Abstract:The controlled release of drugs by biostimuli is highly desirable under physiological conditions for their potential use in advanced applications. The enzyme-inspired controlled release of cucurbituril nanovalves by using magnetic mesoporous silica nanoparticles (MSNs) in near-neutral aqueous solutions is reported for the first time. The encirclement of cucurbit[7]uril (CB[7]) onto the protonated 1,4-butanediamine stalks tethered to the external surfaces of superparamagnetic Fe3O4-embedded mesoporous silica particles leads to tight blocking of the nanopores. The supramolecular nanovalves are activated by the enzymatic decarboxylation products of lysine, cadaverine (in the protonated form), which has a high affinity for CB[7], so that the encapsulated guest molecules, calcein, in the nanopores are released into the bulk solution. The release of calcein can be controlled in small portions on command by alternating changes in enzymatic decarboxylation products and CB[7]. The amino acid derived polyamines have long been associated with cell growth and cancers. The guest molecules released from the delivery system of magnetic MSNs can act not only on sensing probes for levels of decarboxylases and polyamines, but also on efficacious drugs to specific tissues and cells for regulation of polyamine synthesis.
Ph- and Competitor-Driven Nanovalves of Cucurbit[7]uril Pseudorotaxanes Based on Mesoporous Silica Supports for Controlled Release

Abstract:The controlled release of the supramolecular nanovalves of cucurbit[7]uril (CB[7]) pseudorotaxanes based on mesoporous silica MCM-41 driven by dual stimuli were investigated in detail. The encirclement of CB[7] onto the protonated 1,4-butanediamine stalks anchored on the MCM-41 surfaces led to the tight closing of nanopores. The closed pores could be activated by the methods of deprotonation and competitive binding in aqueous media, so that the entrapped model molecules calcein were released from mesopores into bulk solutions. The nanovalve closing and opening were easily detected from the change in fluorescence intensity of calcein and even directly visualized from the change in color of dispersion solutions. The increase of pH resulted in the deprotonation of the initially protonated 1,4-butanediamine units and the dissociation of the supramolecular complexes. The amount of released calcein increased with increasing pH. The competitors cetyltrimethylammonium bromide (CTAB) and 1,6-hexanediamine could activate the nanovalves at near neutral pH through the shift of the supramolecular complex equilibrium. The increases in binding affinity and concentration of the competitors resulted in the increases of release rate and efficiency. Both of the methods could facilitate the nanovalves to realize controlled release on demand. The supramolecular nanovalves driven by a combination of deprotonation and competitive binding under various conditions will have many potential applications in different fields.
Stimuli-responsive Functionalized Mesoporous Silica Nanoparticles for Drug Release in Response to Various Biological Stimuli

Abstract:A silica-based mesoporous nanosphere (MSN) controlled-release drug delivery system has been synthesized and characterized. The system uses l-cysteine derivatized gold nanoparticles (AuNPs), bound to the MSNs using Cu2+ as a bridging ion. The AuNPs serve as removable caps that hinder the release of drug molecules inside the amino functionalized MSN mesoporous framework. The modified MSNs themselves exhibit negligible cytotoxicity to living cells, as revealed using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. The drug delivery system requires one of two biological stimuli to trigger drug release. These stimuli are either: low pH (pH < 5); or elevated levels of adenosine triphosphate (ATP) (concentration > 4 mM). The feasibility of biologically controlled release was demonstrated through the stimuli-induced removal of the AuNP caps over the MSN releasing the anticancer drug doxorubicin. We envisage that this MSN system could play a significant role in developing new generations of controlled-release delivery vehicles.

