Abstract:Drought is one of the main disasters affecting wheat production. As a new type of plant hormone, SLs can effectively regulate the drought resistance of plants. However, so far, there are few studies on its effects on the growth and development of wheat. Aiming at the above situation, this study used foliar spraying to study its effects on the drought resistance metabolism of wheat under different concentration conditions. The results showed that exogenous SLs could not only improve root structure and promote root growth, but also increase the activities of enzymes such as SOD, POD and CAT in leaves, reduce the content of MDA, increase the content of Proline, and then improve the growth of plants. Water content, promote the improvement of photosynthetic pigments, and ensure the effective process of plant water and energy metabolism, thereby improving the drought resistance level of wheat at seedling stage. This study not only provides a theoretical basis for the application of SLs in wheat drought resistance, but also provides some technical support for its field cultivation in wheat.

Strigolactone Alleviates Herbicide Toxicity Via Maintaining Antioxidant Homeostasis in Watermelon (citrullus Lanatus)

Abstract:Strigolactone (SL) plays essential roles in regulating plant growth, development, and stress response. This study was conducted to evaluate the effect of exogenous SL on watermelon resistance against herbicides penoxsulam (PXL) and bensulfuron-methyl (BSM). These herbicides were found to negatively impact watermelon root growth and photosynthetic pigments, probably due to the ultrastructural damage and cell death in leaf and root tissues under herbicide stresses. The activation of SL-related gene expression suggested that the SL pathway may mitigate herbicide toxicity in watermelon. The exogenous SL dose-dependently reversed the PXL- or BSM-induced antioxidant activity, suggesting that SL may participate in maintaining antioxidant enzyme homeostasis under herbicide stresses. The up-regulation of herbicide metabolization and detoxification-related genes (cytochrome P450 and acetolactate synthase) by exogenous SL also in part explained how this phytohormone alleviates herbicide toxicity in watermelon. Our findings will provide valuable information underlying the regulatory effects of SL on herbicide tolerance in Cucurbitaceae crops.

Abstract:Drought poses a significant challenge to global wheat production, and the application of exogenous phytohormones offers a convenient approach to enhancing drought tolerance of wheat. However, little is known about the molecular mechanism by which strigolactones (SLs), newly discovered phytohormones, alleviate drought stress in wheat. Therefore, this study is aimed at elucidating the physiological and molecular mechanisms operating in wheat and gaining insights into the specific role of SLs in ameliorating responses to the stress. The results showed that SLs application upregulated the expression of genes associated with the antioxidant defense system (Fe/Mn-SOD, PER1, PER22, SPC4, CAT2, APX1, APX7, GSTU6, GST4, GOR, GRXC1, and GRXC15), chlorophyll biogenesis (CHLH, and CPX), light-harvesting chlorophyll A-B binding proteins (WHAB1.6, and LHC Ib-21), electron transfer (PNSL2), E3 ubiquitin-protein ligase (BB, CHIP, and RHY1A), heat stress transcription factor (HSFA1, HSFA4D, and HSFC2B), heat shock proteins (HSP23.2, HSP16.9A, HSP17.9A, HSP21, HSP70, HSP70-16, HSP70-17, HSP70-8, HSP90-5, and HSP90-6), DnaJ family members (ATJ1, ATJ3, and DJA6), as well as other chaperones (BAG1, CIP73, CIPB1, and CPN60I). but the expression level of genes involved in chlorophyll degradation (SGR, NOL, PPH, PAO, TIC55, and PTC52) as well as photorespiration (AGT2) was found to be downregulated by SLs priming. As a result, the activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) were enhanced, and chlorophyll content and photosynthetic rate were increased, which indicated the alleviation of drought stress in wheat. These findings demonstrated that SLs alleviate drought stress by promoting photosynthesis through enhancing chlorophyll levels, and by facilitating ROS scavenging through modulation of the antioxidant system. The study advances understandings of the molecular mechanism underlying SLs-mediated drought alleviation and provides valuable insights for implementing sustainable farming practice under water restriction.

