Abstract:Drought is a recurring climatic hazard that reduces the crop yields. To avoid the negative effects of drought on crop production, extensive efforts have been devoted to investigating the complex mechanisms of gene expression and signal transduction during drought stress. Receptor-like kinases (RLKs) play important roles in perceiving extracellular stimuli and activating downstream signalling responses. The rice genome contains >1100 RLK genes, of which only two are reported to function in drought stress. A leucine-rich repeat (LRR)-RLK gene named Leaf Panicle 2 (LP2) was previously found to be strongly expressed in leaves and other photosynthetic tissues, but its function remains unclear. In the present study, it was shown that the expression of LP2 was down-regulated by drought and abscisic acid (ABA). Transgenic plants overexpressing LP2 accumulated less H₂O₂, had more open stomata in leaves, and showed hypersensitivity to drought stress. Further investigation revealed that transcription of LP2 was directly regulated by the zinc finger transcription factor DROUGHT AND SALT TOLERANCE (DST). In addition, LP2 was identified as a functional kinase localized to the plasma membrane and interacted with the drought-responsive aquaporin proteins OsPIP1; 1, OsPIP1; 3, and OsPIP2; 3. Thus, the findings provided evidence that the LRR-RLK LP2, transcriptionally regulated by the drought-related transcription factor DST, served as a negative regulator in drought response.
Osmotic stress-triggered stomatal closure requires Phospholipase Dδ and hydrogen sulfide in Arabidopsis thaliana

Abstract:Osmotic stress is one of the main stresses seriously affects the growth and development of plants. Hydrogen sulfide (H2S) emerges as the third gaseous signal molecule to involve in the complex network of signaling events. Phospholipase Dδ (PLDδ), as signal enzyme, responds to many biotic or abiotic stress responses. In this study, the functions and the relationship of PLDδ and H2S in stomatal closure induced by osmotic stress were explored. Using the seedlings of ecotype (WT), PLDδ deficient mutant (pldδ), L-cysteine desulfhydrase (LCD) deficient mutant (lcd) and pldδlcd double mutant as materials, the Real-time quantitative PCR (RT-qPCR) and the stomatal aperture were analyzed. Osmotic stress induced the expressions of PLDδ and LCD. The H2S content and the activities of PLD and LCD ascended in WT under osmotic stress. The phenotypes of pldδ, lcd and pldδlcd were more sensitive to osmotic stress than WT. Compared with pldδ, the stomatal of lcd showed lower sensitivity to osmotic stress, and the stomatal aperture of pldδlcd was similar to that of lcd. Simultaneous application of PA and NaHS resulted in tighter closure of stomatal than application of either PA or NaHS alone. These results suggested that osmotic stress-triggered stomatal closure requires PLDδ and H2S in A. thaliana. LCD acted downstream of PLDδ to regulate the stomatal closure induced by osmotic stress.

Transcription factor WRKY46 regulates osmotic stress responses and stomatal movement independently in Arabidopsis.

Abstract:Drought and salt stress severely inhibit plant growth and development; however, the regulatory mechanisms of plants in response to these stresses are not fully understood. Here we report that the expression of a WRKY transcription factor WRKY46 is rapidly induced by drought, salt and oxidative stresses. T-DNA insertion of WRKY46 leads to more sensitivity to drought and salt stress, whereas overexpression of WRKY46 (OV46) results in hypersensitivity in soii-grown plants, with a higher water loss rate, but with increased tolerance on the sealed agar plates. Stomatal closing in the OV46 line is insensitive to ABA because of a reduced accumulation of reactive oxygen species (ROS) in the guard cells. We further find that WRKY46 is expressed in guard cells, where its expression is not affected by dehydration, and is involved in light-dependent stomatal opening. Microarray analysis reveals that WRKY46 regulates a set of genes involved in cellular osmoprotection and redox homeostasis under dehydration stress, which is confirmed by ROS and malondialdehyde (MDA) levels in stressed seedlings. Moreover, WRKY46 modulates light-dependent starch metabolism in guard cells via regulating QUA-QUINE STARCH (QQS) gene expression. Taken together, we demonstrate that WRKY46 plays dual roles in regulating plant responses to drought and salt stress and light-dependent stomatal opening in guard cells.