Abstract:As efficient drug carriers, stimuli-responsive mesoporous silica nanoparticles are at the forefront of research on drug delivery systems. An acid-responsive system based on silyl ether has been applied to deliver a hybrid prodrug. Thiol-ene click chemistry has been successfully utilized for tethering this prodrug to mesoporous silica nanoparticles. Here, by altering the steric bulk of the substituent on the silicon atom, the release rate of a model drug, camptothecin, was controlled. The synthesized drug delivery system was investigated by analytical methods to confirm the functionalization and conjugation of the mesoporous silica nanoparticles. Herein, trimethyl silyl ether and triethyl silyl ether were selected to regulate the release rate. Under normal plasma conditions (pH 7.4), both types of camptothecin-loaded mesoporous silica nanoparticles (i.e., MSN-Me-CPT and MSN-Et-CPT) did not release the model drug. However, under in vitro acidic conditions (pH 4.0), based on a comparison of the release rates, camptothecin was released from MSN-Me-CPT more rapidly than from MSN-Et-CPT. To determine the biocompatibility of the modified mesoporous silica nanoparticles and the in vivo camptothecin uptake behavior, MTT assays with cancer cells and confocal microscopy observations were conducted, with positive results. These functionalized nanoparticles could be useful in clinical treatments requiring controlled drug release.

Abstract:New fabrication method of nanostructures is of great importance for the applications of nanoscience and nanotechnology. This review summarizes cucurbit[n]uril (CB[n])-based nanostructure fabrication and modification approaches. These strategies include the use of CB[n]s as building blocks and supramolecular crosslinkers to fabricate nanostructures, to surface modify nanostructures, and as gatekeepers to control the release of encapsulated cargo. These nanostructures are used for drug delivery, bioimaging, chemical sensing, catalysis and other applications. CB[n]s often play a vital role in the fabrication of these nanostructures, and the realization of the applications.

Abstract:A safe, biocompatible, and stimuli-responsive cucurbit[7]uril-mediated supramolecular bactericidal nanoparticle was fabricated by encapsulating a highly bioactive carbazole-decorated imidazolium salt (A1, EC50 = 0.647 μg/mL against phytopathogen Xanthomonas oryzae pv oryzae) into the host cucurbit[7]uril (CB[7]), thereby leading to self-assembled topographies from microsheets (A1) to nanospheroidal architectures (A1@CB[7]). The assembly behaviors were elucidated by acquired single-crystal structures, 1H NMR, ITC, and X-ray powder diffraction experiments. Complex A1@CB[7] displayed lower phytotoxicity and could efficiently switch on its potent antibacterial ability via introducing a simple competitor 1-adamantanamine hydrochloride (AD). In vivo antibacterial trials against rice bacterial blight revealed that A1@CB[7] could relieve the disease symptoms after being triggered by AD and provide a workable control efficiency of 42.6% at 100 μg/mL, which was superior to bismerthiazol (33.4%). These materials can provide a viable platform for fabricating diverse stimuli-responsive supramolecular bactericides for managing bacterial infections with improved safety.

Abstract:A relatively facile and adjustable approach was investigated to synthesize mesoporous silica nanoparticles (MSNs) with a thin controllable solid silica-cap (sSiO(2)) outer layer that prevented unwanted degradation for controlled drug release because the sSiO(2) turned out to degrade gradually. Ibuprofen (IBU) and calcium ions were loaded to evaluate the efficiency of this system. This sSiO(2)-capped degradation strategy for controlled release could easily be extended to other pH- or temperature-responsive MSN systems and applications.

Abstract:Noble metal nanoparticles (NMNPs), which spring up like mushrooms, are gaining momentum owing to their unique physicochemical characteristics. Cucurbiturils, a class of synthetic macrocycles with intriguing and peculiar host–guest properties, have stimulated tremendous research interest in recent years. The marriage of NMNPs with cucurbiturils is expected to integrate and enhance the excellent characteristics of both components, e.g., precisely controlled particle size, stability, assembly, surface functionality, biocompatibility, tunable optical properties, and high catalytic activities. This review systematically outlines the recent progress on the fabricating strategies and important applications of cucurbiturils‐mediated NMNPs in sensing, surface‐enhanced Raman scattering, theranostics, and catalysis. A brief outlook on the future development of cucurbiturils‐mediated NMNPs is also presented.