Effects of Exogenous 24-Epibrassinolide on the Leaf Photosynthetic Characteristics and Polyamines Content of Cucumber Seedlings under Hypoxia Stress

Abstract:A water culture experiment with 1/2 Hoagland solution was carried out to study the effects of exogenous 24-epibrassinolide (EBR) on the leaf photosynthetic characteristics and polyamines (PAs) content of cucumber seedlings under hypoxia stress. Under the stress, the leaf net photosynthetic rate (Pn), stomatal conductance (g(s)), transpiration rate (Tr), and intercellular CO2 concentration (Ci) decreased significantly while the chlorophyll content was in adverse, and the seedling growth was inhibited. Hypoxia stress increased the leaf Put, Spd, and Spm contents and Put/PAs ratio significantly, but decreased the (Spd+Spm)/Put. Under the stress, exogenous EBR not only increased the Pn, g(s), Tr, and chlorophyll content significantly, but also increased the contents of free Spm, conjugated Spd and Spm, and bound Put, Spd, and Spm significantly. Meanwhile, the PAs content and (Spd + Spm)/Put ratio were further increased, and the Put/PAs ratio decreased. The results suggested that exogenous EBR could regulate the formation and form transformation of PAs in cucumber seedling leaf, made the leaf keep a higher photosynthetic performance, promote the increase of leaf area and dry mass, and alleviate the damage of hypoxia stress.

Abstract:Wheat is one of the main crops in northern China, and drought has been a major problem in wheat cultivation. As a new type of plant hormone, Strigolactone (SLs) , its effect on drought resistance of wheat needs to be studied extensively. In this study, the effects of SLs on the growth and development of wheat from florescence stage to maturity stage were determined by plant physiological and enzyme-linked immunoassay techniques. The results showed that, with the progress of reproductive development, leaf spraying exogenous SLs not only delayed the peak of ABA content, promoted the increase of IAA and GA content in the early stage, changed the metabolic balance of hormone in leaves, and thus changed the physiological metabolic process. Especially under the condition of high density, the effects of SLs were more obvious. And, with the change of the blade metabolic balance, its net photosynthetic rate and the level of carbon dioxide fixation increased, and SOD and POD activity enhanced, MDA content decreased, indicating that aging rate of leaves was slowed down and matter accumulation ability enhanced, so as to improve the filling rate and increase the weight of the mature grain, eventually promoted the yield of wheat. Through the above analysis, it can be seen that exogenous SLs can effectively improve the yield of wheat under high density condition, which provides reliable theoretical and technical support for its practical application in the field.

Abstract:Background: Drought is an important factor restricting the development of agriculture. In recent years, plant growth regulators have achieved more results in improving crop stress resistance. The physiological mechanism of exogenous application of mepiquat chloride on soybean seedlings has undergone significant changes under drought stress. Methods: In order to study the effect of exogenous chlorine on the physiology of soybean under drought stress., ‘Heinong 65’ was used as the experimental material, 15% PEG-6000 was used to simulate drought stress and different concentrations of mepiquat chloride solution (100, 300, 500, 700 mg/L) were sprayed at the three-leaf stage of soybean growth by pot method. The dry matter accumulation, auxin (IAA), zeatin (ZA), gibberellin (GA3), abscisic acid (ABA) content and superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), ascorbate peroxidase (APX) activity of soybean were determined. Result: The results showed that the activities of antioxidant enzymes SOD, POD, CAT and APX in soybean leaves under drought stress were significantly increased by the treatment of appropriate concentration of mepiquat chloride solution. IAA, GA3 and ZA endogenous hormone levels increased, dry matter accumulation increased, root-shoot ratio and ABA endogenous hormone levels decreased significantly. It can be seen that mepiquat chloride treatment can improve the antioxidant capacity, promote the growth of soybean seedlings under drought stress and effectively alleviate the inhibition of drought stress on soybean growth and development.