Abstract:Hydrogen sulfide (H2S) nowadays is regarded as a novel gaseous signaling molecule in plants during environmental stress response. The spermidine (Spd), an important plant growth regulator, plays critical roles in regulating stress tolerance in plant species. Objectives of this study were to examine whether Spd could act as a H2S signaling activator to regulate stress-responsive transcription factors (TFs) and antioxidant defense and to further explore the possible crosstalk among Spd-induced hydrogen peroxide (H2O2), nitric oxide (NO), and H2S signaling in leaves of white clover. Results showed that dehydration or exogenous application of Spd could significantly increase L/D-cysteine desulfhydrate (L/DCD) activities and then cause a quick accumulation of H2S, followed by significant improvement of antioxidant enzyme (SOD, CAT, GPDX, APX, GR, DHAR, and MDHAR) activities and transcript levels of TFs (bZIP37, bZIP107, DREB2, DREB4, and WRKY108715) and genes encoding antioxidant enzymes. The inhibitors (AOA and NH2OH) of L/DCD activities could effectively attenuate dehydration- or Spd-induced these effects in leaves. Additionally, dehydration-induced increase in H2S accumulation, L/DCD and antioxidant enzyme activities, and genes expression were remarkably abolished by the application of Spd biosynthesis inhibitor (DCHA), whereas exogenous Spd significantly alleviate the inhibitory effect of DCHA on these parameters under dehydration condition. This indicated that H2S which was derived from L/DCD pathway might act as a downstream signaling of Spd, and Spd-activated H2S signaling was involved in dehydration-regulated TFs and antioxidant defense associated with dehydration tolerance. Further analyses found that Spd-induced H2O, production could not be attenuated by scavengers of NO and H2S, but the scavenger of H2O2 effectively inhibited Spd-induced increases in NO and H2S. The Spd-induced H2S signaling was also substantially inhibited by the application of the NO scavenger. The result implied that Spd-induced H2O2 could be an upstream signal molecule of NO and H2S, and the Spd-induced H2S might act as the downstream signaling of NO in leaves. These obtained results provide the basic evidence to understand Spd-regulated signal transduction in white clover.

Abstract:Hydrogen sulfide (H 2 S) is a crucial signaling molecule in plants. Recent studies have shown that H 2 S plays an equally important role as nitric oxide (NO) and hydrogen peroxide (H 2 O 2 ) in plant signaling. Previous studies have demonstrated the involvement of H 2 S in regulating drought and other stressful environmental conditions, but the exact downstream molecular mechanisms activated by the H 2 S signaling molecule remain unclear. In this study, we conducted a comprehensive genome-wide transcriptomic analysis of both wild type (WT) and double mutant ( lcd/des1 ). Arabidopsis thaliana plants were exposed to 40% polyethylene glycol (PEG) to induce drought stress and 20 μM sodium hydrosulfide (NaHS). The resulting transcriptome data were analyzed for differentially significant genes and their statistical enrichments in the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways. The results indicated significant upregulation of genes related to photosynthesis, carbon fixation, plant secondary metabolite biosynthesis, inositol and phosphatidylinositol signaling pathways, and stress-responsive pathways in mutant plants under drought stress. Mutant plants with impaired H 2 S signaling mechanisms displayed greater susceptibility to drought stress compared to wild-type plants. In summary, all findings highlight the pivotal role of H 2 S signaling in stimulating other drought-responsive signaling pathways.

Abstract:Hydrogen sulfide (H(2)S) plays a crucial role in human and animal physiology. Its ubiquity and versatile properties have recently caught the attention of plant physiologists and biochemists. Two cysteine desulfhydrases (CDes), L-cysteine desulfhydrase and D-cysteine desulfhydrase, were identified as being mainly responsible for the degradation of cysteine in order to generate H(2)S. This study investigated the expression regulation of these genes and their relationship to drought tolerance in Arabidopsis. First, the expression pattern of CDes in Arabidopsis was investigated. The expression levels of CDes gradually increased in an age-dependent manner. The expression of CDes was significantly higher in stems and cauline leaves than in roots, rosette leaves and flowers. Second, the protective effect of H(2)S against drought was evaluated. The expression pattern of CDes was similar to the drought associated genes induced by dehydration, and H(2)S fumigation was found to stimulate further the expression of drought associated genes. Drought also significantly induced increased H(2)S production, a process that was reversed by re-watering. In addition, seedlings after treatment with NaHS (a H(2)S donor) showed a higher survival rate and displayed a significant reduction in the size of the stomatal aperture compared to the control. These findings provide evidence that H(2)S, as a gasotransmitter, improves drought resistance in Arabidopsis.