Abstract:Mesoporous silica has aroused lots of interest in biomedical fields, in which novel kinds of mesoporous silica with tunable mesoporosity and size have attracted much attention, but functionalization or multifunctionalization inside the mesopores of these structures is still rarely detected. Among all the functionalized molecules, beta cyclodextrin hydrate (beta-CD), a kind of hydrophilic non-toxic carrier for hydrophobic drugs, can increase the solubility and bioavailability of drugs, acting as a pH responsive "gate" when functionalized on the surface of mesoporous silica. Herein, we functionalized beta-CD inside novel kinds of mesoporous silica or magnetic mesoporous silica as the physical binding sites for the model drug curcumin through an out-inside two step bifunctionalization process including a vacuum pumping recrystallization drug loading process. According to the physical characterization with XRD, FT-IR, XPS, solid state NMR, BET, TG, DTA, TEM and DLS, the drug delivery nanosystems were successfully fabricated and contained nano-formulations of curcumin. In vitro cell testing, including Cell Counting Kit-8, Hoechst 33342 staining, hemolysis experiments and cell apoptosis, indicated that through the bifunctionalization, the biocompatibility of the mesoporous silica with cells could be improved, and the toxicity of the drug delivery nano-formulations towards two kinds of cancer cells, SK-HEP1 and HepG2, was significantly increased compared with free curcumin. In vitro release studies revealed that the nano-formulations contained more suspended curcumin molecules as described by the Higuchi models and showed enhanced pH responsive release properties, and that the magnetic nano-formulations exhibited alternating magnetically controlled release properties. The confocal microscopy analysis results revealed the obviously increasing intercellular release and uptake of both nano-formulations of curcumin by SK-HEP1 cells and the "on" and "off" stages of the alternating magnetism responsive intercellular release properties of the fabricated magnetic nano-formulations. The bifunctionalization process combined with the nanostar structure has immense potential in the drug delivery field, especially for hydrophobic drugs.

Zein-Functionalized MCM-41 Silica Nanoparticles with Enzyme-Responsive for Controlled Release in Antibacterial Activity

Abstract:This research provided a novel enzyme-responsive antimicrobial carrier aiming at overcoming the volatile loss of active antibacterial components, by employing mesoporous silica nanoparticles (MCM-41) as the matrix of encapsulation and Zein as the molecular gate. Since Zein could be consumed by bacteria, Zein-functionalized MCM-41 acted as an enzyme-responsive gate and improved the controlled-release capacity. The results showed that the amount of capsaicin released from Zein-functionalized MCM-41 without bacteria was quite low compared with the essential oils liberated with bacteria. This validated that the delivery of capsaicin was hampered by Zein and the existence of Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) promoted the release of encapsulated cargo. The release rate of capsaicin in Zein-functionalized MCM-41 climbed with the growth velocity of bacteria. These functions were realized in the form of controlled diffusion of essential oils encapsulated in MCM-41 by electrostatic interaction, and Zein was performed by both covalent bonding interaction and electrostatic interaction. Zein-functionalized MCM-41 was 2.4 times more effective in killing E. coli and 1.2 times more effective in inhibiting S. aureus than an equal amount of free capsaicin, and possessed a long-lasting antibacterial activity. The responsive antimicrobial material might be used as a promising preservative in the food industry for antimicrobial activity enhancement.

Abstract:Core@shell nanoparticles with superparamagnetic iron oxide core, mesoporous silica shell, and crown ether periphery were fabricated toward drug delivery and tumor cell imaging. By the concept of nanovalve based on supramolecular gatekeeper, stimuli-responsive drug delivery nanosystems Fe3O4@SiO2@meso-SiO2@crown ethers were synthesized by (i) modified solvothermal reaction; (ii) sol-gel reaction; and (iii) amide coupling reaction. The successful coupling of the dibenzo-crown ethers onto the mesoporous silica shell was confirmed by thermogravimetric analysis and Infrared spectroscopy. In this system, the "ON/OFF" switching of the gatekeeper supramolecules can be controlled by pH-sensitive intramolecular hydrogen bonding or electrostatic interaction (such as metal chelating). Biological evaluation of the nanoparticles renders them noncytotoxic and can be uptaken by L929 cells. In this work, the antitumor drug (doxorubicin) loading and release profiles which were studied by the UV/visible absorption spectroscopy. The mechanism involves the best-fit binding of crown ethers with cesium or sodium ions at different pH values with ultrasonic wave in phosphate buffered saline (PBS). Magnetic resonance imaging analysis of the particles reveals a high relaxivity, rendering them potentially useful theranostic agents.
Cucurbit[8]uril As Nanocontainer in a Polyelectrolyte Multilayer Film: a Quantitative and Kinetic Study of Guest Uptake.