Abstract:The fodder soybean (Glycine max) is an excellent leguminous forage with a high protein content and hay yield, cultivated comprehensively in alpine regions, but seasonal drought in northern regions severely impacts the growth of seedlings. Melatonin (MT) and strigolactone (SL) are critical in relieving the restraint of plant growth in water-deficient environments, but the mechanisms of MT- and SL-mediated drought resistance in fodder soybean needs to be explored. This study mainly investigated the potential morphophysiological mechanism of MT and SL treatments in protecting fodder soybean from drought stress. The fodder soybean ‘Gongnong 535’ was treated with 100 μM MT or 1 μM SL under normal, moderate, and severe water deficit conditions. The results showed that MT and SL treatments enhanced the plant growth parameters and stomatal aperture under drought stress. Moreover, the observed reductions in superoxide ion (O2.-), malondialdehyde (MDA), and relative electrical conductivity (REC), along with enhancements in the activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT), as well as higher levels of ascorbate (AsA), glutathione (GSH), soluble sugar (SS), soluble protein (SP), and free proline (Pro), indicated that MT and SL application effectively alleviated the oxidative damage and prevented the cell membrane disruption caused by drought stress. Additionally, MT and SL treatments improved photosynthesis and growth in fodder soybean seedlings under water stress by adjusting chlorophyll pigments, gas exchange indexes, and chlorophyll fluorescence parameters, as well as endogenous hormone levels. Simultaneously, MT and SL influenced the expression of genes associated with photosynthesis and antioxidant defenses, as well as phytohormone concentrations. Notably, the protective effect of the SL treatment was superior to that of MT in water-deficient conditions. This study contributes to further understanding the defensive mechanism of MT and SL against drought stress.

Abstract:All around the world, salt stress has severely restricted plant development. Cotton is one of the most important cash crops, but the saline soil negatively influences the quality and output of plants. Strigolactones (SLs) is a new type of phytohormone that plays an important role in the development of plants. However, the underlying mechanism of how SLs alleviate the high salinity stress is unclear in cotton. In this research, we observed that exogenous 10 mu\M GR24 (a synthetic analog of SLs) significantly alleviated the salt stress (200 mM NaCl) in the hydroponically planted seedlings. Under salt stress, the contents of proline, O2 malondialdehyde, and hydrogen peroxide were greatly decreased by 48.40%, 45.57%, 31.03%, and 35.52%. Combined with transcriptome analysis and corresponding verification, it was figured out that with the application of exogenous GR24, the genes encoding antioxidant enzymes, chlorophyll biosynthesis, and photosynthesis system were significantly upregulated, while the activities of antioxidant enzymes, the content of chlorophyll, and the efficiency of photosynthesis were also significantly improved under salt stress in cotton seedlings. Thus, the exogenous application of SLs could enhance the salt tolerance of cotton seedlings, which displayed its great potential for cotton production in salinized land.

Abstract:Abstract This paper is to investigate the physiological mechanism of exogenous abscisic acid (ABA) on alleviating drought stress in Gynura cusimbua The effects of exogenous ABA on physiological characteristics and chlorophyll fluorescence parameters of the leaves of Gynura cusimbua under drought stress were studied with 10% polyethylene glycol 6000 (PEG 6000) in simulated drought stress. The results showed that the activities of antioxidant enzymes and some fluorescence parameters increased slightly after spraying exogenous ABA. Under drought stress, exogenous ABA can increase chlorophyll content, enhance antioxidant enzyme activity, reduce MDA content, increase maximum photochemical efficiency Fv / FM, regulatory energy dissipation Y (NPQ), photochemical quenching QL, actual quantum efficiency Y (II), non-photochemical quenching NPQ fluorescence parameters, and reduce non-regulatory energy dissipation Y (NO) and minimum fluorescence Fo parameters, so as to reduce the damage to seedlings under drought stress. Among them, the ABA concentration of 200 μmol/L was the best to alleviate 10% drought stress.
Foliar application of strigolactones improves the desiccation tolerance, grain yield and water use efficiency in dryland wheat through modulation of non-hydraulic root signals and antioxidant defense