Abstract:Guard cells shrink in response to drought stress and abscisic acid (ABA) signaling, thereby reducing stomatal aperture. Hydrogen peroxide (H 2 O 2 ) is an important signaling molecule acting to induce stomatal closure. As yet, the molecular basis of control over the level of H 2 O 2 in the guard cells remains largely unknown. Here, the leucine-rich repeat (LRR)—receptor-like kinase (RLK) protein HSL3 has been shown to have the ability to negatively regulate stomatal closure by modulating the level of H 2 O 2 in the guard cells. HSL3 was markedly up-regulated by treating plants with either ABA or H 2 O 2 , as well as by dehydration. In the loss-of-function hsl3 mutant, both stomatal closure and the activation of anion currents proved to be hypersensitive to ABA treatment, and the mutant was more tolerant than the wild type to moisture deficit; the overexpression of HSL3 had the opposite effect. In the hsl3 mutant, the transcription of NADPH oxidase gene RbohF involved in H 2 O 2 production showed marked up-regulation, as well as the level of catalase activity was weakly inducible by ABA, allowing H 2 O 2 to accumulate in the guard cells. HSL3 was concluded to participate in the regulation of the response to moisture deficit through ABA-induced stomatal closure triggered by the accumulation of H 2 O 2 in the guard cells.

Abstract:Drought is a global threat that affects agricultural production. Plants have evolved several adaptive strategies to cope with drought. Stomata are essential structures for plants to control water status and photosynthesis rate. Stomatal closure is an efficient way for plants to reduce water loss and improve survivability under drought conditions. The opening and closure of stomata depend on the turgor pressure in guard cells. Three key signaling molecules, including abscisic acid (ABA), reactive oxygen species (ROS), and calcium ion (Ca2+), play pivotal roles in controlling stomatal closure. Plants sense the water-deficit signal mainly via leaves and roots. On the one hand, ABA is actively synthesized in root and leaf vascular tissues and transported to guard cells. On the other hand, the roots sense the water-deficit signal and synthesize CLAVATA3/EMBRYO-SURROUNDING REGION RELATED 25 (CLE25) peptide, which is transported to the guard cells to promote ABA synthesis. ABA is perceived by pyrabactin resistance (PYR)/PYR1-like (PYL)/regulatory components of ABA receptor (RCAR) receptors, which inactivate PP2C, resulting in activating the protein kinases SnRK2s. Many proteins regulating stomatal closure are activated by SnRK2s via protein phosphorylation. ABA-activated SnRK2s promote apoplastic ROS production outside of guard cells and transportation into the guard cells. The apoplastic H2O2 can be directly sensed by a receptor kinase, HYDROGEN PEROXIDE-INDUCED CA2+ INCREASES1 (HPCA1), which induces activation of Ca2+ channels in the cytomembrane of guard cells, and triggers an increase in Ca2+ in the cytoplasm of guard cells, resulting in stomatal closure. In this review, we focused on discussing the signaling transduction of ABA, ROS, and Ca2+ in controlling stomatal closure in response to drought. Many critical genes are identified to have a function in stomatal closure under drought conditions. The identified genes in the process can serve as candidate genes for genetic engineering to improve drought resistance in crops. The review summarizes the recent advances and provides new insights into the signaling regulation of stomatal closure in response to water-deficit stress and new clues on the improvement of drought resistance in crops.

