compounds as keywords to obtain publications from the electronic databases PubMed and Google Scholar using the publish or perish tool. However, priority has been given to the scientific papers, reports, and literature issued within the past 5 years.Several types of research have been conducted on phenolic compounds due to their potential pharmacological activities. Dietary intake of phenolic compounds is also associated with reducing the risk of cardiovascular disease, mainly caused by oxidative stress and behavioral risk factors such as alcohol abuse, tobacco use, a high-fat diet, and a sedentary lifestyle. Dietary Phenolic Compounds: Their Health Benefits and Association with the Gut Microbiota

Abstract:Oxidative stress causes various diseases, such as type II diabetes and dyslipidemia, while antioxidants in foods may prevent a number of diseases and delay aging by exerting their effects in vivo. Phenolic compounds are phytochemicals such as flavonoids which consist of flavonols, flavones, flavanonols, flavanones, anthocyanidins, isoflavones, lignans, stilbenoids, curcuminoids, phenolic acids, and tannins. They have phenolic hydroxyl groups in their molecular structures. These compounds are present in most plants, are abundant in nature, and contribute to the bitterness and color of various foods. Dietary phenolic compounds, such as quercetin in onions and sesamin in sesame, exhibit antioxidant activity and help prevent cell aging and diseases. In addition, other kinds of compounds, such as tannins, have larger molecular weights, and many unexplained aspects still exist. The antioxidant activities of phenolic compounds may be beneficial for human health. On the other hand, metabolism by intestinal bacteria changes the structures of these compounds with antioxidant properties, and the resulting metabolites exert their effects in vivo. In recent years, it has become possible to analyze the composition of the intestinal microbiota. The augmentation of the intestinal microbiota by the intake of phenolic compounds has been implicated in disease prevention and symptom recovery. Furthermore, the "brain-gut axis", which is a communication system between the gut microbiome and brain, is attracting increasing attention, and research has revealed that the gut microbiota and dietary phenolic compounds affect brain homeostasis. In this review, we discuss the usefulness of dietary phenolic compounds with antioxidant activities against some diseases, their biotransformation by the gut microbiota, the augmentation of the intestinal microflora, and their effects on the brain-gut axis.

Abstract:The consumption of phytochemicals, bioactive compounds in fruits and vegetables, has been demonstrated to ameliorate obesity and related metabolic symptoms by regulating specific metabolic pathways. This review summarizes the progress made in our understanding of the potential of phytochemicals as metabolic signals: we discuss herein selected molecular mechanisms which are involved in the occurrence of obesity that may be regulated by phytochemicals. The focus of our review highlights the regulation of transcription factors toll like receptor 4 (TLR4), nuclear factor (erythroid-derived 2)-like 2 (Nrf2), the peroxisome proliferator-activated receptors (PPARs), fat mass and obesity-associated protein (FTO) and regulation of microRNAs (miRNA). In this review, the effect of phytochemicals on signaling pathways involved in obesity were discussed on the basis of their chemical structure, suggesting molecular mechanisms for how phytochemicals may impact these signaling pathways. For example, compounds with an isothiocyanate group or an α, β-unsaturated carbonyl group may interact with the TLR4 signaling pathway. Regarding Nrf2, we examine compounds possessing an α, β-unsaturated carbonyl group which binds covalently with the cysteine thiols of Keap1. Additionally, phytochemical activation of PPARs, FTO and miRNAs were summarized. This information may be of value to better understand how specific phytochemicals interact with specific signaling pathways and help guide the development of new drugs to combat obesity and related metabolic diseases.