Abstract:Abstract Non-hydraulic root signals (nHRS) are affirmed as a unique positive response to soil drying, and play a crucial role in regulating water use efficiency and yield formation in dryland wheat production. Strigolactones (SLs) can enhance plant drought adaptability. However, the question of whether strigolactones enhance grain yield and water use efficiency by regulating nHRS and antioxidant defense systems in dryland wheat remains unanswered. In this study, pot experiments were conducted to investigate the effects of strigolactones on nHRS, antioxidant defense system, and grain yield and water use efficiency in dryland wheat. The results showed that external application of SLs increased drought-induced abscisic acid (ABA) accumulation and activated an earlier trigger of nHRS at 73.4% field capacity (FC), compared to 68.5% FC in the control group (CK). This phenomenon was mechanically associated with the physiological mediation of SLs. The application of SLs significantly enhanced the activities of leaf antioxidant enzymes, reduced ROS production, and mitigated oxidative damage to lipid membrane. Additionally, root biomass, root length density, and root to shoot ratio were increased under strigolactone treatment. Furthermore, exogenous application of SLs significantly increased grain yield by 34.9% under moderate drought stress. Water use efficiency was also increased by 21.5% and 33.3% under moderate and severe drought conditions respectively, compared to the control group (CK). The results suggested that the application of strigolactones triggered earlier drought-sensing mechanism and improved the antioxidant defense ability, thus enhancing grain yield and water use efficiency in dryland wheat production.
Combined analyses of transcriptome and metabolome reveal the mechanism of exogenous strigolactone regulating the response of elephant grass to drought stress

Abstract:Elephant grass is widely used in feed production and ecological restoration because of its huge biomass and low occurrence of diseases and insect pets. However, drought seriously affects growth and development of this grass. Strigolactone (SL), a small molecular phytohormone, reportedly participates in improving resilience to cope with arid environment. But the mechanism of SL regulating elephant grass to response to drought stress remains unknown and needs further investigation. We conducted RNA-seq experiments and identified 84,296 genes including 765 and 2325 upregulated differential expression genes (DEGs) and 622 and 1826 downregulated DEGs, compared drought rehydration with spraying SL in roots and leaves, respectively. Combined with targeted phytohormones metabolite analysis, five hormones including 6-BA, ABA, MeSA, NAA, and JA had significant changes under re-watering and spraying SL stages. Moreover, a total of 17 co-expression modules were identified, of which eight modules had the most significant correlation with all physiological indicators with weighted gene co-expression network analysis. The venn analysis revealed the common genes between Kyoto Encyclopedia of Genes and Genomes enriched functional DEGs and the top 30 hub genes of higher weights in eight modules, respectively. Finally, 44 DEGs had been identified as key genes which played a major role in SL response to drought stress. After verification of its expression level by qPCR, six key genes in elephant grass including PpPEPCK , PpRuBPC , PpPGK , PpGAPDH , PpFBA , and PpSBPase genes regulated photosynthetic capacity under the SL treatment to respond to drought stress. Meanwhile, PpACAT , PpMFP2 , PpAGT2 , PpIVD , PpMCCA , and PpMCCB regulated root development and phytohormone crosstalk to respond to water deficit conditions. Our research led to a more comprehensive understanding about exogenous SL that plays a role in elephant grass response to drought stress and revealed insights into the SL regulating molecular mechanism in plants to adapt to the arid environment.

Abstract:Drought stress is a significant abiotic stressor that hinders growth, development, and crop yield in soybeans. Strigolactones (SLs) positively regulate plant resistance to drought stress. However, the impact of foliar application of SLs having different concentrations on soybean growth and metabolic pathways related to osmoregulation remains unknown. Therefore, to clarify the impact of SLs on soybean root growth and cellular osmoregulation under drought stress, we initially identified optimal concentrations and assessed key leaf and root indices. Furthermore, we conducted transcriptomic and metabolic analyses to identify differential metabolites and up-regulated genes.

Abstract:Salinity-alkalinity stress can remarkably affect the growth and yield of apple. Strigolactone (SL) is a class of carotenoid-derived compounds that functions in stress tolerance. However, the effects and mechanism of exogenous SL on the salinity-alkalinity tolerance of apple seedlings remain unclear. Here, we assessed the effect of SL on the salinity-alkalinity stress response of Malus hupehensis seedlings. Results showed that treatment with 100 μM exogenous SL analog (GR24) could effectively alleviate salinity-alkalinity stress with higher chlorophyll content and photosynthetic rate than the apple seedlings without GR24 treatment. The mechanism was also explored: First, exogenous GR24 regulated the expression of Na+/K+ transporter genes and decreased the ratio of Na+/K+ in the cytoplasm to maintain ion homeostasis. Second, exogenous GR24 increased the enzyme activities of superoxide, peroxidase and catalase, thereby eliminating reactive oxygen species production. Third, exogenous GR24 alleviated the high pH stress by regulating the expression of H+-ATPase genes and inducing the production of organic acid. Last, exogenous GR24 application increased endogenous acetic acid, abscisic acid, zeatin riboside, and GA3 contents for co-responding to salinity-alkalinity stress indirectly. This study will provide important theoretical basis for analyzing the mechanism of exogenous GR24 in improving salinity-alkalinity tolerance of apple.