Abstract:Main conclusion: Jasmonic acid (JA) negatively regulates stomatal development by promoting LCD expression and hydrogen sulfide (H2S) biosynthesis. H2S inhibits the initiation of stomata formation and acts upstream of SPEECHLESS. Stomatal development is strictly regulated by endogenous signals and environmental cues. We recently revealed that jasmonic acid (JA) negatively regulates stomatal development in Arabidopsis thaliana cotyledons (Han et al., Plant Physiol 176:2871–2885, 2018), but the underlying molecular mechanism remains largely unknown. Here, we uncovered a role for H2S in regulating stomatal development. The H2S scavenger hypotaurine reversed the JA-induced repression of stomatal development in the epidermis of wild-type Arabidopsis. The H2S-deficient mutant lcd displayed increased stomatal density and stomatal index values, which were rescued by treatment with sodium hydrosulfide (NaHS; an H2S donor) but not JA, suggesting that JA-mediated repression of stomatal development is dependent on H2S biosynthesis. The high stomatal density of JA-deficient mutants was rescued by exogenous NaHS treatment. Further analysis indicated that JA positively regulates LCD expression, l-cysteine desulfhydrases (l-CDes) activity, and endogenous H2S content. Furthermore, H2S represses the expression of stomate-associated genes and functions downstream of stomate-related signaling pathway components TOO MANY MOUTHS (TMM) and STOMATAL DENSITY AND DISTRIBUTION1 (SDD1) and upstream of SPEECHLESS (SPCH). Therefore, H2S acts downstream of JA signaling to regulate stomatal development in Arabidopsis cotyledons.

Abstract:Sugars play important roles in regulating plant growth, development, and stomatal movement. Here, we found that glucose triggered stomatal closure in a dose- and time-dependent manner in Arabidopsis. Pharmacological data showed that glucose-induced stomatal closure was greatly inhibited by catalase [CAT; a reactive oxygen species (ROS) scavenger], diphenyleneiodonium chloride (DPI; an NADPH oxidase inhibitor), lanthanum chloride (LaCl3; a Ca2+ channel blocker), EGTA (a Ca2+ chelator), and two nitrate reductase (NR) inhibitors, tungstate and sodium azide (NaN3), while it was not affected by salicylhydroxamic acid (SHAM; a peroxidase inhibitor). Moreover, glucose induced ROS and nitric oxide (NO) production in guard cells of Arabidopsis. The ROS production was almost completely removed by CAT, strongly restricted by DPI, and was not affected by SHAM. NO production was partially suppressed by tungstate and NaN3, and the levels of NO were significantly reduced in the nia1-1nia2-5 mutant. Additionally, glucose-triggered stomatal closure was significantly impaired in gin1-1, gin2-1, pyr1pyl1pyl2pyl4, abi1-1, ost1, slac1-4, cpk6-1, and nia1-1nia2-5 mutants. Likewise, the reductions in leaf stomatal conductance (gs) and transpiration rate (E) caused by glucose were reversed in the above mutants. These results suggest that glucose-triggered stomatal closure may be dependent on basal signaling through PYR/RCAR receptors and hexokinase1 (HXK1).

They regulate stomatal movement and enhance drought resistance. However, their physiological relationship is not well understood. Here, their crosstalk involved in osmotic stress resistance in Arabidopsis thaliana was studied. Exogenous H2S and MEL closed stomata under normal or osmotic stress conditions and increased the relative water contents of plants under osmotic stress conditions. At the same time, exogenous H2S and MEL responded to osmotic stress by increasing the content of proline and soluble sugar, and reducing malondialdehyde (MDA) content and relative conductivity. Using mutants in the MEL-associated production of serotonin N-acetyltransferase (snat), caffeic acid O-methyltransferase (comt1) and N-acetylserotonin methyltransferase (asmt), we determined that H2S was partially dependent on MEL to close stomata. Additionally, the overexpression of ASMT promoted stomatal closure. Exogenous H2S increased the transcription levels of SNAT, ASMT and COMT1. Furthermore, exogenous H2S treatments increased the endogenous MEL content significantly. At the post-translational level, H2S sulfhydrated the SNAT and ASMT, but not COMT1, enzymes associated with MEL production. Thus, H2S appeared to promote stomatal closure in response to osmotic stress by increasing the transcription levels of MEL synthesis-related genes and the sulfhydryl modification of the encoded enzymes. These results increased our understanding of H2S and MEL functions and interactions under osmotic stress conditions.

Abstract:Abstract Reactive oxygen species (ROS) are plant metabolic and signaling molecules involved in responses to various external stresses, but the existence of ROS receptors and how plants respond to ROS remain largely unknown. Here we report that the plasma membrane-localized phospholipase D δ (PLDδ) protein is crucial for sensing heat shock-induced ROS to initiate reorganization of guard cell microtubules in Arabidopsis cotyledons. Heat shock of wild-type Arabidopsis cotyledons stimulated ROS production which disrupted microtubule organization and induced stomatal closure, whereas this process was markedly impaired in pldδ mutants. Moreover, wild-type PLDδ, but not the Arg622-mutated PLDδ, complemented the pldδ phenotypes in heat shock-treated plants. ROS activated PLDδ by oxidizing cysteine residues, an action that was required for its functions in ROS-induced depolymerization of guard cell microtubules, stomatal closure, and plant thermotolerance. Additionally, lipid profiling reveals involvement of microtubule organization in the feedback regulation of glycerolipid metabolism upon heat stress. Together, our findings highlight a potential mechanosensory role for PLDδ in regulating the dynamic organization of microtubules and stomatal movement, as part of the ROS-sensing pathway, during the response to external stresses.

