Effects of Exogeneous Hormones on Seed Germination and Seedling Growth of Valeriana officinalis L.under Drought Stress

Abstract:In order to explore the physiological mechanism of exogenous indolebutyric acid(IBA) and abscisic acid(ABA) on the adapation of Valeriana officinalis to drought stress, the effect of different concentrations(0, 5, 10, 50, 100, 200 mg/L) of IBA and ABA on seed germination were studied by soaking seeds. The effects of PEG-6000 solutions with different concentrations(0, 10%, 15%, 20% and 25%) on the seed germination of V. officinalis were studied, and its semi-lethal concentration was determined. Then, the effects of exogenous addition of IBA and ABA on seed germination, seedling growth and development, contents of osmoregulation substances in leaves and antioxidant enzyme activities of V. officinalis under the semi-lethal concentration drought stress were studied. The results showed that 5 mg/L IBA and 100 mg/L ABA treatments could significantly promote the seed germination and increase germination rate. The 20% PEG-6000 stress could inhibit seed germination to a great extent, which could be used as the semi-lethal concentration for drought stress. Exogenous addition of 5 mg/L IBA and 100 mg/L ABA could significantly improve the germination efficiency of V. officinalis seeds under the semi-lethal concentration drought stress, and improve the germination rate, germination potential and germination index. Appropriate concentration of IBA and ABA could increase the content of soluble sugar, soluble protein and free proline in V. officinalis leaves under drought stress, enhance its ability to resist drought stress, improve the activity of protective enzymes such as SOD, POD and CAT, remove excessive reactive oxygen species and free radicals in the leaves in time, and reduce MDA content, and then alleviated the oxidative damage caused by drought stress to the seeds. In summary, exogenous addition of suitable concentrations of IBA and ABA under drought stress could improve the ability of drought resistance of V. officinalis.