Abstract:Exogenous strigolactones (SLs, GR24) are widely used to alleviate drought stress in wheat. The physiological and biochemical mechanisms via which SLs help overcome drought stress in wheat shoots have been reported; however, the mechanisms in wheat roots are unclear. The present study explored the effects of the exogenous application of SLs on wheat roots' growth and molecular responses under drought stress using physiological analysis and RNA-seq. RNA-seq of roots showed that SLs mainly upregulated signal transduction genes ( SIS8 , CBL3 , GLR2.8 , LRK10L-2.4 , CRK29 , and CRK8 ) and transcription factors genes ( ABR1 , BHLH61 , and MYB93 ). Besides, SLs upregulated a few downstream target genes, including antioxidant genes ( PER2 , GSTF1 , and GSTU6 ), cell wall biogenesis genes ( SUS4 , ADF3 , UGT13248 , UGT85A24 , UGT709G2 , BGLU31 , and LAC5 ), an aquaporin-encoding gene ( TIP4-3 ), and dehydrin-encoding genes ( DHN2 , DHN3 , and DHN4 ). As a result, SLs reduced oxidative damage, optimized root architecture, improved leaf-water relation, and alleviated drought damage. Thus, the present study provides novel insights into GR24-mediated drought stress management and a scientific basis for proposing GR24 application.

Abstract:This study comprehensively investigates the physiological and molecular regulatory mechanisms of Camellia oleifera seedlings under drought stress with a soil moisture content of about 30%, where exogenous abscisic acid (ABA) was applied via foliar spraying at concentrations of 50 μg/L, 100 μg/L, and 200 μg/L. The results demonstrated that appropriate concentrations of ABA treatment can regulate the physiological state of the seedlings through multiple pathways, including photosynthesis, oxidative stress response, and osmotic balance, thereby aiding in the restructuring of their drought response strategy. ABA treatment effectively activated the antioxidant system by reducing stomatal conductance and moderately inhibiting the photosynthetic rate, thus alleviating oxidative damage caused by drought stress. Additionally, ABA treatment promoted the synthesis of osmotic regulators such as proline, maintaining cellular turgor stability and enhancing the plant's drought adaptability. The real-time quantitative PCR results of related genes indicated that ABA treatment enhanced the plant's response to the ABA signaling pathway and improved disease resistance by regulating the expression of related genes, while also enhancing membrane lipid stability. A comprehensive evaluation using a membership function approach suggested that 50 μg/L ABA treatment may be the most-effective in mitigating drought effects in practical applications, followed by 100 μg/L ABA. The application of 50 μg/L ABA for 7 h induced significant changes in various biochemical parameters, compared to a foliar water spray. Notably, superoxide dismutase activity increased by 17.94%, peroxidase activity by 30.27%, glutathione content by 12.41%, and proline levels by 25.76%. The content of soluble sugars and soluble proteins rose by 14.79% and 87.95%, respectively. Additionally, there was a significant decrease of 31.15% in the malondialdehyde levels.

Abstract:Drought alters plant physiology, morphology, and biochemical pathways, necessitating effective mitigation strategies. Strigolactones (SLs) are phytohormones known to enhance plant growth under abiotic stress. However, their specific impact on drought stress in maize remains unclear. This study aimed to determine the optimal SL concentration for mitigating drought stress in two maize hybrids (HY-1898, FH-1046). Maize plants were subjected to 60% field capacity drought stress in a pot experiment. After 40 d, different concentrations (0, 0.001, 0.01, and 0.1 mg L −1 ) of the synthetic SL analogue GR24 were applied to evaluate their effects on growth features, photosynthesis attributes, and osmolyte accumulation in the maize hybrids. Results showed that exogenous SL application significantly increased photosynthetic pigments in maize hybrids under drought stress. Chlorophyll content, gas exchange characteristics, net CO 2 assimilation rate, stomatal conductance, water use efficiency, and antioxidant activities were enhanced by GR24. Leaf ascorbic acid and total phenolics also increased with SL application. Organic osmolytes, such as glycine betaine and free proline, were elevated in both maize hybrids under drought stress. Yield-related parameters, including cob diameter, cob weight, number of seeds per cob, and number of seeds per plant, were significantly increased by GR24 under drought stress. Our findings highlight the potential of GR24 foliar application to mitigate drought stress and promote maize growth and grain yield in a concentration-dependent manner. The minimum effective SL concentration against drought stress was determined to be 0.01 mg L −1 . Overall, foliar application of GR24 could serve as a sustainable approach for drought tolerance in agriculture.