Strigolactones (SLs) contribute to regulating shoot branching by targeting the SUPPRESSOR OF MORE AXILLARY GROWTH2 (MAX2)-LIKE6 (SMXL6), SMXL7 and SMXL8 for degradation in a MAX2-dependent manner in Arabidopsis. Although SLs are implicated in plant drought response, the functions of the SMXL6, 7 and 8 in the SL-regulated plant response to drought stress have remained unclear. Here, we performed transcriptomic, physiological and biochemical analyses of smxl6, 7, 8 and max2 plants to understand the basis for SMXL6/7/8-regulated drought response. We found that three D53 (DWARF53)-Like SMXL members, SMXL6, 7 and 8, are involved in drought response as the smxl6smxl7smxl8 triple mutants showed markedly enhanced drought tolerance compared to wild type (WT). The smxl6smxl7smxl8 plants exhibited decreased leaf stomatal index, cuticular permeability and water loss, and increased anthocyanin biosynthesis during dehydration. Moreover, smxl6smxl7smxl8 were hypersensitive to ABA-induced stomatal closure and ABA responsiveness during and after germination. In addition, RNA-sequencing analysis of the leaves of the D53-like smxl mutants, SL-response max2 mutant and WT plants under normal and dehydration conditions revealed an SMXL6/7/8-mediated network controlling plant adaptation to drought stress via many stress- and/or ABA-responsive and SL-related genes. These data further provide evidence for crosstalk between ABA- and SL-dependent signaling pathways in regulating plant responses to drought. Our results demonstrate that SMXL6, 7 and 8 are vital components of SL signaling and are negatively involved in drought responses, suggesting that genetic manipulation of SMXL6/7/8-dependent SL signaling may provide novel ways to improve drought resistance.
Activation of 1-Aminocyclopropane-1-Carboxylic Acid Synthases Sets Stomatal Density and Clustered Ratio on Leaf Epidermis of Arabidopsis in Response to Drought

Abstract:The adjustment of stomatal density and clustered ratio on the epidermis is the important strategy for plants to respond to drought, because the stoma-based water loss is directly related to plant growth and survival under drought conditions. But the relevant adjustment mechanism still needs to be explored. 1-Aminocyclopropane-1-carboxylate (ACC) is disclosed to promote stomatal development, while in vivo ACC levels depend on activation of ACC synthase (ACS) family members. Based on the findings of ACS expression involving in drought response and several ACS activity inhibitors reducing stomatal density and cluster in drought response, here we examined how ACS activation is involved in the establishment of stomatal density and cluster on the epidermis under drought conditions. Preliminary data indicated that activation of ACS2 and/or ACS6 (ACS2/6) increased stomatal density and clustered ratio on the Arabidopsis leaf epidermis by accumulating ACC under moderate drought, and raised the survival risk of seedlings under escalated drought. Further exploration indicated that, in Arabidopsis seedlings stressed by drought, the transcription factor SPEECHLESS (SPCH), the initiator of stomatal development, activates ACS2/6 expression and ACC production; and that ACC accumulation induces Ca 2+ deficiency in stomatal lineage; this deficiency inactivates a subtilisin-like protease STOMATAL DENSITY AND DISTRIBUTION 1 (SDD1) by stabilizing the inhibition of the transcription factor GT-2 Like 1 (GTL1) on SDD1 expression, resulting in an increases of stomatal density and cluster ratio on the leaf epidermis. This work provides a novel evidence that ACS2/6 activation plays a key role in the establishment of stomatal density and cluster on the leaf epidermis of Arabidopsis in response to drought.