Abstract:The host-guest chemistry of cucurbit[8]uril (CB[8]) and the layer-by-layer self-assembly technique are combined to obtain a molecular imprinted polyelectrolyte multilayer film for the recognition and binding of a guest molecule. Cucurbit[8]uril as a ready-made binding site is first associated with a polyelectrolyte and then assembled into a polyelectrolyte multilayer film via layer-by-layer deposition. A cationic guest is subsequently included into the nanocontainer due to specific host-guest interactions. The quantitative analysis of both CB[8] and the included guest molecule in dependence of the surface charge of the multilayer film identifies a high nanocontainer density as well as good to excellent binding efficiencies, therefore yielding a promising imprinted nanomaterial with potential applications in filtration or sensor technology. The investigation of the guest molecule uptake kinetics reveals two processes on different time scales, respectively, which are again related to the charge of the multilayer film surface. The combination of the results obtained from both ultraviolet spectroscopy and dissipative quartz crystal microbalance enables us to describe a full picture of several simultaneous processes initiated by the guest molecule.

Influence of molecular interactions on structure, controlled release and cytotoxicity of curcumin encapsulated chitosan - Silica nanostructured microspheres

Abstract:Curcumin possesses numerous medicinal benefits including anti-cancer and anti-viral properties. However, its wide scale use as a drug is often hindered owing to the dearth of suitable drug-delivery systems which can solubilise it for long-term sustained-release and safeguard its beneficial properties. In this work, a fast, one-step method, employing evaporation induced assembly of colloids, has been employed for the synthesis of curcumin encapsulated organic-inorganic hybrid micron-sized spheres. Detailed physical properties of the microspheres, with scaffolds of silica nanoparticles (∼8.5 nm) cross linked by chitosan, are studied to trace the underlying mechanism of structural assembly in such systems, by tuning the polymer matrix with solubilizing agents, DMSO and Tween 20. A systematic modification in the hydrogen bonding network, conformations and interactions between macromolecules is revealed upon tuning the organic matrix. This in turn is found to control the assembly vis-à-vis the granular morphology, drug entrapment and packing fraction of nanoparticles in the microspheres, which have direct influence on the biological properties. Consequently, the microspheres are found to follow a first order drug release kinetics with a tunable rate constant which follows the ordering of packing fraction of silica nanoparticles in the micro-granules. A sustained curcumin release for a period extending up to 24 h has been achieved. Further studies using human lung and cervical cancer cell lines assert good anti-cancer properties of these nanostructured microspheres in cancer cells, while showing no toxicity towards normal cells. Thus, such hybrid organic-inorganic formulations achieved using multi-component colloidal assembly approach, with enhanced stability against degradation, are promising candidates for future clinical applications of water-insoluble drugs.

Abstract:The control over chemical reactivity and selectivity are always pursued. Using non-covalent interactions to achieve efficient and selective catalysis is an essential goal of supramolecular catalysis. Supramolecular catalysis based on cucurbit[n]urils (CB[n]s) possesses distinct characteristics for the unique structure of CB[n]s. CB[n]s are a family of pumpkin-shaped host molecules with various molecular sizes, rigid structures, electronegative portals and wealthy host-guest chemistry. Herein, we summarize the three major mechanisms of CB[n]s based supramolecular catalysis. Owing to the structural properties of CB[n]s, CB[n]s can serve as nanoreactors and steric hindrance to modulate the reactivity of substrates. They can also catalyze the reactions by modulating the reactivity of ionized intermediates. Recent progresses on the CB[n]s based supramolecular catalysis are introduced in this Minireview and the future development in this field is discussed.