Abstract:Understanding the physiological and biochemical responses of tree seedlings under extreme drought stress, along with recovery during rewatering, and potential intra-species differences, will allow us to more accurately predict forest responses under future climate change. Here, we selected seedlings from four provenances (AH (Anhui), JX (Jiangxi), HN (Hunan) and GX (Guangxi)) of Schima superba and carried out a simulated drought-rewatering experiment in a field-based rain-out shelter. Seedlings were progressively dried until they reached 50% and 88% loss of xylem hydraulic conductivity (PLC) (i.e. P 50 and P 88 ), respectively, before they were rehydrated and maintained at field capacity for 30 days. Leaf photosynthesis ( A sat ), water status, activity of superoxide dismutase (SOD), and proline (Pro) concentration were monitored and their associations were determined. Increasing drought significantly reduced A sat , relative water content (RWC) and SOD activity in all provenances, and Pro concentration was increased to improve water retention; all four provenances exhibited similar response patterns, associated with similar leaf ultrastructure at pre-drought. Upon rewatering, physiological and biochemical traits were restored to well-watered control values in P 50 -stressed seedlings. In P 88 -stressed seedlings, Pro was restored to control values, while SOD was not fully recovered. The recovery pattern differed partially among provenances. There was a progression of recovery following watering, with RWC firstly recovered, followed by SOD and Pro, and then A sat , but with significant associations among these traits. Collectively, the intra-specific differences of S. superba seedlings in recovery of physiology and biochemistry following rewatering highlight the need to consider variations within a given tree species coping with future more frequent drought stress.

Abstract:We exposed seedlings of Cotinus coggygria var. cinerea to drought and exogenous abscisic acid (ABA) under two different light conditions. Two watering regimes (well-watered and drought), two exogenous ABA applications (no ABA and with ABA) and two light regimes (full sunlight and shade) were employed. Compared with well-watered treatment, drought treatment significantly reduced the relative growth rate, relative water content (RWC), net photosynthesis rate (A) and transpiration (E), but increased chlorophyll a (chla), carbon isotope (delta C-13), endogenous ABA, malondialdehyde (MDA) and hydrogen peroxide (H2O2) contents, and guaiacol peroxidase (POD) and catalase (CAT) activities. There was an apparent alleviation of drought effects by shade, as indicated by the lower relative growth rate, and chlorophyll, MDA and H2O2 contents, and increases in indoleacetic acid (IAA) and reduced glutathione (GSH) contents. On the other hand, the exogenous ABA application under shade induced protective effects on drought-stressed seedlings, as visible in RWC, MDA, A, stomatal conductance (g(s)), E, delta C-13, ABA and IAA values. In all, our results suggest that seedlings of C coggygria are more sensitive to drought under full-light than under shade. (C) 2010 Elsevier B.V. All rights reserved.

Abstract:Abstract Neurons of the mammalian central nervous system (CNS) are an essential and largely nonrenewable cell population. Thus, viral infections that result in neuronal depletion, either by viral lysis or by induction of the cytolytic immune response, would likely lead to profound neurologic impairment. However, many viral infections that result in tissue destruction elsewhere in the host produce few overt symptoms in the CNS, despite readily detectable virus expression. This observation has lead to the speculation that neurons possess strategies to limit the replication and spread of otherwise cytopathic viruses. These strategies either favor the clearance of virus in the absence of appreciable neuronal loss or promote the establishment of noncytolytic persistent infections. This review discusses some of these strategies, with an emphasis on how such survival techniques lessen the potential for CNS neuropathology