Abstract:Hydrogen sulfide (H2S) is a newly appreciated participant in physiological and biochemical regulation in plants. However, whether H2S is involved in the regulation of plant responses to drought stress remains unclear. Here, the role of H2S in the regulation of drought stress response in Spinacia oleracea seedlings is reported. First, drought stress dramatically decreased the relative water content (RWC) of leaves, photosynthesis, and the efficiency of PSII. Moreover, drought caused the accumulation of ROS and increased the MDA content. However, the application of NaHS counteracted the drought-induced changes in these parameters. Second, NaHS application increased the water and osmotic potential of leaves. Additionally, osmoprotectants such as proline and glycinebetaine (GB) content were altered by NaHS application under drought conditions, suggesting that osmoprotectant contributes to H2S-induced drought resistance. Third, the levels of soluble sugars and polyamines (PAs) were increased differentially by NaHS application in S. oleracea seedlings. Moreover, several genes related to PA and soluble sugar biosynthesis, as well as betaine aldehyde dehydrogenase (SoBADH), choline monooxygenase (SoCMO), and aquaporin (SoPIP1;2), were up-regulated by H2S under drought stress. These results suggest that H2S contributes to drought tolerance in S. oleracea through its effect on the biosynthesis of PAs and soluble sugars. Additionally, GB and trehalose also play key roles in enhancing S. oleracea drought resistance.

Transcriptome analysis of drought-responsive genes regulated by hydrogen sulfide in wheat (Triticum aestivum L.) leaves

Abstract:Drought is an environmental factor that deeply impacts wheat yield and quality. Hydrogen sulfide (H2S) is a known regulator of drought resistance in plants. To preliminarily elucidate the regulatory mechanisms of H2S on drought tolerance, the effects of H2S on drought-responsive genes were investigated by transcriptome analysis. As a result, a total of 7552 transcripts not only responded to drought stress but also exhibited differential expression relative to the polyethylene glycol (PEG) treatment (P) and the NaHS pretreatment with PEG treatment (SP). GO categories of 'transport' were especially enriched under the SP treatment and ion transport categories (especially 'iron ion transport') were more significantly enriched among up-regulated transcripts in SP versus P treatments (SP.vs.P). Indeed, a higher translocation of iron from root to shoot and iron availability in shoots was detected in SP compared to P. The KEGG pathway of 'ribosome biogenesis in eukaryotes', 'protein processing in endoplasmic reticulum', 'fatty acid degradation', and 'cyanoamino acid metabolism' was induced by H2S under drought stress. Further, H2S was involved in plant hormones signal transduction, and drought-induced transcription factors, protein kinases, and functional genes exhibited higher expression levels under SP relative to P. Additionally, several effectors or master regulatory genes of H2S were identified genome-wide. Summarily, these results showed that H2S alleviated drought damage probably related to transport systems, plant hormones signal transduction, protein processing pathway, fatty acids and amino acids metabolism, which provides a guide for future experimentation to analyze hydrogen sulfide-dependent drought tolerance mechanisms in wheat.
Overexpression of a wheat phospholipase D gene, TaPLDα , enhances tolerance to drought and osmotic stress in Arabidopsis thaliana

Abstract:Phospholipase D (PLD) is crucial for plant responses to stress and signal transduction, however, the regulatory mechanism of PLD in abiotic stress is not completely understood; especially, in crops. In this study, we isolated a gene, TaPLDα , from common wheat ( Triticum aestivum L.). Analysis of the amino acid sequence of TaPLDα revealed a highly conserved C2 domain and two characteristic HKD motifs, which is similar to other known PLD family genes. Further characterization revealed that TaPLDα expressed differentially in various organs, such as roots, stems, leaves and spikelets of wheat. After treatment with abscisic acid (ABA), methyl jasmonate, dehydration, polyethylene glycol and NaCl, the expression of TaPLDα was up-regulated in shoots. Subsequently, we generated TaPLDα -overexpressing transgenic Arabidopsis lines under the control of the dexamethasone-inducible 35S promoter. The overexpression of TaPLDα in Arabidopsis resulted in significantly enhanced tolerance to drought, as shown by reduced chlorosis and leaf water loss, higher relative water content and lower relative electrolyte leakage than the wild type. Moreover, the TaPLDα -overexpressing plants exhibited longer roots in response to mannitol treatment. In addition, the seeds of TaPLDα -overexpressing plants showed hypersensitivity to ABA and osmotic stress. Under dehydration, the expression of several stress-related genes, RD29A , RD29B , KIN1 and RAB18 , was up-regulated to a higher level in TaPLDα -overexpressing plants than in wild type. Taken together, our results indicated that TaPLDα can enhance tolerance to drought and osmotic stress in Arabidopsis and represents a potential candidate gene to enhance stress tolerance in crops.
Transcriptome profiling of Arabidopsis slac1-3 mutant reveals compensatory alterations in gene expression underlying defective stomatal closure

