Abstract:C-H bonds are ubiquitous in organic molecules. Homogenous gold-catalyzed direct functionalization of unsaturated C-H bonds has emerged as a powerful method in our synthetic toolbox. However, C-sp3-H bonds have larger dissociation energy and lower proton acidity, and thus the efficient and exquisitely selective cleavage of this kind of chemical bonds for the formation of new carbon-carbon and carbon-heteroatom bonds is still a great challenge. In this tutorial review, we will highlight the recent achievements of gold-catalyzed oxidative and redox-neutral C-sp3-H bond functionalization, which opens new avenues for economical and sustainable construction of fine chemicals.

Abstract:Compounds containing aryl structures are important organic synthesis intermediates,which are widely present in a large family of natural products and bioactive molecules.In recent years,direct asymmetric functionalization of benzylic C(sp3)-H bond for the efficient construction of arene and hetero-arene motifs with high stereoselectivity has drawn widespread concern from chemical community.Among the various strategies,small molecule-catalyzed metal-free functionalization of benzylic C(sp3)-H bond represents a more challenging but promising transformations.This review is intended to summarize and discuss the most recent advances in this area.

Abstract:C−H bond functionalization has been one of most important subject in chemistry. The carbene transfer reaction is one of the most straightforward and efficient approaches for site‐selective C−H bond functionalization in the presence of transition‐metals. Gold catalysts, which are different with the common‐used transition‐metals (such as Rh, Cu, Ru, Fe), displayed their unique catalytic activity in carbene transfer reactions, especially for C−H bond functionalization. In this focus review, we summarized recent progress of C−H bond functionalization via the decomposition of diazo compounds catalyzed by gold complexes. We also highlighted the specificity and provided the mechanistic rationale to understand these reactions.

Abstract:Abstract The selective functionalization of inert CH bonds is regarded as one of the most attractive research field due to the advantages of expanding diversity, reducing waste and synthetic economy in modifying organic compounds. However, because of the high bond energy, similar bond strengths and steric environments, the CH bond functionalization meets the challenge of controlling the regio‐ and stereoselectivity. Due to the efficient electron transfer and energy transfer, the photocatalysis could provide an accessible alternative for the regioselective functionalization of inert CH bonds. In recent years, the enantioselective functionalization of inert CH bonds by photochemistry has been developed. This article summarized this field as photocatalysis with three main strategies: transition‐metal catalysis, chiral Lewis acid catalysis, and chiral organocatalysis.

Abstract:Direct C-H bond functionalization catalyzed by non-precious transition metals is an attractive strategy in synthetic chemistry. Compared with the precious metals rhodium, palladium, ruthenium, and iridium commonly used in this field, catalysis based on non-precious metals, especially the earth-abundant ones, is appealing due to the increasing demand for environmentally benign and sustainable chemical processes. Herein, developments in iron- and cobalt-catalyzed C(sp3)-H bond functionalization reactions are described, with an emphasis on their applications in organic synthesis, i.e., the synthesis of natural products and pharmaceuticals and/or their modification.

Abstract:Direct C–H functionalization has recently emerged as one of the most efficient strategies to access structurally complex molecules from readily accessible feedstocks in an atom- and step-economic manner. In particular, enantioselective C–H activation has garnered increasing attention by enabling chemists to efficiently assemble valuable chiral compounds by asymmetrically manipulating C–H bonds into useful functionalities. Apart from the extensively studied C–C bond formation, very few endeavors have been focused on the C–X formation analogs. Motivated by the utility of the latter approach in constructing academically and industrially important heteroatom-containing chiral compounds, we provide herein an overview on C–X forming asymmetric C–H activation reactions proceeding through C–H metalation. The advancements are organized according to the employed catalytic systems, which include Pd(II) catalysis, group-9 CpxM(III) catalysis, monovalent group-9 metal catalysis, and multi-boryl/silyl Ir(III) catalysis, with emphasis on the design philosophy, mechanism, and mode of enantiocontrol.

Abstract:Gold allenylidene species have been seldom exploited as reactive intermediates in synthetically versatile catalytic reactions. By employing alkynylbenziodoxoles as the substrates and bifunctional WangPhos as the metal ligand, this work demonstrated ready catalytic access to these intermediates of general substitution patterns and their electrophilic reactivities at the γ-carbon center with a diverse range of nucleophiles. The reaction is driven by the reductive decomposition of the benziodoxole moiety and achieves the replacement of a propargylic proton with a N/O/C-based nucleophile, hence realizing reactivity umpolung. Corroborated by DFT calculations, the reaction mechanism involves a mild propargylic deprotonation. In contrast to prior works employing a tertiary amine functionality, A barely BrØnsted-basic amide group in WangPhos is surprisingly effective in deprotonation at the propargylic position under a gold-ligand cooperation regime.

Abstract:Herein, we have developed a novel and simple protocol to realize the C-H bond functionalization/dearomatization of naphthols and phenols with ortho-alkynylaryl-α-diazoesters under gold(i) catalysis. In this protocol, various spirocyclic molecules could be obtained in good yields with excellent chemo- and regio-selectivity and moderate to good diastereoselectivity.

Abstract:Transition-metal-catalyzed, coordination-assisted C(sp3)-H functionalization has revolutionized synthetic planning over the past few decades as the use of these directing groups has allowed for increased access to many strategic positions in organic molecules. Nonetheless, several challenges remain preeminent, such as the requirement for high temperatures, the difficulty in removing or converting directing groups, and, although many metals provide some reactivity, the difficulty in employing metals outside of palladium. This review aims to give a comprehensive overview of coordination-assisted, transition-metal-catalyzed, direct functionalization of nonactivated C(sp3)-H bonds by covering the literature since 2004 in order to demonstrate the current state-of-the-art methods as well as the current limitations. For clarity, this review has been divided into nine sections by the transition metal catalyst with subdivisions by the type of bond formation. Synthetic applications and reaction mechanism are discussed where appropriate.

Abstract:Here we report the first example of alkyne trifunctionalization through simultaneous construction of C–C, C–O, and C–N bonds via gold catalysis. With the assistance of a γ-keto directing group, sequential gold-catalyzed alkyne hydration, vinyl–gold nucleophilic addition, and gold(III) reductive elimination were achieved in one pot. Diazonium salts were identified as both electrophiles (N source) and oxidants (C source). Vinyl–gold(III) intermediates were revealed as effective nucleophiles toward diazonium, facilitating nucleophilic addition and reductive elimination with high efficiency. Abstract:In past decade, gold revealed more and more unique properties in carbene chemistry. It was disclosed in our recent communication (J. Am. Chem. Soc. 2014, 136, 6904) that gold carbenes have unprecedented chemo- and site-selectivity and ligand effect toward the functionalization of C-H bonds in phenols. In this full article, we report a comprehensively combined theoretical and experimental study on the mechanism of the insertion of gold carbenes into C-H and O-H bonds in phenol. It significantly revealed that the ligands have an important effect on C-H insertion and the reaction proceeds through a pathway involving the formation of an enolate-like intermediate. Moreover, two water molecules serving as a proton shuttle are believed to be the key issue for achieving chemoselective C-H functionalization, which is strongly supported by the DFT calculations and control experiments. It is the first time that a clear explanation is given about the prominent catalysis of gold carbenes toward C-H functionalization based on a theoretical and experimental study.