Abstract:Plants adjust their stomatal aperture for regulating CO 2 uptake and transpiration. S-type anion channel SLAC1 (slow anion channel-associated 1) is required for stomatal closure in response to various stimuli such as abscisic acid, CO 2 , and light/dark transitions etc. Arabidopsis slac1 mutants exhibited defects in stimulus-induced stomatal closure, reduced sensitivity to darkness, and faster water loss from detached leaves. The global transcriptomic response of a plant with defective stimuli-induced stomatal closure (particularly because of defects in SLAC1) remains to be explored. In the current research we attempted to address the same biological question by comparing the global transcriptomic changes in Arabidopsis slac1-3 mutant and wild-type (WT) under dark, and dehydration stress, using RNA-sequencing. Abscisic acid (ABA)- and dark-induced stomatal closure was defective in Arabidopsis slac1-3 mutants, consequently the mutants had cooler leaf temperature than WT. Next, we determined the transcriptomic response of the slac1-3 mutant and WT under dark and dehydration stress. Under dehydration stress, the molecular response of slac1-3 mutant was clearly distinct from WT; the number of differentially expressed genes (DEGs) was significantly higher in mutant than WT. Dehydration induced DEGs in mutant were related to hormone signaling pathways, and biotic and abiotic stress response. Although, overall number of DEGs in both genotypes was not different under dark, however, the expression pattern was very much distinct; whereas majority of DEGs in WT were found to be downregulated, in slac1-3 majority were upregulated under dark. Further, a set 262 DEGs was identified with opposite expression pattern between WT and mutant under light–darkness transition. Amongst these, DEGs belonging to stress hormone pathways, and biotic and abiotic stress response were over-represented. To sum up, we have reported gene expression reprogramming underlying slac1-3 mutation and resultantly defective stomatal closure in Arabidopsis. Moreover, the induction of biotic and abiotic response in mutant under dehydration and darkness could be suggestive of the role of stomata as a switch in triggering these responses. To summarize, the data presented here provides useful insights into the gene expression reprogramming underlying slac1-3 mutation and resultant defects in stomatal closure.

Abstract:Phospholipase D (PLD) is involved in responses to abiotic stress and abscisic acid (ABA) signaling. To investigate the roles of two Arabidopsis (Arabidopsis thaliana) PLDs, PLD alpha 1 and PLD delta, in ABA signaling in guard cells, we analyzed ABA responses in guard cells using Arabidopsis wild type, pld alpha 1 and pld delta single mutants, and a pld alpha 1 pld delta double mutant. ABA-induced stomatal closure was suppressed in the pld alpha 1 pld delta double mutant but not in the pld single mutants. The pld alpha 1 and pld delta mutations reduced ABA-induced phosphatidic acid production in epidermal tissues. Expression of either PLD alpha 1 or PLD delta complemented the double mutant stomatal phenotype. ABA-induced stomatal closure in both pld alpha 1 and pld delta single mutants was inhibited by a PLD inhibitor (1-butanol), suggesting that both PLD alpha 1 and PLD delta function in ABA-induced stomatal closure. During ABA-induced stomatal closure, wild-type guard cells accumulate reactive oxygen species and nitric oxide and undergo cytosolic alkalization, but these changes are reduced in guard cells of the pld alpha 1 pld delta double mutant. Inward-rectifying K+ channel currents of guard cells were inhibited by ABA in the wild type but not in the pld alpha 1 pld delta double mutant. ABA inhibited stomatal opening in the wild type and the pld delta mutant but not in the pld alpha 1 mutant. In wild-type rosette leaves, ABA significantly increased PLD delta transcript levels but did not change PLD alpha 1 transcript levels. Furthermore, the pld alpha 1 and pld delta mutations mitigated ABA inhibition of seed germination. These results suggest that PLD alpha 1 and PLD delta cooperate in ABA signaling in guard cells but that their functions do not completely overlap.
